RESUMEN

<p><b>OBJECTIVE</b>To explore the pathogenesis of tumors by blocking the normal differentiation process of stem cells.</p><p><b>METHODS</b>Bone marrow mesenchymal stem cells (BMSCs) from rats were isolated, cultured and purified by whole bone marrow adherence method. The rat BMSCs were induced to differentiate into adipocytes with dexamethasone, insulin and indomethacin. Blockage of the differentiation process was induced by 3-methylcholanthrene (3-MC).</p><p><b>RESULTS</b>The differentiation experiment showed that at 30 days after the induction, oil red O staining-positive cells occurred with increased intracytolasmic lipid droplets, characteristic for adipocytes. The differentiation blockage experiment showed that at 30 days after induction, the deposits of oil red O staining-cytoplasmic lipid droplets was significantly reduced, indicating that the blocked cells were adipocytes, but not fully differentiated. Morphological identification showed that cell contact inhibition disappeared, abnormal cell nuclei, increased number of micronucleus aberration and karyotype abnormalities, indicating that malignant transformation of the stem cells occurred after the differentiation blockage.</p><p><b>CONCLUSIONS</b>The results of this study show a blockage of the differentiation of that stem cells at the intermediate phase, and a tendency of malignant transformation of the stem cells. The results of our study provide new evidence that cancer stem cells may be originated by suppression of stem cell differentiation.</p>

RESUMEN

Objective To design and build a new dynamic load and circulating perfusion bioreactor system and test its performance. Methods The design principle of the bioreactor system was specified and the dynamic strain loading system and 3D perfusion culture system were designed and built accordingly. A special culture chamber for 3D perfusion and compressive loading was also developed. The sterility of the culture chamber and the accuracy and stability of the strain loading were measured, and the result from the culture of tissue engineering bone was preliminarily observed. Results This bioreactor system could provide compressive strains with different magnitudes and frequencies, as well as perfusions under different flow conditions. It could be controlled accurately and operated easily with a steady performance. No germs were grown in the culture medium after 5 days’ running. The preliminary results showed that after the tissue engineering bones were cultured in the bioreactor for 10 days, cell proliferation and ALP activity in this perfusion culture and loading group were significantly higher than those in the static culture group and the simple perfusion culture group. Conclusions The bioreactor could be an ideal dynamic culture and loading device for biomechanical study of tissue engineering bone.

RESUMEN

Objective To solve the danger and difficulty in transferring seriously injured victims. Methods The operating principle, construction design, electronic control system and software program flowchart of a robot transfer arm for victim-transfer were introduced.Results and Conclusion The victim didn not have to change their body posture during transfer. The procedure is very simple.A push at only one key is enough,without secondary injury.

RESUMEN

<p><b>OBJECTIVE</b>To observe the effect of Fuzheng Yiliu Granule (FZYLG) on cell cycle and nuclear transcription factor-kappa B (NF-kappa B) in tissue of esophageal-gastric carcinoma.</p><p><b>METHODS</b>Seventy-six patients with esophageal gastric carcinoma were randomly divided into two groups, the FZYLG group and the control group. FZYLG was given to the former for 15 days. The tumor tissue in both groups was resected and cell cycle and apoptosis rate as well as NF-kappa B were determined by flowcytometry.</p><p><b>RESULTS</b>Level of NF-kappa B in the treated group was significantly higher than that in the control group (P < 0.05). In the treated group, the percentage of G0/G1 stage cells were significantly increased and that of S stage significantly decreased (both P < 0.05). At the same time, obvious cell apoptosis was found in the treated group, the apoptosis rate of which was significantly higher than that in the control group (P < 0.01).</p><p><b>CONCLUSION</b>FZYLG can increase the NF-kappa B expression, block the proliferation to promote the apoptosis of tumor cells.</p>
