RESUMEN

Objective@#To determine human tear proteins using nanoliter liquid chromatography coupled to quadrupole orbitrap mass spectrometry (NanoLC-Q-Orbitrap-MS), and perform a bioinformatics analysis of main proteins.@*Methods@#Human tear samples were collected with capillary, transferred to 3 kDa ultrafiltration tubes containing 400 μL of superpure water and centrifuged at 12 000×g for 15 min. Repeated extraction of tear proteins were performed four times, and following digestion with trypsin, the proteins were separated using the Waters NanoAcquity peptide BEH C18 column (1.7 μm, 100 μm×100 mm) and determined using NanoLC-Q-Orbitrap-MS with the mobile phase of 0.1% formic acid aqueous solution and acetonitrile (0.1% formic acid) in the full MS/dd-MS2 mode. The types of proteins were characterized in the Uniprot database using the software Proteome Discoverer version 2.1 and verified using bovine serum albumin. The tear proteins were subjected to gene annotation analysis using the String database.@*Results@#A total of (387±160) human tear proteins were yielded, with a relative standard deviation (RSD) of 4.13%, and there were 25 types of proteins with a relative high abundance, including lipocalin 1, lysozyme and lactoferrin. The peptide sequence coverage of bovine serum albumin was (86.08±2.61)%, with a RSD of 3.03%. The 25 major tear proteins were involved in substance transduction among cells, homeostasis process, negative regulation of the endopeptidase activity, detection of chemical stimulants and humoral immune responses, and the 16 proteins had close interactions. Lacritin, lipocalin 1, lactoferrin, lysozyme and zinc-α 2-glycoprotein, which had a relative high abundance, had close biological connections.@*Conclusion@#NanoLC-Q-Orbitrap-MS is stable, reliable and feasible for detection of multiple proteins in tears.

RESUMEN

This study retrospectively analyzed the clinical data of 1 case of spermatic cord leiomyosarcoma admitted to the urology department of the Affiliated Hospital of Chengde Medical College. The clinical characteristics, diagnosis, treatment and prognosis was discussed with the literature review. Radical resection of the left testicle and high ligation of the left spermatic cord were performed. Postoperative pathology was spermatic cord leiomyosarcoma. Its clinical manifestations are painless masses, which are mainly confirmed by pathological examination. The treatment is mainly radical resection, and postoperative radiotherapy can improve the prognosis and reduce recurrence.

@#Objective To investigate the construct and concurrent validity of the Functional Gait Assessment (FGA) as the measure for balance and gait during walking in Parkinson's disease patients. Methods From March to December, 2011, 121 patients with Parkinson's disease (mean aged 61.9 years) were evaluated with FGA by one rater, as well as the other scales for balance and gait, such as Berg Balance Scale, Functional Ambulation Category, Timed Up and Go Test, Activities-specific Balance Confidence Scale, Movement Disorders Society revision of the Unified Parkinson's Disease Rating Scale part 3, modified Barthel Index, maximum walking speed and Modified Hoehn and Yahr Scale. Principal Component Analysis was used to determine construct validity. Spearman correlation coefficients between the FGA and other measures were used to determine concurrent validity. Results One common factor was extracted, which cumulatively explained 64.0% of the total variance. The FGA correlated moderately with the other measures (r = 0.57-0.85). Conclusion FGA is good in validity for Parkinson's disease patients.

Objective To explore the significance of long non-coding RNA (lncRNA) taurine up-regulated gene 1 (TUG1) in hepatocellular carcinoma (HCC),to predict the target gene of TUG1,and to provide a ref-erence for further study of TUG1 in HCC.Methods The differential expression of TUG1 in HCC was ana-lyzed by using the UALCAN database and the survival analysis of TUG1 was performed.The target gene of TUG1 was predicted by RegRNA 2.0 biology software,HMDD,targetscan and microT-CDS,and the regulato-ry network of lncRNA TUG1-microRNAs-mRNAs was constructed.The predicted target gene was analyzed by Gene Ontology (GO) and KEGG signal transduction pathway enrichment by using FunRich platform. Results TUG1 expression in HCC was significantly increased,and the expression level of TUG1 increased generally with the increase of tumor grade.The overall survival of patients with low expression of lncRNA TUG1 was significantly longer than that of lncRNA TUG1 high expression patients.There were four possible binding sites of HCC related microRNAs (hsa-mir-122-5p,hsa-mir-200a-3p,hsa-mir-34c-3p,hsa-mir-629-3p) on TUG1,which regulated 245 downstream target genes and formed the regulatory network of lncRNA TUG1-microRNAs-mRNAs.In the biological process,microRNA target genes were highly enriched in the processes such as the regulation of nucleobase,nucleoside,nucleotide and nucleic acid metabolism.In KEGG pathway analysis,microRNA target genes were highly enriched to the signal pathways mediated by Syndecan and TRAIL.Conclusion TUG1 expression level in HCC increased.Increased expression of TUG1 is associat-ed with poor prognosis in HCC.Bioinformatics methods can be used to explore the mechanism of tumorigene-sis from the molecular level,which can provide valuable information for subsequent experiments and clinical diagnosis and treatment.

Objective To explore the relationship between DNA methyctransferace1(DNMT1)gene polymorphism and cognitive function in the first episode, drug-na?ve schizophrenia. Methods One hundred eighty-six first episode, drug-na?ve schizophrenia patients and 182 healthy controls were examined by using polymerase chain reaction (PCR), denaturing polyacrylamide gel electrophoresis and silver staining to determine genotype and allele of DNMT1 gene. Cognitive function was assessed by using the standardized measurement tools—the Measurement and Treatment Research to Improve Cognition in Schizophrenia (MATRICS) Consensus Cognitive Battery (MCCB) Chinese version. Comparisons were made on the cognitive function and the genotype and allele distribution of SNP rs2114724 and rs2228611 between schizophrenia group and the control group.The relationship between DNMT1 gene polymorphism and cognitive function was analyzed. Results There was no significant difference of the genotype T/T, T/C, C/C and allele T, C distribution of SNP rs2114724 between schizophrenia group and healthy controls group (P>0.05). There was no significant difference of the genotype G/G, G/A, A/A and allele G, A distribution of SNP rs2228611 between schizophrenia group and healthy control group (P>0.05). There was significant difference in Spatial Span Test (SST) scores(P<0.05)among three different genotype T/T,T/C and C/C of rs2114724 in schizophrenic group.The SST scores of the patients with C/C genotype were higher than that of the patients with T/T or T/C genotype (P<0.05). Conclusion DNMT1 gene polymorphisms is not associated with the first-episode, drug-na?ve schizophrenia, but may be associated with the work memory in cognitive function of schizophrenia in the Chinese Han population.

Objective To explore the influence of duration of untreated psychosis (DUP)on cognition and social function in first-episode drug-na?ve schizophrenia. Methods Ninety-three first-episode drug-na?ve schizophrenic were enrolled as the schizophrenia group and 93 normal people served as the control group. The Positive and Negative Symptoms Scale (PANSS) were used to assess the degree of mental symptoms. The Matrics Consensus Cognitive Battery (MCCB) was used to evaluate the cognitive function. Personal and Social Performance Scale (PSP) and the Global Assessment Function Scale (GAF) were used to evaluate the social function. Result There were significant differences in scores of Trail Marking Test, Symbol Coding test, Category Fluency test, Stroop color word test, Continuous Performance Test, Spatial Span test, Brief-Visuospatial Memory Test-Revised (HVLT-R), Brief-Visuospatial Memory Test-Revised (BVMT-R) and Maze test between the schizophrenia group and the control group (all P<0.05). There were significant differences in scores of GAF and PSP between the schizophrenia group and the control group (all P<0.05). DUP was negatively related to the score of HVLT-R2 (r=-0.265, P=0.010) or BVMT-R3 (r=-0.328, P=0.001). DUP was negatively related to the scores of GAF score(r=-0.292,P=0.005)or PSP score(r=-0.397,P<0.001). Conclusion There are social function impairment and a wide range of cognitive function impairment in the first-episode drug-na?ve schizophrenic. The length of the DUP is associated with the severity of the social functional and cognitive functional impairment.

OBJECTIVE:To establish the method for simultaneous determination of epiberberine,jateorhizine hydrochloride, coptisine,palmatine hydrochloride and berberine hydrochloride in Lianshen tonglin tablets. METHODS:HPLC method was adopt-ed. The determination was performed on Thermo Scientific Syncronis C18 column with mobile phase consisted of acetonitrile-0.1%phosphoric acid solution(pH adjusted to 3.0 with triethylamine)(30:70,V/V)at the flow rate of 0.3 mL/min. The detection wave-length was set at 345 nm,and column temperature was 25 ℃. The sample size was 2 μL. RESULTS:The linear ranges were 4.08-163 ng for epiberberine(r=0.9999),5.72-229 ng for jateorhizine hydrochloride(r=0.9998),5.67-227 ng for coptisine(r=0.9999),6.78-271 ng for palmatine hydrochloride(r=0.9996),13.2-526 ng for berberine hydrochloride(r=0.9999). RSDs of pre-cision,stability and reproducibility tests were lower than 2.0%. The recoveries were 96.83%-103.92%(RSD=2.2%,n=9), 97.62%-104.85%(RSD=2.9%,n=9),99.44%-104.18%(RSD=1.8%,n=9), 98.64%-104.42%(RSD=1.8%,n=9), 98.24%-102.40%(RSD=1.5%,n=9),respectively. CONCLUSIONS:The developed method is simple,sensitive,rapid and accu-rate. It is suitable for simultaneous determination of 5 coptis alkaloids in Lianshen tonglin tablets.

Objective:To determine aflatoxin B1 , B2 , G1 and G2 in Shugan pills by HPLC and evaluate their safety. Methods:The product was analyzed by HPLC using a Shiseido C18 column(150 mm × 4. 6 mm,5 μm)with methanol-acetonitrile-water (10 :30 :60) as the mobile phase. An ultraviolet lamp was derivative at 254 nm, and the fluorescence detection was performed with the excitation wavelength at 360 nm and the emission wavelength at 450nm. The samples from nation-wide market were detected by the method. Re-sults:Good linear relationships were obtained within the range of 10-50 pg for aflatoxin B1 and G1 , and 3-15 pg for aflatoxin B2 and G2 . The recovery of aflatoxin B1 , B2 , G1 and G2 was 89. 9%,87. 9%,86. 1% and 87. 1%,respectively with all the RSD below 3. 0%. The results of all samples were satisfactory, and the qualification rate of the samples was 100%, indicating that the samples were safe. Conclusion:The developed method is convenient and accurate, which can be applied in the quantitative determination of Shugan pills.

Objective To explore and analyze the effect of ambroxol hydrochloride combined with budesonide in treatment of children bronchial pneumonia on C-reactive protein level and WBC levels.Methods 100 cases of patients with bronchial pneumonia treated in our hospital from May 2015 to December 2016, were randomly divided into observation group and control group, 50 cases in each group.The control group were given routine treatment, while the observation group were given ambroxol hydrochloride combined with budesonide treatment on the basis of the control group,the course of treatment was 14 days.C-reactive protein and WBC levels were measured before treatment and 14 days after treatment, the Resultswere observed and statistical.ResultsThere were significant differences in the content of CRP between groups and multiple time points(F=11.50, P=0.003;F=4.78, P=0.03;F=5.45, P=0.02).The content of CRP 14 days after treatment in the observation group were significantly lower than the control group(t=14.12,P<0.05), the difference was statistically significant.There were significant differences in the WBC counts between groups and multiple time points(F=11.52, P=0.003;F=4.73, P=0.03;F=5.44, P=0.02),the difference was statistically significant.The count of WBC 14 days after treatment in the observation group after treatment were significantly lower than the control group(t=8.12, P<0.05), the difference was statistically significant.The number of cured, effective and ineffective cases in the control group was 20, 13 and 15, respectively.The observation group was 30, 15, and 5 cases, respectively.By rank sum test, the difference of the effective rate between the 2 groups was statistically significant(u=2.33, P<0.05).And the total effective rate in observation group was 90.0%, which was significantly higher than the control group(70.0%)(x2=6.25, P<0.05), the difference was statistically significant.Conclusion Ambroxol hydrochloride combined with budesonide in the treatment of bronchial pneumonia in children is effective and can effectively reduce the CRP and WBC count, so it is worth popularizing widely in clinic.

Objective To investigate the reliability of Berg Balance Scale (BBS) applied in patients with Parkinson's disease (PD). Methods 121 PD inpatients from March to December, 2011 were assessed with BBS by 2 raters, and the testing procedure was videoed. One of the raters assessed with BBS via video 4 weeks later. The intraclass correlation coefficient (ICC) and Kappa coefficient between raters and between tests were investigated. Results The ICC was 1.00 of the total score between raters and 0.99 between tests, while the Kappa co-efficient were 0.66 to 0.93 and 0.69 to 0.99 of the items. Conclusion BBS is reliable in interrater and test-retest as applied in PD patients.

Objective To discuss medicine using regularity of Yan Shiyun’s treatment for arrhythmia. Methods Medical cases of arrhythmia treated by Yan Shiyun were collected. The method of frequency of SPSS19.0 was used to analyze the frequency of single herb and common prescriptions. Results Totally 112 medical cases were collected. Three different kinds of arrhythmia in totally 315 prescriptions were collected and 18 same herbs were used in the 20 highest frequently used herbs. The first two were Chuanxiong Rhizoma and Persicae Semen. Buyang Huanwu Decoction is the most commonly used prescription, followed by Zhenwu Decoction. Conclusion Most Chinese herbal medicine included in Yan Shiyun’s prescriptions can tonify qi and invigorate the circulation of blood. The medicine use is concentrated, and composition law is clear.

Objective To construct plvx-cyclooxygenase-2(COX-2)-DsRed and establish breast cancer cell line MCF7 which overexpressed COX-2, to explore the effect of COX-2 on breast cancer cell.Methods The full-length COX-2 PCR product was obtained by total COX-2 PCR primers and COX-2 cDNA vector.After the PCR product and lentiviral vector plvx-DsRed-Monomer-N1 were cut simultaneously by restriction enzyme BamH1 and Xholl, they were connected and sequenced, to get lentiviral vector plvx-COX-2-DsRed.After selected by puromycin, overexpressed COX-2 breast cancer cell line MCF7-plvx-COX-2-DsRed was obtained.The stable cell line was verified by real time PCR and Western blot.The differences of proliferation ability between stable cell line and normal one were compared by colony formation test and Western blot.Results The lentiviral vector plvx-COX-2-DsRed and stable cell line MCF7-plvx-COX-2-DsRed after selecting were obtained.COX-2 expression level of the stable cell line was 75.29 times as high as that of MCF7, and 64.91 times as high as that of cell line MCF7-plvx-DsRed-Monomer-N1 by PCR assay (P < 0.05), which was consistent with the results of Western blot and microscope photo.MTT results showed that cell line MCF7-plvx-COX-2-DsRed had grown faster than cell line MCF7 and MCF7-plvx-DsRed-Monomer-N1 from the 2nd day (P < 0.05), which was accordant with colony formation assay.MCF7-plvx-COX-2-DsRed cell line had higher c-myc expression and lower β-catenin expression than MCF7 cell and cell line MCF7-plvx-DsRed-Monomer-N1 detected by Western blot relative quantification (P < 0.05).Conclusion The plvx-COX-2-DsRed lentiviral vector and cell line MCF7-plvx-COX-2-DsRed are successfully constructed.COX-2 can increase proliferation of MCF7 cells through up-regulating the expression of c-myc.

<p><b>OBJECTIVE</b>The effect of sandblasting on the bond strength between 3mol% yttrium-stabilized tetragonal zirconium polycrystal (3Y-TZP) zirconia framework and veneering porcelain was evaluated.</p><p><b>METHODS</b>A total of 21 specimens [(25 ± 1) mm x (3 ± 0.1) mmx (0.5 ± 0.05) mm] were prepared according to ISO 9693. The specimens were then randomly divided into 3 groups. Sandblasting was performed on 2 meshes of Al₂O₃ particles: group A with mesh 110 and group B with mesh 80. Group C, which was not sandblasted, was the control group. The surface roughness of the zirconia framework, as well as the bond strength between 3Y-TZP zirconia framework and veneering porcelain, was measured. The interface microstructure was observed by scanning electron microscope (SEM), and elemental distribution was detected by energy dispersive spectroscopy (EDS).</p><p><b>RESULTS</b>Surface roughness values were (1.272 ± 0.149) μm for group A, (0.622 ± 0.113) μm for group B, and (0.221 ± 0.065) μm for group C. Statistical significance were found among groups (P < 0.05). The bond strength values were (28.21 ± 1.52) MPa for group A, (27.71 ± 1.27) MPa for group B, and (24.87 ± 3.84) MPa for group C. Statistical significance was found between group A and group C (P < 0.05), whereas the other groups had no statistical significance (P > 0.05). Interface adhesion failure was the primary performance. SEM images showed the close interface bonding, and EDS showed that the interface had no obvious element penetration.</p><p><b>CONCLUSION</b>Al₂O₃ sandblasting can slightly enhance the bond strength between zirconia framework and veneering porcelain.</p>

This study was aimed to determine effect of Bu-Shen Kang-Shuai (BSKS) Tablet on HO-1 mRNA and its associated oxidative stress levels among atherosclerotic rabbits. A total of 56 rabbits were randomly divided into the normal group (8 rabbits) and the experimental group (48 rabbits). Normal diet was given to the normal group. Atherosclerotic rabbits models were established in the experimental group. At the eighth week, rabblits in the experi-mental group were randomly divided into the model group, BSKS Tablet group and simvastatin group. Blood samples were collected before medication, 8-, 12-, 16-week after medication from rabbits of each group. Rabbits were sacri-ficed under aseptic conditions at the last blood collection. Expressions of aortic HO-1 mRNA and PPARα mRNA were measured by Q-PCR method. The level of MMP-9 was measured by immunohistochemical assay. Serum HbCO, COX-2 activity and cGMP level were measured by ELISA assay. The results showed that after the intervention of BSKS Tablet, serum HbCO level decreasd, cGMP was obviously increased. However, there was no obvious change on the COX-2 activity. The immunohistochemical assay showed that BSKS Tablet obviously reduced MMP-9 level of rabbits. There was only small amount of aortic HO-1 mRNA expression in the normal group. However, the expres-sion of aortic HO-1 mRNA in the atherosclerosis group was increased. After intervention of BSKS Tablet, the ex-pression of HO-1 mRNA was increased with statistical significance (P < 0.05). Simvastatin had similar antioxidant effect. It was concluded that the compound preparation of traditional Chinese medicine (TCM) BSKS Tablets had an important antioxidant effect in treatment of atherosclerosis. Its protective mechanism may be through the regulation of HO-1 mRNA gene expression and effects of HO-1/CO-cGMP pathway activities of related enzymes while peroxida-tion stability of atherosclerotic plaque.

Objective: To evaluate the rationality and practicability of the provincial directory of antibacterial drugs classification management in China. Methods: The directories at the provincial level published after May 2012 were collected from the government websites of health department in different provinces, cities and autonomous regions. The name, number, grading and classification were counted and anzlyzed. According to the relative national standards, the rationality, practicability and existing problems were ana-lyzed, and some revision sugestions were proposed. Results:Up till now, 30 dirctories were issued with the average recording number-varieties of 147. 1(101-194). The average number of unrestricted, restricted and special use level was 56. 4(37-66), 53. 7(34-68) and 37(24-75), respectively. The top 5 was cephalosporins(29. 5), penicillins(24. 8), antifungi drugs(14. 7), quinolones(12. 8) and aminoglycosides(10). Overall, the directories were rational and practicable. However, 29 kinds of antibacterial drugs were with-out registered number of approval, which was not reasonable or practicable. The other problems were inappropriate classification and grading, name writing mistakes, ect. Conclusion:The directories should be revised in time to correct the existing promblems and im-prove the directories.

RESUMEN

Dilated cardiomyopathy is a disease which manifests with heart enlargement, myocardial contractile dys-function and arrhythmia. The disease cause is unknown. And the disease prognosis is not good with high fatality rate. There is no specific treatment for dilated cardiomyopathy in modern medicine. Currently, a large amount of clinical researches have been conducted by traditional Chinese medicine (TCM) scholars with relatively satisfied therapeutic effects. This article made a review on the etiology, pathogenesis, treatment method (including syndrome differentiation and treatment, modern application of ancient prescription, self-made prescription, and Chinese patent medicine liq-uid) and TCM treatment progress of dilated cardiomyopathy in order to show that TCM has certain advantages in the treatment of dilated cardiomyopathy.

RESUMEN

Objective To explore the difference of sleep quality and the influencing factors in ketamine dependent subjects and methamphetamine dependent subjects.Methods 60 ketamine dependent subjects and 60 methamphetamine dependent subjects with Pittsburgh sleep quality index (PSQI),self-rating depression scale (SDS),self-rating anxiety scale (SAS) were tested.Results Methamphetamine dependent subjects was significantly more likely to elicit poor sleep quality than ketamine dependent subjects (P =0.022).The sleep quality of ketamine dependent subjects had a positive correlation with anxiety(P =0.015),depression(P =0.038),the onset age (P =0.029),and the dose of ketamine use in the last three months (P =0.048),while the sleep quality of methamphetamine dependent subjects had a positive correlation with the total time of ketamine use (P =0.038),anxiety (P =0.041),the dose of ketamine use in the last three months (P =0.011).Conclusion Methamphetamine dependent subjects are prone to a more serious poor sleep quality than ketamine dependent subjects.

RESUMEN

Objective To investigate the prevalence and risk factors of chronic kidney disease (CKD) in Chengdu urban population and the prevalence of CKD in risk population.Methods Questionnaire (anamnesis,smoking,drink) of risk factors of CKD and somatoscopy (blood pressure,body height and body weight) were caried out in railman of Chengdu urban.Their blood and urine indicators (blood sugar,blood lipid,blood uric acid,blood creatinine,uromicroprotein/creatinine ratio,routine urine examination,etc) were measured.The prevalence and risk factors of CKD in Chengdu urban population and the prevalence of CKD in risk population were elucidated.Results Eligible data of 5326 subjects were enrolled in the study.After the adjustment of age and gender component,the prevalence of albuminuria was 11.54%,reduced eGFR was 5.54%,hematuria was 3.87%,and CKD was 18.32%; the recognition was 1.93%.In addition,the prevalence of albuminuria was respectively 23.79%,28.00%,14.08%; prevalence of reduced eGFR was respectively 4.76%,4.53%,3.26%; prevalence of hematuria was respectively 2.94%,3.20%,2.37% in 3098 people with hypertension,diabetes or hyperlipaemia.Independent risk factors of albuminuria were female,hypertension,diabetes,hyperlipemia and high BMI.Independent risk factors of reduced eGFR were female,age,hyperuricemia and hypertension.Drink was negatively correlated with reduced eGFR.Independent risk factors of hematuria were female and age.Conclusions The prevalence of CKD is quite high and the recognition rate is low in the Chengdu urban populaton.Risk factors of CKD are age,female,diabetes,hypertension,hyperlipemia,hyperuricemia and high BMI.Control of the development of metabolic disease can reduce the CKD.

RESUMEN

Cardiovascular disease in peri-menopausal period is a kind of climacteric syndrome which based on the symptoms of the cardiovascular system as the main manifestations. It is manifested as hypertension,pseudo-angina,arrhythmia. Hormone replacement therapy or symptomatic treatment is the common used method of modern medicine,although a certain degree of efficacy,but there are also significant disadvantages. In the treatment of cardiovascular disease in peri-menopausal period,TCM has certain advantages. In this paper,we treat cardiovascular disease in peri-menopausal period from the theory of 'yin deficiency,blood stasis,heat-toxin'and propose the method of 'nourishing yin,promoting blood circulation and detoxification'.
