RESUMEN

This paper aims to investigate the regulatory mechanism of blood-activating and stasis-dissipating drugs on fecal metabolic characteristics of rhubarb-peach kernel in mice with adenomyosis (AM) using fecal metabolome method. Adenomyosis was modeled by pituitary transplantation, and after the end of modeling administration, fecal samples were collected from mice. Non-targeted metabolomics studies were performed using liquid chromatography-mass spectrometry (LC-MS) to compare the metabolic characteristics of the feces of mice in each group and to find intestinal differential metabolites and potential differential metabolic pathways. The results showed that compared with the normal group, 5-hydroxy-L-tryptophan, histidine, L-acetylcarnitine, 16-hydroxy hexadecanoic acid, thromboxane B2, etc. were significantly up-regulated, L-urobilin and prostaglandin D3 were down-regulated in the feces of the model group, and were reversed after treatment with the rhubarb-peach kernel. The results of metabolic pathway enrichment analysis showed that tryptophan metabolism and histidine metabolism were the main intervention pathways of the rhubarb-peach kernel on AM intestinal metabolism. This study found that the underlying mechanism of the rhubarb-peach kernel in the treatment of AM is related to the intervention of intestinal metabolism of tryptophan, histidine, bile acid, choline and arachidonic acid, and the regulation of pro-inflammatory microenvironment and fatty acid metabolic homeostasis. @#Abstract: Objective In order to understand the radon exposure level of homes in Chongqing, this survey was carried out on the indoor radon concentration in 38 districts and counties of Chongqing. Methods According to the population ratio of every 100 000 people, one monitoring site was arranged, and the number of parallel samples was 10% of the distribution sites. The monitoring sites covered 38 districts and counties in Chongqing. A total of 1 019 residential monitoring sites in 38 districts and counties in Chongqing were measured with radon accumulation detectors from July 2020 to June 2021. Results The five districts/counties in Chongqing having the highest average concentration of residential radon in the year were Xiushan County 78.8 Bq/m3, Qianjiang District 78.0 Bq/m3, Yubei District 73.9 Bq/m3, Youyang County 71.4 Bq/m3 and Shapingba District 69.8 Bq/m3. The five districts/counties with the lowest mean concentration of indoor radon were 37.6 Bq/m3 in Zhongxian County, 36.4 Bq/m3 in Changshou District, 33.7 Bq/m3 in Kaizhou District, 33.2 Bq/m3 in Liangping District and 27.3 Bq/m3 in Wushan County. The concentration levels of radon in four seasons were 46.0 Bq/m3, 53.4 Bq/m3, 45.1 Bq/m3 and 59.5 Bq/m3, respectively. The concentration of radon was higher in Summer and Winter, and lower in Spring and Autumn, and the difference of concentration among four seasons was statistically significant (P<0.001). The radon concentration of newly built buildings after 2017 was relatively high, up to 61.8 Bq /m3, but there was no statistical significance in radon concentration in different building ages (P>0.05). The concentration of radon in rooms of buildings with less than 10 floors was higher, up to 63.2 Bq /m3, and there were significant differences in radon concentration among rooms of different floors (P<0.05). The average annual radon concentration in houses in Chongqing was about (51.6±19.5) Bq/m3, and the average annual effective dose of inhaling radon and its progeny by house-related people was about (1.38±0.52) mSv. Conclusion The average annual radon concentration level of houses in Chongqing is within the standard limit value recommended by China, but the prevention and control of radon should be strengthened.

Objective:To study the protective effect of essential oil from Alpiniae Zerumbet Fructus (EOAZF) against high glucose (HG)-induced injury of human umbilical vein endothelial cells (HUVECs) <italic>in vitro</italic>, so as to provide experimental evidence for the treatment of diabetes-induced cardiovascular diseases with EOAZF. Method:The cells were divided into the normal group, model group (25 mmol·L<sup>-1</sup> glucose), positive control group (100 mg·L<sup>-1</sup> vitamin C), and the low- (0.25 μg·L<sup>-1</sup>), medium- (1 μg·L<sup>-1</sup>), and high-dose (4 μg·L<sup>-1</sup>) EOAZF groups. The HUVECs were damaged by HG. The secretion amounts of malondialdehyde (MDA), nitric oxide (NO), and endothelin-1 (ET-1) in HUVECs of different groups were measured to assess the protective effect of EOAZF against HG-induced injury. The effects of EOAZF on the apoptosis and reactive oxygen species (ROS) generation of HUVECs damaged by HG were detected by Annexin V-fluorescein isothiocyanate/propidium iodide (Annexin V-FITC/PI) staining and dichloro-dihydro-fluorescein diacetate (DCFH-DA) assay. The protein and mRNA expression levels of thioredoxin interacting protein (TXNIP) and thioredoxin 1 (Trx-1) were determined by Western blot and Real-time polymerase chain reaction (Real-time PCR), followed by the measurement of total intracellular Trx-1 activity with insulin disulfide reduction method. Result:The comparison with the control group revealed that the proliferation of HUVECs in the model group was significantly inhibited and their shape was damaged. Compared with the model group, EOAZF protected HUVECs against HG-induced injury in a concentration-dependent manner. The secretion amounts of MDA and ET-1 (<italic>P</italic><0.05) in the model group were increased in contrast to those in the control group, while the NO level was decreased (<italic>P</italic><0.01). Compared with the model group, EOAZF at all the three concentrations, especially at 4 μg·L<sup>-1</sup>, obviously reduced the secretion of MDA and ET-1 (<italic>P</italic><0.05), but elevated NO after HG induction (<italic>P</italic><0.05). The cell apoptosis assay and ROS detection results demonstrated that the apoptosis and ROS level in the model group were higher than those in the control group (<italic>P</italic><0.01). Compared with the model group, EOAZF at 4 μg·L<sup>-1 </sup>significantly lowered the ROS level and apoptosis (<italic>P</italic><0.05) of HUVECs damaged by HG. The Western blot assay and Trx-1 activity detection uncovered that the protein and mRNA expression levels of TXNIP in the model group were significantly up-regulated as compared with those in the control group (<italic>P</italic><0.05), whereas the Trx-1 activity was decreased (<italic>P</italic><0.01). Compared with the model group, EOAZF at 4 μg·L<sup>-1 </sup>significantly down-regulated the mRNA and protein (<italic>P</italic><0.05) expression levels of TXNIP and enhanced the total Trx-1 activity (<italic>P</italic><0.05) in HUVECs, thus suppressing the oxidative stress. Conclusion:EOAZF exerts the protective effects against HG-induced injury in HUVECs by improving the endothelial function and reducing intracellular ROS and apoptosis. Its efficacy in anti-oxidative stress may be related to the down-regulation of mRNA and protein expression levels of TXNIP and the enhancement of Trx-1 activity.

Objective:To observe theclinical efficacy of modified Banxia Baizhu Tianmatang combined with acupuncture on migraine with wind phlegm disturbance syndrome, and the regulatory effect on neurovasoactive peptide and vascular endothelial activator. Method:Two hundred and fifty patients were randomly divided into control group (75 cases) and observation group (75 cases). Patients in Two group got acupuncture for 6 times, 1 time/day, after a day of rest, they got placebogranules of Banxia Baizhu Tianmatang, 10 g/time, 2 times/day. Patients in observation group got Banxia Baizhu Tianmatang, 1 dose/day, and also the same acupuncture with the therapyof control group. And the treatment lasted for 4 weeks. At the half, 1<sup>st</sup>, 2<sup>nd</sup>, 6<sup>th</sup>, 12<sup>th</sup>, 24<sup>th</sup>, and 48<sup>th</sup> hour after treatment, VAS were scored, rate of pain relief within 6, 12, and 24 hours, disappearance rate and recurrence rate of pain within 72 hours, migraine attack times, headache duration and headache degree before 4 weeks of treatment, during the treatment and after the treatment were recorded. Before and after treatment, accompanying symptoms, wind phlegm disturbance syndrome, headache impact test version-6 (HIT-6) and the migraine disability assessment questionnaire (MIDAS) were scored. And levels of calcitonin gene-related peptide (CGRP), nitric oxide (NO), endothelin-1 (ET-1), pituitary adenylate cyclase activating peptide (PACAP), S100B protein, substance P(SP), von Willebrand factor (vWF) and fibrinogen (FIB) were detected. And safety was evaluated. Result:VAS in two groups decreased at different time points (<italic>P<</italic>0.01), and VAS in observation group at 6<sup>th</sup>, 12<sup>th</sup>, 24<sup>th </sup>and 48<sup>th</sup> hour after treatment were lower than those in control group (<italic>P<</italic>0.01). The rate of pain relief in observation group at 6<sup>th</sup> and 12<sup>th</sup> hours after treatment and the disappearance rate of pain at 72<sup>th</sup> hour were 67.14%(47/70), 87.14% (61/70) and 92.86% (65/70), which were higher than 50.00% (34/68), 70.59% (48/68) and 79.41% (54/68) in control group. The recurrence rate of pain in observation group was 21.43% (15/70), which was lower than 39.71% (27/68) in control group (<italic>P<</italic>0.05). During the treatment and drug withdrawal, times of migraine attack, headache duration and headache degree were all less than those in control group (<italic>P<</italic>0.01). Scores of accompanying symptoms, wind phlegm disturbance syndrome, HIT-6 and MIDAS were all lower than those in control group (<italic>P<</italic>0.01). The clinical effect was better than that in control group (<italic>Z</italic>=2.106, <italic>P<</italic>0.05). Levels of CGRP, PACAP, S100B protein, SP, ET-1, vWF and FIB were lower than those in control group, while level of NO was higher than control group (<italic>P<</italic>0.01). Conclusion:Modified Banxia Baizhu Tianmatang combined with acupuncture had a better instant analgesic effect, with a significant effect on continuing analgesia and reducing headache recurrence. It can also alleviate migraine symptoms and accompanying symptoms, andreduce the impact of migraine on daily life and the degree of disability. Its mechanism may be related to the regulation of neurovasoactive peptides and vascular endothelial substances. It is worth for further study.

Objective::To clarify the inhibitory effect of essential oil from Alpinia zerumbet rhizome (EOFAZ) on oxidized low-density lipoprotein (ox-LDL)-induced transformation of macrophage into foam cell and explore its possible mechanism. Method::THP-1 monocyte was incubated with 100 μg·L-1 phorbol myristate acetate (PMA) to grow into macrophage, experiment was divided into 4 groups as follows, control group, model group (80 mg·L-1 ox-LDL), EOFAZ at low dose (80 mg·L-1 ox-LDL+ 4 μg·L-1 EOFAZ)and EOFAZ at high dose (80 g·L-1 ox-LDL+ 20 μg·L-1 EOFAZ). Mathye thiazolye telrazliurn (MTT) method was employed to examine the influence of EOFAZ on macrophage viability. Western blot was used to analyze the expression level of cluster of differentiation 36(CD36) and ATP-binding cassette transporter A1(ABCA1) protein in macrophage. Enzyme-linked immunosorbent assay (ELISA) was used to detect cholesteryl ester contents in macrophage. Oil red O staining was applied to determine the accumulation of lipids in macrophage. Result::EOFAZ showed non-toxic effect on macrophage. Compared to control group, macrophage in model group displayed higher level of cholesteryl ester and lipid droplet(P<0.01), as well as significant increasing of CD36 expression (P<0.01), but no effect on ABCA1 expression. EOFAZ notably reduced the contents of lipids and cholesteryl ester(P<0.01), down-regulated expression of CD36 and up-regulated expression of ABCA1 in macrophage in comparison with the model group(P<0.01), indicating that EOFAZ inhibited transformation of macrophage into foam cell. Conclusion::EOFAZ could inhibit ox-LDL-induced transformation of macrophage into foam cell, the underlying mechanism may involves its ability to increase CD36 expression and decrease ABCA1 expression in macrophage.

Objective · To investigate the relationship between the mammographic signs and the pathological features in breast cancer. Methods · From August 2015 to August 2018, 300 patients with primary breast cancer confirmed by operation and pathology in Shanghai First Maternity and Infant Hospital, Tongji University were selected. All patients had mammography before operation. The relation between characteristics of molybdenum target imaging and pathological types were retrospectively analyzed. The correlation between specific imaging signs such as lump, calcification, spiculation sign and important pathological features were further analyzed. Results · Among 300 cases of breast cancer, 251 cases (83.7%) had lumps, of which 183 cases had spiculation sign, and 190 cases had calcification. Besides, 235 cases (78.3%) had calcification; 77 cases (25.7%) had structural disorder with diffuse density; 98 cases (32.7%) had positive signs of vascular symptoms. Lump (95.1%), spiculation sign (82.3%) and calcification (83.7%) were mostly observed in invasive ductal carcinoma. Structural disorder (88.2%) was mostly observed in invasive lobular carcinoma. Calcification (96.1%) and lump (60.8%) were mostly observed in intraductal carcinoma. Calcification (100.0%) was mostly observed in lobular carcinoma in situ. Lump was mostly observed in simple carcinoma, medullary carcinoma and mucinous adenocarcinoma. The positive expression rates of estrogen receptor (ER) and progesterone receptor (PR) were higher in patients with tumor lump>5 cm than those with tumor lump ≤5 cm, but there were no significant differences (P=0.165, P=0.317). The positive rate of ER was 54.5% and the positive rate of PR was 60.8% in patients with calcification, which were lower than those without calcification (P=0.027, P=0.006). The positive rate of PR in patients with spiculation sign was 71.6%, which was higher than those without spiculation sign (P=0.018). But there was no significant difference in the expression of ER in patients with or without spiculation sign (P=0.321). Patients with lumps alone had a lower incidence of lymph node metastasis (29.5% vs 37.9%, P=0.009), and lower histological grade (P=0.043), in comparison with patients with lumps and calcification. But they had higher ER positive rate (70.5% vs 57.9%, P=0.033) and PR positive rate (73.8% vs 65.3%, P=0.014).Conclusion · Breast cancer patients with different pathological types show different signs of mammography which may have certain associations with the expression of immunohistochemical indicators such as ER and PR. Simple lump shadows in mammography may be a predictor in the good prognosis of breast cancer.

BACKGROUND@#Although the use of extra-corporeal membrane oxygenation (ECMO) has been rapidly increasing, the benefit of ECMO in patients with acute respiratory distress syndrome (ARDS) remains unclear. Our objective was to investigate the effect of venovenous ECMO (VV-ECMO) on adult patients with severe ARDS.@*METHODS@#We conducted a multi-center, retrospective, cohort study in the intensive care units (ICUs) of six teaching hospitals between January 2013 and December 2018. Patients with severe ARDS who received VV-ECMO support were included. The detailed demographic data and physiologic data were used to match ARDS patients without ECMO. The primary endpoint was the 28-day mortality.@*RESULTS@#Ninety-nine patients with severe ARDS supported by VV-ECMO and 72 patients without ECMO were included in this study. The acute physiology and chronic health evaluation II score was 23.1 ± 6.3 in the ECMO group and 24.8 ± 8.5 in the control group (P = 0.1195). The sequential organ failure assessment score was 12.8 ± 3.4 in the ECMO group and 13.7 ± 3.5 in the control group (P = 0.0848). The 28-day mortality of patients with ECMO support was 39.4%, and that of the control group was 55.6%. The survival analysis curve showed that the 28-day mortality in the ECMO group was significantly lower than that in the control group (P = 0.0097). Multivariate Cox regression analysis showed that the independent predictors of the 28-day mortality were the requirement of vasopressors before ECMO (hazard ratio [HR]: 1.006; 95% confidence interval [CI]: 1.001-1.013; P = 0.030) and duration of mechanical ventilation before ECMO (HR: 3.299; 95% CI: 1.264-8.609; P = 0.034).@*CONCLUSIONS@#This study showed that ECMO improved the survival of patients with severe ARDS. The duration of mechanical ventilation and the requirement of vasopressors before ECMO might be associated with an increased risk of death.

Objective To explore the factors related to the consumption of sugar-sweetened beverages (SSB) among middle school students in four provinces (municipality) in China.Methods A total of 3 469 middle school students were recruited from the Children health cohort in eight counties within the four provinces (municipality).We collected information regarding demography,lifestyle,and family and school situation as well as the consumption frequency of SSB in the previous week.High SSB consumption was defined as "positive" if the frequency was once a day or more.Mean consumption time was calculated,and then logistic regressions were performed to explore the factors influencing SSB consumption.Results Prevalence of high SSB consumption was 11.18% among the subjects of the eight counties,and the consumption frequency of SSB was 0.21 times per day.Multivariate logistic regression analysis revealed that the following factors were associated with high consumption of SSB:higher grade,rural region,male gender,physical exercise,habit of eating snacks,eating out,and fast-food consumption (P<0.01).Factors influencing high SSB consumption were not completely equal between genders:physical exercise at school was associated with high SSB consumption only in female students (P<0.01),while higher grade and eating out in the past week were associated with high SSB consumption only in male students (P<0.05).Conclusion Factors associated with high SSB consumption among middle school students included grade,region,physical exercise at school,snack consumption,eating out,and fast-food consumption.These factors differed between genders.Intervention programs addressing these factors may be beneficial in reducing SSB consumption in middle school students.

AIM:To investigate the effect of cholesterol metabolite 27-hydroxycholesterol (27-OHC) on the proliferation of lung cancer cells.METHODS:Human lung cancer A549 cells were treated with 27-OHC at different concentrations (0, 0.3125, 0.625, 1.25, 2.5, 5 and 10μmol/L) for 24~48 h.The cell viability, cell cycle, cell proliferation, the intracellular cholesterol levels and cholesterol metabolism-related molecule expression were subsequently assessed by CCK-8 assay, flow cytometry, Ed U staining, tissue total cholesterol detection kit, real-time PCR and Western blot.RESULTS:27-OHC decreased the viability of the A549 cells in a dose-and time-dependent manner (P<0.01) and inhibited the cell proliferation (P<0.05).The expression of typical liver X receptor (LXR) downstream target proteins including ATP-binding cassette transporter A1 (ABCA1) , low-density lipoprotein receptor (LDLR) , and 3-hydroxy-3-methylglutaryl-coenzyme A reductase (HMG-CR) were modulated, which promoted the efflux of intracellular cholesterol, and reduced cholesterol influx and de novo synthesis, resulting in decreased intracellular cholesterol levels and cell viability.Furthermore, the inhibitory effect of 27-OHC on A549 cell viability was significantly attenuated after the LXR pathway was partially blocked by 5μmol/L GSK2033 treatment (P<0.05).CONCLUSION:27-OHC inhibits A549 cell proliferation via activation of LXR signaling pathway.

Colon cancer associated transcript 1 (CCAT1) is a kind of long non-coding RNA (IncRNA) found re-cently, which is aberrant overexpression in colorectal cancer, and it can promote cancer cell proliferation, invasion and metastasis. CCAT1 involves in the regulation of multiple pathophysiological processes, especially the formation and progression of gastrointestinal cancer, through the interaction with microRNA (miRNA) or protein by regulation mechanism like endogenous competition, and it is expected to serve as a valuable biomarker and therapeutic target.

AIM:To investigate the angiogenic effect and mechanisms of astragaloside IV(AS-IV)in rats with myocardial infarction via protein kinase D 1(PKD1)-histone deacetylase 5(HDAC5)-vascular endothelial growth factor (VEGF)signaling pathway.METHODS:The classic model of myocardial infarction by ligation of the left anterior de-scending coronary artery was replicated,and the rats were randomly divided into model group,AS-IV group,and AS-IV+CID755673(PKD1 inhibitor)group.The sham operation control group and DMSO control group were also set up.All the rats were given intravenous injection via caudal vein.The rats were sacrificed 4 weeks later,and segmental heart samples were used for HE staining and Masson staining.The expression of PKD1,HDAC5 and VEGF was analyzed by immunohis-tochemistry,RT-PCR and and Western blot.RESULTS:Compared with sham operation group and DMSO group,the myo-cardium in model group showed disordered arrangement, accompanied with necrotic myocardial cells and obvious fibrosis tissue.After treatment with AS-IV,the morphological changes of myocardium were obviously improved,and the number of new blood vessels increased significantly.However,after treatment with AS-IV+CID755673,the myocardial tissues of the rats became disordered again,with increased necrotic cells and some closed vessels.The mRNA and protein expression of PKD1,HDAC5 and VEGF in myocardial tissue in model group was significantly lower than that in sham operation and DMSO groups(P<0.05).The expression in AS-IV group was significantly higher than that in model group(P<0.01), while that in AS-IV+CID755673 group was significantly lower than that in AS-IV group(P<0.05).CONCLUSION:AS-IV promotes the angiogenesis of myocardial tissues in the rats after myocardial infarction partly by regulating the PKD 1-HDAC5-VEGF signaling pathway.

RESUMEN

[Objective]To investigate the feasibility of making the protocol with computed tomography data for total knee arthroplasty using patient-specific femoral and tibial cutting blocks.[Methods]54 consecutive patients diagnosed with osteoarthritis were divided randomly into two groups. Every patients accepted 3-dimentional computed tomography scan for bilateral legs before operation,the protocol were made for every patients before operation.When making the pre-operative protocol,we accepted the prothesis surpass the osteotomy edge finitely,tibia prothesis rotational orientation according to the outline of the tibial osteotomy. The thickness of the posterior condyle of femur resected was as the same with the prothesis posterior condyle as possible. The patients in trial group undergone total knee arthroplasty(TKA) using patient-specific instruments(PSI)according to the preoperative protocol,while the patients in the control group undergone TKA using custom instruments(CI).The operation time,the times that the scale of the prosthesis were altered intraoperation,the mean thickness of the posterior condyle of femur resected in the operation,AKS score 3 months and 1 year after the operation were recorded and compared between the two groups. Plain film radiography after the operation were analyzed.[Results]No significant differences were found between two groups with respect to the operation time. 8 patients altered to a lesser prosthesis in the CI group during the operation,all in bibia contrast to no one need alteration in the PSI group.The thickness of the osteotomy on the posterior condyle of femur in the PSI group is bigger than that of CI group as a result.The AKS score of the patients in the PSI group is higher than that of CI group 3 months after the opera-tion,whereas no significant difference was found 1 year after the operation.[Conclusion]The reconstructed image by thin layer CT can be used for preoperation protocol for TKA,which might be helpful in deciding the size of femour prosthesis;the implants can surpass the edge of the osteotomy finitely in TKA,which is not necessarily cause any complain.We can orient the tibia prosthesis rotation according to the outline of the osteotomy of the patients'tibia.

RESUMEN

Objective To evaluate the anti-oxidative function of siwu decoction.Methods Using aged mice model and setting three dose groups(0.58 g/kg BW, 1.17 g/kg BW, 3.50 g/kg BW), negative control group, after feeding four groups for 30 days with the lyophilized powder of the siwu decoction product, removing accessories, the content of lipid oxidation product malondialdehyde(MDA), glutathione(GSH)and protein carbonyl, the activity of superoxide dismutase(SOD)were tested. Results In the control group, the activity of SOD was(33.0±3.4)U/mgprot, and the content of GSH and protein carbonyl were(5.56±0.73)mg/gprot,(2.30±0.22)nmol/mgprot, respectively. Compared with the control group, the activity of SOD in the high dose group(39.2±6.3 U/mgprot)and the content of GSH in the medium and high dose group[(7.04±0.86) mg/gprot,(6.81±1.02)mg/gprot, respectively]was significantly higher(P<0.05); the content of protein carbonyl notably in the high dose group[(1.78±0.32)nmol/mgprot]significantly decreased(P<0.05). No obvious difference in the content of MDA were observed between each dose group and the control group (P>0.05). Conclusion Siwu decoction has obvious antioxidant effect on aged mice.

<p><b>OBJECTIVE</b>To explore clinical characteristics of 4 patients with knee discoid medial meniscus under arthroscopic.</p><p><b>METHODS</b>From April 2012 to May 2015, 4 patients(4 knees) with knee discoid medial meniscus were treated with arthroscopic, including 1 male and 3 females, aged from 7 to 56 years old with an average of 26 years old. All patients were performed discoid meniscus plasty under arthroscopic. Postoperative symptoms and signs were observed, and clinical effects were evaluated by Lysholm knee scores.</p><p><b>RESULTS</b>All patients were followed up from 6 to 20 months with an average of 12 months. At the latest follow-up, Lysholm scores were 83.5±10.0;2 knees got excellent results, 1 knee good, and 1 knee moderate.</p><p><b>CONCLUSIONS</b>Knee discoid medial meniscus is easily missed diagnosis for atypical and early clinical symptoms , and seeing a doctor for obvious symptoms with meniscus tear, and could receive good curative effects after diagnosis as soon as possible.</p>

AIM:To determine the role of nuclear receptor subfamily 6,group A,member 1 (NR6A1) in vascular smooth muscle cell (VSMC) apoptosis.METHODS:NR6A1 protein was over-expressed in the VSMCs by infection of adenovirus.The effect of NR6A1 on the viability of VSMCs was measured by MTT assay.DAPI staining,TUNEL staining and caspase activity assay were conducted.DNA microarray was used to quickly screen the target genes of NR6A1.The effect of receptor-interacting serine/threonine-protein kinase 3 (RIPK3) silencing on NR6A1-induced apoptosis of the VSMCs was further analyzed.RESULTS:Adenovirus-mediated over-expression of NR6A1 induced the apoptosis of VSMCs.The RIPK3 gene expression was up-regulated by NR6A1 over-expression in the VSMCs.NR6A1-induced VSMC apoptosis was inhibited by RIPK3 silencing.CONCLUSION:NR6A1 promotes VSMC apoptosis by up-regulating the RIPK3 gene expression.

Objective To investigate the effects of dendrobium officinale leaves on sperm quality and testicular tissue morphology in parent(P)and offspring(F1、F2)rats.Methods P、F1、F2 rats were fed with dendrobium officinale leaves at the dose of 0,2.0,4.0,6.4g/kg·bw for two generation reproduction,and the testicular and epididymal viscera coefficients,the quantity and quality of sperm were examined and the histopathological assessment was carried out. Results There were no statistical differences in the testicular and epididymal viscera coefficients,sperm quantity,sperm motility rate and sperm malformation rate compared with the control group in P,F1,F2 rats(P>0.05). There were no differences between 0 and 6.4 g/kg·bw groups on the testicular and epididymal tissue morphology in P,F1,F2 rats. Conclusion Dendrobium officinale leaves didn't show obvious adverse effects on sperm quality and testicular tissue morphology in P,F1,F2 rats.

RESUMEN

Objective To investigate effects of xylo-oligosaccharides with chitooligosaccharides on alcohol-induced liver injury and immune function in mice. Methods Different doses of chitosan oligosaccharide (0.045 g/kg-0.26 g/kgB.W) and xylo-oligosaccaride (0.055 g/kg-0.32 g/kgB.W) were feed to the mice for 30 days. The mice live-injury model was induced by alcohol. MDA、 GSH、 TG level in liver and T lymphocyte proliferation of spleen cells induced by conA, the antibody-producing cells, and natural killer (NK) activity were detected. Results Compared with the live-injury model group, MDA level of low/middle dose group, TG level of middle dose group and pathological evaluation score of liver steatosis of high dose group in mice liver were decreased because of chitosan oligosaccharide and xylo-oligosaccaride feeding. Compared with the control group, the ability of T lymphocyte proliferation of mouse spleen induced by ConA and the antibody-producing cells were increased in mice of middle and high dose group. The differences had statistical significances (P<0.05) . Conclusion Under this experimental condition, xylo-oligosaccharides in combination with chitooligosaccharides could protect the mice from alcohol-induced liver injury and enhance immune function in spleen of normal mice synergistically.
