Abstract Blood and fecal samples of chukar partridge (Alectoris chukar), albino pheasant (Phasianus colchicus), silver pheasant (Lophura nycthemera), rose-ringed parakeet (Psittacula krameri) and turkeys (Meleagris gallopavo) were analyzed to check parasitic prevalence. To record parasites these five avian species were placed kept in separate cages at Avian Conservation and Research Center, Department of Wildlife an Ecology, University of Veterinary and Animal Sciences, Lahore, Pakistan. 100 fecal and 100 blood samples for each bird species were inspected to analyze internal parasites. During present study, 17 species of endoparasites 14 from fecal samples and three from blood were examined. Two species of ectoparasites i.e. mite Dermanyssus gallinae 42% and fowl ticks Args persicus 41%were studied. Blood parasites included Plasmodium juxtanucleare 50%, Leucoctoyzoon simond having parasitic prevalence 40%, and Aegyptinella pullorum having parasitic prevalence of 40%. Parasitic species recorded from fecal samples included 6 species of nematodes viz. Allodpa suctoria 2%. Syngamus trachea with parasitic prevalence of 60%, Capillaria annulata 37.5%, Ascardia galli 24%, Capillaria anatis 40% and Heterakis gallinarum 28.3%. Similarly, two species of trematodes viz. Prosthogonimus ovatus having parasitic prevalence of 50% and Prosthogonimus macrorchis 21% were also documented from fecal avian samples . Single cestode species Raillietina echinobothrida having parasitic prevalence of 72% and 3 protozoan species i.e. Eimeria maxima having parasitic prevalence of 21%, Giardia lamblia 41% and Histomonas meleagridis 18% were documented during corpological analysis. In our recommendation, proper sanitation, medication and vaccination of birds enclousres are suggested to avoid parasites.
RESUMO Amostras de sangue e fezes de perdiz chukar (Alectoris chukar), faisão-albino (Phasianus colchicus), faisão-prateado (Lophura nycthemera), periquito-de-rosa (Psittacula krameri) e perus (Meleagris gallopavo) foram analisadas para verificar a prevalência de parasitas. Para registrar os parasitas, essas cinco espécies de aves foram colocadas em gaiolas separadas no Centro de Conservação e Pesquisa de Aves, Departamento de Vida Selvagem e Ecologia, Universidade de Veterinária e Ciências Animais, Lahore, Paquistão. Cem amostras fecais e 100 amostras de sangue para cada espécie de ave foram inspecionadas para analisar os parasitas internos. Allodpa suctoria 2%, Syngamus traqueia com prevalência parasitária de 60%, Capillaria annulata 37,5%, Ascardia galli 24%, Capillaria anatis 40% e Heterakis gallinarum 28,3%. Da mesma forma, duas espécies de trematódeos viz. Prosthogonimus ovatus com prevalência parasitária de 50% e Prosthogonimus macrorchis 21% também foram documentados em amostras fecais de aves. Espécies de cestoide único Raillietina echinobothrida com prevalência parasitária de 72% e 3 espécies de protozoários, isto é, Eimeria maxima com prevalência parasitária de 21%, Giardia lamblia 41% e Histomonas meleagridis 18% foram documentadas durante a análise corpológica. Em nossa recomendação, o saneamento adequado, medicação e vacinação de invólucros de pássaros são sugeridos para evitar parasitas.

Introducción. La giardiasis es ocasionada por el protozoario intestinal Giardia lamblia, su transmisión tiene variabilidad geográfica y a pesar de ser muy frecuente, no existe una herramienta de estratificación de riesgo para priorizar las intervenciones. Objetivo. Estimar la prevalencia de G. lamblia en preescolares y escolares peruanos por ecorregiones entre 1990 a 2018. Métodos. A partir de un metaanálisis previo, se realizó un subanálisis por ecorregiones de la prevalencia de G. lamblia en 26 estudios en preescolares y escolares peruanos entre 1990 a 2018. Se extrajeron los datos por distrito y utilizando Google Earth fueron clasificados en ecorregiones. La heterogeneidad fue analizada mediante la prueba de Q de Cochrane y el sesgo de publicación mediante el método de Egger con StatsDirect versión 3.2.7. Se utilizó el método de riesgo absoluto para estratificar la prevalencia a nivel de distrito y se definió 4 estratos en base a los percentiles o arbitrariamente. Resultados. Se identificaron 43 datos de prevalencia a nivel de distrito que incluyeron 7606 participantes. Las prevalencias combinadas más altas por el método de efectos aleatorios de G. lamblia fueron de 47,0% intervalo de confianza (IC) 95%: 40,0 -54,0) en el desierto del Pacífico; 27,9% (IC95%: 22,8 - 33,2) en selva baja y 26,9% (IC95%: 22,5 -31,5) en la Puna. Conclusiones. De cada 100 preescolares y escolares que viven en las ecorregiones del desierto del Pacífico, selva baja y Puna, 47, 28 y 27 menores de edad, respectivamente, están infectados con G. lamblia. Asimismo, se propone 4 estratos de riesgo en función de la prevalencia: esporádico (0 a <1%), hipoendémico (1 a <25%), mesoendémico (≥ 25 a <50%) e hiperendémico (≥50%).
Introduction. Giardiasis is caused by an intestinal protozoan, Giardia lamblia. Despite its high prevalence and geographical transmission variability, there is no risk stratification tool available to prioritize interventions. Objective. To estimate the prevalence of G. lamblia in Peruvian preschoolers and schoolchildren by ecoregion from 1990 to 2018. Methods. Based on a previous meta-analysis, we conducted a G. lamblia prevalence sub-analysis by ecoregions from data of 26 studies in Peruvian preschoolers and school-aged children between 1990 and 2018. The data was extracted by district, a classification by ecoregions was made through Google Earth. Heterogeneity was analyzed using Cochrane Q test and publication bias applying the Egger method with StatsDirect version 3.2.7. The absolute risk method was performed to stratify the prevalence at district level, and 4 strata were defined based on percentiles or arbitrarily. Results. Forty-three district-level prevalence data was estimated, including 7,606 participants. The highest pooled prevalences by the random effects method of G. lamblia were 47.0% (95% CI: 40.0-54.0) in the Pacific desert, 27.9% (95% CI: 22.8-33.2) in the lowland forest and 26.9% (95% CI 22.5-31.5) in the Puna. Conclusions. Of every 100 preschoolers and school-aged children living in the Pacific desert, lowland forest, and in the Puna ecoregions, 47, 28, and 27 minors are infected with G. lamblia, respectively. Likewise, 4 risk strata are proposed based on prevalence: sporadic (0 to <1%), hypoendemic (1 to <25%), mesoendemic (≥25 to <50%) and hyperendemic (≥50%).

Objective To investigate the prevalence and genotypes of Cryptosporidium spp. and Giardia lamblia in dogs and cats from a pet hospital in Shanghai Municipality. Methods A total of 145 fresh fecal samples were collected from pet dogs and cats in a pet hospital in Shanghai Municipality during the period from November 2021 to June 2022, including 99 dog fecal samples and 46 cat fecal samples. The small subunit ribosomal ribonucleic acid (SSU rRNA) gene of Cryptosporidium and the triose phosphate isomerase (TPI) gene of G. lamblia were amplified using nested PCR assay, and the positive amplification products were sequenced from both directions. The sequence assembly was performed using the software Clustal X 2.1, and sequence alignment was conducted using BLAST. A phylogenetic tree was created with the Neighbor-Joining method using MEGA 11.0 to identify parasite species or genotype. Results The overall prevalence of Cryptosporidium and G. lamblia was 20.00% (29/145) in 145 pet dog and cat fecal samples, with the prevalence of 0.69% (1/145) and 19.31% (28/145) in Cryptosporidium and G. lamblia, respectively. G. lamblia was only detected in dog fecal samples, with prevalence of 18.18% (18/99), while the detection rates of Cryptosporidium and G. lamblia were 2.17% (1/46) and 21.74% (10/46) in cat fecal samples. Nucleotide sequence analysis showed that one Cryptosporidium positive sample was characterized as C. felis, and 28 G. lamblia positive samples were all characterized as Giardia assemblage A, which showed 100% sequence homology with human isolates of Giardia. Phylogenetic analysis revealed that the sequences obtained in this study belonged to the same branch with the reported Giardia assemblage A. Conclusions Cryptosporidium and G. lamblia infection was prevalent in pet dogs and cats from the study pet hospital in Shanghai Municipality, and there is a zoonotic risk for the species and genotype. Intensified surveillance of Cryptosporidium and Giardia infection is recommended in pets and their owners, and improved management of pet keeping is required.

@#Abstract: Objective To establish the duplex TaqMan RT-PCR method for detection of Entamoeba histolytica and Giardia lamblia in fecal samples. Methods Primer pairs and probes for Entamoeba histolytica and Giardia lamblia were designed and duplex TaqMan RT-PCR amplification system was constructed. PCR products were inserted into the pUC57 plasmid, and the lower limit of detection of the method was determined. Clinical stool samples were tested in order to evaluated the efficacy of the method. Results The detection limits of duplex TaqMan RT-PCR were 31.6 copies/μL for Entamoeba histolytica and 32 copies/μL for Giardia lamblia, respectively. Of the total of 212 clinical stool samples tested, all 3 samples with E. histolytica-positive patients by microscopy were positive by PCR, while 1 from 209 samples with E. histolytica-negative patients by microscopy were positive by PCR, and the remaining samples were negative. For Giardia lamblia, all 8 samples positive by microscopy were positive by PCR, and 1 from 204 sample with a microscopy-negative patient was positive by PCR, and the remaining samples were negative. The amplification product sequencing and blast analysis were used to confirm that the amplified sequence in the specimen of a patient with negative microscopy but positive PCR belongs to the targeted pathogen, supported by clinical symptoms and laboratory test results. PCR results for other diarrhea-causing pathogens were negative, indicating no cross-reactivity. Conclusions The dual TaqMan RT-PCR method developed in this study can not only detect microscopy-positive samples of Entamoeba histolytica and Giardia lamblia but also can detect samples that were missed by microscopy, with higher sensitivity than the microscopy method. Further, this detection method does not cross-react with other diarrhea-causing pathogens, including cross-react with other diarrhea-causing pathogens including Iodamoeba butschlii, Blastocystis hominis, Plesiomonas, Aeromonas, Salmonella, Shigella, Sphaerozoum fuscum, and Entamoeba hartmani, thus has a good specificity.

BACKGROUND Intestinal parasite Giardia can affect children's physical development mainly stunting even in asymptomatic cases. The protozoa G. lamblia is divided into assemblages A-H. However, it is still unclear whether clinical manifestations and pathogenesis may vary according to the infecting assemblage. OBJECTIVES To investigate whether G. lamblia assemblages influence differently the physical development of preschoolers from a community of Rio de Janeiro, Brazil. METHODS Anthropometric parameters were analysed from children attending a daycare centre and stool samples were obtained for the G. lamblia diagnosis. G. lamblia isolates from positive samples were genotyped. Data were analysed in order to verify whether there is a relationship between G. lamblia infection and the physical development of children according to the assemblage. FINDINGS Herein we demonstrated that although eutrophic, G. lamblia-infected daycare preschoolers from a low-income community presented growth delay compared to non-infected ones. This effect was observed for the three assemblages (A, B or E) found infecting humans. MAIN CONCLUSION G. lamblia causes growth delays on children independent of infecting assemblage (A, B or E).

Objective To investigate the epidemiological characteristics and identify the risk factors of Giardia lamblia infections among patients with colorectal cancer in Henan Province. Methods A cross-sectional study was performed for questionnaire surveys among colorectal cancer patients in Henan Cancer Hospital during the period from March to July, 2021. Patients’ stool samples were collected, and the triosephosphate isomerase (tpi) gene of G. lamblia was amplified in stool samples using nested PCR assay to characterize the parasite genotype. Univariate analysis and multivariate logistic regression analyses were employed to identify the risk factors of G. lamblia infections among colorectal cancer patients. Results A total of 307 colorectal cancer patients were investigated, including 176 males (57.3%) and 131 females (42.7%). PCR assay detected 8.1% [95% confidential interval (CI): (0.056, 0.117)] prevalence of G. lamblia infections among the study subjects, and there was no significant difference in the prevalence between men [9.1%, 95% CI: (0.057, 0.143)] and women [6.9%, 95% CI: (0.037, 0.125)] (χ2 = 0.495, P = 0.482). In addition, there was no age-specific prevalence of G. lamblia infections among the participants (χ2 = 1.534, P = 0.675). Multivariate logistic regression analysis identified use of septic tanks [odds ratio (OR) = 3.336, 95% CI: (1.201, 9.267)], daily use of well water [OR = 3.042, 95% CI: (1.093, 8.465)] and raising livestock [OR = 3.740, 95% CI: (1.154, 12.121)] as risk factors of G. lamblia infections among colorectal cancer patients, and the prevalence of abdominal pain was significantly greater in colorectal cancer patients with G. lamblia infections than in those without infections (P = 0.017). Among the 25 patients with G. lamblia infections, assemblage A was characterized in 24 (96.0%) cases and assemblage B in one case (4.0%). Conclusions The prevalence of G. lamblia is high among colorectal cancer patients in Henan Province, and assemblage A is the dominant genotype of G. lamblia. Use of septic tanks, daily use of well water and raising livestock are risk factors of G. lamblia infections among patients with colorectal cancer.

@#The aim of this study was to detect and characterize Giardia lamblia in animals in the UAE. Eighty-seven fecal samples were tested for G. lamblia using the conserved fragment of small subunit (SSU)-rRNA by nested PCR. Giardia-positive isolates were genotyped for assemblages A and B using assemblage specific primers of the triosephosphate isomerase (tpi) gene. Thirty samples (34.5%) were positive for G. lamblia. Conversely, neither genotype A nor B were detected using tpi genotyping on the studied samples. Further investigations are required using higher number of samples including both human and animals in the country taking into consideration the analysis of other genotypes to provide more detailed understanding about the zoonotic transmission of this parasite.

Aims:The objective was to compare the sedimentation spontaneous in tube technique(SSTT), the direct smear, the zinc sulfhate(surface film and sediment analyzed) methods with the formalin-ethyl acetate method as the reference standard.Study Design:This a cross-sectional study performed in two populations aroundChapala lake, Jalisco, Mexico.Place and Duration of Study:Sample Department of Medical Sciences and Life, University Center of the Ciénega, University of Guadalajara, Ocotlán, Jalisco, Mexico. México, between June 2018 andJuly 2019.Methodology:Sample: We included a total of 297 samples and were analyzed by the direct smear, the zinc sulfhate (surface film and sediment analyzed), the SSTT and formalin-ethyl acetate methods.Results:The SSTT was able to detect 40% of intestinal parasites, very good agreement for Entamoebacomplex, Entamoeba coli, andAscarislumbricoideskappa index=0.697, 0.791, 0.696, respectively and excellent agreement for Giardia lambliakappa index = 0.843. Regarding the isolation ofBlastocystisspp, only a poor agreement was found among all techniques. The SSTT was able to detect multiple parasites with a sensitivity of 82.90% and specificity 83.80 kappa index= 0.649.Conclusion:The SSTT showed a very good agreement for the diagnosis intestinal polyparasitism which could represent another alternative for the concentration and identification protozoans and intestinal helminthes in low-resource settings

Objective To establish a novel nucleic acid assay for detection of Giardia lamblia based on the recombinase-aided isothermal amplification (RAA) assay, and evaluate its sensitivity and specificity for detection of G. lamblia. Methods The specific primer sequences and florescent probes were designed and synthesized based on the G. lamblia β-giardin gene as the target gene, and a fluorescent RAA assay was established. The recombinant plasmids at various copies (containing the β-giardin gene target sequence) and the genomic DNA of G. lamblia at various concentrations were used as templates for the fluorescent RAA assay to assess the sensitivity, and the genomic DNA from G. lamblia, Schistosoma japonicum, Clonorchis sinensis, Cryptosporidium parvum, Ascaris lumbricoides, Salmonella and Shigella was used as templates to assess the specificity of the fluorescent RAA assay. Results A novel fluorescent RAA assay was successfully established for detection of G. lamblia, which allowed the rapid and specific amplification of the target gene fragments at 39 ℃ within 20 min. The sensitivities of the fluorescent RAA assay were 102 copies/μL and 1 pg/μL for detection of the recombinant plasmid and G. lamblia genomic DNA, respectively, and the fluorescent RAA assay was negative for detection of the genomic DNA from S. japonicum, C. sinensis, C. parvum, A. lumbricoides, Salmonella and Shigella, which showed a high specificity. Conclusions A fluorescent RAA assay, which is simple, sensitive and specific, is successfully established for nucleic acid detection of G. lamblia.

Methods: A total of 490 Egyptian children aged 2-16 years were subjected to microscopic stool examination to detect G. lamblia infection, and to exclude other intestinal pathogens. On the basis of their microscopic findings, a group of 80 children were chosen as giardiasis cases, another 80 children were confirmed as Giardia free control group by immunochromatographic test, and the remaining children were excluded. Both giardiasis and control groups were then subjected to blood examination to identify their genetic type of HLA-DRB1 alleles. Results: HLA class-II DRB1∗03:01 and DRB1∗13:01 alleles were significantly associated with G. lamblia infection (P<0.001 for each variable). On the other hand, HLA class-II DRB1∗04:02, DRB1∗10:01, DRB1∗14:01 and DRB1∗15:01 alleles were significantly demonstrated in Giardia free children. However, other HLA-DRB1 alleles did not show any significant association with giardiasis. Conclusions: HLA class-II DRB1∗03, DRB1∗13, DRB1∗04, DRB1∗10, DRB1∗14 and DRB1∗15 alleles may be involved in the establishment of host immune response to G. lamblia infection.

O objetivo desta pesquisa foi verificar a prevalência de enteroparasitoses em pacientes atendidos no Hospital das Clínicas da Faculdade de Medicina de Marília-SP. Desta forma foi realizado um estudo descritivo e analítico, do tipo transversal retrospectivo, com coleta dos dados no Sistema Hospitalar Informatizado do Laboratório de Parasitologia da Faculdade de Medicina de Marília. Entre janeiro de 2012 a dezembro de 2017 foram analisadas 14.343 amostras. Os dados de prevalência foram representados por razão de prevalência e seus respectivos Intervalos de Confiança de 95%. Diferenças significativas entre anos ou faixa etárias foram determinadas pela não intersecção dos limites inferiores e superiores de seus IC95%. Observaram-se 2.149 amostras positivas para parasitas, predominando Blastocystis hominis (10,13%), Giardia lamblia (3,35%) e Strongyloides stercoralis (1,16%). Pode-se inferir que a redução da prevalência das parasitoses é o resultado de políticas públicas pelo uso de medicamentos profiláticos no intuito de interromper a transmissão fecal-oral e não somente pela melhora da infraestrutura sanitária.
The prevalence of enteroparasitosis in patients at the hospital of the School of Medicine in Marília SP Brazil, is analyzed through a descriptive, analytic, retrospective and transversal study. Data were retrieved from the Digitalized Information System of the Laboratory of Parasitology of the School of Medicine in Marília where 14,343 patients were investigated between January 2012 and December 2017. Prevalence data were given in ranking and their respective confidence intervals at 95%. Significant differences between years or age groups were determined by non-intersection of lower and higher limits of IC95%. There were 2,149 positive samples for parasites, with a predominance of Blastocystis hominis (10.13%), Giardia lamblia (3.35%) and Strongyloides stercoralis (1.16%). Results show that the decrease of parasite prevalence is due to public policies through prophylactic medicine to interrupt the fecal-oral transmission rather than by improvement of the sanitary infrastructure.

La Giardiasis ocasionada por Giardia intestinalis (conocida como Giardia lamblia), es una de las infecciones parasíticas más comunes en todo el mundo y con mayor prevalencia en países en desarrollo como el nuestro. Existen muchas drogas para el tratamiento de la giardiasis, de diferente eficacia y efectos adversos como la curcumina que inhibe la polimerización de los microtubulos por un mecanismo distinto al de la colchicina. Sin embargo, la estructura cristalográfica de la Tubulina de G. lamblia (cadenas α y ß) permanece desconocida. El análisis de alineamiento de secuencias (PBLAST) indica una identidad del 86,98 y 88,32 % entre las cadenas α y ß de la Tubulina de G. lamblia y B. taurus (PDB:5NQT). El Modelamiento por Homología de la estructura proteica de la Tubulina de G. lamblia utilizando como molde a la Tubulina de B. taurus mediante el servidor SWISS-MODEL, generó una estructura proteica con los siguientes parámetros: z-core = -1,16 y -1,41, QMEANscore6 = 0,71 y 0,70, % de confiabilidad (de los diagramas de Ramachandran) = 96,08 y 96,95 %, RMS (Root Mean Square) = 0,081 y MOLPROBITYscore = 1,26. Estos parámetros indican que la estructura proteica de la Tubulina de G. lamblia obtenida a partir del Modelamiento por Homología es de buena calidad, por tanto, esta estructura podría ser utilizada en futuras evaluaciones, como el análisis in silico de compuestos antiGiardia.
Giardiasis caused by Giardia intestinalis (known as Giardia lamblia), is one of the most common parasitic infections in the world and with a higher prevalence in developing countries like ours. There are many drugs for the treatment of giardiasis, of different efficacy and adverse effects such as curcumin that inhibits the polymerization of microtubules by a mechanism other than colchicine. However, the crystallographic structure of G. lamblia Tubulin (α and ß chains) remains unknown. The sequence alignment analysis (PBLAST) indicates an identity of 86,98 and 88,32 % between the α and ß chains of the Tubulin of G. lamblia and B. taurus (PDB: 5NQT). Homologous Modeling of the protein structure of G. lamblia Tubulin using the B. taurus Tubulin as a template employing the SWISS-MODEL server, generated a protein structure with the following parameters: z-core = -1,16 and -1,41, QMEANscore6 = 0,71 and 0,70, % of reliability (from Ramachandran plots) = 96,08 y 96,95 %, RMS (Root Mean Square) = 0,081 and MOLPROBITYscore = 1,26. These parameters indicate that the protein structure of G. lamblia Tubulin obtained from Homology Modeling is of good quality, therefore, this structure could be used in further evaluations, such as the in silico analysis of antiGiardia compounds.

To identify the component(s) involved in cell cycle control in the protozoan Giardia lamblia, cells arrested at the G1/S- or G2-phase by treatment with nocodazole and aphidicolin were prepared from the synchronized cell cultures. RNA-sequencing analysis of the 2 stages of Giardia cell cycle identified several cell cycle genes that were up-regulated at the G2-phase. Transcriptome analysis of cells in 2 distinct cell cycle stages of G. lamblia confirmed previously reported components of cell cycle (PcnA, cyclin B, and CDK) and identified additional cell cycle components (NEKs, Mad2, spindle pole protein, and CDC14A). This result indicates that the cell cycle machinery operates in this protozoan, one of the earliest diverging eukaryotic lineages.

Innate lymphoid cells (ILCs) are key players during an immune response at the mucosal surfaces, such as lung, skin, and gastrointestinal tract. Giardia lamblia is an extracellular protozoan pathogen that inhabits the human small intestine. In this study, ILCs prepared from the lamina propria of mouse small intestine were incubated with G. lamblia trophozoites. Transcriptional changes in G. lamblia-exposed ILCs resulted in identification of activation of several immune pathways. Secretion of interleukin (IL)-17A, IL-17F, IL-1β, and interferon-γ was increased, whereas levels of IL-13, IL-5, and IL-22, was maintained or reduced upon exposure to G. lamblia. Goup 3 ILC (ILC3) was found to be dominant amongst the ILCs, and increased significantly upon co-cultivation with G. lamblia trophozoites. Oral inoculation of G. lamblia trophozoites into mice resulted in their presence in the small intestine, of which, the highest number of parasites was detected at the 5 days-post infection. Increased ILC3 was observed amongst the ILC population at the 5 days-post infection. These findings indicate that ILC3 from the lamina propria secretes IL-17 in response to G. lamblia, leading to the intestinal pathology observed in giardiasis.

ABSTRACT BACKGROUND: The prevalence of Giardia lamblia in Pakistani children is currently unknown. The aim here was to evaluate the prevalence and risk factors of Giardia lamblia in children exhibiting diarrhea. DESIGN AND SETTING: Cross-sectional study at different district healthcare hospitals in Pakistan. METHODS: A total of 800 samples were collected from children aged 0-10 years. Information regarding personal data, demographic data and supposed risk factors was collected through a structured questionnaire. Giardia lamblia was detected through direct microscopy and antigens through the enzyme-linked immunosorbent assay (ELISA). RESULTS: The prevalence of Giardia lamblia was 2.75% through direct microscopy and inflated to 9.5% through ELISA. The demographic factors positively associated with occurrences of giardiasis were age (P = 0.035; odds ratio, OR = 1.96; 95% confidence interval, CI = 1.094-3.533), mother's educational level (P = 0.031; OR = 2.67; 95% CI = 1.186-6.045) and father's educational level (P = 0.004; OR = 3.56; 95% CI = 1.612-7.899). Similarly, among the supposed risk factors, rural residency (P = 0.032; OR = 1.76; 95% CI = 1.098- 2.851), absence of proper sewerage system (P = 0.000; OR = 6.60; 95% CI = 4.029-10.841) and unavailability of safe drinking water (P = 0.000; OR = 4.08; 95% CI = 2.207-7.547) were the factors strongly connected with giardiasis. Abdominal discomfort was a prominent clinical sign with 46% frequency. CONCLUSION: Various risk factors were associated with occurrences of Giardia, thus emphasizing the importance of parents' education, safe drinking water and proper sewerage systems for Pakistani children's health.

Se realizó un ensayo clínico en fase IV para evaluar la efectividad del Oleozón® aplicado por vía oral en niños y adolescentes con giardiasis, atendidos en la interconsulta de Pediatría del Policlínico Universitario Julián Grimau García de Santiago de Cuba, desde enero hasta diciembre del 2015. El universo estuvo constituido por 116 pacientes (con igual número de casos y controles) en las edades de 1 a 18 años, infectados por Giardia lamblia, quienes presentaban dolor abdominal, vómito y anorexia como síntomas fundamentales. En la serie se obtuvo que el mayor número de los pacientes del grupo experimental, cuyo esquema terapéutico incluía el Oleozón®, resolvió el dolor abdominal y mejoró su estado general al disminuir el resto de los síntomas. Pudo concluirse que el Oleozón® resultó efectivo en el tratamiento contra la giardiasis, pues las muestras de heces fecales dieron negativo en todos los casos; además, no se produjeron reacciones adversas y su costo es bajo, lo que conlleva un impacto económico, social y medioambiental
A clinical assay in phase IV was carried out to evaluate the effectiveness of oral Oleozón® used in children and adolescents with giardiasis, assisted in the Pediatrics specialty consultation from Julián Grimau García University Polyclinic in Santiago de Cuba, from January to December, 2015. The universe was constituted by 116 patients (with the same number of cases and controls) and aged 1 to 18 years, infected by Giardia lamblia who presented abdominal pain, vomit and anorexy as fundamental symptoms. In the series it was obtained that the highest number in the patients of the experimental group whose therapeutic schedule included Oleozón® , solved the abdominal pain and it improved their general state when decreasing the rest of the symptoms. It could be concluded that the Oleozón® was effective in the treatment against giardiasis, because the samples of faeces were negative in all cases; besides, it produced no adverse reactions and its cost is low, what bears an economic, social and environmental impact

Giardia lamblia, an anaerobic, amitochondriate protozoan parasite causes parasitic infection giardiasis in children and young adults. It produces pyruvate, a major metabolic product for its fermentative metabolism. The current study was undertaken to explore the effects of pyruvate as a physiological antioxidant during oxidative stress in Giardia by cysteine-ascorbate deprivation and further investigation upon the hypothesis that oxidative stress due to metabolism was the reason behind the cytotoxicity. We have estimated intracellular reactive oxygen species generation due to cysteine-ascorbate deprivation in Giardia. In the present study, we have examined the effects of extracellular addition of pyruvate, during oxidative stress generated from cysteine-ascorbate deprivation in culture media on DNA damage in Giardia. The intracellular pyruvate concentrations at several time points were measured in the trophozoites during stress. Trophozoites viability under cysteine-ascorbate deprived (CAD) medium in presence and absence of extracellular pyruvate has also been measured. The exogenous addition of a physiologically relevant concentration of pyruvate to trophozoites suspension was shown to attenuate the rate of ROS generation. We have demonstrated that Giardia protects itself from destructive consequences of ROS by maintaining the intracellular pyruvate concentration. Pyruvate recovers Giardia trophozoites from oxidative stress by decreasing the number of DNA breaks that might favor DNA repair.

Waterborne parasitic protozoa, particularly Giardia lamblia and Cryptosporidium spp., are common causes of diarrhea and gastroenteritis worldwide. The most frequently identified source of infestation is water, and exposure involves either drinking water or recreation in swimming pools or natural bodies of water. In practice, studies on Cryptosporidium oocysts and Giardia cysts in surface water are challenging owing to the low concentrations of these microorganisms because of dilution. In this study, a 3-year monitoring of Cryptosporidium parvum, Giardia lamblia, and Naegleria fowleri was conducted from August 2014 to June 2016 at 5 surface water sites including 2 lakes, 1 river, and 2 water intake plants. A total of 50 water samples of 40 L were examined. Cryptosporidium oocysts were detected in 22% of samples and Giardia cysts in 32%. Water at the 5 sampling sites was all contaminated with Cryptosporidium oocysts (0–36/L), Giardia cysts (0–39/L), or both. The geometric mean concentrations of Cryptosporidium and Giardia were 1.14 oocysts/L and 4.62 cysts/L, respectively. Thus, effective monitoring plans must take into account the spatial and temporal parameters of contamination because they affect the prevalence and distribution of these protozoan cysts in local water resources.
