RESUMEN

Three cancer cell lines including gastric cancer SGC-7901, HGC-27, and MGC-803 cells were employed to evaluate the bioactivity of seven Dendrobium species. Simultaneously, these Dendrobium species were assessed with UPLC-Q-TOF-MS, and 504 common peaks were found. Based on the hypothesis that biological effects varied with differences in components, multivariate relevance analysis for chemical component-activity relationship of Dendrobium, including grey relation(GRA) and partial least squares(PLS) analysis were performed to evaluate the contribution of each identified component. The target peaks were identified by standards toge-ther with databases of Dendrobium, Nature Chemistry, MassBank, etc. Finally, four active components, including 3,5,9-trihydroxy-23-methylergosta-7,22-dien-6-one, diacylglycerol(14∶1/22∶6/0∶0), pipercitine, and 22-tricosenoic acid, might have negative effect on the growth of gastric cancer cells.

RESUMEN

In order to identify the source of Citrus grandis and evaluate its quality originate from two areas comprehensively,DNA barcode was used to identify 26 samples of C. grandis. The content of naringin,rhoifolin,naringenin and apigenin was determined by UPLC method,and the color difference was numerically studied by color difference analyzer,which was related to the effective components of C. grandis. The results showed that samples was the source of C. grandis in both regions. The ITS2 sequence length was about400-500 bp,and the sequence similarity reached 99. 82%. There was only one base deletion in the two groups. There was one base A in some medicinal materials of Guangdong at 330 bp,but no base in Chongqing. The contents of naringin and rhoifolin in Chongqing samples were higher than those in Guangdong samples,and there were statistical differences between naringenin and apigenin. The chroma value showed that L*value of Guangdong was larger,a*value was smaller,L*value of Chongqing was smaller,and a*value was larger,while the b*value of both was not significantly different; The results of correlation analysis showed that naringin,rhoifolin,naringenin were positively correlated with L*,b*value,negatively correlated with a*value,and apigenin had no correlation with L*,a*,b*value. In this study,the scientific identification and evaluation of C. grandis was carried out to provide a new idea for the further study of the rapid identification and evaluation of C. grandis.

RESUMEN

Objective To clone a new NADPH cytochrome P450 reductase gene (GuCPR) (Login number: MH401048) from Glycyrrhiza uralensis and do some bioinformatics analysis. Methods Total RNA was extracted from the roots of G. uralensis and then transcription reversed into cDNA. The GuCPR gene was obtained by screening the transcriptome database. The NCBI ORF finder was used to obtain its open reading frame and translated amino acid sequence, design primers for PCR amplification, construction of recombinant cloned plasmid. Bioinformatics analysis was used to predict the protein properties, structure and model the tertiary structure of the protein, and homologous phylogenetic tree construction was performed. Results cDNA of GuCPR gene was cloned to a total length of 2 118 bp and encoded 705 amino acid residues with a relative molecular mass of 78 450. Electricity (pI) 5.19. GuCPR protein was a non-secretory protein and did not have a signal recognition function. The results of transmembrane prediction showed that the amino acids 44—64 of the protein were transmembrane regions. Meanwhile, the subcellular localization prediction results showed that the protein was located on the endoplasmic reticulum. Conclusion A new GuCPR was cloned and its bioinformatics analysis laid the foundation for further research.

RESUMEN

Endometriosis is a multiple disease that afflicts the health of women at childbearing age,and its incidence rate has been increasing year by year,furthermore,there has been a trend to be younger.At present,the pathogenesis of endometriosis has been not expounded completely,its cure rate is not high with high recurrence rate.In recent years,studies have shown that the human is a commensal body composed of a large number of microorganisms,and especially the microorganisms in the intestinal are closely related to the health of the body.Based on the previous studies on endometriosis,this paper proposes that its pathogenesis may be related to intestinal microbiological disorder,and aims to provide new ideas for the treatment of endometriosis.

RESUMEN

A rapid fluorescence polarization immunoassay (FPIA) has been developed for the determi-nation of aflatoxins in samples of naturally-contaminated herbal teas. The tracers were synthesized by chemical method and determined by thin layer chromatography (TLC) and mass spectroscopy (MS). Fluorescence polarization was evaluated by the detection of polarized light. The results showed that the limit of detection (LOD) of FPIA for aflatoxins was 20 ng·mL-1, the IC50 was 371.80 ng·mL-1, and the linear range of the developed FPIA was 92.76-252.32 ng·mL-1. Compared with conventional HPLC methods, the FPIA developed in this study has the advantages of short analysis time and low cost. This method may be suitable for high- throughput screening of aflatoxins in herbal teas.

RESUMEN

Objective: To observe the anti-fibrotic mechanism of Biejiajian Pill. Methods: SD rats were randomly divided into six groups: control, model, colchicina (0.1 mg/kg, positive), Biejiajian Pill (0.55, 1.1, and 2.2 g/kg) groups. Except for the control group, the rats in the other groups were sc injected with 40% CCl4 olive oil solution twice a week for consecutive six weeks to establish the models of liver fibrosis.. At the same time, drugs were ig administrated at a volume of 10 mL/kg in each group, once daily for consecutive 11 weeks. At the end of the week 11, all rats were sacrificed. The immunohistochemical technique was applied to analyzing the expression of α-smooth muscle actin (α-SMA) in liver tissue, RT-PCR was applied to analyzing the expression of transforming growth factor-β1 (TGF-β1) protein and smad 3 gene in liver tissue. Results: Compared with the control group, the expression levels of α-SMA, TGF-β1, and smad 3 gene increased significantly (P < 0.05) in the model group. Compared with the model group, Biejiajian Pill and colchicina decreased the expression of α-SMA, TGF-β1 and smad 3 gene, the efficacy of high-dose Biejiajian Pill group was more significantly. Conclusion: The antifibrotic mechanism of Biejiajian Pill may be through lowing the TGF-β1/smad 3 signaling pathways and inhibiting hepatic stellate cells activation and proliferation.
