RESUMEN

Objective To investigate the serum Ectodysplasin A(EDA)levels in women with polycystic ovary syndrome(PCOS)and explore the correlation of EDA and insulin resistance(IR).Methods A total of 256 newly diagnosed PCOS patients in Guangzhou First People's Hospital from September 2020 to September 2022 were selected.Taking BMI=25 kg/m2 as the cut-off point,the patients with PCOS were divided into obese subgroup(n=143)and non-obese subgroup(n=113).Among them,43 PCOS received six months of Liraglutide intervention.123 healthy women were normal control group(NC),which were divided into healthy obese subgroup(n=10)and healthy non-obese subgroup(n=113).Serum EDA and related biochemical indexes were detected in both groups.The correlation between serum EDA and metabolic index were analyzed.Results The serum EDA level in PCOS group was higher than that in NC group[(272.59±68.52)vs(214.51±61.36)pg/ml,P<0.05].Serum EDA in obese subgroup was higher than that in no-obese subgroup in PCOS patients[(301.11±55.81)vs(236.84±66.25)pg/ml,P<0.01].After the intervention of Liraglutide,serum EDA in PCOS women markedly decreased[(294.71±57.81)vs(336.0±61.23)pg/ml,P<0.05].EDA levels were correlated positively with body mass index(BMI),waist hip ratio(WHR),fasting blood glucose(FPG),2 hPG,fasting insulin,2 hIns,homeostasis model assessment for insulin resistance(HOMA-IR),TC,LDL-C and testosterone.Multiple stepwise regression analysis showed that BMI and HOMA-IR were independent risk factors of serum EDA.Conclusion Serum EDA is elevated in PCOS patients,especially when combined with obesity,which may be related to IR.Liraglutide intervention can significantly reduce EDA levels in PCOS patients.

RESUMEN

The pulse amplitude of fingertip volume could be improved by selecting the vascular dense area and applying appropriate pressure above it. In view of this phenomenon, this paper used Comsol Multiphysics 5.6 (Comsol, Sweden), the finite element analysis software of multi-physical field coupling simulation, to establish the vascular tissue model of a single small artery in fingertips for simulation. Three dimensional Navier-Stokes equations were solved by finite element method, the velocity field and pressure distribution of blood were calculated, and the deformation of blood vessels and surrounding tissues was analyzed. Based on Lambert Beer's Law, the influence of the longitudinal compression displacement of the lateral light surface region and the tissue model on the light intensity signal is investigated. The results show that the light intensity signal amplitude could be increased and its peak value could be reduced by selecting the area with dense blood vessels. Applying deep pressure to the tissue increased the amplitude and peak of the signal. It is expected that the simulation results combined with the previous experimental experience could provide a feasible scheme for improving the quality of finger volume pulse signal.

OBJECTIVE: Taking polysuccinimide as the main chain, amine side chain and alkyl side chain were grafted to prepare the drug/gene co-delivery vector. The property of the polymers with various side links were investigated to select an optimal vector. METHODS: Poly-D, L-polysuccinimide was synthesized by polymerization reaction of D, L-aspartic acid as monomer. Therefore, N, N-dimethylenedipropyl-triamine and 3, 3'-diaminodipropylamine were grafted with dodecylamine/adecylamine/octadecylamine at different proportions by ring-opening reaction to obtain amphiphilic PEECs. The structure of the material was confirmed by 1H NMR; the particle size and surface potential of the micelles were measured by dynamic light scattering; the critical micelle concentration (CMC) was determined by pyrene fluorescent probe; the RNA blocking ability was characterized by agarose gel electrophoresis; the release behavior of the PEECs was examined and the cytotoxicity, cellular uptake and gene silencing efficiency of the PEECs were studied at the cellular level. RESULTS A series of PEECs with different grafting rates was successfully synthesized. The particle sizes and surface potential of the PEEC derived micelles were between 250 nm and 350 nm and 27 mV and 45 mV, respectively, with a small CMC value. The RNA binding ratio of PEECs was at a mass ratio of about 0.8:1. MTT assay demonstrated that PEEC micelles had certain cytotoxicity. PEECs had excellent micelle formation, drug-loading and gene binding abilities, particularly, PEEC16-2 showed high gene silencing efficiency at the cellular level. CONCLUSIONS PEECs are able to co-delivery drug and gene, and PEEC16-2 micelles have the best ability of drug encapsulation and gene delivery.
