RESUMEN

Objective To observe the effect of Bushen-Huoxue decoction combined with conventional therapy on the clinical symptoms, vertigo degree and cognitive function of elderly patients with chronic cerebral cerebral circulation insufficiency (CCCI) and to explore its mechanism. Methods A total of 70 patients were randomly divided into observation group and control group according to random number table method. The two groups were given antihypertensive, hypoglycemic, lipid and other western medicine treatment. On this basic treatment, the control group added orally flunarizine hydrochloride capsules, 5 mg /day before sleeping, while the observation group with Bushen-Huoxue decoction per day. All the treatment last 30 days. dizziness handicap inventory (DHI) and montreal cognitive assessment (MoCA) were used for the symptoms evaluation, and the mean flow velocity of the following arteries basilar artery (BA), bilateral vertebral artery (VA) and middle cerebral artery (MCA) were assessed by the Ultrasound transcranial doppler blood flow analyzer. The acidity phosphatidic acid, AP were detected by chemical colorimetric method. And the clinical effect rates were compared after treatment. Results The total effective rate of observation group was 94.3% (33/35), higher than 82.9% (29/35) in the control group, and the difference in group 2 was statistically significant (χ2=9.728, P<0.05). After treatment, the TCM symptom scores (6.2 ± 3.3 vs. 9.8 ± 3.7, t=8.920), DHI score (4.4 ± 2.5 vs. 9.3 ± 3.6, t=12.081) and MoCA score (25.7 ± 2.6 vs. 23.2 ± 2.8, t=6.638) improvement of the observation group were significantly better than those of control group (P<0.05). The Vms of LVA (37.2 ± 8.5 cm/s vs. 34.9 ± 7.6 cm/s, t=9.103), LMCA (63.3 ± 9.8 cm/s vs. 60.1 ± 8.4 cm/s, t=7.839), RMCA (62.8 ± 10.5 cm/s vs. 60.9 ± 9.5 cm/s, t=6.583) and plasma AP (3.74 ± 1.08 μmol/L vs. 5.81 ± 1.35 μmol/L, t=9.627) improvement of the observation group were significantly better than those of control group (P<0.05). Conclusions The Bushen-Huoxue decoction can improve the clinical symptoms and vertigo of the elderly and improve the cognitive level of the patients. The mechanism may be related to the improvement of the cerebral blood flow velocity, the decrease of plasma AP, and the state of ischemia and hypoxia.

Objective To investigate the influence of bushenhuoxue drug-containing serum on PI3K and Akt signaling pathway in purified retinal ganglion cell (RGCs) in vitro of Sprague-Dawley (SD) rats,and to explore the protective mechanisms of bushenhuoxue recipe on RGCs.Methods At first,bushenhuoxue drug-containing serum was prepared,and the RGCs of SD rats were purified;after the apoptotic model of pressurized and purified RGCs was established successfully in vitro using open pressure control system,RGCs were dealt with 50 g · L-1,100 g · L-1,200 g · L-1 concentration gradient of bushenhuoxue drug-containing serum.Then the subjected cells were divided into normal culture group (N group),control group (C group),50 g · L-1 bushenhuoxue group (50 g · L-1 BSHX group),100 g · L-1 bushenhuoxue group (100 g · L-1 BSHX group),200 g · L-1 bushenhuoxue group (200 g · L-1 BSHX group).Finally,cell apoptotic rate was detected by Annexin V-FITC/PI staining,while real-time quantitative PCR (qRT-PCR) and Western blot were used to detect the mRNA and protein expression of PI3K and Akt in each group respectively.Results The results of qRT-PCR detection showed that PI3K,Akt mRNA expression level in C group (0.04 ±0.01) was decreased compared with N group (1.00 ± 0.04),and the difference was statistically significant (all P<0.05),while PI3K,Akt mRNA levels in 50 g · L-1,100 g · L-1 and 200 g · L-1 BXHX group (0.18 ±0.01,0.21 ±0.02,0.22 ±0.01,0.36 ±0.01,0.84 ±0.10,1.07 ± 0.17) were increased compared with the C group,and the difference was statistically significant (all P <0.05).The Western blot results of each group showed that PI3K,Akt protein expression level in C group was decreased compared with N group,with statistical difference (all P < 0.05),while PI3 K,Akt protein expression levels in 50 g · L-1,100 g · L-1 and 200 g · L-1 BSHX group were increased compared with C group,with staffstical difference (all P < 0.05).Conclusion Bushenhuoxue drug-containing serum may inhibit the RGCs apoptosis induced by pressure,which may be related to the activation of PBK/Akt signal transduction pathway.

Objective To evaluate the therapeutic effect ofBushen-Huoxue decoction combined with oral calcium and vitamin D for senile osteoporosis.Methods A total of 120 patients with OP were enrolled and randomly divided into aBushen-Huoxue decoction group (60 patients) and a elcatonin group (60 patients). Routine use of calcium and vitamin D was adopted. On this basis, theBushen-Huoxue decoction group was treated byBushen-Huoxue decoction combined with oral calcium and vitamin D, while the elcatonin group was treated by intramuscular injection of elcatonin with oral calcium and vitamin D. VAS of low back pain and bone joint pain was evaluated, the changes of BMD in the lumbar spine and femoral neck was observed, serum calcium and phosphorus, ALP concentration changes were also observed.Results After the treatment, the low back pain in the Bushen-Huoxue decoction group (3.30 ± 0.38vs.4.32 ± 0.51,t=11.148), bone joint pain (3.15 ± 0.22vs.3.87 ± 0.26,t=11.880) VAS were significantly lower than those in the elcatonin group.Bushen-Huoxue decoction group L2-4is BMD (L2: 0.74 ± 0.15 g/cm2vs.0.63 ± 0.14 g/cm2,t=4.153; L3: 0.75 ± 0.13 g/cm2vs. 0.70 ± 0.10 g/cm2,t=2.361; L4: 0.76 ± 0.06 g/cm2vs.0.71 ± 0.10 g/cm2,t=3.321) were significantly higher than those in the elcatonin group(P<0.05).Bushen-Huoxue decoction group serum calcium (2.36 ± 0.45 mmol/Lvs. 2.19 ± 0.17mmol/L,t=2.737), serum phosphorus (1.28 ± 0.12 mmol/Lvs.1.22 ± 0.13 mmol/L,t=2.627) were significantly higher than those in the elcatonin group (P<0.05), ALP (89.20 ± 9.41 U/Lvs.114.00 ± 11.80 U/L, t=12.733) was significantly lower than that in the elcatonin group (P<0.01).ConclusionBushen-Huoxue decoction can effectively improve the clinical symptoms of senile osteoporosis, and elevate BMD content. Its efficiency was better than intramuscular injection of elcatonin.

Objective To investigate the effects of Bushen-Huoxue decoction on the plasma level of brain natriuretic peptide (BNP) and heart function in patients with chronic heart failure (CHF) due to kidney deficiency and blood stasis. Methods A total of 96 patients with CHF due to kidney deficiency and blood stasis were randomized into a treatment group (50 cases) and a control group (46 cases). In the control group, the normal treatment of western medicine was given. In the treatment group, on the basis of the control group, Bushen-Huoxue decoction was added. Both groups were treated for 4 weeks. The plasma BNP level was detected with enzyme-linked immunosorbent assay. The left ventricular ejecting function, New York Heart Association functional classes and score of traditional Chinese medicine syndromes were evaluated before and after the treatment. Results After the treatment, the plasma BNP level in the treatment group was significantly lower than that in the control group (897.3 ± 286.7 pg/ml vs. 1 423.5 ± 458.7 pg/ml;P<0.05);the left ventricular end systolic diameter (4.8 ± 0.8 cm vs. 5.4 ± 0.9 cm) and the left ventricular end systolic volume (57.23 ± 5.30 ml vs. 69.78 ± 5.86 ml) in the treatment group were significantly lower than those in the control group (all P<0.05);the stroke volume (72.56 ± 7.23 ml vs. 62.56 ± 5.86 ml) and the left ventricular ejection fraction (59.75%± 6.47%vs. 49.87%± 6.91%) in the treatment group were significantly higher than those in the control group (all P<0.05). The heart function (90.0%vs. 80.4%;χ2=4.369, P<0.05) and the traditional Chinese medicine syndromes (94.0%vs.82.6%;χ2=3.988, P<0.05) had shown significant improvement in the treatment than those in the control group. Conclusions Bushen-Huoxue decoction can reduced the plasma BNP level, improve the heart function in patients with CHF due to kidney deficiency and blood stasis.

Background The early disorder of diabetic retinopathy (DR) is the damage of retinal neural cells induced by high glucose and lack of oxygen.Previous studies show that bushenhuoxue serum can enhance the activity of glutamine synthetase (GS) in Müller cells under hypoxia,and glutamate-mediated retinal excitotoxicity also can be reduced by bushenhuoxue serum intervention.However,whether the concentration of glycine can be increased by bushenhuoxue serum is not clear.Objective This study was to investigate the protective effects of bushenhuoxue serum on retinal ganglion cells (RGCs) under hypoxia.Methods The Sprague Dawley (SD) rat serum containing bushenhuoxue was prepared.The RGCs of newborn SD rats were purified and identified by a twostep immunopanning procedure.After 72 hours,all RGCs were cultured in 96-well plates and divided into four groups:normal control group (cultured in adult SD rats normal serum),bushenhuoxue group (cultured in bushenhuoxue serum),hypoxia group (cultured in 1 mmol/L sodium dithionite); hypoxia + bushenhuoxue intervention group (cultured in bushenhuoxue serum+sodium dithionite).Glutamate and glycine contents in the extracellular fluid were detected by L-8800 automatic amino acid analyzer,and the content of lactate dehydrogenase (LDH) was assayed using LDH kits in 24,48 and 72 hours after culture.Results Cultured cells showed the green fluorescence under the immnofluorescence microscope.The contents of glutamate,glycine and LDH in the extracellular fluid were (0.0805±0.0010)mg/L,(0.0554±0.001 5)mg/L and (1 626.03 ±122.10)μmol/(min · L) in the normal control group in 24 hours after culture,and those in the hypoxia group were (0.022 5±0.001 1) mg/L,(0.014 6±0.001 1)mg/L and (1 458.68±94.23)μμmol/(min · L),showing significant reducing in the hypoxia group (q =-3.53,P =0.00 ; q =-2.45,P =0.00 ; q =-2.98,P =0.02).Compared with the normal control group,LDH and glycine contents in the extracellular fluid were significant raised in the hypoxia group 48 hours after culture (q =2.55,P =0.01 ;q =4.48,P =0.00).Seventy two hours after culture,the glutamate and glycine contents in the hypoxia group were higher than those of the normal control group (q =2.45,P =0.00 ;q =3.72,P =0.00).In 48 and 72 hours of culture,the contents of glycine were (0.017 4±0.001 5) and (0.019 2±0.001 2) mg/L in the hypoxia+bushenhuoxue intervention group,which were significantly higher than (0.016 0±0.001 2) and (0.018 0±0.000 8) mg/L in the hypoxiagroup (q=2.28,P=0.04;q=2.33,P=0.03),but the LDH level were (1 632.94±264.31) and (1 875.00±137.45)μmol/(min · L) in the hypoxia+ bushenhuoxue intervention group,which were lower than (1 688.49 ± 112.86) and (2 267.86 ± 175.21) μmol/(min · L) of the hypoxia group (q =-2.95,P =0.02 ; q =-2.35,P=0.00).No significant differences were seen in the glutamate content 24,48 and 72 hours after culture (P=0.55,0.28,0.46).A positive correlation was seen between the glutamate content and glycine content in the extracellular fluid (Kendall coefficient =0.519,Spearman coefficient =0.696,both at P =0.000).Conclusions The release levels of glutamate and glycine increase in the hypoxia RGCs,which probably is a compensatory response of RGCs.Bushenhuoxue serum can protect RGCs against injury by increasing the release of glycine and decreasing the LDH leakage from RGCs.

[Methods] The intervention method to retrieve recent Bushenhuoxue premature ovarian failure and decreased ovarian reserve related literature, from theoretical research, clinical research and experimental study three aspects discussed Bushenhuoxue method to improve the effect and mechanism of ovarian reserve. [Results] Invigorating kidney on improving the exact effect of ovarian reserve. [Conclusion] In recent years Bushenhuoxue method research progress in improving ovarian function, mechanism of action has a more in-depth elucidation, but further in-depth exploration and research are stil needed.

Objective To investigate whether Bushen-Huoxue compound adjuvant therapy can be effective on mild cognitive impairment in Parkinson's disease and the influence on plasma phospholipids(PLs) levels. Methods 87 PD patients were recruited and collected for the general information. The patients were evaluated by Unified Parkinson's Disease Rating Scale(UPDRS) Ⅲ and Montreal Cognitive Assessment(MoCA). According to MoCA scores, patients with PD-MCI were divided into a Bushen-Huoxue compound treatment group and a control group(basic L-dopa or dopamine agonist). Reevaluating cognitive function by MoCA and plasma phopholipids levels at posttreatment 12 month and 18 month. Plasma PLs was assayed by measuring its inorganic phosphorus after separation by chromatograph.Results 39 cases in 87 PD patients(44.8%)were with PD-MCI.After 12 months or 18 months treatment period, MoCA scores was significantly higher in Bushen-Huoxue compound treatment group than in controls(P<0.05 or 0.01 respectively). Plasma PLs levels were significantly decreased in Bushen-Huoxue compound treatment group than in controls(P<0.01 or 0.01 respectively). Conclusions Bushen-Huoxue compound could be effective in the adjuvant treatment of PD-MCI, and could delay cognitive rate of decline in patients with PD. Meanwhile, Bushen-Huoxue compound could significantly decrease plasma PLs levels in PD-MCI. The evidence from present study suggested that Bushen-Huoxue compound have neuroprotective effects on patients with PD-MCI.

Objective To discuss the effects of BuShenHuoXue on ovarian granulose cell apoptosis of DOS rats induced by GTW. Methods Sixty 12 -week -old female SD rats were randomly divided into 6 groups:control, model GTW,low dose of BuShenHuoXue Formula, middle dose of BuShenHuoXue Formula, high dose of BuShenHuoXue Formula, and LiuWeiDiHuang soup group. Object To study the feasibility of flocculation procedure on Bushenhuoxue Decoction, using chitosan instead of alcohol. Methods Using chitosan clarifier to the flocculation procedure of Bushenhuoxue Decoction; selecting the optimum content of chitosan and the density of decoction according to the total extract, and the content of icariin, comparing with the flocculation procedure by alcohol sedimentation. Results Chitosan clarifier could make the decoction clarify as alcohol does. On the retaining of active compounds, the former was better than the later. Conclusion Chitosan can be used to the flocculation procedure of Bushenhuoxue Decoction instead of alcohol.
