Abstract Fish is the main source of animal protein for human diet. The aim of this study was to find out prevalence of pathogenic bacteria of two selected economically important fish of Pakistan namely Mahseer (Tor putitora) and Silver carp (Hypophthalmichthys molitrix). Live fish samples from hatcheries and dead fish samples from different markets of study area were randomly collected. The fish samples were analyzed for isolation, identification and prevalence of bacteria. The isolated bacteria from study fish were identified through biochemical test and about 10 species of pathogenic bacteria were identified including the pathogenic bacteria to human and fish namely, Pseudomonas aeruginosa, Escherichia coli, Staphylococcus aureus, Staphylococcus epidermidis, Streptococcus iniae, Serratia spp. Citrobacter spp. Stenotrophomonas spp. Bacillus spp. and Salmonella spp. The bacterial percentage frequency of occurrence in Silver carp and Mahseer fish showed Pseudomonas aeruginosa 21.42%, Staphylococcus epidermidis 17.85%, Escherichia coli 11.90%, Staphylococcus aureus 9.52%, Citrobacter spp. 9.52%, Serratia spp. 8.33%, Streptococcus iniae 7.14%, Stenotrophomonas spp. 5.95%, Bacillus spp. 4.76% and Salmonella spp. 3.57%. The study revealed that Fish samples of Mahseer and Silver carp that were collected from markets have found more isolates (10 bacterial species) than did the fresh fish pond samples (03 bacterial species) of hatcheries. The occurrence of pathogenic bacteria in study fish showed risk factor for public health consumers.


Resumo O peixe é a principal fonte de proteína animal para a alimentação humana. O objetivo deste estudo foi descobrir a prevalência de bactérias patogênicas de dois peixes economicamente importantes selecionados do Paquistão, nomeadamente Mahseer (Tor putitora) e carpa prateada (Hypophthalmichthys molitrix). Amostras de peixes vivos de incubatórios e amostras de peixes mortos de diferentes mercados da área de estudo foram coletadas aleatoriamente. As amostras de peixes foram analisadas quanto ao isolamento, identificação e prevalência de bactérias. As bactérias isoladas dos peixes do estudo foram identificadas através de testes bioquímicos e cerca de 10 espécies de bactérias patogênicas foram identificadas incluindo as bactérias patogênicas para humanos e peixes, nomeadamente, Pseudomonas aeruginosa, Escherichia coli, Staphylococcus aureus, Staphylococcus epidermidis, Streptococcus iniae, Serratia spp. Citrobacter spp. Stenotrophomonas spp. Bacillus spp. e Salmonella spp. A porcentagem de freqüência de ocorrência bacteriana em carpa prateada e peixes Mahseer mostrou Pseudomonas aeruginosa 21,42%, Staphylococcus epidermidis 17,85%, Escherichia coli 11,90%, Staphylococcus aureus 9,52%, Citrobacter spp. 9,52%, Serratia spp. 8,33%, Streptococcus iniae 7,14%, Stenotrophomonas spp. 5,95%, Bacillus spp. 4,76% e Salmonella spp. 3,57%. O estudo revelou que as amostras de peixes de Mahseer e carpa prateada coletadas nos mercados encontraram mais isolados (10 espécies bacterianas) do que as amostras de peixes frescos (03 espécies bacterianas) de incubatórios. A ocorrência de bactérias patogênicas nos peixes do estudo apresentou fator de risco para consumidores de saúde pública.

Fluoride is the organic and inorganic compound of the element fluorine, has detrimental effect to all animals including fish at higher concentration. Ground water with elevated concentration of fluoride is a major global problem including India. On the other hand, Morinda citrifolia (also called ‘Noni’ in Polynesia) is a medicinal plant, has antioxidative, anti-inflammatory and immunostimulatory properties. The ameliorating effect of M. citrifolia fruit extract dietary supplementation against fluoride induced toxicity on the growth performance, haematological parameters, and fluoride accumulation in the muscular tissue was investigated in the freshwater fish, Cyprinus carpio L. for 35 days in triplicate. One hundred and twenty acclimated C. carpio fingerlings were randomly allocated into five experimental groups (Groups I, II, III, IV and V) each with 24 fish divided into three replicates (eight fish/replicate). Group I (Control) and II (Toxic control or negative control) were provided with basal/control diet, whereas groups III, IV, and V were provided with 0.25% (2.5 g/kg), 0.50% (5.0 g/kg) and 0.75% (7.5 g/kg) M. citrifolia fruit extract supplemented diet respectively. All groups except for group I were exposed to 10% of the LC50 of fluoride. Selected growth and haematological parameters were estimated. The fluoride content in the muscular tissue of fish was also estimated following SPADNS method. Results revealed that exposure of sublethal concentration of fluoride significantly (P <0.05) reduced the growth performance and haematological parameters in Gr. II (toxic control) compared to the Gr. I (control). However, M. citrifolia dietary supplementation significantly (P <0.05) improved fluoride induced alteration of growth performance and haematological parameters of fish in the groups III, IV and V compared to Gr. II. Morinda citrifolia fruit extract dietary supplementation also significantly (P <0.05) decreased the accumulation fluoride in the muscular tissue of fish in groups III, IV and V compared to the Gr. II (toxic control). In conclusion, M. citrifolia dietary supplementation ameliorates the fluoride toxicity by preventing the accumulation of fluoride in the muscular tissue of the fish. The M. citrifolia dietary supplementation at 0.25% showed the best response, hence it is considered as the optimum dose.

Abstract Aeromonas hydrophila is a cause of infectious disease outbreaks in carp species cultured in South Asian countries including Pakistan. This bacterium has gained resistance to a wide range of antibiotics and robust preventive measures are necessary to control its spread. No prior use of fish vaccines has been reported in Pakistan. The present study aims to develop and evaluate inactivated vaccines against local strain of A. hydrophila in Pakistan with alum-precipitate as adjuvant. The immunogenic potential of vaccine was evaluated in two Indian major carps (Rohu: Labeo rohita, Mori: Cirrhinus mrigala) and a Chinese carp (Grass carp: Ctenopharyngodon idella). Fish were vaccinated intraperitoneally followed by a challenge through immersion. Fish with an average age of 4-5 months were randomly distributed in three vaccinated groups with three vaccine concentrations of 108, 109 and 1010 colony forming unit (CFU)/ml and a control group. Fixed dose of 0.1ml was applied to each fish on 1st day and a booster dose at 15 days post-vaccination (DPV). Blood samples were collected on 14, 28, 35, 48 and 60 DPV to determine antibody titers in blood serum using compliment fixation test (CFT). Fish were challenged at 60 DPV with infectious A. hydrophila with 108 CFU/ml through immersion. Significantly higher levels of antibody titers were observed from 28 DPV in all vaccinated groups as compared to those in the control group. In challenge experiment the average RPS (relative percent survivability) was 71% for groups vaccinated with 109 and 1010 CFU/ml and 86% for 108 CFU/ml. Vaccine with 108 CFU/ml induced highest immune response followed by 109 and 1010 CFU/ml. The immune response of L. rohita and C. idella was better than that of C. mrigala. In general, normal histopathology was observed in different organs of vaccinated fish whereas minor deteriorative changes were found in fish vaccinated with higher concentrations of the vaccine.

Essa bactéria ganhou resistência a uma ampla gama de antibióticos, e medidas preventivas robustas são necessárias para controlar sua disseminação. Nenhum uso anterior de vacinas para peixes foi relatado no Paquistão. O presente estudo tem como objetivo desenvolver e avaliar vacinas inativadas contra cepa local de A. hydrophila no Paquistão com precipitado de alúmen como adjuvante. O potencial imunogênico da vacina foi avaliado em duas carpas principais indianas (Rohu: Labeo rohita, Mori: Cirrhinus mrigala) e uma carpa chinesa (Grass Carp: Ctenopharyngodon idella). Os peixes foram vacinados por via intraperitoneal, seguido de um desafio por imersão. Peixes com idade média de 4-5 meses foram distribuídos aleatoriamente em três grupos vacinados com três concentrações de vacina de 108, 109 e 1010 unidades formadoras de colônias (UFC) / ml e um grupo de controle. Os peixes foram desafiados a 60 DPV com infecciosa A. hydrophila com 108 CFU / ml por imersão. Níveis significativamente mais elevados de títulos de anticorpos foram observados em 28 DPV em todos os grupos vacinados, em comparação com aqueles no grupo de controle. Na experiência de desafio, o RPS médio (sobrevivência percentual relativa) foi de 71% para os grupos vacinados com 109 e 1010 CFU / ml e 86% para 108 CFU / ml. Abstract The present investigation was aimed to examine the concentrations of trace metals including e.g copper (Cu), manganese (Mn), nickel (Ni), and zinc (Zn) in water samples collected from nursery pond of grass carp (Ctenopharyngodon idella) in Bannu Hatchery of Khyber Pakhtunkhwa during the period from April 2018 to January 2019. The temperature and pH of each water sample were measured for the whole study duration. The concentration of Copper (Cu), Manganese (Mn), Nickel (Ni), and Zinc (Zn) in collected water samples were measured in mg/liter by using Atomic Absorption Spectrophotometer. The blank and standard solutions for device calibration Standard solutions i.e., 2.0 mg, 4.0 mg, and 6.0 were used to measure the concentration of these metals in water samples to verify the measurements. The data was statistically analyzed on descriptive statistics (estimation of proportions and standard deviation) used to summarize mean concentration. The results obtained of both temperature and pH of water samples were found in ranged 10 to 36 0C and 7.0 to 8.44; whereas the size of fry stages was ranged from 4.0 to 56.0 mm in total length. The results of investigated metals found in pond water samples are in order of Zn>Mn>Ni>Cu, respectively. As optimum temperature and pH for grass carp were mostly between 15 0C and 30 0C and pH 6.5 to 8.0. It was concluded from obtained results that temperature, pH, and trace metals were found appropriate for the growth of Ctenopharyngodon idella from fry to fingerling stages, but the highest amount of zinc can cause its mortality. It is a preliminary study on grass carp culturing in Bannu fish hatchery so, it would provide useful information for model fish seed production unit in a hatchery.


Resumo A presente investigação teve como objetivo examinar as concentrações de metais traço, incluindo cobre (Cu), manganês (Mn), níquel (Ni) e zinco (Zn) em amostras de água coletadas de viveiro de carpa-capim (Ctenopharyngodon idella) em Incubatório de Bannu de Khyber Pakhtunkhwa durante o período de abril de 2018 a janeiro de 2019. A temperatura e o pH de cada amostra de água foram medidos durante todo o período do estudo. A concentração de cobre (Cu), manganês (Mn), níquel (Ni) e zinco (Zn) nas amostras de água coletadas foi medida em mg / litro usando espectrofotômetro de absorção atômica. As soluções em branco e padrão para as soluções padrão de calibração do dispositivo, ou seja, 2,0 mg, 4,0 mg e 6,0, foram usadas para medir a concentração desses metais em amostras de água para verificar as medições. É um estudo preliminar sobre a cultura da carpa-capim em incubatório de peixes Bannu, portanto forneceria informações úteis para modelo de unidade de produção de sementes de peixes em um incubatório.

Concluiu-se, a partir dos resultados obtidos, que temperatura, pH e traços de metais foram considerados adequados para o crescimento de Ctenopharyngodon idella desde os estágios de alevinos até alevinos, mas a maior quantidade de zinco pode causar sua mortalidade. É um estudo preliminar sobre a cultura da carpa-capim em incubatório de peixes Bannu, portanto forneceria informações úteis para modelo de unidade de produção de sementes de peixes em um incubatório.

Abstract Aeromonas hydrophila is a cause of infectious disease outbreaks in carp species cultured in South Asian countries including Pakistan. This bacterium has gained resistance to a wide range of antibiotics and robust preventive measures are necessary to control its spread. No prior use of fish vaccines has been reported in Pakistan. The present study aims to develop and evaluate inactivated vaccines against local strain of A. hydrophila in Pakistan with alum-precipitate as adjuvant. The immunogenic potential of vaccine was evaluated in two Indian major carps (Rohu: Labeo rohita, Mori: Cirrhinus mrigala) and a Chinese carp (Grass carp: Ctenopharyngodon idella). Fish were vaccinated intraperitoneally followed by a challenge through immersion. Fish with an average age of 4-5 months were randomly distributed in three vaccinated groups with three vaccine concentrations of 108, 109 and 1010 colony forming unit (CFU)/ml and a control group. Fixed dose of 0.1ml was applied to each fish on 1st day and a booster dose at 15 days post-vaccination (DPV). Blood samples were collected on 14, 28, 35, 48 and 60 DPV to determine antibody titers in blood serum using compliment fixation test (CFT). Fish were challenged at 60 DPV with infectious A. hydrophila with 108 CFU/ml through immersion. Significantly higher levels of antibody titers were observed from 28 DPV in all vaccinated groups as compared to those in the control group. In challenge experiment the average RPS (relative percent survivability) was 71% for groups vaccinated with 109 and 1010 CFU/ml and 86% for 108 CFU/ml. Vaccine with 108 CFU/ml induced highest immune response followed by 109 and 1010 CFU/ml. The immune response of L. rohita and C. idella was better than that of C. mrigala. In general, normal histopathology was observed in different organs of vaccinated fish whereas minor deteriorative changes were found in fish vaccinated with higher concentrations of the vaccine.


Resumo Aeromonas hydrophila é uma causa de surtos de doenças infecciosas em espécies de carpas cultivadas em países do sul da Ásia, incluindo o Paquistão. Essa bactéria ganhou resistência a uma ampla gama de antibióticos, e medidas preventivas robustas são necessárias para controlar sua disseminação. Nenhum uso anterior de vacinas para peixes foi relatado no Paquistão. O presente estudo tem como objetivo desenvolver e avaliar vacinas inativadas contra cepa local de A. hydrophila no Paquistão com precipitado de alúmen como adjuvante. O potencial imunogênico da vacina foi avaliado em duas carpas principais indianas (Rohu: Labeo rohita, Mori: Cirrhinus mrigala) e uma carpa chinesa (Grass Carp: Ctenopharyngodon idella). Os peixes foram vacinados por via intraperitoneal, seguido de um desafio por imersão. Peixes com idade média de 4-5 meses foram distribuídos aleatoriamente em três grupos vacinados com três concentrações de vacina de 108, 109 e 1010 unidades formadoras de colônias (UFC) / ml e um grupo de controle. Foi aplicada dose fixa de 0,1ml em cada peixe no 1º dia e dose de reforço 15 dias pós-vacinação (DPV). Amostras de sangue foram coletadas em 14, 28, 35, 48 e 60 DPV para determinar os títulos de anticorpos no soro sanguíneo usando o teste de fixação de elogio (CFT). Os peixes foram desafiados a 60 DPV com infecciosa A. hydrophila com 108 CFU / ml por imersão. Níveis significativamente mais elevados de títulos de anticorpos foram observados em 28 DPV em todos os grupos vacinados, em comparação com aqueles no grupo de controle. Na experiência de desafio, o RPS médio (sobrevivência percentual relativa) foi de 71% para os grupos vacinados com 109 e 1010 CFU / ml e 86% para 108 CFU / ml. A vacina com 108 UFC / ml induziu a maior resposta imune seguida por 109 e 1010 UFC / ml. A resposta imune de L. rohita e C. idella foi melhor do que a de C. mrigala. Em geral, histopatologia normal foi observada em diferentes órgãos de peixes vacinados, enquanto pequenas alterações deteriorantes foram encontradas no grupo de controle e nos peixes vacinados com concentrações mais altas da vacina.

Abstract The present investigation was aimed to examine the concentrations of trace metals including e.g copper (Cu), manganese (Mn), nickel (Ni), and zinc (Zn) in water samples collected from nursery pond of grass carp (Ctenopharyngodon idella) in Bannu Hatchery of Khyber Pakhtunkhwa during the period from April 2018 to January 2019. The temperature and pH of each water sample were measured for the whole study duration. The concentration of Copper (Cu), Manganese (Mn), Nickel (Ni), and Zinc (Zn) in collected water samples were measured in mg/liter by using Atomic Absorption Spectrophotometer. The blank and standard solutions for device calibration Standard solutions i.e., 2.0 mg, 4.0 mg, and 6.0 were used to measure the concentration of these metals in water samples to verify the measurements. The data was statistically analyzed on descriptive statistics (estimation of proportions and standard deviation) used to summarize mean concentration. The results obtained of both temperature and pH of water samples were found in ranged 10 to 36 0C and 7.0 to 8.44; whereas the size of fry stages was ranged from 4.0 to 56.0 mm in total length. The results of investigated metals found in pond water samples are in order of Zn>Mn>Ni>Cu, respectively. As optimum temperature and pH for grass carp were mostly between 15 0C and 30 0C and pH 6.5 to 8.0. It was concluded from obtained results that temperature, pH, and trace metals were found appropriate for the growth of Ctenopharyngodon idella from fry to fingerling stages, but the highest amount of zinc can cause its mortality. It is a preliminary study on grass carp culturing in Bannu fish hatchery so, it would provide useful information for model fish seed production unit in a hatchery.


Resumo A presente investigação teve como objetivo examinar as concentrações de metais traço, incluindo cobre (Cu), manganês (Mn), níquel (Ni) e zinco (Zn) em amostras de água coletadas de viveiro de carpa-capim (Ctenopharyngodon idella) em Incubatório de Bannu de Khyber Pakhtunkhwa durante o período de abril de 2018 a janeiro de 2019. A temperatura e o pH de cada amostra de água foram medidos durante todo o período do estudo. A concentração de cobre (Cu), manganês (Mn), níquel (Ni) e zinco (Zn) nas amostras de água coletadas foi medida em mg / litro usando espectrofotômetro de absorção atômica. As soluções em branco e padrão para as soluções padrão de calibração do dispositivo, ou seja, 2,0 mg, 4,0 mg e 6,0, foram usadas para medir a concentração desses metais em amostras de água para verificar as medições. Os dados foram analisados estatisticamente em estatística descritiva (estimativa de proporções e desvio padrão) usada para resumir a concentração média. Os resultados obtidos tanto de temperatura quanto de pH das amostras de água variaram de 10 a 36 0C e 7,0 a 8,44, enquanto o tamanho dos estágios de alevinos variou de 4,0 a 56,0 mm no comprimento total. Os resultados dos metais investigados encontrados em amostras de água de lagoas são da ordem de Zn > Mn > Ni > Cu, respectivamente. A temperatura e o pH ótimos para a carpa-capim foram principalmente entre 15 0C e 30 0C e pH 6,5 a 8,0. Concluiu-se, a partir dos resultados obtidos, que temperatura, pH e traços de metais foram considerados adequados para o crescimento de Ctenopharyngodon idella desde os estágios de alevinos até alevinos, mas a maior quantidade de zinco pode causar sua mortalidade. Fluoride is a trace element which has beneficial effect at lower concentration but detrimental at higher concentration. The fluoride contamination in ground water is a major global problem. Similarly, aluminium is the most abundant metal of the earth, highly toxic to all organisms. In the present study, we investigated the acute toxicity of fluoride and aluminium on three month old fingerlings of Cyprinus carpio L. in hard water (Hardness 125 mg/L of CaCO3) under static renewal bioassay. The 96 h LC50 value of the fluoride and aluminium were 675.615 and 224.214 mg/L, respectively. There was a significant correlation (P <0.01) between mortality rate of C. carpio with all concentrations of fluoride as well as aluminium. The exposure time (24, 48, 72 and 96 h) and different concentration of fluoride (680, 684, 686 and 692 mg/L) and aluminium (228, 229, 231 and 232 mg/L) was also significantly correlated (P <0.05). The different abnormal behaviour displayed by the fluoride and aluminium exposed fishes were erratic swimming movements, rapid opercular activity and excessive secretion of mucous. The safe level of concentrations of fluoride and aluminium were 6.75 and 2.24 µg/L, respectively. The LC50 values of fluoride and aluminium of the present study may be useful in deriving water quality standards in West Bengal.

Catla catla (Hamilton) is one of the fastest growing Indian major carps (IMCs) with increasing market demand. Consistent demand and exploitation invites the attention of ichthyologists for its conservation strategies. Age and growth studies play a pivotal role for managing the fishery stocks in different water bodies. Here, we studied the age and growth patterns using opercula of both farm grown as well as the wild grown C. catla adopting standard methodologies. Wild specimens were caught from Harike wetland (Ramsar site) and the cultivated one from a farm at Dhudike in Punjab. High value of correlation coefficient ‘r’ 0.976 (wetland) and 0.983 (farm) reveals the strong relationship in total length and operculum radius of the fish. C. catla achieved average total length from wetland and farm, respectively at 1st (273.44 mm) 2nd (427.44 mm) 3rd (525.49 mm) 4th (624.52 mm) 5th (744.98 mm) 6th (813.62 mm) and 1st (282.1 mm) 2nd (463 mm) 3rd (601.7 mm) year of age of opercular bones study. The growth parameters such as, index of species average size (?h) 135.6 (wetland) and 200.6 (farm), growth constant (Clt) showed two growth phases from both localities i.e. sexual immaturity up to 2 years and sexual maturity afterwards. Whereas, growth characteristic (Cth) has revealed irregular growth pattern at wetland in comparison to growth of fish from farm. The results of this study concluded that the fish experienced more growth from the farm due to better and controlled conditions but, in wetland surrounding conditions were observed to be unfavourable for survival of the fish C. Catla.

Traditionally, bamboo has immense medicinal values owing to their rich bioactive compounds. On the other hand, scientific developments in global aquaculture have revealed the significance of dietary nutraceuticals in aquaculture. In this study, efforts have been made to evaluate the properties of shoot and leaf extracts of Melocanna baccifera (Roxb.) Kurz (Muli bamboo) and demonstrated its immunomodulatory potential. At first, four different extracts of bamboo leaf and shoot were prepared through water and ethanolic solvents followed by phytochemical profiling, antioxidant and antimicrobial activities of extracts. Based on the in vitro evaluation, BLAL (bamboo leaf alcoholic) extract was chosen for further in vivo evaluation. Second experiment was carried to assess the toxicity of BLAL extract on the Indian major carp, Labeo rohita (Hamilton), locally called ‘rohu’. No mortality was observed up to 20 g of extract kg-1 body weight. Additionally, haemolytic assay was also conducted to ascertain the cellular toxicity of extract. Third experiment was conducted to find out the effect of dietary BLAL extract [doses: control (0.0%), T1 (0.01%), T2 (0.1%) and T3 (1%) of extract kg-1 feed] on immune related genes (HSP70, IL-1? and TNF-?) expression in L. rohita. The mean value of total weight of hatchery fishes 345±48.68 and the mean value of brain weight of hatchery reared fishes 0.28±0.047. The mean value of wild fish's total body weight 195.16±52.58 and the mean value of brain weight of wild fishes are 0.45±0.14. Present research calls for the fact that fish in dependent environmental conditions possess brain larger in size as compared to its captive population, it is due to use and disuse of brain in their environmental requirements.

ABSTRACT: The effect of sunflower oil (SO), sunflower oil-pomegranate sauce (PS1) and sunflower oil-plum sauce (PS2) on shelf life of marinated carp fillets was investigated in terms of sensory evaluation, chemical (peroxide value (POV), thiobarbituric acid (TBA), free fatty acids (FFA), total volatile basic nitrogen (TVB-N) and pH), and microbiological properties (Total viable count (TVC), Escherichia coli, Total coliform, Staphylococci/Micrococci, Salmonella spp.) during 30 days of storage. Proximate composition, which includes total crude protein%, fat%, moisture% and ash% of fresh carp fillets before marination process was analyzed and found 18.69±0.86, 4.08±0.19, 74.33±0.63 and 2.17±0.45, respectively. Sensory evaluation analysis showed that total appearance, odor-taste and texture scores decreased during storage. TVB-N significantly increased in all group after 1 month of storage (P<0.05). At the end of storage, the lowest TBA value (1.38 mg MA/kg carp fillet) was determined in the PS1 group. During storage, the highest POV value was observed in the control group (8.49 mEq O2/kg) compared to other groups. Initial TVC of 1.5 log CFU/g, 1.7 log CFU/g and 1.8 log CFU/g increased to 3.7 log CFU/g, 2.9 log CFU/g and 3.2 log CFU/g in SO, PS1 and PS2 groups, respectively. Results showed that the shelf life of marinated carp fillets treated with sunflower oil and in combination with pomegranate and plum sauce was more than 1 month.


RESUMO: O efeito do óleo de girassol (SO), molho de óleo de girassol-romã (PS1) e molho de óleo de girassol-ameixa (PS2) na vida útil de filetes de carpa marinados foi investigado em termos sensorial, químico (valor de peróxido (PV), ácido tiobarbitúrico) (TBA), ácidos graxos livres (AGL), nitrogênio básico volátil total (TVB-N) e pH) e propriedades microbiológicas (contagem viável total (TVC), Escherichia coli, coliforme total, estafilococos/Micrococos, Salmonella spp.). Durante 30 dias de armazenamento. A composição aproximada, que inclui % de proteína bruta total, % de gordura, % de umidade e % de cinzas de filés de carpa frescos antes do processo de marinação foi analisada e encontrada 18,69 ± 0,86, 4,08 ± 0,19, 74,33 ± 0,63 e 2,17 ± 0,45, respectivamente. A análise sensorial mostrou que os escores totais de aparência, odor e sabor diminuíram durante o armazenamento. TVB-N aumentou significativamente (P <0,05) em todos os grupos após 1 mês de armazenamento. Abstract Plant based protein sources are one of the best, cost effective and easily available protein sources being used in fish feed. But due to a lower number of micro-biota in fish gut plant meal based diets cannot be digested and absorbed well in fish body. Probiotics were supplemented at 0, 1, 2, 3, 4 and 5 gkg-1 levels in fish feed for formulating one control and five test diets. In this study, three replicates of each treatment were used and number of fingerlings was 15 in each replicate. The C. carpio (common carp) fingerlings were fed at 5% of live wet weight on their prescribed diet twice daily. The results revealed that supplementation of probiotics in corn gluten meal based diets significantly (p<0.05) improved growth performance, carcass composition and hematological parameters. Most optimum values of growth performance parameters were noted at 2 gkg-1 level of probiotics supplemented diet. C. carpio fingerlings fed corn gluten meal based diet supplemented with 2 gkg-1 level of probiotics indicated significant (p<0.05) improvements in crude protein (17g) crude fat (9g) and gross energy (3 kcalg-1) whereas higher red blood cells (RBCs), white blood cells (WBCs) and hemoglobin (Hb) was also recorded in fish blood when fed 2 gkg-1 probiotics level diet. From these results, it was concluded that 2 gkg-1 probiotics supplementation in corn gluten meal based diet is optimum for improving growth performance, body composition and hematology of C. carpio fingerlings.

RESUMEN

In order to study the effect of light with different wavelengths on the motion behavior of carp robots, phototaxis experiment, anatomical experiment, light control experiment and speed measurement experiment were carried out in this study. Blue, green, yellow and red light with different wavelength were used to conduct phototaxis experiments on carp to observe their movement behavior. By dissecting the skull bones of the carp to determine the appropriate location to carry the light control device, we independently developed a light control carrying device which was suitable for any illumination intensity environment. The experiment of the light-controlled carp robots was carried out. The motion behavior of the carp robot was checked by using computer binocular stereo vision technology. The motion trajectory of the carp robot was tracked and obtained by applying kernel correlation filter (KCF) algorithm. The motion velocity of the carp robot at different wavelengths was calculated according to their motion trajectory. The results showed that carps' sensitivity to different light changed from strong to weak in the order of blue, red, yellow and green, so that using light with different wavelengths to control the speed of the carp robot has certain laws to follow. A new method to avoid brain damage in carp robots control can be provided in this study.

In order to accurately implant the brain electrodes of carp robot for positioning and navigation, the three-dimensional model of brain structure and brain electrodes is to be proposed in the study. In this study, the tungsten electrodes were implanted into the cerebellum of a carp with the aid of brain stereotaxic instrument. The brain motor areas were found and their three-dimensional coordinate values were obtained by the aquatic electricity stimulation experiments and the underwater control experiments. The carp brain and the brain electrodes were imaged by 3.0 T magnetic resonance imaging instrument, and the three-dimensional reconstruction of carp brain and brain electrodes was carried out by the 3D-DOCTOR software and the Mimics software. The results showed that the brain motor areas and their coordinate values were accurate. The relative spatial position relationships between brain electrodes and brain tissue, brain tissue and skull surface could be observed by the three-dimensional reconstruction map of brain tissue and brain electrodes which reconstructed the three-dimensional structure of brain. The anatomical position of the three-dimensional reconstructed brain tissue in magnetic resonance image and the relationship between brain tissue and skull surface could be observed through the three-dimensional reconstruction comprehensive display map of brain tissue. The three-dimensional reconstruction model in this study can provide a navigation tool for brain electrodes implantation.

Cirrhinus mrigala (Hamilton, 1822) is an Indian major carp originally inhabitant of the Ganga river system in Northern India. No information is available on morphometric variation in natural populations of C. mrigala from Ganga river basin. Therefore, to promote management optimization, this study was undertaken to evaluate the population structure on landmark-based morphometric variations of this commercially important species in Ganges. A total of 381 fish samples of C. mrigala were collected from 10 locations of 10 rivers belonging to Ganges basin. A 12-landmark truss network system was used to generate 66 morphometric variables were transformed (standard length was excluded from the final analysis retaining 65 variables). Transformed measurements were employed to univariate analysis of variance, principal component analysis (PCA) and discriminant function analysis (DFA). Univariate analysis of variance showed significant differences in all the 65 transformed morphometric characters studied. Eight principal components were extracted from 65 significant characters accounting for 94.1 % of the variation. Forward stepwise discriminant function analysis of the 65 significant variables produced eight discriminating variables. DFA extracted eight discriminating functions contributed overall to the variance. The first discriminant function (DF) accounted for 37.9 % while second DF accounted for 23.5% of the among-group variability. In DFA, 45.7 % original while 39.1 % of cross-validated grouped cases were correctly classified in the overall assignment of individuals, indicating low distinction among the stocks of all the species i.e. low isolation of the stocks for the morphometric measurements. Common ancestry in the prehistoric period and migration of individuals between the rivers could be the possible reason for the observed low level of morphometric differentiation among wild mrigal populations. In future, the application of molecular genetic markers along with morphometric studies would be an effective method to examine the genetic component of phenotypic relatedness between geographic regions and to facilitate the development of management recommendations.


Cirrhinus mrigala es una carpa originaria del sistema fluvial del río Ganges al norte de India, de cual no hay información sobre la variación morfométrica de sus poblaciones naturales en esta cuenca. Por lo tanto, para promover la optimización de su manejo, se evaluó la estructura poblacional usando morfometría basada en puntos de referencia de C. mrigala. En total se midieron 381 individuos de C. mrigala, recolectados en 10 puntos de 10 ríos de la cuenca del Ganges. Se utilizaron 12-puntos de referencia de sistema de redes truss para generar 66 variables morfométricas, de donde se excluyó la longitud estándar y se utilizaron 65 variables. Las medidas transformadas se utilizaron para un análisis de varianza univariado, análisis de componentes principales y análisis de función discriminante. El análisis de varianza mostró diferencias en todos los 65 caracteres morfométricos estudiados. Se extrajeron ocho componentes principales de los 65 caracteres, que suman el 94.1 % de la variación. El análisis de función discriminante de las 65 variables produjo ocho variables discriminantes que contribuyeron en la varianza a nivel general. La primera función discriminante representó 37.9 %, mientras que la segunda 23.5 % de la variación entre grupos. En el análisis de función discriminante, 39.1 % de lo agrupado por validación cruzada fue correctamente clasificado en la asignación general de individuos, lo que indica baja diferenciación entre las poblaciones, lo que puede ser un bajo aislamiento de las poblaciones para las medidas morfométricas. Otra posible razón para la baja diferenciación morfométrica es la existencia de un ancestro común en el periodo prehistórico y la migración de individuos entre ríos. En el futuro, la aplicación de marcadores genéticos moleculares junto con estudios morfométricos será un método efectivo para examinar el componente genético de la relación fenotípica entre áreas geográficas y facilitar el desarrollo de recomendaciones de manejo.
