RESUMEN

Abstract Bacterial leaf blight (BLB) is one of the major rice diseases in Malaysia. This disease causes substantial yield loss as high as 70%. Development of rice varieties which inherited BLB resistant traits is a crucial approach to promote and sustain rice industry in Malaysia. Hence, this study aims were to enhance BLB disease resistant characters of high yielding commercial variety MR219 through backcross breeding approach with supporting tool of marker-assisted selection (MAS). Broad spectrum BLB resistance gene, Xa7 from donor parent IRBB7 were introgressed into the susceptible MR219 (recurrent parent) using two flanking markers ID7 and ID15. At BC3F4, we managed to generate 19 introgressed lines with homozygous Xa7 gene and showed resistant characteristics as donor parent when it was challenged with Xanthomonas oryzae pv. oryzae through artificial inoculation. Recurrent parent MR219 and control variety, MR263 were found to be severely infected by the disease. The improved lines exhibited similar morphological and yield performance characters as to the elite variety, MR219. Two lines, PB-2-107 and PB-2-34 were chosen to be potential lines because of their outstanding performances compared to parent, MR219. This study demonstrates a success story of MAS application in development of improved disease resistance lines of rice against BLB disease.

Fennel plant (Foeniculum vulgare, Mill.) is one of most widely used plants in food and medicine. Our study aimed to show the response of fennel plants from Egypt and China planted in Egypt to foliar application of benzyladenine (BA) at various concentrations; control, 50, 100 and 150 ppm. The obtained results indicated that treated fennel plants with BA resulted in a significant increase in the vegetative growth, number of umbels/plant, fruit, yield and oil yield per plant and feddan as well as main components, total phenolic contents and antioxidant activity of essential oils compared to untreated plants. In general, the application of benzyladenine was more effective on Egyptian fennel plants than that of Chinese plants. Egyptian fennel plants gave higher values of growth, fruit yield, oil percentage and oil yield than Chinese plants. However, the major components, TPC and antioxidant activity (DPPH %) had higher values in Chinese essential oil than Egyptian essential oil. The highest values of all studied parameters were obtained by using BA at 100 ppm. GC–MS analysis of the volatile oils showed that the major components were trans-anethole followed by fenshone, ?- pinene and D-limonene, respectively for both essential oils. 100 ppm of BA gave the highest values of trans-anethole content and radical scavenging activities of oils. Therefore, we concluded that spraying fennel plants with 100 ppm BA can be used for obtaining higher vegetative growth, fruit and quantity and quality of oil.

RESUMEN

Tagetes minuta L. (Aromatic Tagetes) is an erect and stout summer annual herb. Ocimum basilicum L. (Sweet Basil) is considered a summer aromatic plant. There are several methods for extraction of essential oils i.e. (HD) hydro-distillation, (SD) steam distillation, (SFE) supercritical fluid extraction, and organic solvents. It is now considered the method of extracting essential oil with steam distillation is the commercial method used in many countries that produce essential oils. SFE method consider is the latest extracting essential oils method but the method spread is limited due to the high cost of its establishment. The present study is aimed to compare the two methods SD and SFE to extract the essential oil from sweet basil and aromatic tagetes plants, and its yield on the resulting essential oil yield. SFE method increases oil yield and improves oil quality. SFE method gave a high percentage of the following major’s components for sweet basil oil (linalool (50.22%) and E- Citral (10.44%)) and aromatic tagetes oil ( ?-Ocimene (27.49%) and cis-Tagetone (9.93%)) compared to the steam distillation method.

RESUMEN

@#Trypanosoma brucei parasites are flagellated kinetoplastid protozoan which is responsible for Human African Trypanosomiasis (HAT). Current chemotherapy drugs have a number of side effects and drug resistance has emerged as a major issue in current treatment. Active bisindole alkaloid compound ochrolifuanine was previously isolated from the leaves of Dyera costulata. In vitro antitrypanosomal activity of ochrolifuanine against Trypanosoma brucei brucei strain BS221 showed strong activity with an IC50 value of 0.05 ± 0.01 µg/ml. We compared the effect of ochrolifuanine and reference compound staurosporine in T. b. brucei apoptosis. The apoptosis-inducing activity of ochrolifuanine was evaluated using TUNEL assay and cell cycle analysis. Trypanosoma brucei brucei was shown to undergo apoptotic cells death as demonstrated by the appearance of several conical hallmarks of apoptosis. Ochrolifuanine was found to induce apoptosis in parasites in a dose- and time-dependent manner. The cell cycle study revealed 0.025 and 0.05 µg/ml of ochrolifuanine arrested the growth of T. b. brucei at two different growth phases (G0/G1 and in S phases). While at concentration 0.10 µg/ml arrested at the G2/M phase. In conclusion, the results indicate that ochrolifuanine displayed an antitrypanosomal effect on T. b. brucei by inducing apoptosis cell death and causing the arrest of parasite cells at different growth phases. The results suggested that ochrolifuanine may be a promising lead compound for the development of new chemotherapies for African trypanosomiasis.

ABSTRACT: Idiopathic sleep-related hypoventilation occurs in individuals with hypercapnia during sleep in normal conditions of the respiratory system in the absence of other disorders. Patients with this condition may be asymptomatic or have complaints of morning headache, cognitive deficit and fatigue, and observation of shallow breathing. This report describes the case of a 3-year-old patient with normal physical and neurological exam, appropriate neuropsychomotor development, presenting breathing irregularities, and bradycardia during sleep. The patient was referred to an investigation for sleep respiratory disturbs and was diagnosed with hypoventilation related to sleep. The genetic study, done by enzymatic dosages and molecular genetic tests, showed the deficiency of biotinidase as a possible cause of symptomatology. The drug treatment was initiated early with the resolution of the symptoms. The present clinical report highlights the biotinidase deficiency with an isolated cardiorespiratory condition in a neurologically normal child, which also led to Breath-Holding Spells. This relation was suggested after central hypoventilation resolution following biotin introduction. Besides, Apparent Life-Threatening Events symptoms, which terrify the observer and even professionals, disappeared after the oral intake of biotin. Finally, this case report corroborates the expansion of possibilities for the phenotypic manifestations of late cases from biotinidase deficiency, as the SHC phenotyp. ((AU)

RESUMEN

@#The aim of this study was to detect and characterize Giardia lamblia in animals in the UAE. Eighty-seven fecal samples were tested for G. lamblia using the conserved fragment of small subunit (SSU)-rRNA by nested PCR. Giardia-positive isolates were genotyped for assemblages A and B using assemblage specific primers of the triosephosphate isomerase (tpi) gene. Thirty samples (34.5%) were positive for G. lamblia. Conversely, neither genotype A nor B were detected using tpi genotyping on the studied samples. Further investigations are required using higher number of samples including both human and animals in the country taking into consideration the analysis of other genotypes to provide more detailed understanding about the zoonotic transmission of this parasite.

RESUMEN

@#Rhipicephalus sanguineus, commonly known as brown dog tick is a widespread species with considerable public health and economic importance. Tremendous efforts were performed to control the tick populations with the concern of resistance build-up and environmental issues. Alternative towards microbial control thus emerged as one option to reduce tick populations. In this study, the ovicidal efficacy of a native isolate entomopathogenic hyphomycetes fungi, Metarhizium anisopliae strain HSAH5 was evaluated against eggs of R. sanguineus. Spray applications with three different conidial concentrations of 105, 106 and 107 conidia mL-1; 40 ppm of Flumethrin and a negative control. The M. anisopliae strain was found highly virulent to R. sanguineus eggs by reducing the hatching percentages to ≈30% compared with 8.9% in Flumethrin eggs. The result showed a significantly higher mortality in M. anisopliae group than those of the control groups (F = 42.08, df = 32, P < 0.001) at 30 days post-infection. However, there are no significant differences within the M. anisopliae group, in which the mortality between different conidial concentrations is almost the same. The estimated LC50 of M. anisopliae against eggs of R. sanguineus is 1.36 × 103 conidia ml-1. Thus, these results suggest M. anisopliae strain HSAH5 could be a potential biocontrol agent of R. sanguineus in the integrated approach to managing ticks in the residential landscape by targeting on the eggs.

RESUMEN

Streptococcus pneumoniae, Haemophilus influenzae, and Neisseria meningitidis, which are typically transmitted via respiratory droplets, are leading causes of invasive diseases, including bacteraemic pneumonia and meningitis, and of secondary infections subsequent to post-viral respiratory disease. The aim of this study was to investigate the incidence of invasive disease due to these pathogens during the early months of the COVID-19 pandemic. Methods In this prospective analysis of surveillance data, laboratories in 26 countries and territories across six continents submitted data on cases of invasive disease due to S pneumoniae, H influenzae, and N meningitidis from Jan 1, 2018, to May, 31, 2020, as part of the Invasive Respiratory Infection Surveillance (IRIS) Initiative. Numbers of weekly cases in 2020 were compared with corresponding data for 2018 and 2019. Data for invasive disease due to Streptococcus agalactiae, a non-respiratory pathogen, were collected from nine laboratories for comparison. The stringency of COVID-19 containment measures was quantified using the Oxford COVID-19 Government Response Tracker. Changes in population movements were assessed using Google COVID-19 Community Mobility Reports. Interrupted time-series modelling quantified changes in the incidence of invasive disease due to S pneumoniae, H influenzae, and N meningitidis in 2020 relative to when containment measures were imposed. Findings 27 laboratories from 26 countries and territories submitted data to the IRIS Initiative for S pneumoniae (62 434 total cases), 24 laboratories from 24 countries submitted data for H influenzae (7796 total cases), and 21 laboratories from 21 countries submitted data for N meningitidis (5877 total cases). All countries and territories had experienced a significant and sustained reduction in invasive diseases due to S pneumoniae, H influenzae, and N meningitidis in early 2020 (Jan 1 to May 31, 2020), coinciding with the introduction of COVID-19 containment measures in each country. By contrast, no significant changes in the incidence of invasive S agalactiae infections were observed. Similar trends were observed across most countries and territories despite differing stringency in COVID-19 control policies. The incidence of reported S pneumoniae infections decreased by 68% at 4 weeks (incidence rate ratio 0·32 [95% CI 0·270·37]) and 82% at 8 weeks (0·18 [0·140·23]) following the week in which significant changes in population movements were recorded. Interpretation The introduction of COVID-19 containment policies and public information campaigns likely reduced transmission of S pneumoniae, H influenzae, and N meningitidis, leading to a significant reduction in life-threatening invasive diseases in many countries worldwide. RESUMEN

Aim: Antarctic cyanobacteria may represent a potential resource of new and unique compounds with interesting capabilities. Profiling of fatty acids in Antarctic cyanobacteria can provide an overview of potential fatty acids present in them, that can be utilised in future applicationsMethodology: In total, 4 cyanobacteria previously isolated from Antarctic polar ice was used in this study. Molecular identification using 16S rRNA gene was used to ascertain their identities as Leptolyngbya spp. and their fatty acid profiles were determined using GCMS. Results: Morphologically, these cyanobacteria were found similar to Leptolyngbya sp. Analysis of 16S rRNA gene sequences amplified further confirmed their identity and were designated as Leptolyngbya sp. A, B, D and E. Following fatty acid analysis using GCMS, it was determined that unsaturated fatty acids predominated the fatty acid profiles for Leptolyngbya sp. A, B and D, while saturated fatty acid was found dominant in Leptolyngbya sp. E. Leptolyngbya sp. D contained almost 100% of linoleic acid, whilst Leptolyngbya sp. A and Leptolyngbya sp. B contained 59.35% and 83.33% of linoleic acid, respectively. Besides linoleic acid, palmitoleic acid (18.45%), oleic acid (19.45%) and lauric acid (2.74%) were also present in Leptolyngbya sp. A. As for Leptolyngbya sp. B, other than linoleic acid, only oleic acid (16.67%) was detected. Interpretation: Findings from this study demonstrate that the Antarctic Leptolyngbya spp. A, B and D identified in this study possess high content of unsaturated fatty acids, while only saturated fatty acid was present in Leptolyngbya sp. E. Fatty acid profiles revealed the potential of these Antarctic Leptolyngbya species to be further exploited for other applications.

RESUMEN

Objective: Andrographis paniculata is widely used in Asia for various medicinal purposes. The plant has a major bioactive chemical constituent Andrographolide, which exhibits various essential pharmacological properties. Recently, to enhance immunity against viral fevers especially dengue viral fever during monsoon season, Tamil Nadu state government has advised rural health centres to provide a tea or kashayam made from a mixture containing this plant leaf along with selected plant roots and leaves. However, there is concern among the general public population that this plant extract may have a negative impact on potency. This study is being done to investigate the toxic effects of Andrographis paniculata on fecundity and physiological properties of Drosophila melanogaster. Methods: The flies were treated and mated in pure and mixed form of Andrographis paniculata separately. Results: The results were obtained in the form of egg count, which was compared to control and the conclusion was obtained that Andrographis paniculata might affect the fecundity and at the same time, it was observed that the developmental span from an egg to an adult fly increased upon treatment in successive generations. More importantly, the effect of pure form plant on the fecundity was found to be significant, while that of the mixed form was not. On mating a treated male with an untreated female and an untreated male with a treated female when the treatment is being discontinued, it can be very well seen that there is no effect on the fecundity of the flies. Conclusion: Since the continuity of the treatment played a major role on the effect of fecundity, highlighting the importance of the period of treatment and the exposure time of the compound on the fly system, therefore this research suggests that the conception of tea or kashayam extracted from mixed plant content could not have the same effect as the similar concentration of pure Andrographis paniculata and it does not have any acute effect on the fecundity when being consumed over a short period of time in Drosophila melanogaster model.

ABSTRACT Introduction: Human papillomavirus (HPV) is intimately associated with cervical cancer, and the presence of coinfections, such as with Chlamydia trachomatis, Gardnerella vaginalis and Trichomonas vaginalis, may potentiate or facilitate HPV infection. Female sex workers are considered vulnerable to the acquisition of these infections due to exposure to risk factors. Objective: To determine HPV infection, viral types and coinfections in self-collected samples from female sex workers. Methods: Self-collected samples from female sex workers, of vaginal canal and uterine cervix, were subjected to HPV-deoxyribonucleic acid (DNA) detection, viral genotyping by type-specific polymerase chain reaction (PCR), restriction fragment length polymorphism (RFLP), and the detection of coinfection. Results: HPV-DNA was detected in 19.4% of the samples, and HPV 31, 6, and 53 were the most frequently detected types. There was a predominance of high-risk oncogenic HPV (HR-HPV) and a strong presence of simultaneous infections with multiple HPV types (84.6%). Coinfections with both HPV and C. trachomatis, and HPV and G. vaginalis were detected. The variables that were statistically associated with HPV infection and the presence of multiple infections were non-use of condoms and non-compliance with regular cervical cytology screening. Conclusion: The results highlight the importance of more comprehensive studies among vulnerable populations, aiming to establish measures to raise awareness about the risks of contracting sexually transmitted infections, as well as to support future studies for introducing HPV vaccines with wider coverage of viral types.

Abstract: Human-modified landscapes (HMLs) are composed by small, isolated and defaunated forest fragments, which are surrounded by agricultural and urban areas. Information on species that thrives in these HMLs is essential to direct conservation strategies in local and regional scales. Since HMLs are dominant in the Atlantic Forest, we aimed to assess the mammalian diversity in a HML in southeastern Brazil and to propose conservation strategies. We collected data of terrestrial (small-, medium- and large-sized) and volant mammals in three small forest fragments (10, 14 and 26 ha) and adjacent areas, between 2003 and 2016, using complementary methods: active search, camera trapping, live-traps, mist nets and occasional records (i.e., roadkills). In addition, we used secondary data to complement our species list. We recorded 35 native mammal species (6 small-sized, 16 medium- and large-sized, and 13 bats) and seven exotic species in the HML. The recorded mammal assemblage (non-volant and volant), although mainly composed of common and generalist species, includes three medium- and large-sized species nationally threatened (Leopardus guttulus, Puma concolor and Puma yagouaroundi) and two data deficient species (Galictis cuja and Histiotus velatus), highlighting the importance of this HML for the maintenance and conservation of mammal populations. Despite highly impacted by anthropogenic disturbances, the study area harbors a significant richness of medium- and large-sized mammals, being an important biodiversity refuge in the region. However, this biodiversity is threatened by the low quality of the habitats, roadkills and abundant populations of domestic cats and dogs. Therefore, we stress the need of conservation strategies focusing on the medium- and large-sized mammals as an umbrella group, which could benefit all biodiversity in the landscape. We recommend actions that promotes biological restoration, aiming to increase structural composition and connectivity of the forest fragments, reducing roadkills and controlling the domestic cats and dogs' populations, in order to maintain and improve the diversity of mammals in long-term.
