RESUMEN

OBJECTIVE: To investigate the clinical significance of apoptotic rates of human luteinized granulosa cells in in vitro fertilization and embryo transfer (IVF-ET) program. MATERIALS AND METHODS: From 62 infertile patients undergoing controlled ovarian hyperstimulation (COH) for IVF-ET, luteinized granulosa cells were isolated during oocyte retrieval. The apoptotic rates of luteinized granulosa cells were measured by immunocytochemical staining with TUNEL method, and compared with the various clinical parameters. RESULTS: The apoptotic rates were not significantly different among the groups with the various etiologic factors of infertility. The presence of endometriosis, as well as the extent of endometriosis, did not show any differences in the apoptotic rates. The apoptotic rates were negatively correlated with the embryo quality and serum estradiol levels on hCG day. The apoptotic rates of 2.92% or below in the luteinized granulosa cells could predict a successful pregnancy with the sensitivity of 38.9% and the specificity of 93.2%. CONCLUSION: The apoptotic rates of human luteinized granulosa cells may be used to predict the outcome in IVF-ET program.

RESUMEN

Objective To observe the impacts of the transfection of antisense DNA methyltransferase Ⅰ(DNMT1) gene fragment into SMMC 7721 cell on the changes of cell sensitivities to tumor necrosis factor related apoptosis inducing ligand(TRAIL). Methods The eukaryon expression vector pCl neo was transfected into SMMC 7721 cells by liposomes and the transfection was identified by PCR. Survival cell rate was measured by trypan blue exclusion, apoptotic rate was determined by in situ TdT dUTP terminal nick end labeling(TUNEL) method. Results PCR detection indicated that the eukaryon expression vector pCl neo was successfully transfected into SMMC 7721 cells. Survival cell rate of SMMC 7721 cells transfected with antisense DNMT1 gene fragment was remarkably lower than that of transfected with sense DNMT1 gene fragment and empty vector ( P

RESUMEN

Objective To investigate the inhibitory effect of vitamin E succinate(VES) combined with ~chemotherapeutic drugs on the proliferation of human breast cancer cells. Methods Bcap-37 human breast cancer cells were treated with VES combined with chemotherapeutic drugs for 24h and 36h. The ~concentrations of VES were 10?g/mL and 20?g/mL and those of 5-florouracil, mitomycin and ~cyclophosphamide were 16.9?g/mL and 33.8?g/mL, 1?g/mL and 3.3?g/mL and 100?g/mL and 300?g/mL respectively. The inhibitory effect was measured with MTT method and the cell cycle and cell ~surface Fas expression were analyzed with flow cytometry assay. Results The combination of VES with ~chemotherapeutic drugs had a significant inhibitory effect on the growth of Bcap-37 human breast cancer cells. Flow cytometry assay of cell cycle showed that the natural apoptptic rate of Bcap-37 cells was 0.7%;after treatment with VES 20?g/mL,the apoptotic rate was 19.2%;after treatment with 5-Fu,mitomycin and ~cyclophosphamide the apoptotic rates were 16.2%,16.7% and 12.3%,respectively;after the combined use of VES and the 3 chemotherapeutic drugs,the apoptotic rates were 40.3%,44.8%,39.6%,~respectively .Fas expression in cancer cells increased after the co-administration of VES and chemotherapy drugs. Conclusions VES combined with chemotherapeutic drugs had significant inhibitory effect on the growth of Bcap-37 human breast cancer cells. The mechanism may be related to Fas upregulation on the surface of cancer cells.

We examined eutopic,ectopic endometria and peritoneal fluid macrophages from 22 patients with endometriosis(EMS) and 14 women without EMS.To obtain evidence for the induction of programmed cell death,apoptotic cells were identified using a modified terminal deixynucleotidyltransferasebiotin nick end labeling method(TUNEL).To evaluate cell death repressor activity,bcl 2,bax and fas genes expression was examined using immunohistochemical staining.
