RESUMEN

Objective:To investigate the effect of ginsenoside 20(S),25-epoxydammarane-3β,12β,24α-triol (24-OH-panaxadiol,24-OH-PD) on inhibiting proliferation and inducing apoptosis of tumor cells, and explore its mechanism of action. Method:The inhibitory effect of 24-OH-PD (12.5,25,50,100 mg·L-1) on proliferation of CCRF-CEM, M14, MD-MBA-231 and Jeko-1 cells with different treatment periods (24, 48,72 h) was evaluated by methylthiazolyldiphenyl-tetrazolium bromide (MTT)assay and CellTiter Glo test, and the results were then compared with 20(R)-Rg3 and 20(S)-Rh2.Flow cytometry was used to detect cell apoptosis caused by 24-OH-PD. Besides, the potential anticancer mechanism was studied by docking analysis with 40 cancer related proteins and 24-OH-PD by using drug design platform Schrodinger Maestro 6.7 Software. Result:24-OH-PD inhibited the proliferation of all the 4 cancer cell lines significantly in a time and dosage dependent manner. The IC50 value of 24-OH-PD on CCRF-CEM,Jeko-1,M14,and MD-MBA-231 cell lines was 25.36, 39.29, 21.74, and 19.35 mg·L-1, respectively, similar to 20(S)-Rh2 (IC50 23.35, 65.79, 18.95, 19.67 mg·L-1) and much better than 20(R)-Rg3 (only effective for Jeko-1 cells, IC50 49.5 mg·L-1). Annexin V/PI double staining experiment showed that 24-OH-PD could also induce apoptosis of the 4 kinds of cancer cells (PConclusion:24-OH-PD could inhibit cell proliferation and induce apoptosis for CCRF-CEM, M14, MD-MBA-231 and Jeko-1 cell lines, which may through directly acting on Bcl-2, Bcl-xl, and other proteins.

RESUMEN

<p><b>BACKGROUND</b>Interstitial lung disease (ILD) is a serious lung complication in polymyositis (PM) and dermatomyositis (DM) which affects prognosis and requires a more aggressive approach in therapy. This study investigated the prevalence, characteristics, predictive factors and unfavourable prognostic factors of ILD in newly diagnosed PM, DM and amyopathic DM (ADM).</p><p><b>METHODS</b>From January 2000 to December 2008, the medical records of 197 consecutive PM and DM patients at the Second Affiliated Hospital of Sun Yat-Sen University were reviewed excluding overlapping, juvenile, and malignancy-associated cases. The patients were assigned to an ILD (69 patients) and a non-ILD group (128 patients). The clinical features, laboratory findings, and prognosis were compared.</p><p><b>RESULTS</b>The multivariate analysis indicated that older age at onset (OR 1.033, 95%CI 1.009 - 1.058, P = 0.007), fever (OR 4.109, 95%CI 1.926 - 8.767, P < 0.001) and arthritis/arthralgia (OR 2.274, 95%CI 1.101 - 4.695, P = 0.026) were the independent predictive factors for developing ILD in PM/DM after excluding anti-Jo-1. Regarding anti-Jo-1, fever (OR 4.912, 95%CI 2.121 - 11.376, P < 0.001) was associated with ILD. Poor survival in ILD patients was associated with ILD clinical subset (RR 0.122, 95%CI 0.049 - 0.399, P < 0.001), ADM/DM/PM-ILD (RR 0.140, 95%CI 0.031 - 0.476, P = 0.002), cardiac involvement (RR 4.654, 95%CI 1.391 - 15.577, P = 0.013) and serum albumin level (RR 0.910, 95%CI 0.831 - 0.997, P = 0.042).</p><p><b>CONCLUSIONS</b>Patients who presented with fever tended to have a higher frequency of PM/DM-associated ILD. A Hamman-Rich-like presentation, ADM-ILD, cardiac involvement and hypoalbuminemia were poor prognostic factors in ILD-PM/DM.</p>

<p><b>OBJECTIVE</b>To explore the impact of different warm ischemia time on structure and function of the non-heart-beating donor lung and to find out the feasibility of non-heart-beating donor in rat lung transplantation.</p><p><b>METHODS</b>Sixty Sprague-Dawley rats were randomly divided into 3 groups: heart-beating donor (HBD) group, non-heart-beating donor (NHBD) with 30 minutes of warm ischemia time (WIT) group and NHBD with 60 minutes of WIT group. Each group has 10 pairs (the donors and the recipients). The donor lungs of group HBD were flushed with low potassium dextran (LPD) solution at 4 degrees C after asystolia while the lungs of group NHBD-30 and group NHBD-60 remained ventilated at the room temperature for 30 and 60 minutes after asystolia and then were flushed with LPD solution. All the donor lungs remained inflated when they were stored in LPD solution at 4 degrees C for 4 hours. The recipient rat underwent left thoracotomy, and then orthotopic left lung transplantation. Followed by a right thoracotomy, the right pulmonary hilum were ligated with one-hour reperfusion and ventilation.</p><p><b>RESULTS</b>All the recipients in group HBD and group NHBD-30 survived the observation period of one hour with excellent gas exchange, whereas 4 of recipients in group NHBD-60 survived for 10 minutes after the ligation of right pulmonary hilum and 3 for 20 minutes. The pulmonary compliance, ultrastructure, energy metabolite and other markers revealed no significant differences between group HBD and group NHBD-30 (P > 0.05). But the differences between group NHBD-60 and other two groups were significant (P < 0.05).</p><p><b>CONCLUSIONS</b>The adoption of non-heart-beating donor could be a safe and effective method to expand the lung donor pool. The NHBD lung with 30 minutes of WIT may be suitable for lung transplantation in rat.</p>

OBJECTIVE: To investigate the efficacy and safety of the implantation of phakic anterior chamber intraocular lens for high myopia. METHODS: A consecutive group of 29 eyes in 16 patients with (-7.00 to -30.00) D of myopia were implanted. RESULTS: All of the 29 eyes were implanted successfully and followed-up 3 approximate, equals 6 months. The mean best corrected visual acuity was 0.50+/-0.26 pre-operatively and 0.73+/-0.263 months post-operatively, there was no significant difference t=2.043, P=0.051 . The mean refractive diopter was -18.03+/-5.54 D pre-operatively and -0.82+/-1.54 D post-operatively t=30.899 P=0.000 ; The mean intraocular pressure was 2.091+/-0.380 kPa pre-operatively and (1.734+/-0.572)kPa post-operatively t=1.98 P=0.07 ; The mean counts of endothelial cells was (2704+/-390 /mm2 pre-operatively and (2 519+/-278)/mm2 post-operatively (t=1.16 P=0.26). CONCLUSION: The implantation of phakic anterior chamber intraocular lens for high myopia is predictable, reversible and controllable with simple manipulation. Long- time following-up is still required.

Objective:To explore relationships between life events,social support,coping style and depression.Methods:751 graduate students were asked to complete the ASLEC(adolescent life events),SSRS(social support rating scale),SCSQ(simple coping style questionnaire),and CES-D.Results:(1)21.2 percent of the subjects exhibited definite depression.(2)Academic pressure,pressure of job seeking,homesick,lose of lover,and interpersonal vexation were primary stressors.(3)Life events were positively correlated with depression(0=0.47,P
