RESUMEN

Objective: To study using isotope-labeled relative and absolute quantitative proteomics methodologies to screen for salivary biological markers as a simple, non-invasive tool for identifying hepatitis B-related HCC at an early stage. Methods: Saliva samples were collected to extract salivary proteins. Isotope-labeled relative and absolute quantitative proteomics were used to analyze the differentially expressed proteins between the hepatocellular carcinoma (HCC) and non-HCC groups. Western blotting, immunohistochemistry, and enzyme-linked immunosorbent assays were used to verify differential proteins and identify markers in liver cancer tissues and saliva. Statistical analysis was used to analyze the diagnostic efficiency of salivary biomarkers. Results: 152 differentially expressed salivary proteins were screened out between the HCC and non-HCC groups. Western blot, immunohistochemistry, and enzyme-linked immunosorbent assays validated that the expressions of α-1-acid glycoprotein 1 (ORM1) and alpha-fetoprotein (AFP) were significantly increased in HCC (P < 0.05). There was a significant correlation between salivary AFP and serum AFP (P < 0.05). HCC was diagnosed when salivary α-1-acid glycoprotein 1 combined with AFP. The area under the receiver operating characteristic curve was 0.8726 (95% confidence interval: 0.8104 ~ 0.9347), the sensitivity was 78.3%, and the specificity was 88%. Conclusion: Salivary AFP and α-1-acid glycoprotein 1 can serve as potential biomarkers for hepatitis B-related hepatocellular carcinoma.

RESUMEN

Metabolic homeostasis is a basic function necessary for the survival of the organism. α1-acid glycoprotein (AGP) is an acute phase protein with a glycosylation degree of up to 45%. It has high affinity and low capacity. Although the biological role of AGP is not fully understood, it has been proven that it can regulate immunity and metabolism, and play an important role in transporting drugs and maintaining capillary barrier function. In this review, the structural characteristics, biochemical characteristics and the regulation of AGP expression were reviewed, with emphasis on the regulatory role of AGP in metabolism, suggesting that AGP may be a potential key factor in metabolic pathways, which provides a new research direction for metabolic diseases.

Serum protein concentrations, including acute phase proteins (APPs), of goats and ewes with naturally acquired Sthaphylococcus aureus mastitis were determined by means of SDS-PAGE electrophoresis to evaluate the relevance of these APPs as biomarkers of the disease in these species. Fifteen healthy goats and 5 goats with naturally acquired staphylococci mastitis, as well as fifteen healthy ewes and 5 ewes with staphylococci mastitis were submitted to daily blood sampling during 7 days. In goats, an increase of 570%, 125%, 621%, and 279% in serum concentrations of ceruloplasmin, fibrinogen, haptoglobin and α1-acid glycoprotein, respectively, was observed. In sheep the increase in serum concentrations of ceruloplasmin, fibrinogen, haptoglobin and α1-acid glycoprotein was of 337%, 90%, 461%, and 225%, respectively. Our results indicate that these APPs have considerable potencial as early and sensible biomarkers of mastitis caused by S. aureus in goats and sheep.(AU)


O proteinograma, incluindo proteínas de fase aguda (PFAs), de cabras e ovelhas com mastite de origem natural causada por Staphylococcus aureus, foi determinado por meio de eletroforese em gel de poliacrilamida contendo dodecil sulfato de sódio (SDS-PAGE) a fim de avaliar a importância destas PFAs como biomarcadores da enfermidade nestas espécies. Amostras de sangue foram colhidas diariamente de cinco cabras e cinco ovelhas com mastite estafilocócica naturalmente adquirida, bem como de quinze cabras e quinzes ovelhas saudáveis durante 6 dias consecutivos. Nas fêmeas caprinas, foi verificado aumento dos teores séricos de ceruloplasmina (570%), fibrinogênio (125%), haptoglobina (621%), e α1-glicoproteína ácida (279%). Nas fêmeas ovinas as concentrações de ceruloplasmina, fibrinogênio, haptoglobina e α1-glicoproteína ácida elevaram-se em 337%, 90,9%, 461% e 225%, respectivamente. Os resultados permitem inferir que estas PFAs são marcadores sensíveis e precoces de mastite causada por S. aureus em cabras e ovelhas.(AU)

Este estudo teve por objetivo estabelecer o proteinograma sérico em éguas com placentite induzida e em seus respectivos neonatos. Foram coletadas amostras de sangue das éguas em oito momentos diferentes e dos potros em quatro momentos. Para obtenção da concentração das frações proteicas, utilizou-se eletroforese em gel de acrilaminada contendo dodecil sulfato de sódio (SDS-PAGE). No método utilizado, foram observadas 23 bandas proteicas, cujos pesos moleculares variaram de 16KDa a 245KDa, sendo possível a identificação das seguintes frações: 175KDa, 102KDa, 83KDa, 63KDa, 50KDa, 41KDa, 39KDa e 28KDa. De todas as bandas proteicas encontradas, somente as de 39KDa e 41KDa apresentaram alteração na cinética nos momentos avaliados. De acordo com a solução marcadora, pode-se sugerir que essas proteínas seriam alfa1-glicoproteína ácida (39KDa) e haptoglobina (41KDa). A concentração de imunoglobulinas nos potros apresentou aumento significativo a partir das 12 horas de nascimento. Não está elucidado se estes níveis refletem a persistência do processo inflamatório placentário ou se são alterações fisiológicas do periparto. Não foram observadas alterações na cinética das proteínas nos potros nas primeiras 48 horas.(AU)


The aim of this paper was to identify the serum acute phase protein concentration in mares with induced placentitis and their neonates. Blood samples were collected from the mares in 8 different moments, and from the foals, in 4 moments. To obtain the concentration of protein fractions acrilaminada gel electrophoresis in sodium dodecyl sulfate (SDS-PAGE) was used. In the used method 23 protein bands whose molecular weights ranged from 16kDa to 245kDa were observed, it is possible to identify the following fractions: 175kDa, 102kDa, 83kDa, 63kDa, 50kDa, 41kDa, 39kDa and 28kDa. Of all the protein bands found only the 39KDa and 41KDa have changes in the kinetics in the evaluated times. According to the marker solution, we would suggest that these proteins are alfa1-acid glycoprotein (39kDa) and haptoglobin (41kDa). The concentration of immunoglobulins in foals increased significantly from 12 hours of birth.(AU)

Objective To investigate the clinical value of urinaryα1-acid glycoprotein (AAG)in the early diagnosis of diabetic nephropathy(DN).Methods 266 cases of diabetic patients in Drum Tower Hospital Affiliated to Medical School of Nanjing University from May 2013 to October 2014 were recruited,and patients were divided into four groups based on the urine al-bumin/creatinine (ACR)levels,as follows:non-diabetic nephropathy group (Non-DN)152 cases,low urinary albumin/cre-atinine group (LUA)49 cases,high urinary albumin/creatinine group (HUA)32 cases,and very high urinary albumin/cre-atinine group (VHUA)33 cases.63 cases of healthy subjects (NC group)and 66 cases of fracture patients (stress group) were recruited as control group.The urinary AAG and other biochemical indicators were measured for all subjects.Results The level of urinary AAG in DN group was significantly higher than that in stress group,Non-DN group and NC group (t=7.951,7.985,8.021,P <0.05),and gradually increased from LUA,HUA to VHUA group (P <0.05).The urinary AAG level was positively correlated with UA,Cr,CRP and ACR (r=0.169,0.286,0.373,0.606,P <0.01).Stepwise logistic re-gression analysis showed that the urinary AAG was the independent risk factor of DN (P <0.05).Conclusion The level of urinary AAG has good application value in the early diagnosis and monitoring progression of DN.

Objective To investigate serum alpha 1-acid glycoprotein (AAG)in patients with rheumatoid arthritis (RA)pa-tients with serum drug before and after the treatment,application of evaluation of serum alpha 1-acid glycoproteindetection in RA.Methods 88 cases of newly diagnosed RA patients treated with methotrexate,hydroxychloroquine combined with pyri-dine and the Liu Danhuang administration,in zeroth and June.The patients were taken for determination of serum AAG and rheumatoid factor in the serum of the patients with rate nephelometry (RF)levels,and erythrocyte sedimentation rate (ESR),and record the patients with RA disease the activity index of 28 (DAS28).Results After treatment,RA in the blood of patients with AAG (1 136.30±322.40 mg/L),RF(43.73±4.53 IU/ml),ESR (34.56±9.21 mm/h)and DAS28 (3.10 ±1.0)were significantly decreased (P <0.05 or 0.01),and the AAG,ESR,RF and DAS28 were positively correlated (r=0.19,P =0.02;r=0.24,P =0.0 and r=0.39,P =0.03).Conclusion The serum RA inpatients with AAG changes obvi-ously,had certain curative effect evaluation of RA.

Objective To analysis effect of Bifidobacterium tetravaccine tablets ( live ) combined with cefixime on C-reactive protein, ceruloplasmin, haptoglobin and α1-acid glycoprotein in patients with Bacterial enteritis.Methods 58 patients who were diagnosed with Bacterial enteritis were collected.All patients were randomly divided into experimental group and control group, 29 cases in each group, On the basis of conventional treatment, the control group was treated with cefixime, and the experimental group was treated with Bifidobacterium tetravaccine tablets ( live) on the basis of control group.After treatment, the serum levels of CRP, CER, HPT, AAG and clinical curative effect were detected in all patients.Results After treatment, CRP, CER, HPT, AAG levels were lower than before treatment (P<0.05),compared with control group, CRP, CER, HPT, AAG levels were lower than in the experimental group( P<0.05);the total effective rate was higher in the experimental group (χ2 =4.35, P<0.05).Conclusion Bifidobacterium tetravaccine tablets(live) combined with cefixime can significantly reduce the serum CRP, CER, HPT, AAG levels in patients with bacteria enteritis, improve the clinical efficacy, have guidance significance for clinic.

Objective To investigate the value of serum α1 acid glycoprotein(AAG) and Homocysteine(Hcy) in different stage of renal damage in type 2 diabetes(T2DM ) .Methods a total of 128 patients with T2DM were recruited from January 2013 to Octo‐ber 2013 ,According to 24 h urinary albumin excretion rate (UAER) were divided into the first group ,the second and third groups , and choose 80 healthy cases over the same period as the normal control group .Serum AAG ,Hcy ,creatinine (Cr) and blood urea ni‐trogen (BUN) were detected ,the test results were analyzed statistically .Results There were significant differences in type 2 diabe‐tes mellitus group ,early diabetic nephropathy group ,clinical diabetic nephropathy group of the serum AAG ,Hcy levels(P<0 .05) , there were positively correlated (r=0 .673 ,P<0 .05) between UAER and serum AAG levels ,and were positively correlated (r=0 .35 ,P<0 .05) between UAER and Hcy levels .Conclusion Combined detection of serum AAG ,Hcy have a significant for early diagnosis of diabetic nephropathy and assessing the degree of renal injury in diabetes .

Objective To investigate the significance of single and combined detection of serum alpha1-antitrypsin,alpha1-acid glycoprotein and alpha-fetoprotein in the diagnosis of primary hepatic carcinoma(PHC).Methods Technique of rate nephelometry was used to detect the serum AAT and AAG in 58 PHC patients,47 patients with benign liver disease and 41 healthy people.The serum levels of AFP in the three groups were measured by chemiluminescence immunoassay.Results The levels of AAT,AAG and AFP in PHC group was higher than those in control group (P <0.05).The sensitivity of AAT,AAG and AFP in the diagnosis of PHC was 82.8%,84.5% and 72.4% respectively;The specificity was 85.2%,90.9% and 83.0% respectively.The combined measurement of the 3 makers could elevated the diagnostic specificity of PHC to 98.3%.Conclusion Detection of serum AAT and AAG can be a supplement to AFP.Combined measurement of the 3 makers might increase the diagnostic sensitivity of PHC.

Nas últimas décadas, as proteínas de fase aguda (PFAs) tornaram-se biomarcadores de escolha em medicina humana para identificação e monitoração de doenças. Não há razão para imaginar que tais pesquisas clínicas não sejam igualmente úteis na medicina veterinária. Com o objetivo de verificar a importância das PFAs como biomarcadores de doenças inflamatórias em bovinos, determinou-se o proteinograma sérico, por meio da técnica de eletroforese SDS-PAGE, com interesse especial nas PFAs. Foram utilizados 30 animais, distribuídos em dois grupos: 15 bovinos sadios e 15 bovinos doentes (cinco com mastite estafilocócica, cinco com fotossensibilização e cinco com onfaloflebite). Os animais foram submetidos a colheitas diárias de sangue durante sete dias, enquanto internados no Hospital Veterinário da Unesp, Campus de Jaboticabal. Ceruloplasmina e haptoglobina apresentaram elevação significativa em animais acometidos por mastite, fotossensibilização e onfaloflebite (275,17% e 343,71%; 175,17% e 230,19%; 114,47% e 144,47%, respectivamente). A α1-glicoproteína ácida foi um bom biomarcador apenas em animais com mastite e fotossensibilização, elevando, respectivamente, suas concentrações séricas em 198,14% e 145,89%. Fibrinogênio mostrou-se um indicador confiável apenas em bovinos com mastite, com elevação de 146,5% em relação ao grupo sadio. Ficou clara a diferença na responsividade de distintas PFAs frente a diferentes estímulos inflamatórios. Ceruloplasmina e haptoglobina foram biomarcadores mais sensíveis e, portanto, mais confiáveis entre as PFAs estudadas nessa espécie.


Over the last few decades acute phase proteins (APP) have become the biomarkers of choice in human medicine to identify and monitor inflammation and infection. There is no reason to suppose that clinical investigations in veterinary medicine would not be equally assisted by APP assays. Aiming to verify the importance of APPs as biomarkers of inflammatory diseases in domestic cattle, serum protein profiles, especially APPs, were determined through the SDS-PAGE electrophoresis technique. Thirty animals were allotted in two groups: 15 healthy cattle and 15 clinically ill cattle (5 with staphylococcal mastitis, 5 with photosensitization and 5 with onphalophlebitis). All animals were submitted to daily blood sampling during 7 days, while interned in the Veterinarian Hospital from UNESP, Jaboticabal campus. Ceruloplasmin and haptoglobin were significantly elevated in animals with mastitis, photosensitization and onphalophlebitis (275.17% and 343.71%; 175.17% and 230.19%; 114.47% and 144.47%, respectively). α1-acid glycoprotein behaved as a good biomarker only in animals with mastitis and photosensitization, elevating respectively 198.14% and 145.89% of their serum levels. Fibrinogen was a reliable indicator only in animals undergoing mastitis, with a raise of 146.5%. The diverse responsiveness of different APP under distinct inflammatory stimuli was clear. Ceruloplasmin and haptoglobin were more sensible and, therefore, reliable biomarkers to the diseases studied in this species.

OBJECTIVE: To determine the validity of alpha-1-acid glycoprotein as a novel biomarker for mortality in patients with severe sepsis. METHODS: We prospectively included patients with severe sepsis or septic shock at the emergency department at a single tertiary referral teaching hospital. All of the patients were enrolled within the first 24 hours of emergency department admission, and clinical data and blood samples were obtained. As the primary outcome, we investigated the association of serum levels of alpha-1-acid glycoprotein and 96-hour mortality with logistic regression analysis and generalized estimating equations adjusted for age, sex, shock status and Acute Physiology and Chronic Health Evaluation II score. RESULTS: Patients with septic shock had lower alpha-1-acid glycoprotein levels at the time of emergency department admission compared to patients without shock (respectively, 149.1 ±42.7 vs. 189.8 ±68.6; p = 0.005). Similarly, non-survivors in the first 96 hours were also characterized by lower levels of alpha-1-acid glycoprotein at the time of emergency department admission compared to survivors (respectively, 132.18 ±50.2 vs. 179.8 ±61.4; p = 0.01). In an adjusted analysis, alpha-1-acid glycoprotein levels ≤120 mg/dL were significantly associated with 96-hour mortality (odds ratio = 14.37; 95% confidence interval = 1.58 to 130.21). CONCLUSION: Septic shock patients exhibited lower circulating alpha-1-acid glycoprotein levels than patients without shock. Alpha-1-acid glycoprotein levels were independently associated with 96-hour mortality in individuals with severe sepsis. .

Objective To evaluate the value of haptoglobin (HPT),α1-acid glycoprotein (AAG)and C-reactive protein (CRP) in the differential diagnosis of bacterial and viral infection in children. Methods Serum CRP, AAG and HPT levels were measured in 64 children with bacterial infection and 64 children with viral infection during acute stage. These indicators were also measured in 31 normal control children. The sensitivity ,specificity ,positive and negative predictive values, Youden's index for HPT,AAG and CRP were calculated. These values were compared among three groups. Their early diagnostic value on bacterial infection and virus infection was evaluated. Results (1) These markers were considered positive if HP≥2 g/L,AAG≥2 g/L,CRP≥10 mg/L. (2) The serum CRP [(2.70 ±0.99) g/L vs (1.42 ±0.75) g/L] ,AAG [(0.91 +0.38) g/L vs (0.30+0.08) g/L] and HPT [(6.40 ±0.89) mg/L vs (1.38 +0.80) mg/L]levels were significantly higher in children with bacterial infection compared with those with viral infection during the acute stage (P < 0. 05,P < 0. 01). The serum CRP, AAG and HPT levels were significantly higher in children with bacterial infection compared with normal control children during the acute stage (P < 0. 05);while the levels of these indicators showed no differences between viral infection children and normal control children (P > 0. 05). Conclusion The measurement of serum CRP, AAG and HPT levels has clinical value in the differential diagnosis of bacterial and viral infection in children. Among these indicators, CRP has the highest specificity,positive predictive,negative predictive values and Youden's index.

OBJECTIVE To investigate the value of acute phase protein in the diagnosis and therapy for nosocomial infection monitoring. METHODS The levels of serum C-reactive protein(CRP),serum amyloid A(SAA),?_1-acid glycoprotein(AAG),and ?_1-antitrypsin(AAT) were measured in 71 gynecological patients with nosocomial infection during chemotherapy,Thirty normal controls and 33 chemotherapy patient controls were detected by nephelometry and compared with white blood cell counts. RESULTS The nosocomial infection rate of chemotherapy patients was 16.7%.Main pathogens were Gram-negative bacteria(60.6%).Fungi infection rate was 19.7%.The most frequent hospital infection sites were respiratory tract,gastrointestinal tract and wound.Compared with controls,the levels of serum CRP,SAA,AAG and AAT were significantly higher in chemotherapy patients with bacteria infection(P

OBJECTIVE: We determined the influence of 3-OMD in the protein binding of levodopa to estimate the effect of 3-OMD on the penetration of levodopa into brain. METHOD: P-glycoprotein in the brain may serve to limit drug penetration into the brain. Because it is not available as an experimental substance, but has similar binding properties with alpha 1 acid glycoprotein(AGP), we used AGP in this study. Additionally, we used blood plasma to see the affinity of plasma binding of levodopa. The final concentration of chemicals used in this study were 125, 250, 500, 1000, 2000, 4000 microgram/l for levodopa and 0, 1250, 5000, 10,000 microgram/l for 3-OMD, 1 mg/l for AGP. The free fraction of levodopa in blood plasma and AGP were separated by ultrafiltration method and determined by beta-counter, respectively. RESULTS: We found that levodopa did not bind with AGP, but only 22-24% from 125 microgram/l to 4000 microgram/l of it bound with blood plasma. The addition of 3-OMD to the blood plasma did not significantly change the binding of levodopa. CONCLUSIONS: We can conclude that 3-OMD does not influence the penetration of levodopa into brain. These small amount of the binding does not expect to influence to other drugs on the binding with plasma.

Objective To study the non-covalent binding between the tectoridin(TE) and plasma proteins.Methods The molecular weights of TE,human serum albumin(HSA),?1-acid glycoprotein(AAG) and the protein-drug complexes were measured by the electrospray ion trap mass spectrometry(ESI-MS).The maximum stoichiometric ratios were obtained according to the molecular weight change of the complexes before and after binding reaction.The binding constants(K) of the complexes were calculated by the Scatchard equation.The main sorts of binding force of the complexes were deduced according to the relationship between the reaction temperature and the thermodynamic parameters(?H and ?S).Results The K of the complexes were 1.914?104 mol/L for TE-HSA and 5.893?104 mol/L for TE-AAG,and the number of binding sites were 7.8 and 3.3,respectively.The main sorts of binding force between TE-HSA or TE-AAG were static-electricity gravitation.Conclusion ESI-MS is a good method for studying of the TE-protein non-covalent binding with some advantages in sensitivity,high-speed,and accuracy.

RESUMEN

?-1 acid glycoprotein, also named orosomucoid (ORM), is one kind of serum protein with genetic polymorPhism. Anti-ORM serum is necessary to phenotyping ORM. This communication describes the preparation of the antiORM serum. The anti-ORM sera were produced in three New Zealand rabbits cimmunized with ORM which was isolated and purified from human sera previously in our laboratory. The results of identification showed that the specificity of the home made anti-ORM and the commercial anti-ORM sera (Sigma) were identical. The titer of the home made anti-ORM serum was 128. 2.4?g/ml ORM could be detected by double immunodiffusion with the anti-ORM serum. In addition, the anti-ORM serum did not cross-react with other human serum proteins.

RESUMEN

Serum concentrations Of sialic acid(SA), fucose (Fuc) , ?1-acid glycoprotein (?1-AG), haptoglobi (HP),?1-antitrypsin (?1-AT), Prealbumin (PA) and tranrf-errin (TF)were co comitantly measured in 31 patier's with AFP negative primary hepatocellular carcinoma (HCC) , 33 with AFP positive HCC, 35 with cirrhosis and 58 normal blood donors. The results revealed that serum SA, Fuc, ?1-AG, HP and ?1-AT levels were all significantly higher in patients with AFP negative HCC than in control groups. Among them, Fuc, ?1-AG and HP levels were significantly higher in patients with AFP negative HCC than in AFP positive HCC. Therefore, measurements of these five indices are suggested in the diagnosis of the AFP negative HCC.Furtheremore, the raised concentration of protein bound sugars in HCC usually bear a strong positive correlation with increased concentrations of serum ?1-AG, HP and ?1-AT
