RESUMEN

Pancreatic cancer is among the most malignant cancers, and thus early intervention is the key to better survival outcomes. However, no methods have been derived that can reliably identify early precursors of development into malignancy. Therefore, it is urgent to discover early molecular changes during pancreatic tumorigenesis. As aberrant glycosylation is closely associated with cancer progression, numerous efforts have been made to mine glycosylation changes as biomarkers for diagnosis; however, detailed glycoproteomic information, especially site-specific N-glycosylation changes in pancreatic cancer with and without drug treatment, needs to be further explored. Herein, we used comprehensive solid-phase chemoenzymatic glycoproteomics to analyze glycans, glycosites, and intact glycopeptides in pancreatic cancer cells and patient sera. The profiling of N-glycans in cancer cells revealed an increase in the secreted glycoproteins from the primary tumor of MIA PaCa-2 cells, whereas human sera, which contain many secreted glycoproteins, had significant changes of glycans at their specific glycosites. These results indicated the potential role for tumor-specific glycosylation as disease biomarkers. We also found that AMG-510, a small molecule inhibitor against Kirsten rat sarcoma viral oncogene homolog (KRAS) G12C mutation, profoundly reduced the glycosylation level in MIA PaCa-2 cells, suggesting that KRAS plays a role in the cellular glycosylation process, and thus glycosylation inhibition contributes to the anti-tumor effect of AMG-510.

Objective To calculate the doses and their dose conversion coefficients for the personnel whose organs were under accidental exposure to three types of X-ray machines and two γ radiation sources, and to provide a simple method for rapid estimation of accidental doses. Methods The radiation source models of X-ray machines and two γ sources were established with the FLUKA simulation software and a Chinese reference voxel phantom was imported. The organ absorbed dose, dose conversion coefficient between organ absorbed dose and air Kerma, and conversion coefficient between organ absorbed dose and radiation source were calculated for a simulated scenario where the personnel were under antero-posterior exposure to radiation sources 1 meter away. Results For the lungs, heart, muscles, soft tissue, liver, skin, and brain, the conversion coefficient between organ absorbed dose and air Kerma was 0.30-1.19 (Gy/Gy). For X-ray machines, the conversion coefficient between organ absorbed dose and output for the six organs ranged from 6.12 × 10−3 to 2.90 × 10−2 Gy·m2/(mA·min). For γ radiation sources, the conversion coefficient between organ absorbed dose and activity for the six organs ranged from 1.12 × 10−8 to 7.01 × 10−8 Gy·m2/(GBq·s). Conclusion The conversion coefficient between organ absorbed dose and air Kerma and the conversion coefficient between organ absorbed dose and output or activity of a flaw detector can provide important dosimetric parameters for rapid assessment of similar radiation accidents.

Objective To investigate the association between the interleukin 10 (IL-10) gene promoter region-592A/C (rs1800872) polymorphism and hypertensive disorders of pregnancy (HDP) in Han women of Qinghai province and to determine the expression of this gene in two groups (HDP group and healthy control group) preliminarily. Methods A total of 140 HDP patients (HDP group) and 140 normal pregnant women (control group) in Qinghai Province were selected. Using blood DNA as template, the IL-10-592A/C polymorphism typing of HDP group and control group was detected by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) and verified by sequencing. The expression of IL-10 mRNA in the placental tissues of the two groups was detected by Real-time PCR. Plasma IL-10 levels of the two groups were detected by ELISA. Results The frequencies of AA, AC and CC genotypes of IL-10 gene in HDP group and control group were 24. 29%, 44. 29%, 31. 42% and 13. 57%, 41. 43%, 45. 00% respectively, the difference in genotype distribution between the two groups was statistically significant (P<0. 05);AA genotype frequency in HDP group(24. 29%)was higher than that of control group(13. 57%)(P<0. 05), CC genotype frequency in HDP group (31. 42%) was lower than that in control group (45. 00%) (P < 0. 05), while there was no significant difference in genotype frequency of AC between the two groups (P<0. 05); The distribution of A and C allele frequencies of IL-10592A/C polymorphism was different between the two groups, and the A allele frequency of HDP group was higher than that of control group (

[ Abstract] Objective To investigate the relationship between single nucleotide polymorphism (SNP) Fok (rs2228570 / rs10735810) of vitamin D receptor (VDR) gene and hypertensive disorder complicating pregnancy (HDCP) in Han nationality women of Qinghai province. Methods A total of 137 Han nationality HDCP subjects (HDCP group) and 146 Han nationality normal pregnant subjects (control group) were selected from Qinghai province. The Fok polymorphism typing in HCDP group and control group was analyzed by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) . The mutation was confirmed by sequencing. SPSS 19. 0 statistical software was used to test whether there were significant differences between two groups in general clinical data, genotype and allele frequency distribution. Results The frequency of FF Ff ff genotype of Fok in HDCP group and control group were 51. 82%, 37. 96%, 10. 22% and 34. 93%, 43. 15%, 21. 92% respectively (

Objective To compare the measurement differences between the skull 3D printed model and the real specimen under different CT scan slice thicknesses, and to explore the effect of slice thickness on the accuracy of the 3D printed model. Methods Eight normal skull specimens (marked as Nos. f-8) (group N) were used for CT scanning with different slice thicknesses, specifically 0.625 mm (group A),1.25 mm (group B) , and 2.5mm (group C) ,3.75 mm (group D) , and 5 mm (group E) , and then earned out 3D reconstruction and 3D printing respectively, and compared the anatomical reduction degree of the foramen magnum diameter, anterior clinoid distance, and butterfly wing distance of the 3D printed skull model. Results The reduction degree of anatomical structure of 3D printed skull model decreased with the increase of CT slice thickness. There was no significant difference in the accuracy of 3D model among groups A, B and C (P >0.05 ) . There was a high correlation between group A, B and C and group N ( P < 0 .05 ).The size indexes and statistical values of group A, B and C were similar. Conclusion CT slice thickness has a significant effect on the accuracy and reduction of the 3D printed skull model. The 3D printed model with thin slice data (0.625 mm,1.25 mm,2.5 mm) has higher accuracy and less difference.

Objective To evaluate the effect of mesenchymal stem cell (MSC) coated-islets on instant blood-mediated inflammatory reaction (IBMIR) after islet transplantation. Methods MSC labeled with tracer and human islets were placed into an ultra-low adsorption cell culture dish, shaken and mixed twice at an interval of 0.5 h, and then incubated at 37 ℃ and 5% CO2 for 24 h to obtain MSC-coated islets. The coating effect of MSC and in vitro function of the islets were assessed. A blood circulation tube-shaped model was established in vitro. In the blank control group, 0.2 mL of islet culture solution was added. In the islet group, 800 islet equivalent quantity (IEQ) of uncoated islets were supplemented. In the MSC-coated islets group, 800 IEQ of MSC-coated islets were added, and circulated for 60 min at 37 ℃. A portion of 0.5 mL blood sample was taken for routine blood test at 0, 30 and 60 min, respectively. After 60 min circulation, the blood sample was filtered with a 70 μm filter to collect plasma, blood clots and islets. Blood clots and islets were subject to hematoxylin-eosin (HE) staining and immunohistochemical staining. Morphological changes and the aggregation of CD11b-positive cells surrounding the islets were observed. The contents of plasma thrombin-antithrombin complex (TAT), tissue factor (TF), C3a, C5b-9, interleukin (IL)-1β, IL-6, tumor necrosis factor (TNF)-α, monocyte chemoattractant protein (MCP)-1 and IL-8 were determined by enzyme-linked immune absorbent assay. Results After 24 h co-incubation, the islets were coated by MSC, with a coating degree of approximately 80%. In the islet and MSC-coated islet group, a large quantity of neutrophils and monocytes were observed surrounding the blood clots and islets, and the quantity of CD11b-positive cells in the MSC-coated islet group was less compared with that in the islet group. After co-incubation with the whole blood for 0, 30 and 60 min, the quantity of platelets, neutrophils and monocytes was declined in the MSC-coated and islet groups, and gradually decreased over time. Compared with the blank control group, the quantity of platelets, monocytes and neutrophils was lower, whereas the TF content was higher in the MSC-coated islet group. Compared with the islet group, the quantity of platelets, monocytes and neutrophils was higher, whereas the TAT and TF contents were less in the MSC-coated islet group, the differences were statistically significant (all P < 0.05). Compared with the blank control group, the expression levels of C3a, C5b-9, IL-6, TNF-α and IL-8 were up-regulated in the MSC-coated islet group. Compared with the islet group, the expression levels of C3a, C5b-9, IL-1β, IL-6, TNF-α, IL-8 and MCP-1 were down-regulated in the MSC-coated islet group, and the differences were statistically significant (all P < 0.05). Conclusions MSC-coated islets may reduce the exposure of islet TF in the blood and prevent the incidence of IBMIR during the coagulation response stage, thereby mitigating the injury and loss of islet allograft in the early stage of islet transplantation.

Objective To investigate the correlation between the polymorphism of 5,10-methylenetetrahydrofolate reductase ( MTHFR) gene rs!801131 and hypertensive disorders complicating pregnancy ( HDCP ) in Qinghai Han nationality. Methods The polymorphism of MTHFR rsl801131 in 120 pregnant women with HDCP (HDCP group) and 120 normal pregnant women ( control group) were detected by restriction fragment length polymorphism polymerase chain reaction (PCR-RFLP) and verified by sequencing. Results The frequencies of AA, AC, and CC genotype of MTHFR gene in the HDCP group were 56. 67% , 32. 50% , and 10. 83% respectively, and those in the control group were 74.17%, 23.33% and 2. 50% respectively (P<0. 05, the distribution of genotype was different significantly between the two groups). The frequency of AA genotype of HDCP group (56. 67%) was lower than that of control group (74. 17%, P<0. 05) , the frequency of CC genotype of HDCP group ( 10. 83%) was higher than that of control group ( 2. 50% , P< 0. 05) , while there was no significant difference in the frequency of AC genotype between HDCP group and control group ( P<0. 05). The frequency distribution of alleles A and C of MTHFR rsl801131 polymorphism was significantly different between the HDCP group and the control group (P<0. 001) , and the frequency of allele C in the HDCP group was significantly higher than that in the control group (X2 = 12. 229, 0R=L 574, 95% C/= 1. 181-2. 099, P<0. 001). Conclusion The polymorphism of MTHFR rsl801131 is related to the occurrence of HDCP in Qinghai Han population. The C gene might be the susceptibility gene of HDCP, and CC genotype might be the susceptibility genotype of HDCP.

Objective To explore the somatotype characteristics and rules of Lisu and Dulong adolescents aged 13-18 years, and to analyze their annual somatotype differences. Methods According to the method of human bod)' measurement, through the measurement of 10 related indexes, using Heath-Carter bod)' type evaluation method, 634 cases (328 boys, 306 girls) Lisu and Dulong teenagers bod)' type evaluation. Results The average bod)' size of male and female students of Lisu ethnic group was of balanced mesoderm type (2. 9-4. 8-3. 3) and endoderm type (4. 7-4. 8-2. 8) . The average body size of male and female students of Dulong nationality is mesoderm type (3 . 1-3. 9-2. 1) and mesoderm type (5 . 2 - 4 . 6 - 1 . 6) . Among the 10 test indexes, there were significant differences in height, body weight, intercondylar diameter of humerus and intercondylar diameter of femur (P < 0. 01) . There were significant differences in height, intercondylar diameter of humerus and intercondylar diameter of femur among female students (P<0. 0 1) . Conclusion In Yunnan Province, adolescents of Lisu and Dulong ethnic groups generally have the body type characteristics of southern ethnic groups, but the body type difference between the two ethnic groups is great, and only has the characteristics of similar stage, age and body type.

RESUMEN

Gut microbiota is a bridge between the metabolism and health state of the host,which plays a very important role in maintaining homeostasis. Natural polysaccharides,widely existed in nature,are a kind of biological macromolecules with prebiotics effects,which can improve a degree of physiological status by selectively changing the gut microbiota structure and function,enhancing the content of short chain fatty acids(SCFAs)and decreasing the level of inflammatory cytokines. In addition,the majority of polysaccharides can be degraded by gut microbiota to enhance their bioavailability and to promote the health state of the host. In this paper we discuss the interaction among polysaccharides and gut microbiotanatural,degradation mechanism and review gut microbiota as a target in the treatment of metabolic diseases,so as to provide future prospects of natural polysaccharides as " prebiotics " functional factors in the field of biological medicine and health products.

Objective:To explore the clinical features and treatment strategies of the transsyndesmotic ankle fracture dislocation.Methods:Data of 26 patients of transsyndesmotic ankle fracture dislocation who were treated in our hospital from December 2013 to November 2020 were retrospectively analyzed. There were 16 men and 10 women with an average age of 49.54±12.81 years (range, 26-68 years). Open injuries in 17 cases, of which the Gustilo-Anderson II type in 6 cases, IIIA type in 11 cases, closed injuries in 9 cases. According to the AO/OTA fracture classification, 44B type in 4 cases, 44C type in 22 cases. According to the Lauge-Hansen classification, there were 16 cases of pronation-abduction, 10 cases of pronation-external rotation, including 4 cases of Maisonneuve fractures, and of the 4 cases of Maisonneuve fractures, there were 3 cases of double Maisonneuve fracture. The talar dislocation was anterior, neutral, and posterior within the distal tibiofibular joint in 10 cases, 7 cases, and 9 cases. Fibular fractures in 26 cases, medial malleolar fractures in 24 cases, deltoid ligament rupture in 2 cases, posterior malleolar fractures in 13 cases, and anterior malleolar fractures in 8 cases. All closed injuries were closed reduction and plaster fixation and all open injuries were emergently debridement and reduced under the tibial plafond in the emergency department. Surgical treatment was taken until the soft tissue conditions to be allowed. The reduction quality was assessed by postoperative radiography according to the criteria proposed by Burwell-Charnley. The function of the ankle joint was assessed by the ankle-hindfoot rating system of the American Orthopaedic Foot and Ankle Society (AOFAS), and the posttraumatic arthritis and objectively quantified was assessed using the Kellgren-Lawrence grading scale.Results:There were 4 cases were unreduced due to the tibial posterior tendon to flip through the ankle joint and dislocate anterior to the tibia through the interosseous membrane. Stabilization of fibular fractures were achieved with plate in 25 cases. There were 24 cases of medial malleolar fractures, and the fixation were achieved with cannulated screws in 23 cases and with K-wire fixation in 1 case. There were 12 cases of posterior malleolar fractures treated with open reduction and internal fixation including cannulated screws in 9 cases and antiglide plates in 3 cases. There were 7 cases of anterior malleolar fractures treated with open reduction and internal fixation including suture anchors in 1 case and cannulated screws in 6 cases. Stabilization of syndesmosis was achieved with syndesmotic screws in 14 cases and with TightRope in 2 cases. All patients were followed up for 20.23±9.70 months (range, 12-60 months). According to the Burwell-Charnley criteria of reduction quality, anatomic reduction was obtained in 22 cases, and satisfactory reduction was gained in 4 cases. All fractures healed in 16.31±3.64 weeks (range, 10-24 weeks). Functional examination of the ankle joint (angle measurement method): dorsiflexion average angle 10.38°±6.66°, plantarflexion average angle 34.04°±7.20°. At latest follow up, the AOFAS score was 83.30±13.94 (range, 24-100). Ten (38%) of 26 patients had radiographic evidence of posttraumatic ankle arthritis. According to the Kellgren-Lawrence grading scale criteria, there were grade I in 5 cases, II in 2 cases, III in 2 cases, and IV in 1 case. 2 cases of wound dishence were recovered through changing dressing and 2 cases of skin necrosis were recovered by skin graft and flap transposition respectively. There were no significant complications such as infection, nonunion, or implant failure.Conclusion:The transsyndesmotic ankle fracture dislocation, represents an exceptional pattern of high-energy fractures with significant syndesmotic disruption, and potential soft tissue compromise. Careful attention to radiographic findings can identify unique fracture characteristics relative to operative decision-making. Tibialis posterior tendon dislocation, a rare complication in the transsyndesmotic ankle fracture dislocation injuries, can impede anatomical reduction of the ankle mortise. The open reduction and internal fixation may be an optimal approach to treat transsyndesmotic ankle fracture dislocation injuries. However, the rate of posttraumatic arthritis is relatively high.

Objective:To explore the changes of peripheral blood Th17/Treg and serum cytokines in AECOPD patients with secondary pulmonary fungal infection.Methods:Selected the clinical data of 27 AECOPD patients who were admitted between January 2018 to March 2020 in the Department of Respiratory Medicine, Hai'an People's Hospital Affiliated to Nantong University with fungal infection (fungal infection group), and 58 AECOPD patients without fungal infection (non-fungal infection group) who received treatment in the hospital during the same period. Compared the general clinical data, peripheral blood Th17 and Treg cell levels, Th17/Treg ratio, interleukin-17 (IL-17), interleukin-23 (IL-23), interferon-γ (interferon-γ, IFN-γ), and transforming growth factor-β (TGF-β) levels. Meanwhile, compared the levels of Th17 and Treg cells in peripheral blood, the ratio of Th17/Treg and serum cytokines in patients with different infection severity in fungal infection group. The measurement data with normal distribution were compared by independent samplet t-test between the two groups, one-way ANOVA between multiple groups, LSD-t test for pairwise comparision, and χ 2 test for counting data. Results:In the 27 AECOPD patients with fungal infection group, the pathogen distribution was 65.52% (19/27) of candida albicans, 10.34% (3/27) of candida tropicalis,10.34% (3/27) of candida albicans, and 6.90% (2/27) of Aspergillus. The level of Th17 [(16.18±3.15) % and (12.34±2.64) %, t=5.87, P<0.001)], the ratio of Th17/Treg [(4.70±0.85) and (2.41±0.51), t=22.87, P<0.001] in Patients with fungal infection group were higher than those in the non-fungal group. The level of Treg [(3.42±0.42) % and (5.13±0.51) %, t=20.77, P<0.001] in Patients with fungal infection group was lower than those in the non-fungal group. The levels of IL-17 [(85.67±21.51) μg/L and (53.64±14.36) μg/L, t= 8.12, P<0.001], and IL-23 [(61.38±16.58) μg/L and (38.29±12.60) μg/L, t=7.10, P<0.001] in Patients with fungal infection group were higher than those in non-fungal infection group, but the levels of IFN-γ ((47.75±17.72) μg/L and (62.37±19.06) μg/L, t=3.37, P=0.001) and TGF-β ((110.34±26.03) μg/L and (131.40±35.03) μg/L, t=2.87, P=0.007) were lower than those in non-fungal infection group, and the differences were statistically significant. There were statistically significant differences in the ratio of Th17/Treg, and the levels of Th17, Treg cells and cytokine among patients with different infection severity in the fungal infection group. With the increase of infection severity, the levels of Th17 ((13.06±1.98)%, (15.94±2.11)%, (17.75±2.20)%, F=10.19, P<0.001), the ratios of Th17/Treg ((5.01±0.60), (5.66±0.69), (6.52±0.65), F=10.77, P<0.001), the levels of IL-17 ((63.39±11.64) μg/L,(78.66±12.82) μg/L, F=9.01, P=0.001), and IL-23 ((42.52±13.11) μg/L, (59.97±15.25) μg/L, (69.75±14.30) μg/L, F=7.41, P=0.003) were increase, the levels of Treg ((4.33±0.39)%, (3.32±0.42)%, (2.50±0.35)%, F=44.42, P<0.001), IFN-γ ((57.78±10.52) μg/L, (48.82±10.39) μg/L, (38.90±10.56) μg/L, F=6.50, P=0.006), TGF-β ((126.62±18.94) μg/L, (115.34±13.66) μg/L, (102.52±17.73) μg/L, F=4.25, P=0.026) were significantly decreased. Conclusion:The imbalance of Th17/Treg ratio and related serum cytokines play an important role in the process of lung fungal infection in AECOPD patients, and their imbalance is related to the severity of fungal infection. Therefore, the levels of Th17/Treg and serum cytokines should be closely monitored in AECOPD patients.

@#AIM: To investigate the difference of the optic vessel density in early primary open angle glaucoma(POAG), ocular high tension subjects and healthy subjects. <p>METHODS: A cross-sectional study. From January 2019 to April 2021, patients were selected in the Department of Glaucoma in Fuzhou Southeast Eye Hospital. There were 45 patients(70 eyes)of early POAG group, including 32 males(49 eyes)and 13 females(21 eyes)with age of 48.50(26.75, 64.50)years. There were 37 patients(65 eyes)of ocular high tension group, including 17 males(29 eyes)and 20 females(36 eyes)with age of 37.00(27.00, 47.00)years. There were 51 patients(94 eyes)of healthy group, including 23 males(39 eyes)and 28 females(55 eyes)with age of 46.00(34.50, 56.50)years. Ophthalmic examinations including best-corrected visual acuity(BCVA), intraocular pressure, central corneal thickness(CCT), mean deviations(MD), retinal nerve fiber layers thickness(RNFL)were measured in three groups. The center area, the inner circle area, the outer circle area, and the whole area of the optic vessel density in three groups were measured <i>via</i> optical coherence tomography angiography(OCTA)device.<p>RESULTS: The difference of the intraocular pressure in early POAG group-ocular high tension group was not significant(<i>H</i>=146.876,<i>P</i><0.001), while significantly in early POAG group-healthy group and ocular high tension group-healthy group(all <i>P</i><0.01). Significant differences in BCVA, MD, and RNFL were observed in early POAG group-ocular high tension group(<i>P</i>=0.005, <i>P</i>=0.01, <i>P</i><0.01)and early POAG group-healthy group(<i>P</i>=0.013, <i>P</i><0.01, <i>P</i><0.01), while the difference was not found in ocular high tension group-healthy group(<i>P</i>=1.000, <i>P</i>=0.660, <i>P</i>=1.000). There was no difference of CCT in early POAG group-healthy group(<i>P</i>=0.074), and significant differences were observed in early POAG group-ocular high tension group and ocular high tension group-healthy group(<i>P</i>=0.006, <i>P</i><0.01). The optic vessel density comparison in the center area, the inner circle area, and the whole area were significantly different in early POAG group-ocular high tension group(all <i>P</i><0.01)and in early POAG group-healthy group(all <i>P</i><0.01), which indicated that the optic vessel density was obviously less in the early POAG group than the other two groups. The optic vessel density had no significant difference in ocular high tension group-healthy group in these areas(all <i>P</i>=1.000). In the outer circle area, there were no significant differences of the optic vessel density in early POAG group-ocular high tension group and early POAG group-healthy group(<i>P</i>=0.067, <i>P</i>=0.877), while significant difference was observed in early POAG group-healthy group(<i>P</i>=0.001).<p>CONCLUSION: The optic vessel density was decreased in early POAG comparing with ocular high tension subjects and healthy subjects, which was agree with the changes of MD, RNFL. The optic vessel density decreased firstly from the center area and the inner circle area for early POAG.

Objective To investigate the effect of compound Fufangteng mixture-containing serum on the proliferation of bone marrow mesenchymal stem cell (BMSC) and its mechanism. Methods Rat BMSC were isolated, cultured and purified in vitro by direct adherence method. Cell morphology was observed. Surface markers were identified by flow cytometry. The rats were treated with compound Fufangteng mixture at a dose of 3 mL/(kg·d) by gavage for 14 d, and then the drug-containing serum was collected. BMSC were divided into the blank control group, drug-containing serum group, Notch1 small interfering ribonucleic acid (siRNA) group and Notch1 siRNA+drug-containing serum group. The proliferation rate of BMSC was detected and the relative expression levels of Notch1 signaling pathway-associated messenger ribonucleic acid (mRNA) and proteins were measured in each group. Results Microscopic observation showed that the first generation BMSC were seen in the long spindle shape, and grown in the parallel or spiral pattern. The third generation BMSC positively expressed CD90 and CD44, whereas were negative for CD45. Compared with the blank control group, the proliferation rate of BMSC in the drug-containing serum group and Notch1 siRNA+ drug-containing serum group was significantly increased, whereas that of BMSC was significantly decreased in the Notch1 siRNA group (all P < 0.05). Compared with the Notch1 siRNA group, the proliferation rate of BMSC was significantly increased in the Notch1 siRNA+drug-containing serum group (P < 0.05). Compared with the blank control group, the relative expression levels of Hey1 and Delta-like ligand (DLL)1 mRNA and proteins were significantly up-regulated in the drug-containing serum group, whereas those were significantly down-regulated in the Notch1 siRNA group and Notch1 siRNA+drug-containing serum group (all P < 0.05). Compared with the Notch1 siRNA group, the relative expression levels of Hey1 and DLL1 mRNA and proteins were significantly up-regulated in the Notch1 siRNA+drug-containing serum group (all P < 0.05). Conclusions Compound Fufangteng mixture-containing serum may promote the proliferation of rat BMSC, and its mechanism is probably associated with the activation of Notch1 signaling pathway.

Objective To establish a sourceless efficiency calibration method by coupling Monte Carlo simulation with analytical calculation. Methods Monte Carlo simulation was used to calculate the point-to-point detection efficiency of specific detectors to establish a detection efficiency grid. The detection efficiency of point source, disc, cylindrical, beaker, spherical, U-tube and Marlin cup samples was analyzed using numerical integration method after detection efficiency grid interpolation. Results The above coupling method was used for sourceless efficiency calibration. Within the energy range of 0.2–3 MeV, the relative deviation of calibration between coupling method and Monte Carlo simulation was mostly less than 10%, the maximum relative deviation was 18.06%, and the computation time was reduced by at least 86%. The above coupling method was used for sourceless efficiency calibration of an HPGe detector manufactured by ORTEC for point source detection, which was in good agreement with the experimental calibration, and the relative deviations were less than 10%. Conclusion This method can be generalized and used in the sourceless efficiency calibration of HPGe, LaBr3, and NaI detectors.

OBJECTIVE@#To explore if acupoint injection can improve analgesic effects or delivery outcomes in parturients who received combined spinal-epidural analgesia (CSEA) and patient-controlled epidural analgesia (PCEA) for labor analgesia.@*METHODS@#A total of 307 participants were prospectively collected from July 2017 to December 2019. The participants were randomized into the combined acupoint injection with CSEA plus PCEA group (AICP group, n=168) and CSEA plus PCEA group (CP group, n=139) for labor analgesia using a random number table. Both groups received CSEA plus PCEA at cervical dilation 3 cm during labor process, and parturients of the AICP group were implemented acupoint injection for which bilateral acupoint of Zusanli (ST 36) and Sanyinjiao (SP 6) were selected in addition. The primary outcome was Visual Analogue Scale (VAS) score, and the secondary outcomes were obstetric outcomes and requirement of anesthetics doses. Safety evaluations were performed after intervention.@*RESULTS@#The VAS scores were significantly lower in the AICP group than in the CP group at 10, 30, 60, and 120 min after labor analgesia (all P<0.05). The latent phase of the AICP group was shorter than that of the CP group (P<0.05). There were less additional anesthetics consumption, lower incidences of uterine atony, fever, pruritus and urinary retention in the AICP group than those in the CP group (all P<0.05).@*CONCLUSION@#Acupoint injection combined CSEA plus PCEA for labor analgesia can decrease the anesthetic consumption, improve analgesic quality, and reduce adverse reactions in the parturients. Long non-coding RNAs (LncRNAs) , as regulators of a class of gene expression, play a key role in the development of various types of tumor.We analyzed the TCGA database and found that the expression of LncRNA AC009686.2 in breast cancer tissues was significantly higher than that in normal tissues, and was positively correlated with the poor prognosis of breast cancer patients.qRT-PCR analysis showed that the expression of LncRNA AC009686.2 in breast cancer cells was significantly up-regulated, and the expression level of LncRNA AC009686.2 in MCF7, T47D, ZR7530, BT549, HCC1937, MDA- MB-231 and SKBR3 eells was 6.58, 5.66, 7.29, 9.06, 6.89, 11.17 and 5.38 folds of that in MCF10 A eells, respectively.LncRNA AC009686.2 knockdown in MDA-MB-231 and BT549 cells which expressed relatively high LncRNA AC009686.2 significantly inhibited cell proliferation, colony formation and invasion, and induced cell G,/S phase arrest.The clone inhibition rates of MDA-MB-231 and BT549 cells with LncRNA AC009686.2 knockdown were 0.496%, 0.438% and 0.495%, 0.353% of the control group, respectively.LncRNA AC009686.2 knockdown also down-regulated protein levels of cyclinD2 and ZEB1.However, overexpression of ZEB1 could significantly reverse the decrease of cell invasion ability caused by LncRNA AC009686.2 knockdown.We further analvsed in the software JASPAR database and found that LncRNA AC009686.2 promoter had ZEB1 binding site, and overexpression of ZEB1 could down-regulate the expression level of LncRNA AC009686.2 in breast cancer cells.In conclusion, LncRNA AC009686.2 which highly expressed in breast cancer, promotes cell proliferation and invasion by up-regulating cyclinD2 and ZEB1 expression, while ZEB1 positively regulates LncRNA AC009686.2 expression.This study will provide a theoretical basis for elucidating the role of LncRNA AC009686.2 in breast cancer and related molecular mechanisms.

RESUMEN

Objective:To estimate the physical dose of an over exposed person working for industrial radiography.Methods:The main exposure parameters were obtained. The exposure duration was 8 min. The exposure pattern was external exposure by isotropic point radioactive source. The radioactive activity was 2.183 TBq. In the present calculation, the Chinese reference adult voxel phantom was used, and the Monte Carlo simulation was performed using the program based on the secondary development of Geant4 to obtain the absorbed dose of each part of the victim.Results:The dose distribution in the victim′s hands was obtained. The doses to most areas of the palm were 2-10 Gy, and the doses to the fingers were 10-20 Gy. The equivalent doses to 23 tissues or organs of the exposed person were estimated to be in the range of 0.012-0.207 Gy.Conclusions:The physical dose estimation method could evaluate rapidly the local dose distribution of the victim′s key exposed body parts, and thus provide an important reference for medical treatment.

Objective:The Chinese pediatric mesh-type reference phantoms were developed and applied in the dose assessment of X-ray radiography.Methods:The 5- and 10-year-old Chinese pediatric mesh-type reference phantoms were developed based on the CT data. Based on the phantoms, the Monte Carlo method was applied to simulate the X-ray radiography to calculate the organ dose conversion coefficient and the effective dose conversion coefficient in different radiography conditions.Results:The 5- and 10-year-old Chinese pediatric mesh-type reference phantoms were developed, and the physical parameters were consistent with the national standard. The differences of the organ mass between the established phantoms and reference data were within 2%. The database of the pediatric chest posteroanterior projection and abdominal anteroposterior projection, tube voltage 60-90 kVp, total filtration 2.5~4 mmAl were simulated, and the difference between the effective dose conversion factor and the literature result was within 3%.Conclusions:The established Chinese pediatric mesh-type reference phantoms can be applied in the studies of radiation protection and clinical medicine and their result can provide an important reference for the dose assessment of the pediatric X-ray radiography.

Objective:To investigate the efficacy of modified Zhigancao decoction on Qi/ Yin-deficiency acute viral myocarditis and its effect on serum level of high mobility group box chromosomal protein 1 (HMGB-1). Methods:Eighty-eight patients with Qi/ Yin-deficiency acute viral myocarditis who received treatment in Chun'an Hospital of Traditional Chinese Medicine from July 2017 to December 2018 were included in this study. They were randomly assigned to undergo either standard conventional intervention (control group, n = 43) or treatment with modified Zhigancao decoction based on standard conventional intervention (observation group, n = 45) for 4 weeks. After treatment, serum levels of cardiac troponin I (cTnI) and HMGB1, traditional Chinese medicine symptom score, and clinical efficacy were compared between the two groups. Results:After 2 and 4 weeks of treatment, serum level of cTnI in the observation group was (590.22 ± 78.42) ng/L and (461.36 ± 52.44) ng/L, respectively, which was concurrently significantly lower than that in the control group, respectively [(621.41 ± 86.12) ng/L, (587.62 ± 67.94) ng/L, ( t2 weeks = 1.574, t4 weeks = 9.785, both P < 0.05). After treatment, traditional Chinese medicine symptom score in the observation group was significantly lower than that in the control group ( tfever = 27.650, tsore throat = 27.210, tpalpitation = 14.560, tvexation and chest tightness = 15.370, all P < 0.01). Total effective rate in the observation group was significantly higher than that in the control group [93.33% (42/45) vs. 72.09% (32/43), χ2 = 8.41, P < 0.05). After 2 and 4 weeks of treatment, serum level of HMGB1 in the observation group was concurrently significantly lower than that in the control group ( t2 weeks = 9.144, t4 weeks = 11.534, both P < 0.01). Conclusion:Modified Zhigancao decoction combined with conventional western medicine intervention for the treatment of Qi/ Yin-deficiency acute viral myocarditis exhibits great therapeutic effects possibly through downregulating serum level of HMGB1.
