RESUMEN

Recent innovations in nanomaterials inspire abundant novel tumor-targeting CRISPR-based gene therapies. However, the therapeutic efficiency of traditional targeted nanotherapeutic strategies is limited by that the biomarkers vary in a spatiotemporal-dependent manner with tumor progression. Here, we propose a self-amplifying logic-gated gene editing strategy for gene/H2O2-mediated/starvation multimodal cancer therapy. In this approach, a hypoxia-degradable covalent-organic framework (COF) is synthesized to coat a-ZIF-8 in which glucose oxidase (GOx) and CRISPR system are packaged. To intensify intracellular redox dyshomeostasis, DNAzymes which can cleave catalase mRNA are loaded as well. When the nanosystem gets into the tumor, the weakly acidic and hypoxic microenvironment degrades the ZIF-8@COF to activate GOx, which amplifies intracellular H+ and hypoxia, accelerating the nanocarrier degradation to guarantee available CRISPR plasmid and GOx release in target cells. These tandem reactions deplete glucose and oxygen, leading to logic-gated-triggered gene editing as well as synergistic gene/H2O2-mediated/starvation therapy. Overall, this approach highlights the biocomputing-based CRISPR delivery and underscores the great potential of precise cancer therapy.

RESUMEN

Polysomnography (PSG) monitoring is an important method for clinical diagnosis of diseases such as insomnia, apnea and so on. In order to solve the problem of time-consuming and energy-consuming sleep stage staging of sleep disorder patients using manual frame-by-frame visual judgment PSG, this study proposed a deep learning algorithm model combining convolutional neural networks (CNN) and bidirectional gate recurrent neural networks (Bi GRU). A dynamic sparse self-attention mechanism was designed to solve the problem that gated recurrent neural networks (GRU) is difficult to obtain accurate vector representation of long-distance information. This study collected 143 overnight PSG data of patients from Shanghai Mental Health Center with sleep disorders, which were combined with 153 overnight PSG data of patients from the open-source dataset, and selected 9 electrophysiological channel signals including 6 electroencephalogram (EEG) signal channels, 2 electrooculogram (EOG) signal channels and a single mandibular electromyogram (EMG) signal channel. These data were used for model training, testing and evaluation. After cross validation, the accuracy was (84.0±2.0)%, and Cohen's kappa value was 0.77±0.50. It showed better performance than the Cohen's kappa value of physician score of 0.75±0.11. The experimental results show that the algorithm model in this paper has a high staging effect in different populations and is widely applicable. It is of great significance to assist clinicians in rapid and large-scale PSG sleep automatic staging.

RESUMEN

The paper summarizes the definition, location and main indication of qicimai points recorded in Huangdi Neijing (Yellow Emperor 's Inner Classic). It is found that qicimai points are the "upward moving points" in reference to the meridian distribution rule of "rooting, running, infusing and moving"; and corresponding to the sites of "running outwards and inwards" of the meridians' "separating, meeting and running outwards and inwards". It also includes the infusing points for the sea of qi and marrow. The new idea, "selecting qicimai points for the treatment of qi obstruction in the neck gate", is proposed. Based on the systematic application of the acupoints on the nape region, it is anticipated that a new approach will be provided to the treatment of the diseases in the neck, shoulder, head, face, the five sensory organs, mental disorders and zangfu qi dysfunction.

Objective:To determine the expression of transglutaminase 2 (TGM2) in peripheral blood mononuclear cells (PBMCs) from patients with atopic dermatitis (AD), and to analyze its correlation with AD-related inflammatory factors and disease severity.Methods:A total of 29 AD patients and 15 healthy controls were collected from the First Affiliated Hospital of Fujian Medical University from July 2020 to January 2021. Ten milliliters of peripheral blood samples were collected from each subject, so was the clinical information, including age, gender, course of disease, eosinophil counts, basophil counts, total IgE levels, Scoring AD index (SCORAD), etc. PBMCs were isolated by density gradient centrifugation. Fluorescence-based quantitative PCR was performed to determine the mRNA expression of TGM2 and AD-related inflammatory factors (interleukin [IL]-1β, IL-4, IL-6, IL-8, IL-10, IL-13, IL-17, thymic stromal lymphopoietin [TSLP], P2RX7 [purinergic receptor P2X, ligand-gated ion channel, 7], etc.) in PBMCs from 29 AD patients and 15 healthy controls, and flow cytometry to determine TGM2 protein expression on PBMCs. Mann-Whitney U test was used to analyze differences between groups, and Spearman correlation analysis to evaluate the correlation. Results:The relative mRNA expression of TGM2 in PBMCs did not differ between the AD group and control group ( M[ Q1, Q3]: 0.509 [0.325, 0.958] vs. 0.475 [0.328, 1.051], U = 210.50, P = 0.872). Compared with the control group, the AD group showed significantly decreased IL-4 mRNA expression (0.171[0.049, 0.449] vs. 0.824 [0.397, 1.378], P < 0.001), but significantly increased mRNA expression of IL-8 and IL-13 ( P = 0.011, 0.006, respectively). Spearman correlation analysis showed that the mRNA expression level of TGM2 in PBMCs was positively correlated with the mRNA expression levels of IL-4 and P2RX7 in the AD group ( rs = 0.42, 0.40, P = 0.024, 0.034, respectively), while there were no correlations between TGM2 mRNA expression and AD severity-related indicators (all P>0.05), such as age (21[16, 29] years), course of disease (4[1,10] years), eosinophil counts (0.33[0.18, 0.65] × 10 9/L), basophil counts (0.04[0.03, 0.06] × 10 9/L], SCORAD scores (60.5[46.98, 66.13] points), and serum total IgE levels (373 [40, 1 815] IU/ml). The relative protein expression levels of TGM2 on the surface of PBMCs did not differ between the AD group and control group (54.9 [47.6, 62.8] vs. 55.55 [51.5, 60.25], U = 112.00, P = 0.922) ], and no correlations were observed between the protein expression of TGM2 on PBMCs and AD severity-related indicators in the AD group (all P > 0.05) . Conclusion:No significant differences were observed in TGM2 mRNA expression in PBMCs or TGM2 protein expression on the surface of PBMCs between the AD patients and healthy controls, and there were no correlations between the TGM2 mRNA and protein expression and AD severity.

Aims@#Expression of recombinant proteins across a range of different host organisms has profound contribution to the advancement in biotechnology. In this study, we aimed to construct a highly versatile broad host range (BHR) expression vector, designated as pYL101C.@*Methodology and results@#The Golden Gate cloning approach was used to construct pYL101C. Key features of pYL101C include a strong integron promoter (PINTc), a BHR pBBR1 origin of replication (ori), gentamycin resistance gene (GmR) as a selectable marker and a multiple cloning site (MCS) downstream of the promoter for easy-cloning purpose. To verify the functionality of pYL101C, we cloned the superfolder green fluorescent protein (sfGFP) reporter gene into pYL101C and transferred the resultant recombinant plasmid pYL101C::sfGFP into various Gram-negative bacteria. Transformants obtained stably expressed strong green fluorescence under blue light excitation even without selection after four passages. @*Conclusion, significance and impact of study@#The constructed BHR expression vector, pYL101C and recombinant pYL101C::sfGFP are stable and can be used to monitor the presence of Gram-negative bacteria, such as endophytes and pathogens in their hosts and environment.

RESUMEN

Objective To discuss the value of E/e′index measured by dual gate Dopper predicting recurrence after radio frequency catheter ablation ( RFCA ) in persistant atrial fibrillation ( PeAF) patients . Methods Fifty‐three patients with PeAF w ho had successful RFCA and 25 controls were prospectively enrolled . T he patients with PeAF were divided into AF recurrence group ( n =21) and AF non‐recurrence group ( n =32) with median follow‐up time of ( 25 .94 ± 2 .78) months . All patients with PeAF underwent echocardiography in the preoperative 7 days . With dual gate Doppler ,transmitral flow peak velocity ( E) and mitral annular septal or lateral peak velocity e′( S ) , and e′( L ) in early diastolic were measured simultaneously in the same cardiac cycle ,then E/e′( S ) and E/e′( L ) were automatically calculated .With traditional method ,E ,e′( S) and e′( L ) were measured in different cardiac cycles ,then E/e′( S) and E/e′( L ) were manually calculated .T he time of w hole analysis process with each method was recorded . Results PeAF patients had bigger E/e′( S ) and E/e′( L ) with both dual gate Doppler and traditional method than controls . Compared with those in controls and AF non‐recurrence group ,E/e′( S) and E/e′( L ) increased in AF recurrence group with both dual gate Doppler and traditional method ( all P < 0 .05 ) . No statistical difference was found between the data acquired by dual gate Doppler and traditional method ( all P >0 .05) . Cut‐off values of 10 .90 ( sensitivity of 71 .4% ,specificity of 87 .5% ) for E/e′( S) was obtained by dual gate Doppler and 10 .70 ( sensitivity of 81 .0% ,specificity of 62 .5% ) was obtained by traditional method for predicted AF recurrence . Areas under curves between the two methods in ROC analysis was not significant ( 0 .819 vs 0 .728 , P >0 .05) . T he w hole analysis time of dual gate Doppler was less than that of traditional method . Between inter‐and intraobservers ,the interclass correlation coefficient with dual gate Doppler was higher and 95% confidence interval range was smaller . Conclusions E/e′( S ) is a valuable predictor for PeAF recurrence after RFCA in patients with PeAF . Dual gate Doppler can noninvasively assess single‐beat E/e′with less analysis time and better reproducibility compared with traditional method .

Objective • To generate a doxycycline (Dox)-inducible multiplexed CRISPR interference (CRISPRi) system for multiple gene inhibition in human embryonic stem cells (hESCs) to explore the function of gene families and model multigene diseases. Methods • A Dox-inducible multiplexed CRISPRi system was developed by Golden Gate assembly in hESCs. This system consisted of two plasmids, one expressing modified repressive nuclease-deactivated CRISPR-associated protein 9 (dCas9) and Krüppel-associated box (KRAB) transcriptional repressor domain under the control of Dox, the other carrying eight independent guide RNA (gRNA) expression cassettes. PCR was conducted using total genomic DNA as a template to confirm whether these two plasmids were integrated into genome. Western blotting was performed to confirm whether the expression of dCas9-KRAB could be induced by Dox treatment. Results • Using this tunable CRISPRi system, multiple genes were successfully silenced simultaneously in hESCs. The silence of genes and related to hESC self-renewal caused obvious cell differentiation in terms of changed cell morphology, decreased activity of alkaline phosphatase, and reduced expression of stage-specific embryonic antigen 4 (SSEA4), a marker of undifferentiated hESCs. Conclusion • This Dox-inducible multiplexed CRISPRi system can be used for quick and efficient silence of multiple genes in hESCs in a highly controlled manner.

The following article presents a review of the evolution of the gate and neuromatrix theories as well as their contribution in the development of neuromodulation to the present day. Nowadays the mechanism of action of neuromodulation is better to understand by dividing it according to the level where it acts (local, central or peripheral). Patient selection is crucial to optimize the effectiveness of the device; although the indications for pain that support current guidelines are failed back syndrome, complex regional pain syndrome and lower limb ischemic pain and refractory angina; neuromodulation is already being extended to multiple fields of action that affect the future, such as the improvement of human capacities.


El siguiente artículo consiste en una revisión no sistemática de la evolución de la teoría de la compuerta y la neuromatriz, así como su aporte en el desarrollo de los dispositivos de neuromodulación. Para entender mejor el mecanismo de acción de la neuromodulación se divide según el nivel en que actúe sea local, central o periférico. La selección de pacientes es crucial para optimizar la eficacia del dispositivo; aunque las indicaciones para manejo del dolor que soportan las guías actuales son síndrome de espalda fallida, síndrome doloroso regional complejo y dolor tipo isquémico en miembros inferiores y dolor por angina refractaria; actualmente la neuromodulación se está extendiendo a múltiples campos de acción entre los cuales se encuentra el mejoramiento de las capacidades humanas.

PURPOSE: This study aims to compare Monte Carlo-based codes' characteristics in the determination of the basic parameters of a high-resolution single photon emission computed tomography (HiReSPECT) scanner.METHODS: The geometry of this dual-head gamma camera equipped with a pixelated CsI(Na) scintillator and lead hexagonal hole collimator were accurately described in the GEANT4 Application for the Tomographic Emission (GATE), Monte Carlo N-particle extended (MCNP-X), and simulation of imaging nuclear detectors (SIMIND) codes.We implemented simulation procedures similar to the experimental test for calculation of the energy spectra, spatial resolution, and sensitivity of HiReSPECT by using 99mTc sources.RESULTS: The energy resolutions simulated by SIMIND, MCNP-X, and GATE were 17.53, 19.24, and 18.26%, respectively, while it was calculated at 19.15% in experimental test. The average spatial resolutions of the HiReSPECT camera at 2.5 cm from the collimator surface simulated by SIMIND, MCNP-X, and GATE were 3.18, 2.9, and 2.62 mm, respectively, while this parameter was reported at 2.82 mm in the experiment test. The sensitivities simulated by SIMIND, MCNP-X, and GATE were 1.44, 1.27, and 1.38 cps/µCi, respectively, on the collimator surface.CONCLUSIONS: Comparison between simulation and experimental results showed that among these MC codes, GATE enabled to accurately model realistic SPECT system and electromagnetic physical processes, but it required more time and hardware facilities to run simulations. SIMIND was the most flexible and user-friendly code to simulate a SPECT camera, but it had limitations in defining the non-conventional imaging device. The most important characteristics like time and speed of simulation, preciseness of results, and user-friendliness should be considered during simulations.

RESUMEN

In 1905, Henry Head first suggested that transmission of pain-related protopathic information can be negatively modulated by inputs from afferents sensing innocuous touch and temperature. In 1965, Melzak and Wall proposed a more concrete gate control theory of pain that highlights the interaction between unmyelinated C fibers and myelinated A fibers in pain transmission. Here we review the current understanding of the spinal microcircuits transmitting and gating mechanical pain or itch. We also discuss how disruption of the gate control could cause pain or itch evoked by innocuous mechanical stimuli, a hallmark symptom for many chronic pain or itch patients.

Objective To evaluate the changes of intra-and interatrial synchronization with age in normal adults by dual gate Doppler and discuss the feasibility of dual gate Doppler in assessing atrial synchronization.Methods Fifty-seven healthy volunteers were divided into three groups according to age:group A (20-39 years old,n =20),groupB(40-59 years old,n =21),groupC (60-79 years old,n =16).The time interval of the onset of a'wave between mitral annular septal and lateral site was T1 by dual gate Doppler,which was used to evaluate left atrial synchronization.The time intervals from the onset of a'wave at tricuspid annular right ventricular free wall site to a'wave at tricuspid annular septal site and mitral annular lateral site were T2 and T3,which were respectively used to evaluate right atrial and interatrial synchronization.With traditional Doppler technique,the time intervals from the onset of P wave to the onset of a'wave at the mitral annular lateral site (P-L),the mitral annular septal site (P-S),and the tricuspid annular right ventricular free wall site (P-RVFW) were measured.The time differences between P-L and P-S,between P-S and P-RVFW,and between P-L and P-RVFW were t1 (left atrial synchronization),t2 (right atrial synchronization),t3 (interatrial synchronization).The greater time interval predicted the worse synchronization.Results ① Systolic and diastolic blood pressure elevated with age among three groups (P<0.05).Compared with group A,the ratio (E/e') of early diastolic transmitral flow velocity (E) and mitral annular velocity of septal site (e'),and late diastolic transmitral flow velocity increased in group B and C.In contrast with group A and B,left atrial volume (LAV) increased,and E decreased in group C (all P <0.05).②Compared with group A,T1,T2,T3 and t1,t2,t3 increased in group B and C (all P <0.05).③T1,T2 and T3 were positively correlated with age,body mass index,systolic blood pressure,left atrial systolic antero-posterior diameter and E/e'(all P <0.05).Besides,T1 was positively correlated with LAV (P <0.05).T3 was positively correlated with LAV and right atrial upper-inferior diameter.④ The parameters of atrial synchronization in dual gate Doppler were concordant with that in traditional Doppler technique.And the measurements of two methods were correlated (r =0.78,P <0.01).For inter-and intraobservers,the interclass correlation coefficient with dual gate Doppler was higher and 95 % confidence interval range was smaller.Conclusions The intra-and interatrial synchronization reduces with age in normal adults.Dual gate Doppler is feasible in evaluating atrial synchronization,which can provide more repeatable measurements and can be a new prospective method to assess atrial synchronization.

Nanopore / nanochannel sensing technique drawing more attention in analytical chemistry due to its unique advantages and the fabricated electrochemical sensors and electrochemical responsive gates have been widely used for more target detection, including single molecule protein and DNA sequencing. Nanopore /nanochannel that used for fabricating electrochemical detection system is mainly divided into biological nanopore and solid state nanopore, and among them, solid state nanopore / nanochannel has a wide range of application due to its inherent properties, such as easy for modification, good mechanical property and stability. Resistive pulse sensing and current-voltage curves (ion current rectification) are two main methods of nanopore / nanochannel sensing technique used for target analysis, so in this review, we introduced the fundamentals and applications of nanopore sensing technique based on the above two methods. In addition, we concluded the application and development of single state nanopore / nanochannel in recent years, and also gave a brief look at the future challenges and prospects in the development of this field.

Objective To discuss whether atrial synchronization measured by dual gate Dopper can predict recurrence after radiofrequency catheter ablation ( RFCA) in paroxysmal atrial fibrillation ( PAF) patients . Methods Forty-five patients with PAF who had successful RFCA and 26 controls with sinus rate were prospectively enrolled . During 6-month follow-up , the patients with PAF were divided into AF recurrence group ( n = 16) and AF non-recurrence group ( n = 29) . All patients with PAF underwent echocardiography in the preoperative 7 days . The time interval of the onset of the late diastolic ( a′) wave between mitral annular septal and lateral site was T 1 by dual gate Doppler ,which was used to evaluate left atrial synchronization . The time intervals from the onset of a′ wave at tricuspid annulus right ventricular free wall site to a′wave at tricuspid annular septal site and mitral annular lateral site were T 2 and T3 ,which were respectively used to evaluate right atrial and interatrial synchronization . The greater time interval predicted the worse synchronization . Results Compared with controls ,left atrial diameter ,volume and volume index ,right atrial transverse diameter ,volume and volume index ,T 1 ,T2 and T3 ( standardized before and after) increased in patients with PAF ( all P <0 .05) . In patients with PAF ,left atrial volume index and T2 ( standardized before and after) had no differences between AF recurrence group and AF non-recurrence group( P >0 .05) . But there was a trend toward lager left atrial diameter in the AF recurrence group ( P < 0 .05) . T1 and T3 ( standardized before and after) increased significantly in AF recurrence group (all P < 0 .05) . Multivariate logistic regression analysis indicated standardized T 1 was the only independent predicator of AF recurrence after RFCA ( OR = 1 .060 ,95% CI 1 .002 -1 .121 , P = 0 .044) . The area under curve of standardized T1 for predicting recurrence was 0 .709% , the sensitivity and specificity were 62 .5% and 75 .9% using a cutoff value of standardized T1≥39 .38 ms . Conclusions T1 significantly prolongs in patients with AF recurrence . Dual gate Doppler as a new technique to evaluate atrial synchronization can provide predictive value for 6-month recurrence after RFCA .

PURPOSE: This study aimed to prove the effects of the net-step exercise (NSE) on gait ability, depression, cognitive function and activities of daily living (ADL) in older adults. METHODS: The study employed a non-equivalent control group non-synchronized design. A total of 64 community-dwelling older adults were recruited and divided equally into two groups; 32 subjects for an experimental group and 32 subjects for a control group. In the experimental group, the NSE was applied to an hour, two times per week for 4 weeks. The level of gait ability, depression, cognitive function and ADL were measured before and after NSE. The study conducted from July to August, 2016. Data were analyzed with descriptive statistics, χ² test, Fisher's exact test, t-test, ANCOVA, and Pearson correlation coefficients using SPSS/WIN 22.0 version. RESULTS: Gait ability, depression, cognitive function were significantly better in the experimental group than the control group. However, the difference in ADL was not significant between the two groups. CONCLUSION: These findings in this study showed that the NSE was an efficient intervention for older adults. Nurses could apply non-pharmacological interventions to avoid pharmacological side-effects.

Objective To evaluate intra-and interatrial synchronization in patients with paroxysmal atrial fibrillation (PAF) by dual gate Doppler and discuss the predictive value of atrial asynchrony for detecting PAF.Methods Twenty-nine patients with PAF as AF group and 24 healthy volunteers as control group underwent echocardiography.The time interval of the onset of a'wave between mitral annular septal and lateral site was T1 by dual gate Doppler,which was used to evaluate left atrial synchronization.The time intervals from the onset of a'wave at tricuspid annular right ventricular free wall site to a'wave at tricuspid annular septal site and mitral annular lateral site were T2 and T3,which were respectively used to evaluate right atrial and interatrial synchronization.The greater time interval predicted the worse synchronization.Compared these indexes between two groups,the most valuable index was obtained and combined with left atrial volume index (LAVI) to predict PAF,its optimal values were found out by ROC curve.Results ①There were no significant differences in many other basic indexes between two groups (P >0.05).However,compared with control group,the ratio of early diastolic transmitral flow velocity (E) and mitral annular velocity of septal site (e'),the diameters and volume indexes of atria were significantly increased in AF group.②T1,T2 and T3 (standardized before and after) increased significantly in AF group (P <0.01).Standardized T1 had the largest area under curve (AUC) for detecting PAF.The AUC of standardized T1 and LAVI were 76.6% and 84.1%.Standardized T1 ≥29.55 ms and LAVI≥ 21.60 ml/m2 were combined for detecting PAF (sensitivity 82.8 %,specificity 79.2%,AUC 87.1%).③ Standardized T1 was positively correlated with age,left atrial systolic diameter,LAVI,RAVI and E/e'(P< 0.05).Conclusions Patients with PAF have worse intra-and interatrial synchronization and left atrial asynchrony can provide predictive value for detecting PAF.Dual gate Doppler is expected to be a new technique for evaluating intra-and interatrial synchronization accurately and detecting the risk of AF.

RESUMEN

Objective To explore the feasibility of prospective ECG-gated dynamic volume CT coronary angiography performed on patients with heart rate greater than 70 beats per minute.Methods 1 18 patients with heart rate greater than 70 beats per minute were involved.All patients general data were collected.All the patients were scanned by 320 volume CT with prospective ECG-gated mode.Scanning was triggered by 250 ms-400 ms after R wave.100 kVp tube voltage,tube current automatically adjustment,and iodine contrast agent (370 mg I/mL)were used.All patients’radiation dose were recorded.Coronary artery can be divided into 1 5 segments,coronary CT values were measured and evaluated by two radiologists.The results are statistically analyzed.Results There was no significant difference of gender,age and weight between the groups.The image quality of met diagnostic rate was 83.05%.Image subjective scores of groups(heart rate > 70 beats per minute group,> 75 beats per minute group and > 85 beats per minute group) was 4.23 ± 0.72,3.54±1.22,1.87±0.74 espectively.There was no statistical difference in the groups (F =5.125,P =0.050 3).Radiation dose was increaseed with heart rate increasing.Conclusion Image quality of CT coronary angiography of patients with heart rate greater than 70 beats per minute,performed by 320 row volumetric CT and using prospective ECG-gated scanning mode can satisfy clinical diagnosis requirement .

Objective To explore the sluice gate technology in the control of nutrient vessel retrograde island skin flap with sural nerve small saphenous vein blood flow.Methods Totally 32 cases were divided into group A and group B,group A flap with no special treatment to the return of small saphenous vein,group B applied river gate principle,in operation around the small saphenous vein reserved suture,postoperative through a rubber band traction control flap blood circulation and skin flap swelling. Observation and comparison of two groups of skin flap necrosis.Results There had 4 cases in group A flap incomplete necrosis,the incidence rate was 25.0%,1 cases of skin flap incomplete necrosis in B group,the incidence rate was 6.2%.Conclusion The use ofsluice gate technique to control the small saphenous vein reflux of blood,relieve skin flap swelling,improve the survival rate of the flap is simple and effective.

Objective: To analyze FPGA technology in diagnostic ultrasound B signal processing system in order to provide a theoretical basis for the further development of diagnostic ultrasound signal processing. Methods:Analysis of FPGA technology in diagnostic ultrasound signal processing system for the study through literature study to analyze the relevant principles. Results: FPGA to be more widely used in ultrasonic diagnostic apparatus in full. Conclusion: It comprehensively improved the reliability and flexibility of ultrasound diagnostic signal processing system, making the modular, miniature, diversification, serialization and the rapid development of digital direction.

Background & objectives: Integrated vector management (IVM) emphasizes sustainable eco-friendly methods and minimal use of chemicals. In this context, the present study highlights the environmental control of breeding of Anopheles fluviatilis, the primary malaria vector, through water management in a natural stream in Koraput district, Odisha, India. Methods: The District Rural Development Agency (DRDA), Koraput, constructed two bed-dams across streams, one in Barigaon and the other in Pipalapodar village. The bed-dam in the former village was fitted with two sluice gates whereas the bed dam constructed in the latter village was without the sluice gate. tThe sluice gates were opened once in a week on a fixed day to flush out the water from the dam. Anopheles immatures were sampled systematically in the streams using a dipper for density measurement and species composition. Results: There was a reduction of 84.9 per cent in the proportion of positive dips for aAnopheles larvae/pupae and a reduction of 98.4 per cent in immature density (number/dip) of An. fluviatilis in the experimental downstream compared to the control following opening of the sluice gates. Interpretation & conclusions: oOur findins showed that opening of sluice gates of the bed-dam regularly once in a week resulted in the control of vector breeding in the downstream due to the flushing effect of the water released with a high flow from the bed-dam that stagnated water in the upstream. The outcome of the study encourages upscaling this measure to other areas, wherever feasible.
