RESUMEN

【Objective】 To investigate the inhibitory effect of magnesium citrate (MgCit) on hyperphosphorus-induced VSMCs calcification and its mechanisms. 【Methods】 VSMCs were divided into the following groups: normal control group, high-phosphorus group, low-dose MgCit group, high-dose MgCit group and high-dose MgCit+NPS2143 (calcium-sensitive receptor inhibitor) group. Alizarin red staining was used for semi-quantitative analysis of VSMCs calcification and the calcium content in VSMCs was detected by the commercial kit. The osteogenic transdifferentiation parameters including alkaline phosphatase (ALP) activity, as well as mRNA and protein levels of smooth muscle 22α (SM22α), runt-related transcription factor 2 (RUNX2) and bone morphogenetic protein 2 (BMP2), were detected in each group. 【Results】 Compared with those in the control group, the calcium content of VSMCs in the model group was increased, ALP activity, and the mRNA and protein expressions of RUNX2 and BMP2 were increased, and the mRNA expression of SM22α was decreased (P<0.05). MgCit could reduce VSMCs calcification, decrease ALP activity, decrease mRNA and protein levels of RUNX2 and BMP2, and increase mRNA levels of SM22α induced by high-phosphorous (P<0.05). The above effects of MgCit were reduced when NPS2143 was administered simultaneously (P<0.05). 【Conclusion】 MgCit can reduce VSMCs calcification and osteogenic transdifferentiation induced by high phosphorus through activating of CaSR.

【Objective】 To explore the correlation between serum trace elements and systolic and diastolic blood pressure in healthy population and patients with hypertension. 【Methods】 The health examination data of the subjects from our hospital from September 2018 to May 2021 were selected. They consisted of 3430 healthy people with no history of chronic diseases and 216 patients with hypertension previously diagnosed. We measured the levels of serum zinc, iron, calcium, magnesium, lead, copper and cadmium and analyzed their correlation with systolic blood pressure (SBP) and diastolic blood pressure (DBP). 【Results】 The incidence of low zinc (9.5%) and hypomagnesemia (1.8%) was relatively low, while hypomagnesemia (27.8%) was relatively common in the healthy population. Logistic regression analysis showed that age, BMI and serum albumin were associated with increased SBP and DBP. Fasting blood glucose (OR=1.095, 95% CI: 1.007-1.191) and blood lead (OR=1.006, 95% CI: 1.000-1.012) were risk factors for SBP. Serum total cholesterol (OR=1.244, 95% CI: 1.095-1.412), serum iron (OR=1.275, 95% CI: 1.114-1.460) and blood lead (OR=1.010, 95% CI: 1.004-1.015) were risk factors, while serum magnesium (OR=0.488, 95% CI: 0.266-0.894) acted as a protective factor for DBP. After adjusting for age, gender, BMI, waist-hip ratio, and smoking history, there was no significant difference in serum trace element levels between the hypertension and healthy control groups. 【Conclusion】 SBP is correlated with serum lead, while DBP is positively correlated with serum iron and lead, but negatively correlated with serum magnesium in normal healthy population. There was no significant difference in serum trace element content between hypertensive patients and healthy control group, but the conclusion still needs to be further verified.

【Objective】 To explore the potential mechanism of Salvia miltiorrhiza and its active monomers in patients with coronary heart disease (CHD) complicated with depression or anxiety disorder based on network pharmacology. 【Methods】 We collected the clinical data of 147 patients with CHD admitted to our hospital from March to April 2021 and investigated biochemical examination, drug treatment and the use of Chinese herbal compound. The downstream targets of active monomers of Salvia miltiorrhiza were screened based on TCMSP, PubChem and SwissTargetPrediction databases, and depression or anxiety-related genes were mining in OMIM, GeneCards and TTD databases. Protein-protein interaction network and GO and KEGG gene enrichment analyses were performed by matching Chinese herbal compound with disease targets. Chinese herbal compound-target-disease network was built by Cytoscape. 【Results】 Shuxuening (44.9%), Salviae miltiorrhiza (27.9%), and Sofren injection (21.8%) were the most commonly used Chinese herbal or injections in the clinical treatment of CHD, and there was no difference in the frequency of use between genders. Network pharmacology showed that the active components of Salvia miltiorrhiza might act on neuroactive ligand-receptor binding, 5-hydroxytryptamine synapses and dopaminergic synapses and participate in the regulation of neurotransmitter transport, dopamine and catecholamine metabolism and stress response. Target-disease network analysis showed that many active components of Salvia miltiorrhiza interacted with catechol-O-methyltransferase, monoamine oxidase, 5-hydroxytryptamine receptor and opioid receptor, which may be the potential targets of Salvia miltiorrhiza in improving depression and anxiety. 【Conclusion】 Salvia miltiorrhiza is involved in the regulation of neurotransmitters and synaptic activities, thus making it a potential drug for the clinical treatment of CHD complicated with depression and anxiety disorder.

【Objective】 To explore the correlation of the intake of different nutrients with the levels of serum lipids and uric acid. 【Methods】 Based on the data of China Health and Nutrition Survey (CHNS2009), a total of 1 343 individuals were finally included by ID matching with demographic data, serological tests and dietary nutrition results. We compared serum lipids and uric acid levels between groups of different nutrients intake (group Q1-Q4) and conducted the correlation analysis. 【Results】 The total cholesterol (TC), triglyceride (TG), and low-density lipoprotein (LDL-C) in the men were significantly higher than those in the women, while high-density lipoprotein (HDL-C) was significantly lower in the men. The average intakes of daily energy, carbohydrate, fat and protein were significantly higher in the males than in the females. Intergroup comparisons and correlation analyses showed that total energy and protein intakes were positively correlated with TG (r=0.070 9,P=0.008 4;r=0.051 7,P=0.012 3), while fat intake was positively correlated with TC (r=0.086 1,P=0.001 4). The blood uric acid level was positively correlated with the intake of fat and protein (r=0.127 3,P<0.001;r=0.138 0,P<0.001). The results of subgroup analysis of the males were consistent with those of the whole population. However, there was a significantly negative correlation between carbohydrate intake and blood uric acid level in the females (r=-0.107 9,P=0.002 2). Multiple linear regression analysis showed that TC, TG and uric acid were correlated with gender, age and intake of different nutrients. 【Conclusion】 The levels of blood uric acid and lipids are significantly higher in men than in women. Different types of dyslipidemia and hyperuricemia are related to different nutrient intakes in different genders and ages.

Objective:To investigate and analyze the distribution of pathogens and to explore the related risk factors of infections in burn patients in order to provide theoretical basis for effective prevention and treatment of wound infections in burn patients.Methods:A total of 245 burn patients, admitted to the PLA 926 Hospital from January 2016 to December 2020, were selected. The pathogens and antimicrobial susceptibility of the wound secretions were tested and the clinical data of burn patients were collected. The risk factors of burn wound infection were screened by single factor comparison method and multivariate logistic regression.Results:Among the 245 burn patients, 184 patients were with bacterial infections, the infection rate was 75.10% (184/245), and 79 patients were multi-drug resistant, accounting for 42.93%(79/184). A total of 367 strains of pathogens were isolated from wound secretions in the 184 patients, among which 72 (19.62%, 72/367) strains were gram-positive bacteria, mainly Staphylococcus aureus (11.44%, 42/367), 283 (77.11%, 283/367) strains were gram-negative bacteria, mainly Pseudomonas aeruginosa (20.16%, 74/367) and Acinetobacter baumannii (19.89%, 73/367), and 12 (3.27%, 12/367) strains were fungi, mainly Aspergillus (1.91%, 7/367). The results of drug sensitivity tests showed that the pathogens of burn patients were highly resistant to common antibiotics. Monofactor analysis showed that age(χ 2=8.629, P=0.013), burn area (χ 2=21.504, P=0.002), wound depth (χ 2=17.139, P=0.000), hypovolemic shock (χ 2=21.112, P=0.000) and length of hospital stay (χ 2=21.967, P=0.000) were the related risk factors for wound infections. Multivariate logistic regression analysis showed that ages below 4 years old or over 69 years old ( OR=8.414,95% CI:2.971-23.830, P=0.000), burn area>30% ( OR=5.672,95% CI:1.029-31.272, P=0.046), Ⅲ degree wound ( OR=4.069,95% CI:1.396-11.858, P=0.010), length of hospital stay≥15 days ( OR=2.593,95% CI:1.091-6.162, P=0.031) were the independent risk factors of wound infections in burn patients. Conclusions:The incidence of wound infection and multi-drug resistance is relatively high in burn patients, and the pathogens are mainly gram-negative bacteria. Age, burn area, burn wound depth and length of hospital stay are the high-risk factors for wound infections, and taking effective preventive measures according to risk factors can reduce the occurrence of wound infections and hospital infections.

【Objective】 To investigate the negative emotions among university students during home quarantine under the epidemic of coronavirus disease 2019 (COVID-19) so as to provide scientific basis for psychological counseling. 【Methods】 We conducted an online questionnaire survey to collect 405 undergraduates’ and postgraduates’ DASS-21 scale scores and responses to epidemic prevention knowledge, and analyzed the university students’ negative emotions during the epidemic and influencing factors. 【Results】 Of the 405 students surveyed, 178 individuals (44.0%) had depression, 171(42.2%) felt anxious, and 119(29.4%) felt stressed. Individuals in the DASS21+ group spent much time on entertainment on the cell phone daily than those in the DASS21- group (P<0.001), and the degree of impact by COVID-19 on daily life was significantly higher (P<0.001). Correlation analysis showed that entertainment time on the cell phone, the severity of COVID-19, and the impact of the epidemic were positively related to DASS-21 scores, with correlation coefficients of 0.231, 0.143 and 0.259, respectively (all P<0.01). Binary logistic regression analysis found that mobile entertainment time for over 5 hours per day (OR=3.370, 95% CI: 1.369-8.294, P=0.008) was a risk factor for negative emotions during quarantine at home. 【Conclusion】 During quarantine at home in the epidemic of COVID-19, nearly half of university students are prone to breed negative emotions, like depression, anxiety or stress, which may be related to long-term mobile phone entertainment.

<p><b>OBJECTIVE</b>To explore the interaction between P311 and transforming growth factor beta 1 (TGF-β1) in murine fibroblasts and its effect on the function of fibroblasts.</p><p><b>METHODS</b>Skin fibroblasts obtained from five neonatal P311 wild-type C57BL/6 mice and P311 gene knock-out C57BL/6 mice were cultured. The second passage of fibroblasts were used in the following experiments. All experiments were repeated for 3 times. (1) The fibroblasts of P311 wild-type mice were divided into blank control group and P311 over-expression group according to the random number table (the same grouping method below), with 36 wells in each group. Fibroblasts in blank control group were transfected with 10 μL control vector, and fibroblasts in P311 over-expression group were transfected with equal efficiency P311 expression adenovirus vector. After being cultured for 48 hours, the mRNA expression level of P311, and the mRNA and protein expression levels of TGF-β1, α-smooth muscle actin (α-SMA), and collagen type I of fibroblasts in both groups were determined with real-time fluorescent quantitative RT-PCR and Western blotting (the same detection methods below), respectively. (2) After cultured reaching the cell density of 80%-90%, the mRNA and protein expression levels of TGF-β1, α-SMA, and collagen type I of the fibroblasts of P311 wild-type mice and P311 gene knock-out mice, with 4 flasks in each type of fibroblasts, were determined. (3) The fibroblasts of P311 wild-type mice were divided into blank control group and 5, 10, 15, 20, and 25 ng/mL TGF-β1 groups after being starved treatment with DMEM medium containing 1% FBS for 3 hours, with 2 flasks in each group. Fibroblasts in blank control group were routinely cultured, while fibroblasts in the latter five groups were treated with 5, 10, 15, 20, and 25 ng/mL TGF-β1, respectively. After being cultured for 48 hours, the mRNA expression levels of P311 in fibroblasts of the six groups were determined. Another fibroblasts of P311 wild-type mice were divided into blank control group and 10 ng/mL TGF-β1 group, with 6 wells in each group, and the protein expression levels of P311 in both groups were determined by immunofluorescence staining. (4) The fibroblasts of P311 wild-type mice were divided into blank control group and 10 ng/mL TGF-β1 group after being starved treatment as above, with 2 flasks in each group, and fibroblasts in blank control group were routinely cultured, while fibroblasts in the latter group were treated with 10 ng/mL TGF-β1. After being cultured for 48 hours, the mRNA and protein expression levels of α-SMA and collagen type Ⅰ were determined. The fibroblasts of P311 gene knock-out mice were grouped and treated as above, and the mRNA and protein expression levels of α-SMA and collagen type I were determined. Data were processed with one-way analysis of variance and t test.</p><p><b>RESULTS</b>(1) The mRNA expression level of P311 of fibroblasts in P311 over-expression group was increased nearly 300 000-fold compared with that in blank control group (t=9.942, P<0.001). The mRNA expression levels of TGF-β1, α-SMA, and collagen type I of fibroblasts in P311 over-expression group, and the protein expression levels of pro-TGF-β1, activated TGF-β1, α-SMA, and collagen type I of fibroblasts in P311 over-expression group were significantly higher than those in blank control group (with t values from 8.192 to 49.090, P values below 0.01). (2) The mRNA expression levels of TGF-β1, α-SMA, and collagen type I in fibroblasts of P311gene knock-out mice were significantly lower than those in fibroblasts of P311 wild-type mice (with t values from 8.157 to 22.270, P values below 0.01). The protein expression levels of pro-TGF-β1, α-SMA, and collagen type I in fibroblasts of P311 gene knock-out mice were significantly lower than those in fibroblasts of P311 wild-type mice (with t values from 2.995 to 12.600, P<0.05 or P<0.01), and the protein expression levels of active TGF-β1 were similar in two types of fibroblasts (t=1.070, P>0.05). (3) The mRNA expression levels of P311 of fibroblasts in blank control group and 5, 10, 15, 20 and 25 ng/mL TGF-β1 groups were 1.28 ± 0.44, 3.61 ± 0.91, 6.64 ± 0.92, 6.58 ± 1.04, 1.79 ± 0.31, 0.16 ± 0.06, respectively. Compared to the mRNA expression level of P311 of fibroblasts in the blank control group, the mRNA expression levels of P311 of fibroblasts in 5 and 20 ng/mL TGF-β1 groups were similar (with t values respectively 2.302 and 0.955, P values above 0.05), while they were significantly higher in 10 and 15 ng/mL TGF-β1 groups (with t values respectively 5.630 and 4.710, P values below 0.001), and they were significantly lower in 25 ng/mL TGF-β1 group (t=2.509, P<0.01). The protein expression level of P311 of fibroblasts in 10 ng/mL group was higher than that in blank control group. (4) The mRNA and protein expression levels of α-SMA and collagen type I of fibroblasts of P311 wild-type mice in 10 ng/mL TGF-β1 group were significantly higher than those in blank control group (with t values from 3.523 to 14.290, P<0.05 or P<0.01). The mRNA and protein expression levels of α-SMA and collagen type I of fibroblasts of P311 gene knock-out mice in 10 ng/mL TGF-β1 group were significantly higher than those in blank control group (with t values from 4.895 to 14.870, P<0.05 or P<0.01).</p><p><b>CONCLUSIONS</b>The interaction between P311 and TGF-β1 in murine fibroblasts exists and it may enhance the differentiation of fibroblasts in combination.</p>

RESUMEN

Objective To investigate of the effect and mechanisman of SERCA2 on the phenotype modulation of HASMCs. Methods HASMCs were starved for 5 days and divided into different groups ,then we observed morphology change of the cells from the microscope and detected a-actin、SERCA2 and p-ERK by Western Blot,cells proliferation was observed by CCK-8 method. Results Compared with the control group,PDGF could reduce a-actin of HASMCs and increased the cells proliferation ability ,TSG could significantly inhibit the effect (P<0.01), PDGF could also significantly inhibit SERCA2 protein and increased the expression p-ERK (P<0.01), while U0126 significantly inhibited the effect (P < 0.01). Conclusion PDGF may induce HASMCs phenotype modulation through the regulation of SERCA2 and p-ERK.

Objective To identify the biological and molecular biological characteristics of SN7 virus isolated from Niviventer Confucianus in Sichuan province. Methods Monoclonal antibody, PRNT and PCR antigenicity analysis and genotyping of SN7 strain were performed. M and S segments of SN7 genome were also cloned and sequenced. The sequences were compared with those of other strains of Hantavirus. Results It was difficult to identify SN7 by using monocloncal antibody, PRNT and PCR. With sequence comparison, we found that strain SN7 had high homology(80.2%~87.1% of M segment and 76.6%~92.0% of S segment) with HTN type strains, and relatively low homology(70.0%~71.6% of M segment and 71.0%~72.2% of S segment) with SEO type strains. Strain SN7 was believed to belong to HTN type. Conclusions SN7 is a new subtype strain of HTN type viruses. It is possible that Hantavirus has immune escape in its natural hosts.
