RESUMEN

ObjectiveTo investigate the mechanism of Huazhuo Jiedu Huoxue Tongluo prescription in alleviating cerebral ischemia-reperfusion injury via regulating nerve cell autophagy based on c-Jun N-terminal kinase(JNK)signaling pathway . MethodSixty SD rats were randomly divided into 6 groups: sham group, middle cerebral artery occlusion/reperfusion (MCAO/R) group (model group), Huazhuo Jiedu Huoxue Tongluo prescription group [traditional Chinese medicine (TCM) group(25.0 g·kg-1)], JNK inhibitor SP600125 (SP) group(5 mg·kg-1), TCM+SP group and JNK agonist Anisomycin (Ani) group(15 mg·kg-1). After 24 h of modeling, TCM group and TCM+SP group were given TCM decoction (ig) for 3 consecutive days, and the other groups were given equal volume of normal saline (ig). Neurological deficit was evaluated by neurological function score and cerebral infarct volume was determined by 2,3,5-triphenyltetrazole chloride (TTC) staining. Hematoxylin-eosin (HE) staining and Nissl staining were used to observe the structural changes of brain tissue and the damage of neurons, respectively. Terminal deoxynucleotidyl transferase-mediated dUTP-biotin nick end labeling assay (TUNEL) was performed to detect cell apoptosis. The ultrastructure of autophagosomes was observed by transmission electron microscope. Western blot was employed to detect the protein expressions of microtubule-associated protein 1 light chain 3A/B (LC3A/B), autophagy related 5 (Atg5), the ortholog of yeast Atg6 (Beclin1), p62, B-cell lymphoma 2 (Bcl-2), JNK, phosphorylated (p)-JNK and c-Jun in brain tissue. The mRNA expressions of LC3A/B, Beclin1, p62, Atg5, Bcl-2, JNK and c-Jun were detected by Real-time fluorescence quantitative polymerase chain reaction (Real-time PCR). ResultCompared with the sham group, the model group had elevated neurological deficit score (P<0.05), enlarged cerebral infarct volume (P<0.05)and typical infarction manifestations formed in hippocampal region and its surrounding brain tissue. In addition, there were a large number of neuronal cell degeneration, necrosis, liquefaction, nucleus pyknosis and deep staining, and inflammatory cell infiltration in the cortex in the model group, and severe swelling of mitochondria, lysosomes, autophagosomes and autophagolysosomes were clearly seen under electron microscope. TUNEL positive cells were increased (P<0.05), and cell apoptosis was severe. The nuclear membrane and nucleolus of neurons in brain tissue were blurred with discontinuous processes, and Nissl bodies in cytoplasm were stained light with reduced number. Western blot revealed that the model group had up-regulated protein expressions of LC3A/B, Beclin1, Atg5, JNK, p-JNK and c-Jun in brain tissue (P<0.05), while down-regulated protein expressions of p62 and Bcl-2 (P<0.05)as compared with the sham group. Real-time PCR indicated that the mRNA expressions of LC3A/B, Beclin1, Atg5, JNK and c-Jun in the model group were higher (P<0.05) while the mRNA expressions of p62 and Bcl-2 were lower (P<0.05) than those in the sham group. Compared with the conditions in model group, the neurological deficit scores of TCM, SP and TCM+SP groups were lowered (P<0.05), and the cerebral infarct volume was reduced (P<0.05), with improved pathological status of brain tissue, especially in the TCM group. Furthermore, there were abundant and basically normal mitochondrial cristae, slightly dilated endoplasmic reticulum, slightly swollen golgi apparatus, slightly fused nuclear membrane, and few visible lysosomes, autophagosomes and autophagolysosomes. TUNEL positive cells were decreased (P<0.05), displaying reduced apoptosis, especially in the TCM group. The nucleolus and nuclear membrane of neurons in brain tissue were discernible, and Nissl bodies in cytoplasm was reduced to a certain degree as compared with those in the model group. Western blot showed a decrease in the protein expressions of LC3A/B, Beclin1, Atg5, JNK, p-JNK and c-Jun in the TCM group ,the SP group,and the TCM+SP group(P<0.05),while an increase in the protein expressions of p62 in the TCM group and SP group(P<0.05),and an increase in the protein expressions of Bcl-2 in the TCM group and TCM+SP group. By Real-time PCR, the mRNA expressions of LC3A, LC3B, Beclin1, Atg5, JNK and c-Jun had a down-regulation(P<0.05) while the mRNA expression of p62 a up-regulation in the TCM group ,the SP group,and the TCM+SP group(P<0.05),and the mRNA expression of Bcl-2 a up-regulation in the TCM group and the TCM+SP group(P<0.05).Scores of the Ani group were raised (P<0.05), and infarct volume was increased significantly, with aggravated neuronal cell necrosis and obvious inflammatory infiltration. Moreover, there were neuronal nuclear membrane fusion with abnormal protrusion, increased heterochromatin aggregation in edge, severe mitochondrial swelling, endoplasmic reticulum expansion, increased lysosomes, increased intracytoplasmic vesicles, and visible autophagosomes and autophagolysosomes. TUNEL positive cells were increased (P<0.05), representing severe apoptosis. The number of Nissl bodies dropped with light staining, and the nucleolus and nuclear membrane were blurred. Real-time PCR found that the mNRA expressions of Atg5, c-Jun, JNK were up-regulated (P<0.05),while Beclin1, p62, Bcl-2 were were down-regulated in the Ani group (P<0.05). Compared with the TCM group and SP group,the protein expressions of LC3A/B, Beclin1, Atg5, JNK, p-JNK, c-Jun were decreased,and p62, Bcl-2 were increased in the Ani group(P<0.05). Compared with the TCM group,the mRNA expressions of JNK mRNA had a down-regulation in the SP group and TCM+SP group,while LC3A, LC3B, Atg5, c-Jun, JNK had an up-regulation(P<0.05) and Bcl-2 had a down-regulation in the Ani group(P<0.05). Compared with the SP group,the mRNA expressions of Atg5, c-Jun, JNK had an up-regulation(P<0.05), and Beclin1, p62, Bcl-2 had a down-regulation in the Ani group(P<0.05). ConclusionHuazhuo Jiedu Huoxue Tongluo prescription significantly up-regulates the protein and mRNA expressions of LC3A/B, Beclin1, Atg5, JNK, p-JNK and c-Jun, and down-regulates the protein and mRNA expressions of p62 and Bcl-2, suggesting that the prescription can inhibit autophagy through JNK signaling pathway to reduce ischemia/reperfusion injury in rats.

Objective:To discuss the mechanism of Yueju Pill in the treatment of functional dyspepsia (FD) based on network pharmacology and molecular docking.Methods:The chemical components and action targets of Yueju Pill were screened out by TCMSP platform and HERB, BATMAN-TCM database combined with literature were used to supplement effective components of Vietnam bow. The targets of FD were screened out by GeneCards database and OMIM database, and the intersection of the two targets was used to analyze the protein interactions using the STRING platform to construct the PPI network. Metascape platform was used for GO and KEGG pathway enrichment analysis, and Cytoscape 3.7.2 software was used to construct a network of "Yueju Pill components-functional dyspepsia targets-pathways". Online mapping tools were used to obtain the Venn plot of the intersection targets of Yueju Pill, FD and its related pathogenesis. Finally, AutoDock software was used for molecular docking.Results:The main active components of Yueju Pill in the treatment of FD are quercetin, wogonin, luteolin, kaempferol, etc. The main targets are AKT1, MAPK1, JUN, RELA, IL6, BCL2, BAX, MAPK8, EGFR, ESR1, etc. Molecular docking shows that the targets and the active components of the Yueju Pill have better binding abilit. The GO enrichment analysis result shows that there are 2 273 biological processes, 152 molecular functions and 91 cell components. KEGG enrichment analysis shows that there are 344 pathways associated with FD. According to literature review, the pathways related to FD include PI3K-Akt signaling pathway, AGE-RAGE signaling pathway, IL-17 signaling pathway, etc.Conclusion:Yueju Pill might act on AKT1, MAPK1, JUN, RELA, IL6, BCL2, BAX, MAPK8, EGFR, ESR1 and other targets to regulate PI3K-Akt signaling pathway, AGE-RAGE signaling pathways and IL-17 signaling pathway and it could treat FD.

RESUMEN

Objective:We aimed to analyze the spectrum of diseases of Experienced Ten Acupoints in clinical studies, in order to provide reference for further clinical trials.Methods:The Chinese academic journals full-text database (CNKI),Chinese science and technology journals full-text database (VIP),Wanfang database and Chinese biomedical database (SinoMed), PubMed, Cochrane Library and Web of Science Database were searched from the incecption to October, 2021. The quantitative analyses were conducted on the characteristics of the clinical studies, including years of publication, journals, affiliations of authors, interventions, diseases, types of studies, evaluation of efficacy, rate of citations.Results:A total of 81 articles were included in 44 journals. The majority types of studies were RCTs, the main intervention was acupuncture as well as acupuncture combined with Traditional Chinese Medicine or medicine. For types of diseases, digestive system diseases account for the major part (66.67%), followed by nervous system diseases (17.28%).Conclusions:The studies associated with Experienced Ten Acupoints were relatively few, the corresponding spectrum of diseases was in limit range, and experimental studies lacks. This study suggested that the clinicians should pay attention to the classical acupoints prescriptions, and the research progress of clinical trials should be more standardized.

RESUMEN

Objective:To explore the effect of Plantaginis semen on the expressions of AQP4 gene and protein in small intestine of diarrhea rats. Methods:Sixty Sprague-Dawley male rats were randomly divided into normal control group, model control group, hydrochlorothiazide group, and low-, medium-, and high-dose Plantaginis semen groups. Intragastric administration of Senna (20 ml/kg) was implemented in the morning for 5 groups except the normal group. The rats in the low-, medium-, and high-dose Plantaginis semen groups were intragastrically administered with 0.95, 1.9, 3.8 g/kg Plantaginis semen formula granule solution, while the rats in hydrochlorothiazide group were given hydrochlorothiazide suspension 9 mg/kg by gavage according to 1 ml/100 g body weight for 14 days. The loose stool rate, average ranking of loose stool, and diarrhea index were compared according to the fecal traits and stool times after 14 days of treatment, and the small intesine tissue were collected. Hematoxylin-eosin (HE) staining to observe the pathological morphological changes in small intestine, and Quantiative Real-time PCR and Western blot were applied to detect the gene and protein expressions of AQP4. Results:Compared with the model group, the loose stool rate (50.89% ± 6.17%, 41.14% ± 4.48%, 36.37% ± 4.81 % vs. 67.45% ± 7.35%), the average ranking of loose stool (2.16 ± 0.34, 1.73 ± 0.28, 1.52 ± 0.25 vs. 2.63 ± 0.29), and the diarrhea index (1.10 ± 0.19, 0.71 ± 0.11, 0.57 ± 0.12 vs. 1.77 ± 0.24) of rats in each group of Plantaginis semen significantly decreased ( P<0.05); the degree of intestinal mucosal injury, hyperemia and neutrophil infiltration were alleviated; the expressions of AQP4 mRNA (0.48 ± 0.10, 0.69 ± 0.12, 0.97 ± 0.15 vs. 0.21 ± 0.03), and the protein of AQP4 (0.59 ± 0.08, 0.64 ± 0.09, 0.78 ± 0.11 vs. 0.32 ± 0.05) in the small intestine tissue of Plantaginis semen groups significantly increased ( P<0.05). Conclusions:Plantaginis semen has antidiarrheal effect, and its mechanism is related to up-regulation the gene and protein expressions of AQP4, addition of water absorption and promotion of water and fluid metabolism.

Objective To observe the effect of Chaijin-Huayu decoction combined with endoscopic minimally invasive for gallbladder polyposis and its effect on quality of life.Methods A total of 91 patients with gallbladder polyposis were selected from March 2014 to early March 2017 outpatients and wards of Hebei Provincial Traditional Chinese Medicine Hospital.According to the method of random number table,the paitents were randomly divided into the observation group (n=46) and the control group (n=45).After three courses of treatment,the total effective rate,TCM symptom score and quality of life were observed in both groups,and the postoperative complications of the two groups were compared.Results After three courses of treatment,the total effective rate of the observation group was 91.3% (42/46),while that of the control group was 88.9% (40/45).There was no significant difference between the two groups (x2=0.149,P=0.592).In the observation group,the scores of subcardiac pycnosis and belching,right flank distended pain and burp were lower than those of the control group (t were 2.431,3.560 and 2.891 respectively,all P<0.05).In the SF-36 scale,the scores of mental health,social function,physical pain,physiological function,emotional function,vitality,physiological function and overall health of the observation group were higher than those of the control group (t were 2.342,2.831,2.765,2.545,2.485,2.650,2.582,2.325 respectively,all P<0.05);The incidence of surgical complications of the observation group was 2.2% (1/46),while that of the control group was 17.8% (8/45),and the difference between the two groups was statistically significant (x2=0.149,P>0.05).Conclusions Chaijin-Huayu decoction combined with minimally invasive choledochoplasty could relieve symptoms,improve quality of life and reduce postoperative complications in patients with gallbladder polyposis.
