RESUMEN

Objective:To investigate and compare the predictive effects of two postoperative nausea and vomiting (PONV) prediction models in elderly patients with thoracoscopic partial pneumonectomy.Methods:The total of 227 elderly patients who underwent thoracoscopic partial pneumonectomy in Brain Hospital Affiliated to Nanjing Medical University from October 2021 to January 2022 were collected. Apfel risk score and Koivuranta risk score were used to predict the risk of PONV in each patient. The area under the ROC curve was used to evaluate the discrimination ability of the two prediction models; Hosmer-Lemeshow goodness of fit test was used to evaluate the calibration of the two prediction models.Results:Among 227 patients, 74 had PONV, and the incidence of PONV was 32.6%. The area under the ROC curve of Apfel and Koivuranta risk scores in the risk prediction of PONV in elderly patients with thoracoscopic partial pneumonectomy were 0.640 and 0.683 respectively. There was no significant difference between the two ( Z=1.54, P>0.05). The Hosmer-Lemeshow goodness of fit test showed that the accuracy of the two models in predicting PONV risk in elderly patients with thoracoscopic partial pneumonectomy was better ( P>0.05). Conclusions:The difference between the two models in PONV risk prediction in elderly patients with thoracoscopic partial pneumonectomy is general, Koivuranta model is more suitable for predicting PONV in elderly patients with thoracoscopic partial pneumonectomy, but its applicability is still insufficient.

Objective: To study the antioxidation activities in vitro of a comment flavonoid component named vicenin Ⅱ(Apigenin 6,8-di-C-glucoside) in Dendrobii Officinalis Caulis from different origin places and investigate its effects on apoptosis of HepG2 cells. Method: The antioxidation activities in vitro of vicenin Ⅱ (0.005-1 g·L-1) were evaluated by 2,2-diphenyl-1-picrylhydrazyl (DPPH), salicylic acid and 2-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid(ABTS) and copper ion reduction assays. Methye thiazolye telrazlium(MTT) assay was used to test the inhibitory effect of vicenin Ⅱ(12.5~100 μmol·L-1) on proliferation of 6 tumour cells in vitro. In subsequent apoptosis experiment, the concentration of vicenin Ⅱ was 75 μmol·L-1. The morphological changes of HepG2 cells were evaluated by Hoechst 33258 under fluorescence microscope; and the cell apoptosis rate was detected by flow cytometry with AnnexinV/PI apoptosis assay kit. The mRNA expressions of mitogen activated protein kinase (MAPK) pathway related apoptotic genes were detected by Real-time PCR assay. Result: The 1 g·L-1 vicenin Ⅱ showed 48.82% and 22.01% for DPPH scavenging rate and Cu2+ reduction rate respectively(P-1 vicenin Ⅱ showed 86.88% for ABTS scavenging rate(P-1 Vicenin Ⅱ, the cells survival rate was 45.69%(PPN-terminal kinase (JNK), and nuclear transcription factor (NF)-κB were increased(PConclusion: The general flavone glycosides component vicenin Ⅱ of Dendrobii Officinalis Caulis from different origins has a certain antioxidation effect and significant inhibitory effect on proliferation, and could induce apoptosis on HepG2 cells probably by regulating the expression of related genes in MAPK pathway and Bax/Bcl-2.

To investigate the effect of turmeric volatile oil (TVO) on the apoptosis and proliferation of human skin SCC A431 cells, A431 cells were incubated with different concentrations (5-80 mg•L⁻¹) of TVO in vitro.The proliferation and cell cycle were assessed by CCK8 assay. The change of morphology was observed with inverted microscope. Apoptosis was evaluated with AO/EB double staining and flow cytometry (FCM); cell cycle was analyzed with FCM .Western blot method was used to detect caspase-3 and caspase-9 protein expression. Results indicated that TVO has significant inhibitory effects on the growth of A431 cells in a dose dependent relationship, the difference between groups has statistically significant (P<0.05). TVO group compared with control group, concentrations in cells shrivel and broken phenomenon, cell apoptosis rate increased, and a dose dependent and increased the expression of caspase-3 and caspase-9. The experiment results suggested that TVO could restrain skin squamous carcinoma A431 cells proliferation, and induce its apoptosis. The mechanism may be related to increase the expression of caspase-3 and caspase-9.

<p><b>OBJECTIVE</b>To observe the expression of hypoxia-inducible factor-lalpha subunit (HIF-1alpha), HIF prolyl hydroxylase domain-containing protein(PHDs) and factor inhibiting HIF-1(FIH) in pulmonary arteries of patient with chronic obstructive pulmonary disease (COPD).</p><p><b>METHODS</b>Pulmonary specimens were obtained from patients undergoing lobectomy for lung cancer, 12 had concurrent COPD (COPD group) and 14 without COPD (control group). The ratio of vascular wall area to total vascular area (WA%) and pulmonary artery media thickness (PAMT) was observed, and HIF-1alpha and its hydroxylases(PHD1, PHD2, PHD3, FIH) mRNA and protein were detected by in situ hybridization and immunohistochemistry respectively.</p><p><b>RESULTS</b>WA% and PAMT of COPD patients(50 microm +/- 9 microm, 40% +/- 5%, were statistically different from those of the control subjects (39 microm +/- 6 microm, 31% +/- 4%, P < 0.01). Relative quantification of mRNA and protein levels (absorbance, A) showed that HIF-lalpha mRNA and protein levels in COPD group (0.230 +/- 0.036,0.275 +/- 0.039) were statistically higher than those of the control subjects (0.174 +/- 0.029, 0.102 +/- 0.015, P < 0.01 ), and that the protein level increased more markedly. PHD1 mRNA in COPD subjects (0.180 +/- 0.030) was comparable to that in control group (0.191 +/- 0.029, P > 0.05); PHD2 and PHD3 mRNA levels in COPD (0.245 +/- 0.044, 0.252 +/- 0.023) were significantly higher than those in control group(0.182 +/- 0.028, 0.127 +/- 0.017, P < 0.01). On the other hand, in COPD subjects PHD1 protein (0.104 +/- 0.015) was significantly lower(P < 0.01), whereas PHD2 protein (0.274 +/- 0.044) was significantly higher(P < 0.01) than those in control group(0.209 +/- 0.023, 0.219+/- 0.043). As for PHD3 protein, no significant changes were observed between the two groups (0.161+/- 0.023 in COPD, 0.146 +/- 0.021 in control, P > 0.05). FIH mRNA and protein both showed no differences between the two groups. Linear correlation analysis showed that HIF1alpha protein was positively correlated with WA%, PAMT, PHD2 mRNA and protein, PHD3 mRNA, and that HIF1alpha protein was negatively correlated with PHD1 protein.</p><p><b>CONCLUSION</b>PHDs may be involved in the process of hypoxic pulmonary vascular remodeling in COPD via regulation of HIF-1alpha gene expression</p>

<p><b>OBJECTIVE</b>To evaluate the changes of plasma levels of the excitatory amino acid neurotransmitter aspartic acid (Asp), inhibitory neurotransmitter glycine (Gly) and asparagine (Asn) in patients with major depressive disorder (MDD).</p><p><b>METHODS</b>Plasma samples were collected from 15 MDD patients (9 males and 6 females, aged 32-64 y) and 14 healthy subjects (7 males and 7 females, aged 30-65 y); and also collected from 7 MDD patients (5 males and 2 females) 2 months after antidepressant treatment. The plasma levels of amino acids were determined by high performance liquid chromatography with fluorescence detection method.</p><p><b>RESULTS</b>Plasma Asp and Gly levels were significantly lower in MDD patients than those in controls (P<0.04). There were positive correlations between plasma levels of Gly and Asp, and between Gly and Asn (P<0.005) in the control group; while in MDD patients, a significant positive correlation was found only between plasma levels of Gly and of Asp (P<0.001). MDD patients did not show significant changes in plasma Asp, Asn and Gly levels after antidepressant treatment compared to those before treatment.</p><p><b>CONCLUSION</b>The reduced plasma Asp and Gly levels may serve as a clinical biomarker for MDD.</p>

Objective: To compare the clinical efficacies and complications between the posterior lumbar interbody fusion (PLIF) using bilateral pedicle screw fixation and modified PLIF using unilateral pedicle screw fixation. Methods: A total of 96 patients who underwent lumbar spinal fusion operation during Feb. 2007 to May 2009 were divided into 2 groups. Group A included 40 patients (14 males and 26 female, average age being [52.23±9.75] years) receiving unilateral modified PLIF; Group B included 56 patients (22 males and 34 female, average age being [56.02±10.25] years) receiving bilateral PLIF. Oswestry disability index (ODI), visual analog scale (VAS), operating time, blood loss, length of hospital stay, hospitalization cost, fusion rates, complication rates, intervertebral height and medical expenses were all compared between the two groups. Results: The short-term ODI, VAS indices showed no significant difference between the two groups. Blood loss, operating time, and hospitalization cost of group A were significantly less than those in group B (P<0.05). Fusion rate was 100% in both groups 12 months after the operation. Conclusion: Modified PLIF using unilateral pedicle screw fixation is effective for lumbar spinal fusion, and it has the less trauma, risk and hospitalization cost compared with PLIF using bilateral pedicle screw fixation; but its long-term efficacy still needs to be further observed.

RESUMEN

It is critical to generate marker gene free transgenic plants for retransformating or eliminating the potential harmfulness of marker gene and its product. In this study, Ac/Ds transposon system was developed for removal of hpt selection marker gene to obtain marker-free transgenic plants in rice ( Oryza sativa L.). Ds element containing the interesting gene bar was constructed next to the selection marker gene hpt to get Ds-T-DNA. Rice plants were transformed by Agrobacterium tumefaciens EHA105 containing Ac-T-DNA and Ds-T-DNA respectively. Rice plant containing single copy Ac-T-DNA was crossed with plant containing single copy Ds-T-DNA to obtain the F1 plant containing both Ac and Ds elements. F1 plant was self-crossed to produce F2 progeny in which T-DNA insert and transposed Ds element segregated independently. Two plants contained Ds element but no hpt marker gene in total 100 F2 plants. The result indicated that Ac/Ds transposon system could be used as a vector system for generating marker gene free transgenic plants in rice.
