ABSTRACT: Leishmania infantum causes canine leishmaniasis. Using parasitological and molecular analyses, we identified L. infantum in the reproductive organs of male and female dogs. Using histochemistry, immunohistochemistry, and PCR, we examined tissue samples from the reproductive organs of 8 male dogs and 16 female dogs diagnosed with leishmaniasis. Despite the absence of macroscopic or microscopic lesions in these organs, we observed L. infantum amastigotes in tissue samples from the testis and the uterus. PCR and sequencing of these tissues revealed sequences that matched 100% with L. infantum DNA available at GenBank. The presence of L. infantum amastigotes and DNA in testicular and uterine tissue samples suggested that these organs can harbor the parasite without associated macroscopic or microscopic lesions, and this can be especially important in the vertical and venereal transmission of leishmaniasis in dogs.


RESUMO: Leishmania infantum é agente etiológico da leishmaniose canina. Por meio de análises parasitológicas e moleculares, a presença do parasita foi investigada em órgãos reprodutivos de cães machos e fêmeas. Amostras de tecidos dos órgãos reprodutivos de 8 cães machos e 16 fêmeas diagnosticados com leishmaniose foram avaliadas por histoquímica, imunohistoquímica e PCR. Apesar de não terem sido observadas lesões macroscópicas ou microscópicas nos órgãos reprodutivos desses cães, formas amastigotas de L. infantum foram observadas em amostras teciduais do testículo e útero. A PCR e o sequenciamento do DNA extraído desses tecidos revelaram sequências 100% idênticas a L. infantum depositadas no GenBank. Nossos resultados sugerem que os testículos e o útero podem abrigar o parasita, sem associação com lesões macroscópicas ou microscópicas, o que pode ter uma grande importância na transmissão venérea e vertical da leishmaniose entre cães.

Abstract Clusia grandiflora belongs to an important botanical family which is known for its medicinal value, however there are few reports in literature about the species, highlighting the relevance of this study. Anatomical studies with leaves and stems were performed using traditional techniques. In this investigation it was identified particularities of the species such as the presence of vascular system arranged in an opened arc-shaped with the flexed ends towards the inside of the arch with accessory bundles in the petiole. In histochemical studies, performed with different reagents, alkaloids, phenols substances, carbohydrates and lipids were located. The cytotoxic activity of the extracts was performed by tetrazole salt and showed promising results for ethanolic extracts of stems (IC50 human colon cells of 24.30 μg/ mL) and leaves (IC50 ascites gastric cells of 44.15 μg/ mL), without cell membrane disruption of erythrocytes. The antibacterial activity was evaluated by tryptic soy agar and minimal inhibitory concentration assays and showed positive results for Pseudomonas aeruginosa and Escherichia coli, with better result for adventitious roots (32 μg/ mL and 16 μg/ mL, respectively), stems (64 μg/ mL and 32 μg/ mL, respectively) and leaves (64 μg/ mL and 32 μg/ mL, respectively) ethanolic extracts. Thus, these studies were able to characterize the species and show its potential as promising source of active substances.

RESUMEN

The aim of the research was to investigate the gross morphological, histological and histochemical structure of epididymis in local pig of Mizoram. The epididymis was collected from six apparently healthy adult Zovawk pigs. The epididymis appeared as a straight elongated tubular structure with reddish-white colour comprises of three regions; head, body and tail. The biometrical parameter including the weight, length and thickness (head, body and tail) were found to be higher in the left side of epididymis than the right side. Histologically, the duct of epididymis was surrounded by irregularly arranged tissue. The parenchyma of epididymis consists of tubules with distinct boundaries of connective tissues. The collagen and reticular fibers were abundant in tunica albuginea and connective tissue present between the tubules. However, few elastic fibers occurred in the capsule and only in the blood vessels in both the left and right epididymis of Zovawk pig. Histochemical studies revealed that basement membrane of tubules, blood vessels and stereocilia shows strong Periodic acid Schiff activity, whereas the basement membrane of tubules and stereocilia showed more concentration of acidic mucopolysaccharides. This is the first study to provide the detailed morphological, histological and histochemical structure of Zovawk epididymis

Angiokeratomas consist of ectasias of dermal capillaries associated with an acanthotic and hyperkeratotic overlying epidermis. These dark red-to-purple, papular vascular anomalies can vary considerably in size, depth, and location. It is a skin disorder that rarely involves oral cavity. It can occur in localized or generalized form and is often associated with underlying metabolic disorder such as Fabry's disease and fucosidosis. It has many clinical variants with the same underlying histopathology. Mucosal involvement, including the oral cavity, is occasionally found either as a component of the systemic variety, called angiokeratoma corporis diffusum, or associated with cutaneous lesions in more locations. Isolated oral involvement seems to be rather infrequent, and only eighteen cases have been described in the world literature thus far. Isolated multiple angiokeratomas of tongue without plaque formation have been reported only four times before this. Here, we report a fifth case of isolated multiple angiokeratomas of tongue in a 16-year-old female which was confirmed by immunohistochemical pattern in consonance with a blood vessel origin, with expression of CD31, CD34, and von Willebrand factor. The lesion did not express D2-40 and CD45. No other malformation or metabolic disorder was found in the patient.

DcCDPK8 involved in abiotic stress such as low temperature and signal transduction of hormones ABA and MeJA,but the transcriptional regulation is still unclear. In order to study the core promoter region of DcCDPK8 gene in Dendrobium catenatum and explore its transcriptional regulation mechanism,the DcCDPK8 gene promoter sequence was cloned by PCR from D. catenatum. Promoter sequence function was studied by fusion of 5 'terminal deletion and GUS gene. The results showed that the promoter sequence of DcCDPK8 gene has a low-temperature responsive element( LTR) between~(-1) 749 bp and-614 bp,two MeJA responsive elements between~(-1) 749 bp and-230 bp,and one ABA responsive elements between-614 bp and-230 bp. Three 5'-end different deletion fragments were constructed to fuse the eukaryotic expression vectors p BI121 with GUS,which were transformed into tobacco leaves. The GUS activity under cold stress treatment was DcCDPK8-p1>DcCDPK8-p2>DcCDPK8-p3. GUS activity under exogenous ABA induction was DcCDPK8-p1>DcCDPK8-p2>DcCDPK8-p3,and GUS activity under exogenous MeJA induction was DcCDPK8-p1>DcCDPK8-p2>DcCDPK8-p3. It is speculated that the ABA response element( ARE) in the promoter sequences of DcCDPK8 is positive regulatory role in response to exogenous ABA,the MeJA cis-acting element plays a negative role in response to exogenous MeJA.

Objective To explore the cell viability in the ablation area of thyroid nodules at 6 months after microwave ablation by enzyme histochemical staining. Methods Twenty-four ablation areas of thyroid nodules were selected from 20 patients who underwent histopathological assessment of the ablation area by core needle biopsy at 6 months after microwave ablation between Dec. 2017 and Sep. 2018. Core needle biopsy was performed at the central and marginal regions of the ablation area with a cutting biopsy needle. The specimens were obtained and placed in liquid nitrogen to make frozen sections. Enzyme histochemical staining was used to detect the activities of succinate dehydrogenase (SDH) and nicotinamide adenine dinucleotide phosphate diaphorase (NADPH-d), and the difference of cell morphology and histological structure was compared with H-E staining results. Results The specimens of the central and marginal regions of 24 ablation areas were successfully obtained. The histochemical staining of SDH and NADPH-d in the central region of ablation area had good consistency, and the negative rates were both 95.83% (23/24). The histochemical staining of SDH and NADPH-d in the marginal region of ablation area also had good consistency, and the negative rates were both 91.67% (22/24). H-E staining of 23 central regions and 22 marginal regions showed pink amorphous mass of necrosis. H-E staining of 1 central region and 2 marginal regions showed partly necrotic and fibrous tissue hyperplasia. The location of fibrous tissue hyperplasia was consistent with the location of the positive region of enzyme histochemical staining. Conclusion At 6 months after microwave ablation, the tissue in the ablation area of thyroid nodules is consistent with coagulative necrosis, and is still inactivated. SDH or NADPH-d enzyme histochemical staining combined with H-E staining can objectively evaluate the old ablation area.

A babesiose é uma doença hemolítica transmitida por carrapatos e causada por protozoários intraeritrocitários do gênero Babesia. Esta é uma doença de incidência elevada na Região Sul do Brasil e responsável por perdas econômicas consideráveis. O diagnóstico clínico-patológico pode ser feito através da demonstração do parasito pelo exame de esfregaços sanguíneos ou de tecidos frescos. Entretanto, com frequência, somente órgãos fixados em formol são remetidos para laboratórios de patologia, o que impossibilita a realização do esfregaço. O principal objetivo deste estudo foi encontrar técnicas histoquímicas alternativas e capazes de aprimorar a evidenciação de Babesia bovis intraeritrocitária em tecidos fixados em formol. Para este estudo, foram analisadas retrospectivamente amostras de tecidos de 50 casos de necropsias de bovinos, as quais haviam sido fixadas em formol e processadas rotineiramente para histopatologia. Os casos foram divididos em um grupo controle, constituído por 12 casos de babesiose cerebral com substância cinzenta encefálica róseo-cereja característica (grupo A), e 38 casos sugestivos de tristeza parasitária bovina (grupo B), conforme os protocolos de necropsias. Foram testadas as técnicas histoquímicas de Azul Alciano, Azul de Metileno, Azul de Toluidina, Giemsa, Gram (método de McCallum-Goodpasture), Grocott, Ácido Periódico de Schiff e Ziehl-Neelsen. Dentre estas, observou-se que as técnicas de Azul de Metileno e Azul de Toluidina permitiram observar características morfológicas e tintoriais de maneira mais nítida, auxiliando na identificação de B. bovis. Adicionalmente, foram estabelecidos vários parâmetros clínico-epidemiológicos e anatomopatológicos da babesiose por B. bovis.(AU)


Babesiosis is a hemolytic tick-borne disease caused by intraerythrocytic protozoal parasites of the genus Babesia. This is a disease of high incidence in the southern Brazil and responsible for considerable economic losses. Clinical-pathological diagnosis can be made by demonstrating the parasite by examining blood smears or fresh tissues. However, frequently, only formalin-fixed organs are sent to pathology laboratories, which makes it impossible to perform the smears. The main objective of this study was to find alternative histochemical techniques capable to improve the identification of intraerythrocytic Babesia bovis in histological sections. For this study, tissue samples from 50 bovine necropsy cases were retrospectively analyzed, which had been fixed in formalin and routinely processed for histopathology. The cases were divided into a control group, consisting of 12 cases of cerebral babesiosis with characteristic pink-cherry gray matter (group A), and 38 cases suggestive of cattle tick fever (group B), according to necropsy protocols. Histochemical techniques of Alcian Blue, Methylene Blue, Toluidine Blue, Giemsa, Gram (McCallum-Goodpasture method), Grocott, Periodic Acid of Schiff and Ziehl-Neelsen were tested. Among these, it was observed that the techniques of Methylene Blue and Toluidine Blue allowed to observe morphological and dye characteristics in a clearer way, aiding in the identification of B. bovis. In addition, several clinical-epidemiological and anatomopathological parameters of babesiosis caused by B. bovis were established.(AU)

ABSTRACT Pollen morphology characterization is important in taxonomy, conservation and plant breeding, and pollen viability studies can support breeding programs. This study investigated pollen morphology and male fertility in 18 species of Bromeliaceae with ornamental potential. For morphological characterization, pollen grains were acetolyzed and characterization of exine was done using scanning and transmission electron microscopy. Pollen viability was investigated by in vitro germination and histochemical tests. Species belonging to Aechmea and Ananas genera presented medium size pollen, except for Ae. fasciata, with large pollen. Al. nahoumii, P. sagenarius and the Vriesea species analyzed showed large pollen, except for V. carinata, with very large pollen. Pollen of Aechmea, Ananas and P. sagenarius presented bilateral symmetry, diporate, exine varying from tectate to semitectate. Al. nahoumii and Vriesea species presented pollen with bilateral symmetry, monocolpate; exine was semitectate, reticulate and heterobrochate. Germination percentage and tube growth were greater in SM and BKM media. Histochemical tests showed pollen viability above 70% for all species, except for Ananas sp. (40%). Pollen morphology is important for the identification of species, especially in this family, which contains a large number of species. High rates of viability favor fertilization and seed production, essential for efficient hybrid production and conservation.

A study on the leaf anatomy of Cordiera sessilis (Rubiaceae), a native medicinal shrub from Brazilian Cerrado was carried out to identify features that may be useful in species recognition. Leaves were collected, fixed and processed by usual techniques, and studied by light and electron microscopy. Quantitative analyzes of stomata and trichomes were performed. In addition to the typical anatomical characteristics of Rubiaceae leaves, two types of vascular patterns were identified in the petiole: in distal part, the vascular system is arranged cylindrically surrounded by sclerenchyma sheath and in proximal part the vascular system is arranged in U-shape coupled to sclerified cells. The micromorphological organization of leaf surface, epicuticular wax types, the petiole pattern and histochemical characteristics as the presence of druses, crystal sand and alkaloids and absence of raphides in the mesophyll, midrib and petiole are considerate representative characteristics of C. sessilis and may be useful in the species recognition.


Um estudo da anatomia foliar de Cordiera sessilis (Rubiaceae), um arbusto medicinal nativo do Cerrado brasileiro foi realizado com o objetivo de identificar características úteis no reconhecimento da espécie. As folhas foram coletadas, fixadas e processadas por meio de técnicas usuais e estudadas em microscopia de luz e eletrônica. As análises quantitativas dos estômatos e tricomas foram realizadas. Além das características anatômicas típicas das folhas de Rubiaceae, dois padrões vasculares foram identificados no pecíolo: na porção distal, o sistema vascular disposto cilindricamente, contornado por uma bainha esclerenquimática e na porção proximal, o sistema vascular organizado em forma de U associado a células esclerenquimáticas. A organização micromorfológica da superfície foliar e da cera epicuticular, o padrão do pecíolo e as características histoquímicas como a presença de drusas, areia cristalina e alcaloides e a ausência de cristais do tipo ráfides no mesofilo, nervura central e pecíolo são consideradas características representativas de C. sessilis e podem ser úteis para o reconhecimento da espécie.

Abstract Myrciaria glomerata O. Berg., Myrtaceae, popularly known as "cabeludinha", has high content of ascorbic acid and anti-inflammatory property and is used in folk medicine. The objectives of this study were the morphological, anatomical and histochemical characterization of the leaves. Leaf studies were made with optical, scanning electron and confocal microscopy. The collection of botanical material was held at the Tijuca Forest, Rio de Janeiro, RJ. Histochemical tests aimed the identification of lipids, starch grains, phenolic compounds and crystals. The leaves are simple, opposite, lanceolate, pinnate, hairy, with involute margins, hypostomatic and dorsiventral. The stomata are anomocytic. The epidermis presents simple trichomes. Epidermal cells show uneven thickening of their periclinal outer walls, mainly on the adaxial side of the leaf. Secretory cavities of essential oils are subepidermal and exceed, in height, the palisade parenchyma, formed by one cell layer. Four to five cellular layers, rich in phenolic compounds and lipids form the spongy parenchyma. The bundles are collateral and there are many crystals of calcium oxalate spread throughout the mesophyll. In the midrib and petiole the bundles are bicollateral. Analysis by scanning electron revealed epicuticular wax rod-shaped and as grains. In confocal microscopy, the adaxial epidermis, the fibers and the secretory epithelium of the cavities show autofluorescence. The data obtained are important in quality control exams of samples of this species.

Abstract Amphilophium crucigerum (L.) L.G. Lohmann, known as “pente-de-macaco” is a species of Bignoniaceae native to Brazil, and whose seeds are used in folk medicine. This study aimed to describe morphoanatomical features of this species of fruit to aid in its correct identification and pharmacognostic analysis. Samples of ovary, pericarp and seed were fixed with 3% glutaraldehyde, sectioned on a rotary microtome and analyzed by stereomicroscope. The results are shown in three parts: (1) The ovary presents peltate trichomes, long non-glandular trichomes and emergences in the epidermis; it is 2-carpellate and unilocular with two intruding parietal placenta; ovules are numerous on the placenta; it presents a large quantity of crystals. (2) The pericarp is woody, densely echinate and elliptic shape; it presents a 2-valved capsule and is septicidal; it presents emergences, stomata, lenticels, crystals and a large quantity of clustered stones cells. (3) Seeds are alate, exalbuminate and exotestal; there is a large amount of crystals in the exotestal region; it presents an endothelium and remnant endosperm. Histochemical tests showed the presence of lipophilic substances, polysaccharides, phenolic substances, alkaloids and a small quantity of starch. These pharmacobotanical features described for A. crucigerum are essential for the pharmacognostic analysis of the drug plant.

Objective:To investigate the expression of AEG-1 gene in NSCLC and its clinical significance. Methods:Selected our hospital cardiothoracic surgical resection of 83 cases of postoperative cancer tissues of NSCLC patients and 20 paracancer to study, immunohistochemical staining was used to detect the expression level of AEG-1 protein in two groups,the clinical and pathological of AEG-1 protein in patients with NSCLC was analyzed. Results:NSCLC tissues AEG-1 protein expression 46 cases ( 55. 42%) was sig-nificantly higher than 2 cases ( 10. 00%) of paracancer ( P0. 05 ) . AEG-1 high expression of NSCLC in patients with a median survival time of 15. 0 months was significantly lower than that of 19. 0 months (log-rankχ2=4. 119 P<0. 05,) in patients with low expression of AEG-1. Conclusion:AEG-1 gene expression has been up-regulated in NSCLC tissue,which was related to the clinical stage and distant metastasis of the patients.

A 4-year-old female captive-bred snake of the genus Bothrops showed swelling on the left side of the oral cavity, suggesting the development of neoplasia. The mass was removed surgically and sent for pathological examination. Two months later a new increase in volume in the same site was observed, suggesting recurrence. The lesion was completely removed and sent for pathological analysis. Histologically, the two-samples consisted of a mass with highly-cell density composed of spindle-shaped anaplastic cells arranged in interwoven bundles, distributed throughout the tissue extension and, occasionally, polygonal cells arranged in irregular fascicles. The Masson trichrome staining showed modest amount of collagen supporting the neoplastic cells. PAS-positive content was not observed in the cytoplasm of neoplastic cells. Histological and histochemical findings indicated that it was a spindle cell neoplasm, but the classification was not possible. Immunohistochemistry was requested and performed using the streptavidin-biotin-peroxidase method. The markers used were anti-vimentin, anti-PCNA, anti-EMA, anti-melan A and anti-melanosome, anti-desmin, anti-actin, anti-CD68 and anti- S100protein. The neoplastic cells were immunoreactive for vimentin and PCNA and negative for the other antibodies. The morphology characterization, histochemical and immunohistochemical analysis of neoplastic cells allowed the definitive diagnosis of oral fibrosarcoma...


Uma serpente de cativeiro, fêmea, quatro anos de idade, do gênero Bothrops apresentou aumento de volume no lado esquerdo da cavidade oral, sugerindo tratar-se de neoplasma. A massa foi removida cirurgicamente e enviada para exame anatomopatológico. Dois meses depois foi observado novo aumento de volume no mesmo local, sugerindo recidiva. A lesão foi removida por completo e também enviada para análise. Histologicamente, as duas amostras consistiam de massa altamente celular, composta por células anaplásicas fusiformes organizadas em feixes entrelaçados e distribuídos por toda extensão tecidual e, ocasionalmente, células poligonais arranjadas em fascículos irregulares. A coloração de tricrômico de Masson apresentou quantidade modesta de colágeno sustentando as células neoplásicas. Não foi observado conteúdo PAS-positivo no citoplasma das células neoplásicas. Os achados histológicos e histoquímicos indicavam tratar-se de neoplasma de células fusiformes, porém não era possível sua classificação. A imuno-histoquímica foi requisitada e realizada pelo método streptavidina-biotina-peroxidase, utilizando os anticorpos anti-vimentina, anti-PCNA, Anti-EMA, anti-melan A, anti-HMB45, anti-desmina, anti-actina, anti-CD68 e anti-proteína S-100. As células neoplásicas foram imunorreativas para vimentina e PCNA e, negativas para os demais anticorpos. A caracterização morfológica, histoquímica e imuno-histoquímica das células neoplásicas permitiu o diagnóstico definitivo de fibrossarcoma oral...

OBJECTIVE: To investigate the structural features of vegetative organs, the location of alkaloids and saponins, and the contents of sipeimine-3β-D-glucoside, sipeimine and peiminine to clarify the similarities and differences in the bulbs of medicinal plants of Fritillaria from Xinjiang. METHODS: Anatomy, histochemistry and phytochemistry approaches were used. RESULTS: All the bulbs consisted of epidermis, parenchyma cells and closed collateral vascular bundles. The basic structure of bulbs was similar to medicinal plants of Fritillaria, but no feature could characterize the species. The histochemical test revealed that alkaloids and saponins were present in the bulbs, but the intensity and parts of histochemical reaction differed among different medicinal plants. The alkaloids existed in the parenchyma cells and vascular bundle of bulbs, while the saponins were present in the epidermis and parenchyma cells surrounded vascular bundle of bulbs. The phytochemical analysis revealed that sipeimine-3β-D-glucoside, sipeimine and peiminine were commonly present in the bulbs of F. verticillata, F. tortifolia, F. verticillata var. albidoflora and F. yuminensis, and the content of peiminine was higher than those of sipeimine-3β-D-glucoside and sipeimine except F. yuminensis. Peiminine was absent in the bulbs of F. walujewii and F. pallidiflora. CONCLUSION: When choosing the species of Fritillaria, the fact that parenchyma cells have a large percentage and the total number of vessel is high in the bulbs of the medicinal plants should be fully considered. Data obtained in this research can be used to help identify and standardize the plant material and improve the output and quality of medicinal plants of Fritillaria from Xinjiang.

A dexametasona, um glicocorticóide sintético, tem a capacidade de atravessar a placenta aumentando o nível de circulação de corticosteróides da mãe para o feto durante a prenhez. Quando administrada nas fases finais da prenhez pode produzir efeitos indesejáveis na formação da placenta e em vários órgãos da prole. Assim, o presente estudo objetivou investigar o efeito da administração da dexametasona (0,8mg/dia/animal) nos cinco primeiros dias da prenhez, sobre o desenvolvimento placentário de ratas. Utilizou-se 30 ratas albinas, divididas em dois grupos: Grupo I -ratas prenhes sem aplicação de dexametasona, sacrificadas ao 7º e 14º dia; Grupo II -ratas submetidas à aplicação de dexametasona nos cinco primeiros dias de prenhez, sacrificadas ao 7º e 14º dia. Os resultados mostraram que a dexametasona não afetou o número e a histologia dos sítios de implantação, porém, promoveu alteração no disco placentário ocasionando hipertrofia na camada de células trofoblásticas gigantes. Não foram evidenciadas alterações no teor de colágeno, porém houve interferência no metabolismo do glicogênio no espongiotrofoblasto trofospongio. Na morfometria de linhas houve diferença entre os grupos na região de labirinto e células trofoblásticas gigantes, porém a morfometria de pontos só ratificou as alterações percebidas na região do labirinto.


The dexamethasone, a synthetic glucocorticoid, has the ability to cross the placenta by increasing the level of movement of corticosteroids from mother to fetus during pregnancy. When administered in the late stages of pregnancy can produce effects undesirable on placental formation. The present study aimed to investigate the effect of administration of dexamethasone (0.8mg/day/animal) in the first five days of pregnancy, on placental development in rats. We used 30 albino rats, divided into two groups: Group I -pregnant rats without the application of dexamethasone, sacrificed to the 7th and 14th day. Group II -rats subjected to the application of dexamethasone in the first five days of pregnancy, sacrificed to the 7th and 14th day. The results showed that dexamethasone did not affect the number and histology of the implantation sites, but promoted changes in the disk placental causing hypertrophy in trophoblastic giant cell layer. No changes were found in the content of collagen, but there was interference with the metabolism of glycogen in spongiotrophoblast. The morphometry of lines showed, a difference between groups in the region of labyrinth and trophoblast giant cell. However, in morphometry of points there was a difference between groups in the region of labyrinth.

Descreveram-se os sinais clínicos e achados anatomopatológicos da intoxicação crônica por cobre em um ovino da raça Texxel e definiu-se a conduta diagnóstica correta para confirmação da enfermidade. Um ovino foi encaminhado ao setor de patologia com histórico de apatia, hemoglobinúria e morte em dois a três dias. No exame necroscópico, observaram-se icterícia e edema subcutâneo, fígado aumentado de volume e amarelado e rins escuros. No exame histológico, observaram-se necrose zonal aleatória e acentuada no fígado, necrose epitelial tubular, gotas hialinas e cilindros marrom-alaranjados em túbulos coletores dos rins. O histórico alimentar, a sensibilidade de espécie/raça, o quadro clínico, as alterações macroscópicas e microscópicas sugeriram o quadro de intoxicação crônica por cobre. A confirmação diagnóstica somente foi possível após a marcação de pigmentos de cobre pela técnica histoquímica de Ulzmann e pela quantificação de cobre em matéria seca de fígado e rins, cujos valores foram mais altos que o normal.


The present work describes the clinical signs and anatomopathological findings of chronic copper toxicities in a Texxel breed sheep and defines the optimal diagnostic procedure for confirmation of the disorder. A sheep was sent to pathology analysis service with a history of apathy, hemoglobinuria and death within two to three days. Necropsy showed jaundice and subcutaneous edema, enlarged yellow liver and dark kidneys. The histologic examination showed random zonal necrosis, marked necrosis in the liver and tubular epithelial and orange-brown spotted hyaline cylinders in the collecting tubules of the kidneys. The dietary history, sensitivity of species/breed, clinical, macroscopic and microscopic alterations suggested the framework of chronic copper poisoning. Diagnostic confirmation was only possible after staining copper pigments trough the Ulzmann technique and quantification of copper in the dry liver and kidney, which were higher than normal levels.

O presente estudo avaliou a permeabilidade dentinária em toda a extensão do canal radicular após o uso do EDTA associado a diferentes concentrações de NaOCl. Foram utilizados cinquenta incisivos bovinos divididos em cinco grupos experimentais (n=10) de acordo com as soluções irrigantes empregadas: G1- NaOCl 1%; G2- NaOCl 5,25%; G3- NaOCl 1% + EDTA; G4- NaOCl 5,25% + EDTA; G5- Água destilada (grupo controle). Após o preparo biomecânico, as raízes foram submetidas ao método de coloração histoquímico com sulfato de cobre a 10% e ácido rubeânico a 1% para identificação da permeabilidade dentinária. Para a análise morfométrica as amostras foram seccionadas transversalmente, obtidas três secções de cada terço, desidratadas e clarificadas. Analisadas em microscopia óptica para quantificar a penetração dos íons cobre na dentina radicular. Após a análise estatística dos dados, os resultados indicaram que, houve diferença significativa entre os terços analisados seguindo uma ordem de efetividade do terço cervical maior que o médio e este maior que o apical. Entretanto, nenhuma diferença significante em relação à permeabilidade foi encontrada quando se comparou as diferentes soluções estudadas. Concluiu-se que o uso do EDTA associado com diferentes concentrações de hipoclorito de sódio, como irrigante final, não promoveu alteração na permeabilidade dentinária no canal radicular em relação as soluções usadas isoladamente.


This study evaluated the dentine permeability on the length of the root canal after the use of EDTA associated with different concentrations of NaOCl. Fifty bovine incisors were used divided into five experimental groups (n = 10) according to the irrigating solutions used: G1- 1% NaOCl; G2- 5.25% NaOCl; G3- 1% NaOCl + EDTA; G4- 5.25% NaOCl + EDTA; G5- distilled water (control group). After biomechanical preparation, the roots were subjected to histochemical staining method with 10% copper sulfate and then in 1% rubeanic acid alcohol solution for identification of dentin permeability. For the morphometric analysis the samples were cross sectioned, obtained three sections of each third, dehydrated and clarified. Analyzed with light microscopy to quantify the penetration of copper ions in the root dentin. After statistical analysis, the results indicated that there were significant differences among thirds analyzed by an order of effectiveness of the cervical third larger than the middle and this larger than the apical. However, no significant difference in relation to the permeability was found when comparing the different solutions studied. In conclusion, the use of EDTA associated with different concentrations of sodium hypochlorite, as final rinse, did not cause changes in dentin permeability in relation solutions used alone.

Polygonum hydropiperoides Michaux é uma espécie conhecida popularmente como "erva-de-bicho", amplamente utilizada na medicina tradicional como anti-hemorroidal, antiinflamatória e antidiarréica. O presente trabalho tem como objetivo a caracterização anatômica e histoquímica da folha, caule e raiz, que constituem a droga vegetal, visando estabelecer parâmetros para o controle de qualidade. O material vegetal foi fixado e submetido às técnicas usuais de microscopia de luz e aos testes histoquímicos. A folha é anfiestomática, dorsiventral e com estômatos paracíticos e anisocíticos. É comum a presença de estruturas secretoras como: tricomas glandulares capitados e glândulas epidérmicas e subepidérmicas em ambas as faces da lâmina foliar e também no caule. O material secretado pelas glândulas apresenta composição heterogênea de lipídios e flavonóides, segundo análises histoquímicas. Cristais de oxalato de cálcio e grãos de amido são freqüentes em células parenquimáticas da folha e do caule. Compostos fenólicos estão presentes na folha (parênquima paliçádico), no caule (parênquima cortical e floema) e na raiz (parênquima cortical).


P. hydropiperoides is popularly known as "erva-de-bicho", and it is used in traditional medicine as anti-hemorroidal, anti-inflammatory and antidiarrhoeic. The present work has as objective the anatomical and histochemical characterization of leaf, stem and root that are used as drug in order to offer elements for quality control. The botanical material was prepared for the usual optical and histochemical microtechniques. The leaf is amphistomatic, dorsiventral and paracytic and anisocytic stomata. It is common to find the presence of secretory structures as: capitate glandular trichomes and secretory glands on both epidermis, that are also present in stem. The secretion material is heterogeneous composition of lipids and flavonoids. Druses of calcium oxalate and starch are common in parenchyma cells of leaf and stem. Phenolic compounds are present in leaf (palisade parenchyma), in stem (cortical parenchyma and phloem) and root (cortical parenchyma).

To determine the prevalence of goblet cell metaplasia in endocervical and endometrial adenocarcinomas by histochemial staining and to investigate the most sensitive histochemical staining method to detect this metaplasia, a total of 90 tissue blocks representing 30 non-neoplastic cervix, 30 non-neoplastic endometrium, 30 endocervical and endometrial adenocarcinoma cases were obtained for histochemical staining with Toluidine Blue (TB), Methylene Blue (MB), Mucicarmine (MUC), Periodic Acid Schiff before and after Diastase digestion (PAS, PAS-D), Alcian Blue pH 2.5 (AB), and Periodic Acid Schiff after Alcian Blue pH 2.5 (PAB). The cases were blinded and evaluated by a pathologist [NHO] for the presence of goblet cell metaplasia, the amount of goblet cells present and the histochemical differentiation of the goblet cells compared with its surrounding glandular epithelium. Goblet cell metaplasia was present in 2 out of 30 cases in non-neoplastic cervix, 0 out of 30 cases in non-neoplastic endometrium, 7 out of 15 cases in endocervical adenocarcinoma and in 2 out of 15 cases in endometrial adenocarcinoma. Relatively few goblet cells were seen in endometrial adenocarcinoma, few to moderate amounts were seen in endocervical adenocarcinoma and relatively more goblet cells were seen in non-neoplastic cervix. The differentiation of the goblet cells with its surrounding glandular epithelium was moderate to strong in non-neoplastic cervix and endocervical adenocarcinoma, while the differentiation in endometrial adenocarcinoma was weak to moderate. The various staining methods showed differences in presence, amount anddifferentiation of the goblet cells. Goblet cell metaplasia of the reproductive organs is not as rare as previously reported. There was no statistical difference in presence, amount and differentiation of goblet cells according to the various cases. The must optimum staining methods for staining goblet cells in non-neoplastic cervix, endocervical adenocarcinoma and endometrial adenocarcinoma were PAS, PASD and AB.

OBJECTIVE: In order to establish the index of degeneration, the authors performed a histochemical study with Safranin-O staining and investigated the occurrence of apoptosis in the human intervertebral disc. METHODS: Eighteen intervertebral disc specimens surgically extracted from the patients and two additional specimens from the autopsied cases were stained with Safranin-O for proteoglycan according to a standard protocol. Terminal deoxynucleotidyl transferase-mediated deoxyuridine triphosphate-biotin nick end labeling(TUNEL) was used to detect the fragmented DNA known to be associated with apoptotic cell death and classification scheme was formulated for categorization of the degree of Safranin-O staining (normal, moderate reduction, faint) by modification of Makin's histological-histochemical grading. The Kruskal-Wallis H test and Chi-square test were used for statistical analysis. RESULTS: The statistical results showed a significant difference in the mean age between "normal" Safranin-O staining group and the others (19.3 versus 55, 43.4, p=0.021). However, there was no statistically significant correlation between Safranin-O staining and MR grading of disc degeneration. Only six of eighteen surgical specimens and none in autopsies showed positive apoptotic cells in TUNEL staining. CONCLUSION: The determination of the degree of degeneration in surgically obtained disc tissue per se by histochemical staining or by the degree of apoptosis that corresponds to its morphologic change was not feasible.
