RESUMEN

Aim: To isolate and identify Alcaligenes aquatilis PJS_1 from slaughter house soil samples for production of enzymatic fibrinolytic agent productionMethodology: Fibrinolytic enzyme producing bacterium was isolated from slaughter house soil samples and identified by biochemical tests and 16S rRNA sequencing. The fibrinolytic enzyme production media was optimized by various factors like energy sources, pH and temperature. Bioreactor used in the experiment was designed with suitable parameters for effective production and purification is by gel filtration chromatography. Blood clotting assay was performed to determine its anticoagulant property. Results: The isolated enzyme producing bacterium was identified as Alcaligenes aquatilis PJS_1. The medium with fructose and urea at pH 7.0 was found to have optimum production when incubated for 24 hr at 37ºC. The crude enzyme was purified by acetone precipitation followed by gel filtration chromatography. The enzyme showed a final specific activity of 629.32 Umg-1 with of 88.24% yield Interpretation: The present study provides information that the enzyme produced by Alcaligenes aquatilis PJS_1 acts as an effective fibrinolytic agent

Aim: Potential microbial isolates for bioremoval of copper were collected from phenolic and heavy metal-contaminated soils and screened in copper-containing medium for determining the maximum tolerance level for copper. Methodology: Bioremoval of copper was assessed using sodium diethyl dithiocarbamate assay. Physical and cultural conditions influencing copper bioremoval such as initial concentration, biomass dosage (inoculum volume), temperature and pH were optimised via one-factor-at-a-time. Results: The highest maximum tolerance level was displayed by Serratia sp. AQ5-03 at 600 mg l-1, while for Alcaligenes sp. AQ5-02 and Pseudomonas sp. AQ5-04 it was 500 mg l-1. The highest bioremoval for Alcaligenes sp. AQ5-02, Serratia sp. AQ5-03 and Pseudomonas sp. AQ5-04 was achieved at 20, 50 and 75 mg l-1, respectively. The optimum biomass dosage (inoculum volume) for both Serratia sp. AQ5-03 and Pseudomonas sp. AQ5-04 were 15%, whereas it was 10% for Alcaligenes sp. AQ5-02. The results also revealed that maximum bioremoval was achieved at pH 6 at an optimum temperature of 20°C for both Alcaligenes sp. AQ5-02 and Pseudomonas sp. AQ5-04, while Serratia sp. AQ5-03 showed optimum removal at pH 7 and at 30°C temperature. The maximum bioremoval efficiency for Alcaligenes sp. AQ5-02, Serratia sp. AQ5-03 and Pseudomonas sp were found to be 62, 57 and 70%, respectively. Interpretation: The results indicate that Alcaligenes sp. AQ5-02, Serratia sp. AQ5-03, Pseudomonas sp. AQ5-04 can be utilised as bioremoval agent since it possessed the highest bioremoval efficiency for copper remediation.

Aims@#Erwinia psidii was first reported in 2017 to be the causal pathogen of papaya dieback disease (PDD) in Sabah, Malaysia. The present study aimed to isolate potential biocontrol agents against this pathogen. @*Methodology and results@#Out of the 20 samples collected from Crocker Range of Sabah, 154 bacteria and 55 fungi isolates were isolated and screened for their antagonistic activity against E. psidii. The fungi and bacteria which gave the highest inhibition to E. psidii were identified using molecular technique as Alcaligenes faecalis and Lecanicillium sp., respectively. Both isolates were selected for extraction of their secondary metabolites to determine their bioactivity against E. psidii. Micro-well dilution method was used to determine minimum inhibitory concentration (MIC) for each microbes’ extract. GC-MS analysis was carried out to determine their secondary metabolites. @*Conclusion, significance and impact of study@#Lecanicillium sp. (Diethyl ether, chloroform and ethyl acetate) extracts and A. faecalis (Diethyl ether extract) showed positive inhibition against E. psidii. GC-MS analysis revealed that both A. faecalis and Lecanicillium sp. had secreted some secondary metabolites such as N-formylmaleamic acid, Oleamide and D-1-Piperideine-2-carboxylic acid that may relate to the growth inhibition. Alcaligenes faecalis and Lecanicillium sp. are potential to be further investigated as biocontrol against E. psidii.

Aims@#Metal transcriptional regulators controlled the regulation of metal ion homeostasis in bacteria genera. Cd(II)/Pb(II) transcriptional regulator is one of the member of MerR family found in Alcaligenes faecalis SF-S1-60 (PbrT-AF). @*Methodology and results@#The PbrT-AF gene with 432 bp open reading frame was successfully isolated from genomic DNA of A. faecalis using polymerase chain reaction (PCR) analysis. This gene was phylogenetically grouped with A. alcaligenes species using PHYLIP version 3.69 by the neighbor-joining method with 1000 bootstrap replicates. Phylogeny analysis shows that these proteins have distinct amino acids compared to Cd(II)/Pb(II) regulators from different species. The structure of PbrT-AF shows similar conformation with other members of MerR family using MODELLERv9.17. We also demonstrated that the expression of Pbrt-AF in Escherichia coli BL21 were able to increase the bacteria tolerance towards Pb up to 1000 ppm. @*Conclusion, significance and impact of study@#This result suggests that PbrT-AF promotes cell adaptation and tolerance towards Pb toxicity.

Heavy metal contamination, particularly of cultivable lands, is a matter of concern. Bioremediation helps in reversing such contamination to certain extent. Here, we report isolation, polyphasic identification and the role of siderophore producing rhizobacteria Alcaligenes feacalis RZS2 and Pseudomonas aeruginosa RZS3 in bioremediation of heavy metal contaminated soil and plant growth promotion activity in such contaminated soil. Siderophore produced by A. feacalis RZS2 and P. aeruginosa RZS3 strains chelated various heavy metal ions like MnCl2.4H2O, NiCl2.6H2O, ZnCl2, CuCl2 and CoCl2 other than FeCl3.6H2O at batch scale. Their bioremediation potential was superior over the chemical ion chelators like EDTA and citric acid. These isolates also promoted growth of wheat and peanut seeds sown in heavy metal contaminated soil. Effective root colonizing ability of these isolates was observed in wheat and peanut plants.

A. xylosoxidans infection can result in potentially severe sequelae, especially in unhealthy populations. We present a rare case of infectious skin ulceration with scar contracture due to A. xylosoxidans in a healthy patient. A 53-year-old male patient without underlying disease and trauma history visited our department for an irritating skin ulcer with purulent discharge on the right lower leg. The chronic wound on the leg exists since he got contact burn 30 years ago. The wound did not respond to treatment, and the wound culture results indicated A. xylosoxidans. Given the incurable character of A. xylosoxidans, we performed a wide excision and split thickness skin graft with collagen-elastin matrix (Matriderm(R)). A. xylosoxidans is an opportunistic, aerobic, gram-negative, rod-shaped bacterium that is oxidase positive. According to reports, A. xylosoxidans is not sensitive to many antibiotics and can cause bacteremia. It is suggested that the blood supply is compromised near a scar lesion despite the fact that blood flow is rather increased during the initial scar formation phase. Therefore, we presumed that the infection was most likely caused by low blood circulation due to scar contracture. Thus, when a scar contracture is present, A. xylosoxidans infection can manifest in a healthy patient.

Descreve-se um caso de infecção por Alcaligenes faecalis em peixe ornamental Betta splendens, incluindo avaliação clínica, patológica e bacteriológica. Um peixe fêmea, com peso de 16 g, inicialmente demonstrou discreta lesão escoriativa no tegumento, inapetência, emagrecimento progressivo, letargia, dispneia e aumento de volume na região lateral esquerda. Constataram-se em necropsia a presença de exoftalmia bilateral, opacidade de córnea, ascite e abscesso no subcutâneo provocando distensão da cavidade celomática. As alterações histológicas da pele se caracterizaram por abscesso, necrose, hemorragia e infiltrado inflamatório constituído principalmente de heterófilos. No exame microbiológico com exsudato e sangue venoso isolou-se exclusivamente A. faecalis.(AU)


This paper aimed to describe one case of infection by Alcaligenes faecalis in an ornamental fish Betta splendens, including clinical, pathological, and bacteriological evaluation. A female fish weighing 16 g initially showed discrete ulcerative lesions in the integument, inappetence, progressive weight loss, lethargy, dyspnea, and increased volume in the left lateral region. Necropsy revealed bilateral exophthalmia, corneal opacity, ascites, and subcutaneous abscess causing distension of the celomatic cavity. Histological changes of the skin were characterized by abscess, necrosis, hemorrhage, and inflammatory infiltrate consisting mainly of heterophils. Microbiological assays using the exudates and venous blood isolated exclusively A. faecalis.(AU)

Aims: The focus of this study was to evaluate the antimycobacterial activity of Alcaligenes faecalis BW1 extract and to purify it partially. Study design: Partial purification of A. faecalis BW1 extract was performed by using thin layer chromatography and active substances responsible for the biological activity were localized. Place and Duration of Study: The study was carried out at laboratory of Microbial Biotechnology, Department of Biology, Faculty of Sciences and Technical, University Sidi Mohamed Ben Abdellah, BP 2202, Road of Immouzer, Fez, Morocco, during the period from January 2011 to July 2011. Methodology: Crude extract of A. faecalis BW1 was obtained by using ethyl acetate as an organic solvent and its antimycobacterial effect was investigated by agar discs diffusion method. The extract was then fractionated by thin layer chromatography and the bioactivity was assessed with a bioautography technique followed by spots elution tests. Results: The results showed that A. faecalis BW1 produced compounds with antimycobacterial activity. All the detected spots by thin layer chromatography inhibited the growth of M. smegamtis. Conclusion: Various metabolites of A. faecalis BW1 are responsible for the sought effect or they could act synergistically to inhibit mycobacterial growth. These compounds could be used after their total purification in further work against mycobacterial infections.

Aims: To highlight whether metabolites of Alcaligenes faecalis BW1 extract can be administered orally for their possible antimycobacterial effects. Study Design: Study of the influence of certain parameters on the extract of Alcaligenes faecalis by using either discs or well diffusion methods against M. smegmatis. Place and duration of study: Laboratory of Microbial Biotechnology, Department of Biology, Faculty of Sciences and Technical, University Sidi Mohamed Ben Abdellah, BP 2202, Road of Immouzer, Fez, Morocco. From April to August, 2012. Methodology: The impact of acidic pH of gastric juice, bile, hydrogen peroxide, pancreatic enzymes and lysozyme on the antimycobacterial activity of Alcaligenes faecalis BW1 extract was evaluated by agar diffusion method. Detection whether or not antibacterial metabolites having a synergistic effect with rifampicin against M. smegmatis was also explored. Results: Antibacterial metabolites of Alcaligenes faecalis BW1 extract resist to the action of gastric pH, gallbladder bile and hydrogen peroxide. In addition, they are not affected by pancreatic enzymes and lysozyme. Moreover, they have a synergistic effect with rifampicin against M. smegmatis. Conclusion: Anti-mycobacterial metabolites of Alcaligenes faecalis BW1 extract are compatible with rifampicin and could be administered orally as antitubercular agents after their purification, identification in further work.

We describe a rare case of extraocular inflammation secondary to scleral buckle infection with Alcaligenes xylosoxidans. A 60‑year‑old female with a history of retinal detachment repair with open‑book technique of scleral buckling presented with purulent discharge and irritation in the right eye that had begun 4 weeks earlier and had been treated ineffectively at another hospital. Conjunctival erosion with exposure of the scleral buckle was noted. The scleral buckle was removed and cultured. The explanted material grew gram‑negative rod later identified as A. xylosoxidans. On the basis of the susceptibility test results, the patient was treated by subconjunctival injection and fortified topical ceftazidime. After 4 weeks of treatment, the infection resolved.

Mounier-Kuhn syndrome is a rare entity characterized by abnormal dilatation of the trachea and main bronchi (tracheobronchomegaly). Alcaligenes xylosoxidans is a non fermenting gram-negative pathogen common in extra-and intra-hospital environment, which may be related to immunosuppression states. We describe the case of a 75 years old male, ex-smoker with moderate functional obstruction, chronic respiratory failure and chronic colonization by Pseudomonas aeuriginosa. He had an infectious exacerbation of his disease, reason that previously required several hospital admissions. The patient was treated with antibiotics and his evolution was favourable with negativization in cultures of the pathogen. This is the first description of the isolation of Alcaligenes xylosoxidans as a cause of respiratory infection in a patient with Mounier-Kuhn syndrome.

Bacterial peritonitis is a well-recognized complication of continuous ambulatory peritoneal dialysis (CAPD) in patients with end-stage renal failure. Achromobacter xylosoxidans subsp. xylosoxidans is a catalase and oxidase positive, motile, nonfermentative and gram-negative rod bacterium that is a rare pathogen in humans and has rarely been reported as an opportunistic human pathogen. We present a case of peritonitis due to unusual pathogens, Achromobacter xylosoxidans subsp. xylosoxidans. A 49-year-old diabetic man undergoing CAPD for 90 days developed the first peritonitis due to Achromobacter xylosoxidans subsp. xylosoxidans. A. xylosoxidans was detected from a culture of peritoneal fluid. Susceptible antibiotic treatment was provided.

Alcaligenes latus, isolated from industrial effluent, was able to grow in mineral salts medium with 50 ppm (0.15 mM) of H-acid as a sole source of carbon. Immobilization of Alcaligenes latus in Ca-alginate and polyurethane foam resulted in cells embedded in the matrices. When free cells and immobilized cells were used for biodegradation studies at concentration ranging from 100 ppm (0.3 mM) to 500 ppm (1.15 mM) degradation rate was enhanced with immobilized cells. Cells immobilized in polyurethane foam showed 100 percent degradation up to 350 ppm (1.05 mM) and 57 percent degradation at 500 ppm (1.5 mM). Degradation rate of Ca-alginate immobilized cells was less as compared to that of polyurethane foam immobilized cells. With Ca-alginate immobilized cells 100 percent degradation was recorded up to 200 ppm (0.6 mM) of H-acid and only 33 percent degradation was recorded at 500 ppm (1.5 mM) of H-acid. Spectral analysis of the products after H-acid utilization showed that the spent medium did not contain any aromatic compounds indicating H-acid degradation by A. latus.

PURPOSE: To report a case of corneal ulcer due to Alcaligenes xylosoxidans in a soft contact lens wearer. CASE SUMMARY: A 20-year-old female patient presented with a corneal ulcer. The patient had been wearing soft contact lenses over 6 months. The ulcer had a typical appearance, similar to that of immune infiltration due to contact lenses. However, the corneal scraping and culture yielded Alcaligenes xylosoxidans, which is susceptible to ceftazidime and levofloxacine in the antibiotic susceptibility test. After treatment with topical antibiotics, her eye improved, with only a mild corneal scar remaining. CONCLUSIONS: Alcaligenes xylosoxidans should be considered as a potential causal pathogen of keratitis upon wearing soft contact lenses. Additionally, any lesion should be suspected as a bacterial corneal ulcer, even if appearing as an immune infiltration due to contact lenses.

OBJECTIVE To study 39 kinds of resistant-related genes in a pan-resistant Alcaligenes xylosoxidans subsp xylosoxidans(AXXxx) in the sputum isolated from a severe hepatitis B patient.METHODS The susceptibility to antimicrobial agents were detected by MIC.16S rRNA and 39 resistant-related genes including 29 ?-lactamases genes,6 aminoglycoside-modifying enzymes(AMEs)genes,1 chlorhexidine-sulfadiazine resistant gene(qacE△1-sul1)and 3 intergron genes separated(intⅠ1,2,3) an AXXxx strain in the sputum of a severe hepatitis B patient were measured by PCR,and verified by DNA sequencing.RESULTS Among the strains,7 kinds of resistant-related genes(blaTEM-116,blaCARB-8,aac(6′)-Ⅱ,aac(3)-Ⅱ,ant(3″)-Ⅰ,qacE△1-sul1,and intI1)detected out.But other 27 kinds of ?-lactamases genes,3 kinds of AMEs(aac(6′)-Ⅰb,aac(3)-Ⅰ,ant(2″)-Ⅰ) genes,and 2 kinds of intⅠ(intⅠ2 and intⅠ3) genes were negative.CONCLUSIONS The pan-resistant A.xylosoxidans,mainly relates to 7 kinds of resistant-related genes(blaTEM-116,blaCARB-8,aac(6′)-Ⅱ,aac(3)-Ⅱ,ant(3″)-Ⅰ,qacE△1-sul1,and intⅠ1).

PURPOSE: To report a case of corneal ulcer due to Alcaligenes faecalis in a patient with a preexisting corneal ulcer. CASE SUMMARY: A 58-year-old male patient presented with a corneal ulcer without a history of any trauma. The patient had a history of corneal ulcer 9 months earlier. The patient had previously been diagnosed with diabetic retinopathy and neovascular glaucoma, and his visual acuity was no light perception. Corneal scraping and culture yielded Alcaligenes faecalis susceptible to most antibiotics in the antibiotic susceptibility test. After treatment with empirical systemic antibiotics and eyedrops, his eye improved with a remaining corneal scar. CONCLUSIONS: Alcaligenes faecalis should be considered as a causal pathogen of corneal ulcer in patients with suspicious compromised ocular surface, such as previous corneal ulcer.

RESUMEN

This paper reports the isolation and characterization of a new o-nitrobenzaldehyde (ONBA)-degrading bacterium, Alcaligenes sp. ND1. ND1 degraded almost all ONBA (100 mg L-1) in M9 medium within 36 hours. The key enzyme(s) involved in the initial biodegradation was a constitutively intracellular enzyme(s). This bacterium has great potential utility for bioremediation.


Esse trabalho relata o isolamento e a caracterização de uma nova bactéria degradadora de o-nitrobenzaldeido (ONBA), Alcaligenes sp ND1. A bactéria ND1 decompôs todo o ONBA (100 mg.L-1) do meio M9 em 36 horas. A enzima-chave envolvida na biodegradação inicial foi uma enzima constitutiva intracelular. Esta bactéria apresenta um potencial de aplicação para bioremediação.

We report two cases of postoperative Alcaligenes xylosoxidans endophthalmitis. A 78-year-old woman in good general health developed A. xylosoxidans endophthalmitis one month after an uncomplicated phacoemulsification procedure with posterior chamber intraocular lens. It was performed on her left eye, at another institution. Removal of the intraocular lens and capsule was performed because of recurrent inflammation after vitrectomy and intravitreal antibiotic injections. Her son, a 55-year-old man, developed A. xylosoxidans endophthalmitis in his left eye, two months after an uncomplicated phacoemulsification procedure with posterior chamber intraocular lens (it also was performed by the same surgeon). He also required vitrectomy with removal of the intraocular lens and capsule because of persistent disease. A. xyloso - xidans can cause chronic low-grade and progressive endophthalmitis after cataract extraction that is often resistant to corrective antibiotic therapy. This kind of case is rare. There have been only previously reported seven cases worldwide to the best of our knowledge.


Relatamos dois casos de endoftalmite pós-operatória causada por Alcaligenes xylosoxidans. Uma mulher de 78 anos de idade e saúde geral boa desenvolveu endoftalmite por A. xylosoxidans um mês após ser submetida a facoemulsificação com implante de lente intra-ocular de câmara posterior, sem complicações. A cirurgia foi realizada no olho esquerdo em outra instituição. A remoção da lente e da cápsula intra-ocular foi realizada em conseqüência da inflamação recorrente após vitrectomia e injeções intravítreas de antibiótico. Seu filho, de 55 anos, desenvolveu endoftalmite por A. xylosoxidans, no olho esquerdo, dois meses após facoemulsificação com implante de lente intra-ocular de câmara posterior (realizada também pelo mesmo cirurgião). Foi necessária também realização de vitrectomia com remoção da lente intra-ocular e da cápsula posterior em função da doença persistente. A. xyloso - xidans pode causar a endoftalmite crônica, branda e progressiva após a extração da catarata e é freqüentemente resistente à terapia antibiótica. Este tipo de caso é raro. De acordo com nosso conhecimento, existem apenas sete casos relatados previamente.

lcaligenes xylosoxidans is a catalase and oxidase positive, motile, nonfermentative and gram-negative rod bacterium. A. xylosoxidans infection is a rare cause of pulmonary infection and little information concerning treatment is available. The majority of patients that develop A. xylosoxidans infection belong to a high-risk group due to an immunocompromised condition or due to pulmonary cystic fibrosis. We report two rare cases of immunocompentent patients that developed a pulmonary infection due to A. xylosoxidans. A 77-year-old man was admitted with a lung abscess. The patient denied having any prior medical illness. A culture of bronchial washing fluid showed the presence of A. xylosoxidans. Despite appropriate antibiotic treatment, the patient died from acute respiratory distress syndrome (ARDS). Another patient, a 61-year-old man without an underlying disease, was admitted with empyema. Under the condition of a closed thoracostomy, a high fever persisted and the empyema was also aggravated. A. xylosoxidans was detected from a culture of pleural fluid. Susceptible antibiotic treatment was provided and surgical intervention was performed. We report these cases with a review of the literature.

The purpose of the present work was to inform about the first case of meningitis associated to the bacteria Alcaligenes xylosoxidans in a patient with HIV/AIDS. The patient was a 46-year-old male, with the antecedent of have been diagnosed for HIV/ AIDS, who attended in the Hospital Universitario de Colima, Mexico, with fever, shock and meningismus. The study of the cerebrospinal fluid showed pleocytosis, elevated protein levels and hypoglycorrhachia. The culture yielded the presence of Alcaligenes xylosoxidans with sensitivity to ciprofloxacin. After 14 days of treatment with this antibiotic, the patient showed neurologic improvement and was able to continue with his outpatient antiretroviral treatment. The present case shows the importance of the inclusion of this bacterium in the differential diagnosis of the neurological infections in HIV/AIDS patients and emphasizes the importance of considering the bacterial meningitis in this population.
