RESUMEN

Objective To investigate the expressions of tumor suppressor gene FEZ1 and hypoxia inducible factor (HIF)-1α in oral squamous cell carcinoma (OSCC), and their correlation with clinicopathological features and prognosis. Methods A total of 146 specimens of OSCC and 80 normal oral mucosa were used to detect the expressions of FEZ1 and HIF-1αby immunohistochemistry. Thirty specimens of fresh OSCC tissues, 30 samples of the paracancerous tissues and 30 normal mucosa tissues were also included in the study. The relationship between the expressions of FEZ1 and HIF-1α, the clinicopathological features and the prognosis of the patients were analyzed. Western blot assay was used to detect the expres?sions of FEZ1 and HIF-1αin 30 fresh OSCC tissues, 30 samples of the paracancerous tissues and 30 normal mucosa tissues. Results In OSCC tissues, the positive rate of FEZ1 protein (30.14%) was significantly lower than that in normal oral muco?sa (81.25%), and the positive rate of HIF-1αprotein (71.23%) was significantly higher than that in normal mucosa (12.50%,χ2=54.076 and 71.317, P<0.01). There was a correlation between the expression of FEZ1 and clinical stage, tumor size and differentiation. The expression of HIF-1αwas correlated with gender, lymph node metastasis, tumor size and differentiation. There was no significant difference in 5-year survival rate between positive and negative FEZ1 expression groups. The 5-year survival rate was significantly lower in HIF-1α positive expression group than that of HIF-1α negative expression group (P<0.05). The expression level of FEZ1 was decreased by the sequences in normal oral mucosa, paracancerous tissue and OSCC (P<0.01), but the expression level of HIF-1αwas increased in turn (P<0.01). Conclusion FEZ1 and HIF-1αmay be involved in the occurrence and progress of OSCC. The expressions of FEZ1 and HIF-1αcan be used as a reference for the biological behavior of OSCC.

RESUMEN

Objective To investigate the expressions and the relationship of FEZ1 and p16 in cervical intraepithelial neoplasia (CIN).Methods The expressions of FEZ1 and p16 in 93 cases of CIN and 10 cases of normal cervical specimens were detected by immunohistochemistry. Results The positive expression rates of FEZ 1 were 90.0% (9/10) of normal cervical specimens,67.9% (19/28) of CIN I,36.0% (9/25) of CIN Ⅱ,22.5%(9/40) of CIN Ⅲ.The positive expression rates of p16 were 10.0%(1/10) of normal cervical specimens,32.1%(9/28) of CIN I,76.0%(19/25) of CIN Ⅱ,92.5%(37/40) of CIN Ⅲ.There were significant differences in the positive expression rates of FEZ1 and p16 between CIN Ⅰ and CIN Ⅲ (P < 0.01),there was no significant difference in the positive expression rates of FEZ1 and p16 between CIN Ⅱ and CIN Ⅲ.The expression of FEZ1 and p16 was negatively correlation in CIN (r =-0.712,P< 0.01).Conclusion The abnormal high expression of p16 and abnormal low expression of FEZ1 in CIN may be involved in the occurrence and development of CIN,detecting the expressions of the two indexes may be helpful for clinical diagnosis.

RESUMEN

Aberrations on the short arm of chromosome 8 (8p) are frequently observed in several human cancers. In this study, 20 squamous cell carcinoma (SCC) specimens from the tongue were examined in order to evaluate the role of 8p in SCC of the tongue. Microsatellite analysis using 14 markers demonstrated two commonly deleted regions (CDRs) on 8p. Reverse transcription-polymerase chain reaction (RT-PCR) revealed frequent down-regulation of the FEZ1 gene, mapped to 8p22, and frequent over-expression of the cathepsin B gene, mapped to 8p-21-22. These results suggested that genetic aberrations are involved in the development of SCC of the tongue. However, no significant relationship was observed to be established between the genetic alterations and clinicopathological features. Thus, further investigation is necessary in order to clarify the clinical role of 8p in carcinoma of the tongue.
