RESUMEN

OBJECTIVE: To investigate inhibitory effects of lupeol on the proliferation of human breast cancer MCF-7 cells and its possible mechanism. METHODS: Taking MCF-7 cells as research object, MTT assay was used to detect the proliferation of MCF-7 cells after treated with different doses of lupeol (7.5, 15, 30, 60, 90 mg/L) for 24 h. Survival rate and IC50 of MCF-7 cells were calculated. The inverted microscope and cell cloning experiment were used to observe and detect the morphological characteristics of MCF-7 cells and clonal colony formation after treated with different doses of lupeol (15, 30, 60 mg/L) for 24 h. The rate of clonal colony formation was calculated. MTT method and Western blotting assay were used to detect the proliferation of MCF-7 cells and the expression of related regulatory proteins (ERK1/2, p-ERK1/2, JNK, p-JNK, p38 MAPK, p-p38 MAPK) after additionally treated with MAPKs signaling pathway-related regulation protein inhibitors PD98059, SP600125 and SB203580. RESULTS: After treated with 15, 30, 60, 90 mg/L lupeol, survival rates of MCF-7 cells were decreased significantly (P<0.05 or P<0.01). IC50 value of the compound was 52.94 mg/L. After treated with 15, 30, 60 mg/L lupeol, the morphological characteristics of cells in each group changed, and the phenomena of cell exfoliation, floating, solid shrinkage, roundness, volume reduction and necrosis were observed. The formation of clonal colony decreased and the rate of clonal colony formation decreased significantly (P<0.05 or P<0.01). When lupeol used alone, compared with control group, survival rate (60 mg/L lupeol)of MCF-7 cells was decreased significantly; the expression of p-ERK1/2 (15, 30, 60 mg/L lupeol), p-JNK (30, 60 mg/L lupeol) and p-p38 MAPK (30, 60 mg/L lupeol) were increased significantly (P<0.05 or P<0.01). After additional use of relevant inhibitors, compared with 60 mg/L lupeol group, survival rates of MCF-7 cells in combination groups were increased significantly, while relative expression of p-ERK1/2, p-JNK and p-p38 MAPK were decreased significantly (P<0.05 or P<0.01). CONCLUSIONS: Lupeol can inhibit the proliferation of human breast cancer MCF-7 cells, the mechanism of which may be related to the phosphorylation of MAPKs signaling pathway-related regulatory proteins.

Objective To discuss the treatment efficacy and radiotherapy side effects of the preoperative long-course radiochemotherapy and preoperative short-course radiotherapy.Methods 64 patients with local advanced middle and low rectal cancer who got the treatment from April 2004 to April 2010 were analyzed retrospectively.40 patients got the preoperative long-course radiochemotherapy under the dose of DT 45-50 Gy/25 F,1.8-2.0 Gy/F,5 F/W,combining with the synchronous capecitabine chemotherapy (1 650 mg/m2,2 F/d,d1-14/d21-35),and accepted operation 4-6 weeks after the radiotherapy.The rest 24 patients underwent the preoperative short-course radiotherapy under the dose of DT 25 Gy/5 F,5 Gy/F,5 F/W,and got the operation in 2 weeks after the radiotherapy.Results The radical and anus reservation rates in preoperative long-course radiochemotherapy group [85.0 % (34/40),65.0 % (26/40)] were higher than those in preoperative short-course radiotherapy group [58.3 % (14/24),33.3 % (8/24)] (x2 =5.689,P =0.019;x2 =6.040,P =0.041).There were no significant differences between the two groups on the index of remission rates,radiation injury,surgical complications,and overall survival rate of 1,3,5 years (all P > 0.05).Conclusions The remission rate and overall survival time between the preoperative long-course radiochemotherapy group and preoperative short-course radiotherapy have no significant difference.But the preoperative long-course radiochemotherapy may improve the anus reservation rate and the radical resection rate,without increasing the radiation injury and surgical complications.

Objective There are a few reports on rats with spontaneous congenital cataract in China .The purpose of this study was to investigate the morphological changes of lens in rats with spontaneous congenital cataract . Methods 24 d, 1-year rats with cataract and microphthalmos cataract and normal rats (n=5) were selected as research objects .Their lens were observed by a slit lamp microscope and taken photos in front of them , followed by examination through light micrograph and transmission electron micros-copy. Results Rats with microphthalmos cataract showed narrowed palpebral fissure and broaden nucleus while rats with cataract showed normal palpebral fissure and narrowed nucleus .As for 24 d,1-year rats with microphthalmos cataract , the fibers of their lens showed derangement and vacuole-like degeneration by light microscope , in addition, the abnormal connection between fiber cells were observed by electron microscopy .As for 1-year normal rats , the fibers were in consistent structure and regular arrangement without cell ingredient . Conclusion The appearance and morphological changes of the lens in rats with spontaneous congenital cataracts are in consistence with the pathological changes of cataracts , which is appli-cable in further research on the pathogenesis of cataract .

Objective To study whether the green fluorescent protein ( GFP) gene can be successfully expressed in green fluorescent nude mice and the tissue distribution characteristics.Methods Small animal imaging system and RT-PCR assay were used to detect the GFP tissue distribution and fluorescence expression level.Results The GFP can be expressed in multiple tissues in green fluorescent nude mice.A higher expression was observed in the pancreas, heart, brain, and skin.Conclusion Exogenous GFP can be stably expressed and inherited in green fluorescent nude mice, with the highest expression in the pancreas.

Objective To measure the range of routine blood counts and blood biochemical parameters ,to analyze hemorheology of the rats with congenital cataract .Methods Blood samples were taken from 90 rats with congenital cataract weight about 185 ~211 g.Routine blood analysis was performed and blood biochemical and hemorheology paramenters were determind using an automatic blood biochemical and hemorheology analyzer .Results There were no significant difference ( P >0.05 ) between the cataract rats and the normal rats in Blood test results; but there were significant difference between the microphthalmos cataract rats and the normal same -sex rats ( P <0.01 or P <0.05 ) . The biochemical results is the cataract rats and the normal rats were different significantly in ALB group ( P <0.01 or P<0.05), and the female microphthalmos cataract rats compared with the control rats had significant difference in Ure (P<0.01) , the female cataract rats ompared with the normal rats were very significant difference in Cr group ( P <0.05 or P <0.01).The erythrocyte counts of the male cataract rats and male microphthalmos cataract rats were significantly lower than that in the female ones, respectively(P <0.05, P <0.01).The platelet counts of the male cataract rats and the male microphthalmos cataract rats were significantly higher than that in the female ones , respectively(P <0.01), and the creatinine of the male cataract rats and the male microphthalmos cataract rats were significantly lower then that in the female ones, respectively(P <0.01).There were no significant difference in every group on hemorheology .Conclusions There were significant differences in some blood indexes between the congenital cataract rats and the normal rats .These data may become useful reference for biomedical researcher in this field .

Objective To study the extracranial scalp electroencephalography ( EEG ) and intracranial electrocorticography (ECoG) of closed colony New Zealand white rabbits .Methods To record the extracranial scalp EEG and intracranial ECoG of closed colony New Zealand white rabbits , and to compare and analyze the results of those two scanning methods .Results EEG was characteristic of 9-12 c/sαwave and 16-20 c/sβwave with an amplitude of 30-100μV as the basic rhythm .ECoG showed 10-12 c/s αwave and 16-20 c/s βwave with an amplitude of 200-300 μV as the basic rhythm.Anesthesia could attenuate the electrocerebral activity , cause brain tissue hypoxia , and induce δ wave and slow θ wave in ECoG .Conclusions EEG method is a simple , non-invasive and convenient operation , and can be made in rabbits without anesthesia .The recorded EEG waveform is highly consistent with that of ECoG , and may be used as an alternative to the traditional ECoG in neurofunctional studies .

RESUMEN

Objectives:To observe the effect of intrahepatic cholestasis on the development and growth of fetus in pregnant rats. Methods: The animal models of intrahepatic cholestasis were made by injecting benzogynestryl (5 mg/kg body weight) or cholate (2.4 mg/kg body weight) for 5 days starting from 16th day of pregnancy. Biochemical parameters of fetal plasma and amniocent were determined at the 21st day of pregnancy. The pathological lesion of placenta in rats was also observed. Results: Cholic acid group showed lower body weight and high level of estrodiol in fetal plasma with high metaphase mortality of fetus. The level of TBA in amniocent was high in estrodiol group compared with normal control group. Pathologically, placenta edema and chorion denaturalization and necrosis with infiltration of neutrocyte were observed in both groups. Conclusions: Intrahepatic cholestasis is detrimental to the development and growth of fetal rats in pregnancy rats.

RESUMEN

Objective To explore an effective method of repairing the old rupture of patellar tendon. Methods We designed a new 井-shaped apparatus for traction and fixing which was to be used, under the dynamic conditions, to reduce the patella, loosen the quadriceps m. of thigh, repair the tendon of knee cap and facilitate postoperative exercises with the fixing apparatus on. Results We used the apparatus to repair the tendon of knee cap in 6 cases. The follow up of 8 to 56 months (averaging 26 months) showed that their functions of the knee joint were all satisfactory. Conclusions The 井-shaped external fixation device can be used under the dynamic conditions to repair the old rupture of patellar tendon with the advantages of little contracture of the knee cap and quadriceps m. of thigh, easy fixation after repair and suture, and early exercise of the knee joint. This novel method is simple and recommendable.
