RESUMEN

Aim To investigate the effects of gastrodin on SH-SY5Y cell autophagy induced by methamphet-amine (METH) and the underlying mechanisms. Methods SY5Y cells were treated by METH with the concentration of 0.5,1.0,1.5,2.0,2.5,3.0 mmol·L-1for 24 h. The morphological changes were ob-served by microscopy,the expression of LC3-Ⅱ,Bec-lin-1,Akt,p-Akt,mTOR and p-mTOR were detected by Western blot. Gastrodin was added to the medium 1 h before METH treatment. Results The SY 5 Y cells were morphologically featured by shrinkage and den-drite disruption after exposed to METH(0~3 mmol· L-1),and autophagic vacuoles occurred in cytoplasm. The expression of LC3-Ⅱ increased over METH dose. Confocal results showed that LC3-Ⅱsignificantly in-creased in METH group as compared with control, while decreased in METH+ Gastrodin group. The ex-pression levels of LC3-Ⅱand Beclin-1 significantly in-creased (P<0.01) in METH group, p-mTOR and p-Akt decreased, and mTOR and Akt showed no signifi-cant difference as compared with control. However, the gastrodin could decrease the expression of LC3-Ⅱand Beclin-1 and increase the expression of mTOR,p-mTOR,Akt and p-Akt as compared with METH-trea-ted groups. Conclusions METH can induce SY5Y cells autophagy. The protective effect of gastrodin a-gainst METH-induced autophag may be related to gast-rodin regulation mTOR and Akt signaling pathway.

BACKGROUND: Current serologic tests for Syphilis(STS) in the blood donors are Veneral Disease Reseach Laboratory(VDRL), Rapid Plasma Reagin(RPR) test or Groupamatic Automated Syphilis Test(GAST) using modified VDRL antigen as screening method, and Treponema Pallidum Hemagglutination(TPHA) test as a confirmatory method in Korean Red Cross Blood Centers. This study was carried out to evaluate the usefulness of Enzyme Immunoassay(EIA) as STS in blood donors. METHODS: A total of 11,335 donors s serum samples were tested by RPR and GAST. We analyzed 138 samples including 6 samples of anti-treponema pallidum panel with TPHA and EIA to compare as a confirmatory test. RESULTS: The positive rate of RPR and GAST in 11,335 samples were 0.68%, 0.24%, respectively. Confirmed positive rates by TPHA was 0.26%, and by EIA was 0.27%. False negative results of GAST were 0.11% and 0.13%, respectively according to the results of TPHA and EIA. The agreement between TPHA and EIA was 98.5%(130/132). CONCLUSION: The EIA results were comparable with RPR, GAST and TPHA test. It is considered that EIA method for STS would be alternative one for TPHA as a conformative test because there was excellent agreement between TPHA and EIA method, and EIA method showed almost same results as that of TPHA test.

RESUMEN

Objective To study the action of Shuganwan(SHGW) on experimental gastric ulcer in mice. Methods The anti-ga stric ulcer action of SHGW was observed on the gastric ulcer induced by water im mersion stress, reserpine and absolute ethyl alcohol. The effect on gastric secr etion in rats was studied by pyloric ligation. The effect on gastrointestinal mo tility was observed by gastric emptying in mice. The analgesic effect was tested by the hot-plate test and writhing method. Results SHGW m arkedly inhibited gastric ulcer induced by water immersion stress, reserpine and absolute ethyl alcohol. SHGW significantly inhibited the secretion of gastric a cid and pepsin and promoted the secretion of gastric mucus. SHGW markedly delaye d gastric emptying in normal mice and when gastric emptying was stimulated by ne ostigmine. SHGW mildly strengthend the effect of atropine on gastric emptying. S HGW enhanced hot pain threshold and decreased the number of twisting body in mic e. Conclusion Shuganwan may have anti-gastric ulcer ac tion. The mechanism of the action might be associated with its inhibition of the secretion of gastric juice, promotion of the secretion of gastric mucus,inhibit ion of activity of gastric smooth muscle and analgetic effect.
