RESUMEN

OBJECTIVE To study the effects of the active component 17-hydroxy-jolkinolide B (HJB) of Euphorbia fischeriana on the proliferation and apoptosis of human triple-negative breast cancers (TNBC) MDA-MB-231 and MDA-MB-468 cells. METHODS MTT assay was adopted to detect the inhibitory rate of MDA-MB-231 and MDA-MB-468 cells proliferation after treated with 0 (blank control),5,10,20,40,80 μmol/L HJB for 24, 48 and 72 h. Laser confocal microscope and flow cytometry were adopted to detect the apoptosis, mitochondrial membrane potential(MMP) and reactive oxygen species (ROS) of above 2 kinds of cells after treated with 0 (blank control), 10,20,40 μmol/L HJB for 24 h. Western blot assay was used to detect the expressions of B cell lymphoma-2( Bcl-2), Bcl-2-associated X protein (Bax), cytochrome-C (Cyt-C), caspase-3, cleaved caspase- 3, caspase-9 and cleaved caspase-9. RESULTS Compared with blank control group, 5,10,20,40,80 μmol/L HJB could significantly increase the inhibitory rate of MDA-MB-231 and MDA-MB-468 cells proliferation (P<0.05), in dose- and time- dependent trend. After 24 h treatment of HJB (10,20,40 μmol/L), the apoptosis of above 2 kinds of cells increased, and the total apoptotic rate increased significantly (P<0.05); the mitochondrial membrane potential decreased significantly (P<0.05); the level of ROS increased significantly (P<0.05); the protein expressions of Bcl-2, caspase-3 and caspase-9 were decreased significantly (P< 0.05), while the protein expressions of Cyt-C, Bax, cleaved caspase-3 and cleaved caspase-9 were increased significantly (P<0.05). CONCLUSIONS HJB can inhibit the proliferation of MDA-MB-231 and MDA-MB-468 cells, and induce their apoptosis.

RESUMEN

Objective:To investigate the prevention and treatment for deviation of costal cartilage dorsal onlay grafts in rhinoplasty.Methods:From January 2010 to October 2020, a total of 588 patients (83 male cases, 505 female cases, age range from 25 years to 55 years, 32±4 years in average) accepted costal cartilage rhinoplasty in Shenzhen Mylike Medical Plastic Aesthetic Hospital. During the operation, various methods were used in the process of catilage selection, water bath, sculpture, treatment of nasal dorsal, graft fixation and fascial modification to prevent and treat the deformation and displacement of the costal cartilage dorsal onlay grafts.Results:The immediate postoperative photograph of 588 cases showed that costal cartilage dorsal onlay graft was put on the middle of dorsum. After a follow-up period of 396 cases from 6 to 60 months, the average follow-up period was 12.1 months, there were 44 cases happened with the deviation of dorsal onlay grafts, and deviation was managed after the second rhinoplasty surgery. There was no prolonged function sequela such as ventilation dysfunction, abnormal sensation, or hyposmia occured. 362 cases were satisfied with the aesthetic effect.Conclusions:It is particularly important to grasp the principles of managing costal cartilage in rhinoplasty and to learn how to prevent and treat postoperative complications of costal cartilage dorsal onlay graft.

RESUMEN

To explore the application effect of team-based learning (TBL) model in functional experiment teaching. A clinical case related to the animal experiment was selected and distributed to students through Internet before class. Individual tests for students were conducted in class to assess their understanding for the case. Then group tests were carried out, which were completed by in-group discussion. Finally, the clinical case was analyzed within group. The teaching process possesses several characteristics and advantages in helping students retrieve literature, cultivate their spirit of cooperation, their ability of clinical practice and so on, but it also has some shortcomings and deficiencies. Therefore, this paper summarizes the advantages and disadvantages of TBL teaching in functional experiment teaching based on existing experience so as to promote a broader application of TBL in our medical education.

OBJECTIVE: To study the inhibitory effect of dictamnine on the viability of mice spleen lymphocyte in vitro and explore its potential mechanism. METHODS: The primary spleen lymphocytes of mice were isolated and cultured. The cells were treated with 0 (blank control), 50, 100, 150 μmol/L dictamnine for 24 h. MTT assay was used to determine the cell viability; Lactic dehydrogenase (LDH) assay was used to determine release rate of LDH. Early apoptosis rate was detected by flow cytometry. Necrosis rate was detected by Hoechst 33342 and PI double staining; Western blot assay was used to detect the protein expressions of Caspase 3 and Cleaved-Caspase 3 in cells. Comet assay was used to detect DNA damage in cells (reflected in the proportion of DNA tail area). RESULTS: Compared with blank control, 100, 150 μmol/L dictamnine could significantly inhibit the viability of lymphocytes (P<0.01). 150 μmol/L dictamnine could significantly increase the release of LDH (P<0.05), and release rate reached 79.37%. 50, 100, 150 μmol/L dictamnine could improve the early apoptotic rate of lymphocyte, but there was no statistical significance (P>0.05). 150 μmol/L dictamnine could significantly increase the necrosis rate (P<0.05), and necrosis rate reached 78.64%. 50, 100, 150 μmol/L dictamnine could increase the protein expression of Caspase 3, but there was no statistical significance (P>0.05), while 50, 100 μmol/L dictamnine could improve the protein expression of Cleaved-Caspase 3 significantly (P<0.05). DNA damage was induced in a dose-dependent manner by dictamnine, in which 100 and 150 μmol/L dictamnine could significantly increase DNA tail area (P<0.01). CONCLUSIONS: Dictamnine can inhibit spleen lymphocyte viability, and the mechanism may be related to inducing spleen lymphocyte necrosis and DNA damage.
