RESUMEN

Objective:To observe the effect of self-designed Buqi Huoxue Decoction on the rehabilitation of patients with stroke sequelae and qi deficiency and blood stasis syndrome.Methods:A total of 116 patients with stroke sequelae treated from October 2019 to October 2020 were selected and divided into observation group and the control group by randomized digital tables, 58 patients in each group. The control group was given the conventional treatment, and the observation group combined self-designed Buqi Huoxue Decoction and conventional treatment. Both groups were treated for 3 months. The soluble vascular cell adhesion molecule-1 (sVCAM-1), nito-oxide (NO), endothelin (ET-1), endothelial-derived hyperpolarization factor (EDHF), insulin-like growth factor (IGF-II), interleukin-1β (IL-1β), interleukin-6 (IL-6) levels were detected by ELISA. The National Institutes of Health Stroke Scale (NIHSS) was used to evaluate the degree of neurological deficit, motor function was evaluated by the Fugl-Meyer motor function scale (FMA), patients' daily living ability was assessed by daily living ability assessment table (ADL), the QLQ-C30 was used to evaluate the QoL of patients. The TCM syndrome scores were evaluated. The clinical response was compared between the two groups.Results:The total response rate was 94.8%(55/58) in the observation group and 81.0%(47/58) in the control group. There was significant difference between the two groups ( χ2=3.98, P=0.046). After treatment, the scores of TCM syndrome and NIHSS of pruritus in the observation group were significantly lower than those in the control group ( t values were 5.61, 5.21, respectively, all Ps<0.01). After treatment, the scores of FMA, ADL QLQ-C30 of pruritus in the observation group were significantly higher than those in the control group ( t values were 3.61, 3.13, 9.38, respectively, all Ps<0.01). After treatment, the NO[(87.61±19.18) μmol/L vs. (77.93±17.26) μmol/L, t=2.81], EDHF [(21.14±6.14) μmol/L vs. (16.61±4.52) μmol/L, t=4.45] levels in the observation group were significantly higher than those in the control group ( P<0.01). After treatment, the sVCAM-1[(309.58±30.11) μg/L vs. (354.16±33.04) μg/L, t=7.46], ET-1 [(50.07±18.23)ng/L vs. (66.31±17.89)ng/L, t=4.76] levels in the observation group were significantly lower than those in the control group ( P<0.01). After treatment, IL-1β, IGF-Ⅱ, IL-6 levels in the observation group were significantly lower than those in the control group ( t values were 4.66, 8.14 7.43, respectively, all Ps<0.01). Conclusion:The treatment of self-designed Buqi Huoxue Decoction can improve the vascular endothelial function, inhibit the inflammation, reduce the nerve function damage, improve the limb movement function, daily life ability, quality of life and the clinical symptoms.

Objective@#To detect the expression of GREM1 gene in gastric cancer cells, investigate its effects on the biological characteristics of gastric cancer cells and evaluate its application value in the diagnosis and prognosis of gastric cancer. @*Methods@#The expression difference of GREM1 in gastric cancer tissues and adjacent normal tissues was analyzed by the database, and the correlation of GREM1 expression levels with the prognosis of gastric cancer patients was evaluated. The expression levels of GREM1 protein in gastric cancer cell lines were detected by western blot. After GREM1 gene in AGS cells was silenced, its effects on the proliferation, migration, epithelial-mesenchymal transition (EMT) and Wnt/β-catenin pathway of AGS cells were detected by the colony formation assay, Transwell and Western blot, respectively. @*Results@#Kaplan-Meier analysis showed that the patients with high expression of GREM1 gene had low overall survival (OS) and progression-free survival (PFS). The expression level of GREM1 protein in AGS cells was the highest in all gastric cancer cell lines (1.967 ± 0.056). The analysis of colony formation assay, Transwell and Western blot showed that the silencing of GREM1 gene could decrease the proliferation and migration of gastric cancer cells (t=22.00; t=29.60; P<0.01), increase the expression of E-cadherin (t=10.65, P<0.01), and decrease the expressions of ZEB1 and MMP2 (t=10.74; t=13.67; P<0.01) and the expressions of β-catenin, Cyclin D1, c-myc, p-GSK3β and PCNA in the Wnt/β-catenin pathway (t=12.65; t=16.21; t=8.74; t=7.75; t=8.42; P<0.01). @*Conclusion@#GREM1 may induce EMT by activating the Wnt/β-catenin pathway, [JP2]and promote the metastasis and growth of tumors, which may be used as a new molecular diagnostic and prognostic marker for gastric cancer.

Objective@#To understand the current situation and impacts of household factors on of unhealthy dietary habits among preschool children, and to provide reference for preventing the occurrence of unhealthy dietary habits of preschool children.@*Methods@#By using stratified random cluster sampling method, the study selected 1 070 children aged 3-6 years in 7 kindergartens in Urumqi. Household general information , children’s dietary behavior and parenting environment was collected through parent questionnaire survey.@*Results@#The prevalence rate of unhealthy dietary habits among preschool children was 32.71%. Multifactor Logistic regression analysis showed that the unhealthy dietary habits of preschool children were negatively correlated to the following factors, including age of children [β=-0.32, 95%CI=(0.62,0.86)] , gender[β=-0.33,95%CI=(0.53,0.97)], educational level of mothers [β=0.94, 95%CI=(1.79,3.68)] , type of fanmily [β=0.64, 95%CI=(1.414,2.522)] and the scores of social adaptation/selfcare and environmental atmosphere in the family nurturing environment [β=-0.07, 95%CI=(0.90,0.97); β=-0.21, 95%CI=(0.76,0.87)].@*Conclusion@#The cultivation of the dietary habits should pay more attention on younger children and boys, and the enhancement of health education on the children’s eating behaviors among mothers with lower education background and the primary child caregivers in the stem family. Family nurturing environment should be actively improved, in order to prevent the occurrence of children’s unhealthy dietary habits.

RESUMEN

We have developed a rapid and high throughput lipase-ANS (8-Anilino-l-naphthalenesulfonic acid) assay to evaluate the thermo-stability of lipases based on the ANS fluorescence signal's increasing and shifting when this small fluorescence probes binds to lipase. The testing lipase samples were incubated at a temperature range of 25 degrees C to 65 degrees C for 30 min before mixed with ANS solution (0.20 mg/mL lipase and 0.05 mmol/L ANS in the buffer of 20 mmol/L Tris-HCl, 100 mmol/L NaCl, pH 7.2) in a cuvette or microplate. Fluorescence signals of the samples were measured at EX 378 nm, EM 465 nm with a fluorescence photometer or a plate reader, and Tm was calculated with the software of GraphPad Prism5.0. The Tm values of several mutants of Penicillium expansum lipase (PEL) were measured with this ANS assay and conventional method simultaneously and the results show that Tm values are comparative and consistent between these methods, suggesting that the lipase-ANS assay is a reliable, rapid and high throughput method for lipase thermo-stability measurement.

@#ObjectiveTo investigate the relationship between high-sensitivity C reactive protein (hs-CRP) and oxygen uptake efficiency slope (OUES) in patents with cardiovascular disease. MethodsExercise tests, following a symptom-limited standard Bruce protocol with simultaneous respiratory gas-exchange measurements, were performed on a treadmill in 17 post-PCI patients, 21 stable angina pectoris and 24 patients with essential hypertension (EH). The oxygen uptake (VO2), minute ventilation (VE) were determined. The OUES was derived from the relation between VO2 (ml/min) and VE (L/min) during incremental exercise and was determined by VO2=a×lgVE+b, where a = OUES. The OUES was calculated from data of the first 75% (OUES75) and 100% (OUES100) of exercise duration. The serum hs-CRP was measured by immunoassays, and the left ventricular ejection fraction (LVEF) was measured with echocardiography. ResultsOUES75 (r=-0.506,P<0.001), OUES100 (r=-0.567,P<0.001) and LVEF (r=-0.286,P<0.01) were correlated with lg(hs-CRP). In stepwise multivariable linear regression models, lg(hs-CRP) (β=-0.374,P=0.006), body height (β=-1.854,P=0.036), body weight (β=-5.033,P=0.034)were independent risk factors of lower OUES75. lg(hs-CRP) (β=-0.396,P=0.003), body height (β=-2.157,P=0.013), body weight (β=-5.912,P=0.011) were independent risk factors of lower OUES100. ConclusionThe serum leveles of hs-CRP may be the useful marker that reflecting cardiopulmonary functional reserve and left ventricular function.

@#Objective To investigate the characteristics of pulmonary function after completed cervical spinal cord injury (CSCI) and the effect of pulmonary rehabilitation with abdominal pressure. Methods 32 male patients with CSCI were measured with pulmonary function test (PFT), and 10 of them accepted pulmonary rehabilitation with 3~5 kg pressure on their abdomen 30 min per day for 8 weeks. Results Their forced expiratory volume in one second (FEV1), vital capacity (VC), forced vital capacity (FVC), peak expiratory flow (PEF) were all less than 50% prediction. After 8 weeks, the pulmonary function of 10 patients who received exercise significantly improved (P<0.01). Conclusion The pulmonary function impaired in patients with CSCI. The pulmonary rehabilitation with pressure on abdomen can improve the pulmonary function.
