RESUMEN

Echocardiogram is vital for the diagnosis of cardiac disease. The heart has complex hemodynamics requiring an advanced ultrasound imaging mode. Cardiac ultrasound vector flow imaging is capable of measuring the actual magnitude and direction of the blood flow velocity, obtaining the quantitative parameters of hemodynamics, and then providing more information for clinical research and diagnosis. This study mainly reviewed several different vector flow imaging techniques for cardiac flow and presented the implementation difficulties, and proposed a diverging wave based high frame rate cardiac ultrasound vector flow imaging. The study discussed the limitation of current ultrasound technology used in the cardiac flow measurement, analyzed and demonstrated the specific reasons for these implementation difficulties and the potential future development.

RESUMEN

<p><b>OBJECTIVE</b>To investigate the inhibitory effect of classic demethylating drug 5-aza-2'-deoxycytidine (5-Aza-CdR) on the growth of human lung adenocarcinoma cells in nude mouse xenograft models, and to observe its effect on methylation status and expression of TFPI-2 gene in the nude mouse xenograft tissues.</p><p><b>METHODS</b>The nude mouse xenograft model was established by subcutaneous inoculation of human lung adenocarcinoma A549 cells. According to different doses of 5-Aza-CdR, the tumor-bearing nude mice were randomly divided into experimental groups (0.5 mg/kg group, 1 mg/kg group, 2 mg/kg group) and control group (0 mg/kg group). The tumor growth in the nude mice was observed. The methylation status and the expression of TFPI-2 gene mRNA and protein were detected by methylation specific polymerase chain reaction, real-time fluorescent quantitative polymerase chain reaction and Western blot assay.</p><p><b>RESULTS</b>The nude mice were euthanized at 28 days after intraperitoneal injection of 5-Aza-CdR. The body weight of tumor-bearing nude mice was (27.12 ± 0.38) g in the 0 mg/kg group, (26.80 ± 0.18) g in the 0.5 mg/kg group, (26.67 ± 0.28) g in the 1 mg/kg group, and (26.50 ± 0.26) g in the 2 mg/kg group, showing no significant difference among them (P > 0.05). The volume of xenograft tumors in the 0 mg/kg group was (709.22 ± 2.87)mm³, (400.67 ± 2.68)mm³ in the 0.5 mg/kg group, (285.71 ± 2.91)mm³ in the 1 mg/kg group, and (230.44 ± 3.15)mm³ in the 2 mg/kg group, showing a significant difference (P < 0.05). There were complete methylation of TFPI-2 gene in the 0 mg/kg group, incomplete methylation in the 0.5 and 1 mg/kg groups, and unmethylation in the 2 mg/kg group. The relative mRNA level in the 0, 0.5, 1, 2 mg/kg groups were 1.00 ± 0.00, 1.67 ± 0.07, 3.40 ± 0.24, and 5.55 ± 0.61, respectively (P < 0.05). The relative expression level of TFPI-2 protein in the 0, 0.5, 1, 2 mg/kg groups was 0.18 ± 0.02, 0.36 ± 0.01, 0.64 ± 0.02, and 0.81 ± 0.20, respectively (P < 0.05).</p><p><b>CONCLUSIONS</b>5-Aza-CdR suppresses the tumor growth of human lung adenocarcinoma cells in nude mouse xenograft models, and induces expression of TFPI-2 gene in the xenograft tumor cells. The mechanism might be that 5-Aza-CdR induces re-expression of demethylated TFPI-2 gene by demethylation, and thus inhibits the growth and proliferation of human lung adenocarcinoma cells.</p>

RESUMEN

Objective To evaluated clinical effects and effectiveness of dual-port video-assisted thoracic surgery(VATS)for secondary spontaneous pneumothorax.Methods The clinical data of 48 pa-tients with secondary spontaneous pneumothorax who underwent open thoracotomy(n =18)or dual-port VATS(n =30)were reviewed retrospectively.Mean operation time,mean postoperative drainage period and mean postoperative hospital stay between the two groups were compared.Results All surgeries were suc-cessfully performed without reoperation and severe complications.No patient was converted to thoracotomy in the VATS group.There were significant differences in mean operation time [(67.9 ±7.2)min vs (73.3 ±6.4)min],mean postoperative drainage period [(3.2 ±0.9)d vs(5.0 ±1.3)d],and mean postoperative hospital stay [(7.2 ±1.4)d vs(8.7 ±1.5)d]between the dual-port VATS and open thora-cotomy(P <0.05 ).Conclusion Dual-port video-assisted thoracic surgery can reduce postoperative drainage period and postoperative hospital stay and increase satisfaction.It has little influence on upper limb movement and improves postoperative recovery.

RESUMEN

Objective To discuss the curative effect between percutaneous vertebroplasty ( PVP) and percu-taneous kyphoplasty ( PKP) to treat osteoporotic vertebral compression fractures .Methods 70 cases with osteoporotic vertebral compression fractures were selected and divided into the observation group and control group at random ,who were treated with PKP and PVP respectively .Postoperative vertebral compression rate ,Cobb's angle recovery,relief of pain and occurrence rate of untoward effect of patients in the two groups were observed and compared .Results The vertebral compression rate and Cobb's angle recovery of patients in the two groups one month after the operation were improved than before(t=2.39,2.21,3.52,2.89,all P0.05).The postoperative complication occur-rence rate of patients in the observation group was much lower than that in control group (χ2 =4.20,P<0.05). Conclusion The application of PVP and PKP can obviously relieve the pain of patients with osteoporotic vertebral compression fractures ,but PKP treatment is more favorable for the recovery of vertebral height and Cobb 's angle with little negative effect and high security .

Objective To study the effects of hypoxia-inducible factor-1α (HIF-1α) on radiationinduced autophagic cell death in breast cancer cells.Methods MCF-7 cells were divided into four groups:control (normoxia,21% Oxygen),irradiation (8 Gy X-rays),hypoxia (Cobalt chloride,CoCl2)and irradiation with hypoxia (CoCl2).150 μmol/L CoCl2 was utilized to induce hypoxic conditions.Western blot was applied to detect the expression of HIF-1α and MAPLC3.MDC and Hoechst staining were used to detect autophagy and apoptosis.Radiosensitivity was detected by cloning formation.The short hairpin interfering RNA (shRNA) retroviral transduction particles targeting HIF-1α was transfected into MCF-7 cells to establish HIF-1 α knockdown cells,then the radiosensibility,autophagy and apoptosis were detected.Results Compared with control group and irradiation group,the protein level of HIF-1 increased obviously in the normaxia,irradiation,hypoxia and irradiation with hypoxia groups,and the values were 0,0,1.00,1.89,respectively.The expression levels of MAPLC3 were markedly up-regulated in irradiation,hypoxia and irradiation with hypoxia groups as compared with control,and the ratios of LC3Ⅱ/LC3Ⅰ were 1.15,1.73,2.38 and 3.60,respectively.The radiosensitivity of MCF-7 cells decreased in the following order:normoxia with 3MA > normoxia > hypoxia with 3MA > hypoxia.HIF-1α knockdown cell (pSUPER-HIF-1α Ri) and vector control were constructed.After treatment with CoCl2,survival fraction of MCF-7-pSUPER was significantly higher than that of control (t =3.080,6.946,6.658,6.380,P <0.05),and radiosensitivity was down-regulated after irradiation,but there was no significant difference between normoxia and hypoxia in survival fraction of MCF-7-pSUPER-HIF-1α Ri.After treatment of irradiation or hypoxia,the autophagic fractions in MCF-7-pSUPER-HIF-1α Ri significantly decreased,reduced by 21.1%,25.5%,15.5%,respectively(t =4.635,4.738,6.354,P <0.05) as compared with MCF-7-pSUPER,but there was no change in apoptosis.Conclusions HIF-1α may increase radiationinduced autophagy and decrease radiosensitivity,but have no influence on apoptosis.

Objective:To investigate the effect of neonatal dog peripheral mononuclear cells on left ventricular function, haemodynamic, and infarcted size when transplanted into infarcted myocardium. Methods: Mononuclear cells from neonatal dogs were isolated and purified. The descending coronary arteries of adult dogs were ligated and the cells were injected into the border zone of the infarcted region. The control group was injected with non serum culture medium. Left ventricular pressure, d p /d t max , and power of LV were measured during and 30 d after operation. Echocardiogram(HP Sonos 5500, 2.5 MHz) were performed promptly and 30 d after operation to determine the changes of left ventricular function. Infarcted size, capillary density and cardiac muscle width were measured 30 d after operation. Results: LV pressure decreased in both groups, there were no marked differences between the 2 groups. LV pressure and d p /d t max of transplant group were greater than those of control group 30 d after operation. The systolic and diastolic functions were preserved after mononuclear cell transplantation. Infarcted sizes were lower and capillary density was greater in transplant group than those of control group. Conclusion: Transplantation of neonatal dog peripheral mononuclear cells into the infarcted regions can increase the capillary density and decrease infarcted size, preserving left ventricular function.
