@#Abstract: Objective To develop and validate a reverse phase⁃high performance liquid chromatography(RP⁃HPLC) method for determination of residual N⁃hydroxy succinimide(NHS)content in semaglutide. Methods A RP⁃HPLC method was developed based on the screening of chromatographic column and optimization of mobile phase(phosphate concentration and the ratio of acetonitrile),validated for specificity,suitability,accuracy,reproducibility and stability, and determined for linear range,limit of quantitation(LOQ)and limit of detection(LOD). The NHS contents in three batches of semaglutide were determined by the developed method. Results The optimal condition for RP⁃HPLC was as follows:CAPCELL PAK ADME column(4. 6 mm × 150 mm,3 μm)was adopted,serving 0. 05 mol/L potassium dihy⁃ drogen phosphate solution⁃acetonitrile(98∶2)as mobile phase A,and 70% acetonitrile as mobile phase B with gradient elution(0 min,0% B;10 min,0% B;19 min,90% B;19. 1 min,0% B;25 min,0% B)at a flow rate of 0. 8 mL/min. The detection wave length was set at 260 nm,while the column temperature was 30 ℃. The developed method showed good specificity and systemic suitability,of which the linear range was 0. 2 ~ 3. 0 μg/mL(R2 = 1. 000 0),while the LOD and LOQ were 4. 8 and 9. 6 ng respectively. The RSD of recovery rates of NHS samples at three concentrations was 0. 58%, indicating a high accuracy. The RSD of NHS contents in six test samples was 0. 16%,indicating a high reproducibility. The RSD of peak areas of NHS after storage at room temperature for 0,4,8,12,16,20 and 24 h was 0. 34%,indicating a high stability. No NHS was detected in three batches of semaglutide by the developed method. Conclusion The developed RP⁃HPLC method is simple and sensitive,which may be used for the determination of NHS content in semaglutide.

RESUMEN

XueBiJing is an intravenous five-herb injection used to treat sepsis in China.The study aimed to develop a liquid chromatography-tandem mass spectrometry(LC-MS/MS)-or liquid chromatography-ultraviolet(LC-UV)-based assay for quality evaluation of XueBiJing.Assay development involved identifying marker constituents to make the assay therapeutically relevant and building a reliable one-point cali-brator for monitoring the various analytes in parallel.Nine marker constituents from the five herbs were selected based on XueBiJing's chemical composition,pharmacokinetics,and pharmacodynamics.A selectivity test(for"similarity of response")was developed to identify and minimize interference by non-target constituents.Then,an intercept test was developed to fulfill"linearity through zero"for each analyte(absolute ratio of intercept to C response,<2%).Using the newly developed assays,we analyzed samples from 33 batches of XueBiJing,manufactured over three years,and found small batch-to-batch variability in contents of the marker constituents(4.1%-14.8%),except for senkyunolide I(26.5%).

Abstract Objective: To assess the effectiveness and safety of fast-track cardiac anesthesia using the short-acting opioid sufentanil in children undergoing intraoperative device closure of ventricular septal defect (VSD). Methods: This retrospective clinical study included 65 children who underwent intraoperative device closure of VSD between January 2017 and June 2017. Patients were diagnosed with isolated perimembranous VSD by transthoracic echocardiography. Then, they were divided into two groups, group F (n=30), whose patients were given sufentanil-based fast-track cardiac anesthesia, and group C (n=35), whose patients were given conventional cardiac anesthesia. Perioperative clinical data were analyzed. Results: No significant differences were found between the preoperative clinical parameters and intraoperative hemodynamic indices between the two groups. In group C, compared with group F, the postoperative duration of mechanical ventilation, the length of stay in the intensive care unit, the length of hospital stay, and the hospital costs were significantly increased. Conclusion: In this retrospective study at a single center, sufentanil-based fast-track cardiac anesthesia was shown to be a safe and effective technique for minimally-invasive intraoperative device closure of VSD in children, which was performed with reduced in-hospital costs.

Abstract Objectives: To compare the advantages and disadvantages of perventricular and percutaneous procedures for treating isolated ventricular septal defect (VSD). Methods: A total of 572 patients with isolated VSD were selected in our hospital between January 2015 and December 2016. The patients' median age and weight were five years (1-26 years) and 29 kg (9-55 kg), respectively. The median diameter of VSD was 6.0 mm (5-10 mm). Patients were divided into two groups. In group A, perventricular device closure was performed in 427 patients; in group B, 145 patients underwent percutaneous device closure. Results: Four hundred twelve patients in group A and 135 patients in group B underwent successful closure. The total occlusion rate was 98.5% (immediately) and 99.5% (3-month follow-up) in group A, which were not significantly different from those in group B (97.7% and 100%, respectively). Patients in group A had longer intensive care unit (ICU) stay than those in group B, but patients in group B experienced significantly longer operative times than those in group A. The follow-up period ranged from 8 months to 1.5 year (median, 1 year). During the follow-up period, late-onset complete atrioventricular block occurred in two patients. No other serious complications were noted in the remaining patients. Conclusion: Both procedures are safe and effective treatments for isolated VSD. The percutaneous procedure has obvious advantages of shorter ICU stay and less trauma than the perventricular procedure. However, the perventricular procedure is simpler to execute, results in a shorter operative time, and avoids X-ray exposure.

·AIM:To investigate the clinical effect of scleral buckling with nondrainage of subretinal fluid in the treatment of rhegmatogenous retinal detachment.·METHODS: A total of 111 patients ( 113 eyes ) with rhegmatogenous retinal detachment in our hospital from January 2012 to January 2017 were retrospectively reviewed. All the patients were identified as fulfilling the criteria of rhegmatogenous retinal detachment. Localization and scleral cryotherapy of retinal breaks with binocular indirect ophthalmoscopy in all eyes were performed during the operations. Simple scleral pressure were performed in 98 eyes. Intravitreal injection of sterile air were performed in 15 eyes. Scleral cryotherapy were performed in all cases with retinal degeneration and breaks. Anterior chamber puncture were performed before ligation suture. The final retinal reattachment rate, best corrected vision acuity, intraocular pressure and complications were observed.·RESULTS: The mean follow-up period was 6mo after operation. The final retinal reattachment rate at 6mo was 97. 4%. There were 91 eyes (80. 5%) with improved visual acuity after operation. High intraocular pressure can be controlled after operation. Complications were occurred in 7 eyes in the operation, 3 eyes postoperatively.·CONCLUSION: The surgery of scleral buckling with nondrainage of subretinal fluid have few complications and is effective.

This study was aimed to establish ELISA for recombinant bovine IFN-gamma (BovIFN-gamma) detection and provide a new method for diagnosis of pathogenic infection. The total RNA was isolated from peripheral blood leucocytes cultured with PHA mitogen stimulation. Then bovine IFN-gamma (BovIFN-gamma) gene cDNA was amplified by RT-PCR and cloned into pET28a to obtain the expression plasmid designated as pETBovIFN-gamma. The pETBovlFN-gamma was further transformed into competent E. coli BL21 cells and a 18kD His-tagged protein as expected was expressed after IPTG induction. By using purified recombinant BovIFN-gamma as antigen and lymphocyte-hybridoma technique, four hybridoma cell lines which stably secreted monoclonal antibodies against rBovIFN-gamma were generated, designated as A7, A10, G6, and G10. The immunoglobin subset was identified as IgG1 . Western-blotting analysis and ELISA demonstrated that the monoclonal antibodies secreted by all the four hybridoma cell lines could react specifically to the recombinant BovIFN-gamma, but not irrelative proteins such as Ag85B, ESAT-6-CFP-10 and GM-CSF, suggesting that the four hybridoma cell lines were rBovIFN-gamma specific monoclonal antibodies. A sandwich ELISA was established by using A10 secreted monoclonal antibody and rabbit polyclonal antibodies against BovIFN-gamma, HRP labeled goat anti-rabbit IgG. The results indicated that the sensitivity was 2ng/mL. This sandwich ELISA to detect BovIFN-gamma paved the way to develop a sensitive method for specific infection detection such as bovine tuberculosis diagnosis.
