RESUMEN

PURPOSE: To evaluate the level of cytokines and keratinocyte growth factor (KGF) or Fibroblast Growth Factor 7 (FGF-7) in the culture medium of cultured human dermal fibroblasts from patients with large burn in comparison to small burn. METHODS: Fibroblasts of 10 patients (four large burns, four small burns and two controls) were initiated by the enzymatic method using collagenase. Cytokines and KGF in the supernatant of the culture medium was measured by, respectively, flow cytometry using Cytometric Bead Array Human Inflammation kit (CBA, BD Biosciences, USA) and the enzyme immunoassay method using the Quantikine (r) Human KGF. The experiments were performed in triplicate. RESULTS: The expression of IL-12 protein in patients with large burns showed a tendency to increase. IL- 6, IL- 10, and IL- 1beta were observed no difference. For IL - 8, TNF - alpha and KGF was observed a significant difference between the expression in large and small burned patient. CONCLUSION: That IL-8, TNF-alpha and KGF showed higher expression in cultured fibroblasts of large burned patients. .

RESUMEN

Objective To study the effect of TNF-α antagonist (etanercept) treatment on the peripheral T cell reactivity of ankylosing spondylitis (AS) patients. Methods Peripheral blood mononuclear cells (PBMC) were collected from 40 patients with AS at baseline, after two and six weeks of etanercept treatment or placebo and from healthy controls. The number of cells that secret TNF-α,IL-2 and IFN-γwas respectively detected by ELISPOT. CD+/CD8+ T cell proliferation was assayed with WST-1 live cell staining method. Results After 2 and 6 weeks of etanercept treatment, the number of TNF-α secreting monocytes was decreased. Although the T cell proliferation rate was not reduced, the number of T cells secreting IL-2 and IFN-γ under anti-CD3/anti-CD28 stimulation was significantly decreased. Conclusion The anti-TNF-α therapy suppresses the functions of effector T cells. The reduced T cell reactivity may contribute to the efficacy of the TNF-α antagonist therapy in AS patients.

RESUMEN

Urea soluble antigens of Trichinella spiralis infective larva were prepared and the efficiency of the antigens in diagnosis of trichinosis by ELISA are studied. Specific IgG antibodies are detected by ELISA using urea soluble antigens in 91.67%(22/24) of 24 sera and 91.67% (ll/12)of 12 blood spots of filter paper of patients of trichinosis. Specific IgM antibodies are seen in 95.24% (20/21) of 21 sera of patients using the antigens. Sera of patients infected with Schistosoma japonicum have some cross reactions on the antigens. Sera from mice infected with - Schistosoma japonicum, Ancyclostoma caninum larva, and Plasmodium yoelii are negative. The urea soluble antigen has a relatively high sensitivity, specificity and good reproducibility in diagnosis of trichinoisis by ELISA, and has a practical and economic value.
