RESUMEN

This study was conducted to test the protective activity of ethanol extract of Herba Scutellariae Barbatae (SE) against hepatotoxicity induced by Rhizoma Dioscoreae Bulbiferae in mice and its mechanism. SE was orally given to mice at various doses, and ethyl acetate fraction of Rhizoma Dioscoreae Bulbiferae (EF, 450 mg·kg-1) was also orally given at the same time. After 11 days, serum levels of alanine/aspartate aminotransferase (ALT/AST), alkaline phosphatase (ALP), total protein (TP) and albumin (ALB) were measured, and liver histological examination was conducted. Liver glutathione (GSH) amount, myeloperoxidase (MPO) activity and serum tumor necrosis factor-α (TNF-α), interleukin-6 (IL-6) and interferon-γ (IFN-γ) levels were measured. The expression of heme oxygenase-1 (HO-1), inhibitor of kappa B (IκB) and nuclear factor κB (NF-κB) p65 were determined by Western blot. The results showed that SE (200 mg·kg-1) reversed EF-induced changes of serum ALT, AST, ALP, TP and ALB. Liver histology also suggests the protection of SE against EF-induced liver injury. SE reduced the increased MPO activity in liver and TNF-α, IL-6, IFN-γ contents in serum, and blocked the decrease in IκB expression and subsequent increase in phosphorylation and nuclear translocation of p65 induced by EF. EF increased liver GSH amount and heme oxygenase-1 (HO-1) protein expression in mice. SE increased liver GSH amount, but decreased the expression of HO-1. All those results suggest that SE alleviates liver injury induced by consecutive administration of EF by alleviating inflammatory injury via inhibiting NF-κB signaling pathway and elevating antioxidant capacity.

RESUMEN

Objective To compare the content of polysaccharide in Herba Scutellariae Barbatae derived from different places. Method Anthrone-sulphuric acid colrimetry was used. Result The contents of polysaccharide in Herba Scutellariae Barbatae derived from Mojiang Yunnan, Nantong Jiangsu, Huaning Hunan, Yongding Fujian, Lixin Anhui, Jiaoling Guangdong, Nanchang Jiangxi, Longsheng Guangxi, Honghu Hubei and Qingyuan Zhejiang were 8.12%, 7.61%, 6.61%, 6.07%, 5.84%, 4.61%, 3.15%, 2.69%, 2.65% and 2.34%, respectively. Conclusion The content of polysaccharide in Herba Scutellariae Barbatae from different places is quite different, and the highest is the one from Mojiang Yunnan.

RESUMEN

Objective To compare the content of total flavonoids in Herba Scutellarlae Barbatae derived from different places.Method UV metric method was used to determine the contents of total flavonoids of Herba Scutellarlae Barbatae from different regions.Result The contents of total flavonoids in Herba Scutellariae Barbatae derived from Nantong Jiangsu,Juaoling Guangdong,Lixin Anhui,Huaning Hunan, Mojiang Yunnan,Yongding Fujian,Nanchang Jiangxi,Longsheng Guangxi,Honghu Hubei and Qingyuan Zhejiang were 4.16%,3.55%,2.89%,2.78%,2.41%,2.22%,2.16%,1.98%,1.61% and 1.51%,respectively. Conclusion The content of total flavonoids in Herba Scutellariae Barbatae from different places is quite different,and the highest is the one from Nantong Jiangsu.

RESUMEN

[Objective] To compare the effects of heat-clearing and detoxifying herbs with that of tonifying herbs on mice hepatoma cells line H22, and to explore their immunologic mechanisms. [Methods] Herba Rabdosiae Rubescentis (HRR) and Herba Scutellariae Barbatae (HSB) were selected as the heat-clearing and detoxifying herbs, and Radix Astragali (RA) as the representative of tonifying herbs. Fifty KM mice were inoculated with hepatoma cells line H22 and then were randomized into five groups: HRR group (30 g?kg-1?d-1 by gavage for 12 days), HSB group (15 g?kg-1?d-1 by gavage for 12 days), RA group (15 g?kg-1?d-1 by gavage for 12 days) , heat-shock group (treated 20 min at 43℃, once every 4 days and three times in all) and model group. Twelve days later, the mice were executed and the ascites smears were prepared. The cell feature in ascites was observed under light microscope and the expression of CD4+ and CD8+ on the cellular membrane of the T lymphocyte in ascites was detected with immunohistochemical method. [Results] A large amount of damaged and apoptotic H22 cells were found in HRR, HSB and heat-shock groups, and the changes of cell feature in RA group were not obvious. The expressions of CD4+ and CD8+ on the membrane of T cells in ascites was increased in HRR and HRB groups, but the expression of CD4+ was increased in RA group, and CD8+ expression was increased in heat-shock group.[Conclusion] Heat-clearing and detoxifying herbs can induce the necrosis and apoptosis of mice hepatoma cells line H22 and the mechanism may be related to the increase of cellular immunity, thus enhancing the body immunity to kill the tumor cells. RA can not kill the tumor cells directly and its antitumor action may be related to the increase of CD4+ expression.
