RESUMEN

@#Objective To study the mechanism of extremely low frequency electromagnetic field (ELF EMF) promoting the recovery of neural function loss in ischemic stroke by regulating the miRNA23 (miR-23)/Loch signal pathway. Methods An adult male SD rat model of ischemic stroke was established. ELF EMF intervention was performed and NC miR sequence or miR-23 inhibitor was injected into the lateral ventricle. The degree of neurological deficit,the volume of cerebral infarction,the expression level of miR-23 and the expression level of Notch1,Jagged1 and Hes1 proteins were evaluated. Results Compared with the sham operation group,the Zea Longa score,cerebral infarction volume and apoptosis rate of the model group were increased,and the expression levels of miR-23,Notch1,Jagged1 and Hes1 were decreased (P<0.05);Compared with the model group,the Zea Longa score,cerebral infarction volume and apoptosis rate of ELF EMF group were decreased,and the expression levels of miR-23,Notch1,Jagged1 and Hes1 increased (P<0.05);Compared with miR-NC+ELF EMF group,the Zea Longa score,cerebral infarction volume and apoptosis rate of miR-23+ELF EMF group were increased,and the expression levels of miR-23,Notch1,Jagged1 and Hes1 were decreased (P<0.05). Conclusion ELF EMF can promote the recovery of neurological deficit in ischemic stroke rats,which is related to the increase of miR-23 expression and activation of downstream Notch1 pathway.

Objective To investigate the effect of extremely low frequency-electromagnetic field (ELF-EMF) on bone mesenchymal stem cells (BMSCs) differentiating into neuron like cells in vitro and research its mechanism.Methods BMSCs were collected from rats by means of whole bone marrow adherent.Flow cytometry was used to assay cell surface marker at passage 3.And then,BMSCs were assigned into four groups:ELF-EMF group,ELF-EMF+U0126 (inhibitor) group,U0126 group and control group;cells were induced by medium (2% DMSO and 200 μmol/L BHA) for 5 h.In the process of neural induction,ELF-EMF group and ELF-EMF+U0126 group were received 10 Hz,500 GS ELF-EMF stimulation.Besides,ELF-EMF+U0126 group and U0126 group were pretreated with 50 μmol/L extracellular signal-regulated kinase (ERK)1/2 inhibitor U0126.The morphology of BMSCs was observed under inverted microscope.The expression of nestin was detected by immunofluorescent staining and Western blotting to identify and determine the differentiation.Western blotting was applied to detect the preotein level of phosphorylase-ERK1/2 after ELF-EMF exposure.Results BMSCs presented a single long spindle morphology,growing with close whirlpoor-like arrangement;CD90 expression rate was up to 97.9%,while that of CD45 only 4.7%.After induction,each group of cells showed similar shape with neuron-like cells gradually.As compared with the other three groups,ELF-EMF group had significantly higher expression levels of nestin and phosphorylatd ERK1/2 detected by immunofluorescent staining and Westem blotting,respectively (P<0.05).Meanwhile,the expression levels of nestin among the ELF-EMF+U0126 group,U0126 group and control group were not statistically significant (P>0.05).Conclusion ELF-EMF could promote neuronal differentiation of mesenchymal stem cells via activation of ERK1/2 signaling pathways.

Se realizó un ensayo clinicoterapéutico aleatorizado y controlado en fase III en 52 pacientes con psoriasis vulgar, atendidos en la consulta de Dermatología del Hospital Clinicoquirúrgico Docente “Dr. Joaquín Castillo Duany” de Santiago de Cuba, del 2005 al 2010, quienes fueron asignados a 2 grupos (de 26 integrantes cada uno): el activo, que fue tratado convencionalmente con pomada cádica a 20 %; yel experimental, al que se le aplicó el campo electromagnético de extremada baja frecuencia, con vistas a determinar la eficacia de este último en los afectados durante los períodos de crisis e intercrisis. La población estudiada fue caracterizada clínicamente mediante un análisis de frecuencias, en tanto, se utilizaron las pruebas de hipótesis para la evaluación de proporciones, cuyas hipótesis nulas se aceptaron con 5 % de error como máximo. En la serie el campo empleado resultó eficaz para tratar la afección en la etapa de crisis y más eficaz que la crema cádica para prolongar la fase de intercrisis, sin producir reacciones adversas, por lo que constituyó una terapéutica alternativa novedosa, que puede ser generalizada en los servicios dermatológicos del país, a fin de mejorar la calidad de vida de los que padecen esa dermatosis.


A controlled and randomized clinical therapeutical assay in phase III was carried out in 52 patients with vulgar psoriasis, who were assisted in the Dermatology department of "Dr. Joaquín Castillo Duany" Clinical Surgical Hospital in Santiago de Cuba, from 2005 to 2010 who were divided into 2 groups (26 members each): the active group which was conventionally treated with 20% cade ointment; and the experimental group, to which the extremely low frequency electromagnetic field was applied, with the objective of determining the effectiveness of this field in those affected during the periods of crisis and intercrisis. The studied population was clinically characterized by means of a frequency analysis, while the hypothesis tests were used for the evaluation of proportions which null hypothesis were accepted with 5 % as maximum error. In the series the used field was effective to treat the disorder during crisis and more effective than the cade ointment to prolong the intercrisis stage, without causing adverse reactions, so that it constituted a new alternative therapy that can be generalized in the dermatological services of the country, in order to improve the life quality of those who suffer that dermatosis.

BackgroundThe effects of extremely low frequency electromagnetic fields (ELF-EMFs) on public health have attracted wide attentions.The association of the thermal effect of ELF-EMFs with cancer and ocular tissue damage has been of concern.However,the pathological changes of scleral tissue after exposure to ELF-EMFs as well as the relationship between these changes and myopia are still poorly understood.ObjectiveThe present study was to investigate the molecular pathological changes of human fetal scleral fibroblasts (HFSFs) after exposure to ELF-EMFs in vitro and to explore the possible mechanism in the occurrence and development of myopia.MethodsHFSFs were cultured and passaged and then exposed to 50 Hz electromagnetic fields,and HFSFs that did not receive the irradiation of ELF-EMFs were used as the control group.The expression of collagen type Ⅰ (COL1A1 ) mRNA and matrix metalloproteinase-2 (MMP-2) mRNA in cultured HFSFs were detected by real-time qualitative polymerase chain reaction (real-time PCR) under different magnetic field intensites (0,0.1,0.2,0.5,1.0 mT) and different exposure time (0,6,12,24,36,48 hours).Cell proliferation assay of HFSFs was detected by the cell counting kit 8 ( CCK8 ) assay.The expression levels of COL1 A1 and MMP-2 proteins in HFSFs were further confirmed by immunofluorescence staining.Results The expression of COL1A1 mRNA was significantly down-regulated under the exposure of 0.2 mT ELF-EMFs for 6 hours,in comparison with the control group;moreover,it decreased in parallel with the increased of flux density (0.099±0.008 vs.0.050±0.004) (P =0.009 ).The expression of MMP-2mRNA was up-regulated conspicuously after exposure to 0.1 mT ELF-EMFs for 24 hours,and it increased with exposure time in comparison with the control group ( 0.009 ±0.001 vs.0.018±0.003 ) ( P =0.038 ).Proliferation of HFSFs (A450) was inhibited following the exposure to 0.2 mT ELF-EMFs for 24 hours in comparison with the control group (P =0.009 ).The expression of COL1 A1 in the experimental group was decreased,compared with the control group,but the expression of MMP-2 was increased.ConclusionsELF-EMFs inhibit the proliferation of HFSFs and expression of COL1 A1 in HFSFs,which might be one of the reasons for the development of myopia.

Objective To investigate the mechanism of extremely low frequency electromagnetic field (ELF-EMF) and X-ray irradiation on liver carcinoma cell lines BEL-7402. Methods Liver carcinoma cell lines BEL-7402 had been incubated with ELF-EMF (100 Hz, 0.7 mT, 30 min, 3 days) after X-ray irradiation at different doses (0, 2, 4, 6, 8,10 Gy). The cells were observed on morphologic changes with scanning electron microscope. Flow cytometry and gene microarray were used to investigate the mechanism of cell apeptosis. Results ELF-EMF plus X-ray induced apoptosis on BEL-7402 was observed under scanning electron microscope.When X-irradiation was 2, 4, 6, 8 and 10 Gy, the apoptosis ratios of combined group and only X-irradiation group were 10.0%, 14.5%, 4.3%, 5.1%, 7.1% and 0.1%, 8.1%, 0.1%, 0.4%, 2.2% (P < 0.05) on flow cytometry. The result of microarray indicated that 1465 genes were up-regulated and 1108 down*regulated in the ELF-EMF plus X-ray group in comparison with the control group. The change rates of 110 apoptosis related genes were above 2 times, which including 71 up-regulated and 39 down-regulated. Gene microarray showed that ELF-EMF and X-ray irradiation had a mainly effect on different gene of apoptosis paths (CDC25 and CHKI, ATM, p38, PTEN, p53, G1/S, Fas, G2/M, Cell Cycle, Apoptosis and Caspase). The same genes of ELF-EMF plus X-ray group were showed 13 in ELF-EMF group and 42 in X-ray group. Conclusions Apoptosis paths were significantly different between ELF-EMF and X-irradiation. ELF-EMF cooperates with X-irradiation on inducing BEL-7402 cell apoptosis.

RESUMEN

With the scientific and technological progress and rapid development of the social economy, the extensive application of various electronic equipments has gradually intensified the possibility of ELF-EMF(extreme low frequency electromagnetic fields) exposure, the relationship between ELF-EMF exposure and health effects attracted more and more eyes and many corresponding researches have been finished. In recent years, a lot of epidemiological researches about the relationship between ELF-EMF and leukemia, brain tumor, breast cancer and other tumors have been done at home and abroad, in the present paper the research results were reviewed and the possible mechanism of the effect of ELF-EMF exposure on tumors was discussed.
