RESUMEN

Objective To investigate the effect of interfering Hiwi gene on the apoptosis of MDA-MB-231 cells. Methods The mRNA and protein expression of Hiwi mRNA and its target protein were analyzed by qRT-PCR and Western Blot after transfection. MDA-MB-231 cells were divided into 6 groups according to the experimental design. Interference effects were screened as siRNA interference group (Hiwi10330 group), and then divided into 3 groups according to the experimental design: interference group, negative control group/NC, blank control group/Blank. The cell apoptosis rate was detected by flow cytometry after transfection. Results The expression of mRNA in the interference group was significantly lower than that in the siRNA group (P < 0.05), the expression of target protein of Hiwi gene was also significantly inhibited (P < 0.05). The apoptosis rate of MDA-MB-231 cells was significantly higher than that of NC and Blank groups (P<0.05). Conclusion The apoptosis rate of breast cancer cells MDA-MB-231 was significantly increased after siRNA targeting hiwi gene silencing.

Objective To explore the expression differences of long noncoding RNA (LncRNA) LOC100288637 in liver canc‐er ,the effect of LOC100288637 on liver cancer cell proliferation and the relevant mechanisms .Methods Based on the analysis of the GEO database data sets GSE58043 and GSE10694 ,we found that both LOC100288637 and hsa‐miR‐101‐3p has obvious expression differences in liver cancer tissues .RNA hybrid revealed the possibility of combination between LOC 100288637 and hsa‐miR‐101‐3p;RT‐PCR was performed to measure the expression level of LOC100288637 in tissues and cells ;fluorescence in situ hybridization was used to observe LOC100288637 localization in cells ;cell proliferation was determined by CCK8 experiment after LOC100288637 siRNA knock down .The expression of LOC100288637 in cells were measured after treated with hsa‐miR‐101‐3p mimics .Results Relative quantitative expression of LOC100288637 in liver cancer tissues group was significantly higher than that in no tumor tis‐sues group (P<0 .05);relative quantitative expression of LOC100288637 in liver cancer cell lines were significantly higher than that in normal liver cell line(P<0 .05) .LOC100288637 was located in both cytoplasm and nucleus of HepG2 cells ,and mainly in cy‐toplasm .Cell proliferation vitality of HepG2 reduced after treated with LOC100288637‐siRNA(P<0 .01) .Relative quantitative ex‐pression of LOC100288637 in HepG2 reduced after treated with hsa‐miR‐101‐3p mimics ( P< 0 .05 ) .Conclusion LncRNA LOC100288637 may play an important role in liver cancer development ,it can be down regulated by hsa‐miR‐101‐3p and affect the proliferation of liver cancer cells .

Objective To quantitatively analyze the dynamic posture in patients with early stage Pakinson disease (ESPD) by using dynamic posture equilibrium equipment. Method Thirty two ESPD patients were recruited in this study, in which, 10 was at Hoehn-Yahr stage 1, 9 at Hoehn-Yahr stage 1.5, 13 at Hoehn-Yahr stage 2. Thirty two age and gen?der matched healthy subjects served as a control group. The sensory organization test (SOT), motion coordination test (MCT), adaptive test (ADT), walk across were performed in those two groups using NeuroCom smart dynamic posture equipment. Results The average composite scores for SOT were 64 and 79 in ESPD patients and control group(P=0.032). The eqilibrium ratio of vision and vestibular sense were obviously lower in ESPD patients than in the control group ( 73.55±3.15 vs. 82.45±3.53, P=0.037;45.25±4.25 vs. 66.43±6.98, P=0.021). MCT showed that ESPD patients had a higher abnormality ratio of centre-of-gravity position compared with controls ( 70.3%vs. 7% )(P<0.001). ESPD pa?tients had 33% and 57% abnormality in ADT and Walk Across, which were obviously higher compared with control group (P<0.001). Conclusions ESPD patients have different degrees of posture disorders and posture equilibrium assess?ment at early stage is beneficial to rehabilitation training.

Objective To study the clinical effect of ultrasound guidance interventional radiofrequency ablation (RFA) in treatment of liver tumor.Methods One hundred and eighteen patients with liver tumor were treated with interventional RFA under ultrasound guidance,the number of tumors and pain conditions before and after treatment were observed and recorded.Results Before treatment,there were 67 cases with single tumor,24 cases with double tumors,7 cases with triple tumors,20 cases with more tumors;after treatment,tumor necrosis were observed in 29 cases,51 cases with single tumor,25 cases with double tumors,0 case with triple tumors,13 cases with more tumors,the number of tumors before and after treatment had significant difference (P < 0.05).Visual analog scale (VAS) score,Oswestry disability index questionnaire (ODI) score after treatment 1 day,3 days,3 months,6 months respectively (3.0 ± 1.8),(3.1 ± 2.3),(3.5 ± 2.4),(3.0 ± 2.5) scores and (15.6 ± 1.5),(16.2 ± 2.3),(15.6 ± 3.5),(16.2 ± 2.5)scores,compared with before treatment 1 day [(7.5 ± 1.9) scores and (38.9 ± 3.5) scores],there were significant differences (P < 0.05).Conclusion Ultrasound guidance interventional RFA can improve the effect of surgery in treatment of liver tumor,relieve patients' pain,which is worthy of reference for clinical application.

<p><b>BACKGROUND AND OBJECTIVE</b>Exercise can improve circulation, muscular strength and happiness of cancer survivors. But more data were needed to demonstrate both the exercise ability of cancer suivivors after pulmonary lobectomy and the influences of exercise on their survivals. The aim of this study was to examine changes of exercise and its clinical effects among eldly non-small cell lung cancer survivors.</p><p><b>METHODS</b>Elderly non-small cell lung cancer survivors who had progression-free disease after surgery, chemotherapy, radiation therapy or tyrosine kinase inhibitors were included. Their exercises and participation rates per week before cancer diagnosis, after 3 months anticancer therapy and 1 year after diagnosis as well as their exercise motivations and prevalences were investigated retrospectively.</p><p><b>RESULTS</b>Forty-eight elderly non-small cell lung cancer survivors were selected. Moderate-vigorous intensity exercise had by the elderly progressin-free non-small cell lung cancer survivors after diagnosis decreased, but the participation rate of light intensity exercise was higher in 1 year after diagnosis than before diagnosis. 75.9% (14/58) patients had exercise up to the standard and the cancer recurrence rate was 20.0% (7/35). The recurrence rate of the other group was 35.7% (5/14), and the risk ratio of recurrence was 2.14 (95% CI: 0.81-5.68, P = 0.26). The most common motivations of exercise were improving health, increasing physical activity, maintaining healthy life style and improving immunity. And the main disturbances were fatigue, discomfort and lack of motivation.</p><p><b>CONCLUSION</b>The exercise participation rate during anticancer treatment among the elderly non-small cell lung cancer survivors decreased and did not return to prediagnosis levels after treatments were completed. The relationship between exercise and recurrence of cancer was not clear and needed further work.</p>

RESUMEN

Objective To study the factors influencing the recurrence of aggressive fibromatosis(AF) patients.Methods Analyze clinical features and find out factors affecting AF recurrence with univariate and multivariate analysis. Results One hundred and three AF patients were analyzed. Diameter of all the masses was (7.84?5.62)cm. 98.2% of all patients received surgery and radical resection rate is 79.4%. First recurrent time after surgery of male and female is (1563?377) and (2117?3704) days for male and female cases respectively,it's (2723?461),(657?262),(2090?499),(812?220) and (721?234) days for that of abdomen wall、head and neck(H & N),deep mass,joints and chest wall. Recurrent time is (2232?271) and (1347?267) days for those with or without surgery history patients. Univariate analysis showed that gender,tumor site and surgery history of tumor site are prognostic factors of tumor recurrence. Multivariate analysis suggest that only surgery history of tumor site is an independant prognostic factors of AF recurrence after surgery. Conclusion Gender,tumor location and surgery history of tumor site can be valuable to predict postsurgery recurrence of AF.

<p><b>OBJECTIVE</b>To investigate the relationship between subsets of lymphocytes and between its activated status and the clinical manifestations in patients with PNH, and to unfold immunological mechanism in the pathogenesis of PNH.</p><p><b>METHODS</b>The peripheral blood mononuclear cells (PBMNC) from 18 PNH patients and 20 controls were separated into two subpopulations using anti-CD(59) monoclonal antibody combined with goat-anti-mouse IgG immunomagnetic beads. CD(3)(+), CD(4)(+) and CD(8)(+) lymphocyte subsets were detected by flow cytometry. In 6 newly diagnosed patients, phenotypes associated with T cell activation such as CD(28)(+)/CD(4)(+) or CD(8)(+) cells, CD(8)(+) CD(38)(+) cells, and HLA-DR(+)/CD(4)(+) or CD(8)(+), and NK (CD(3)(-) CD(16)(+)) cells were detected in the peripheral blood.</p><p><b>RESULT</b>Patients with PNH showed significantly increased CD(3)(+) CD(8)(+)/CD(3)(+) CD(4)(+) ratio as compared with controls (1.22 +/- 0.51 vs 0.86 +/- 0.27, P < 0.05), and the CD(3)(+) CD(8)(+)/CD(3)(+) CD(4)(+) ratio in CD(59)(-) PBMC was higher than that in CD(59)(+) PBMC (2.31 +/- 1.56 vs 0.62 +/- 0.27, P < 0.05). The ratios of CD(4)(+) CD(28)(+)/CD(4)(+) markedly decreased and CD(8)(+)HLA-DR(+)/CD(8)(+) increased.</p><p><b>CONCLUSION</b>Patients with PNH appear to have abnormalities in their lymphocytes. Increased ratios of CD(3)(+) CD(8)(+)/CD(3)(+) CD(4)(+) and HLA-DR(+) CD(8)(+)/CD(8)(+) lymphocytes as well as declined ratio of CD(4)(+) CD(28)(+)/CD(4)(+) lymphocytes might be involved in the pathogenesis of PNH.</p>
