RESUMEN

Background: The incidence of ulcerative colitis (UC) has gradually increased in China in recent years. The pathogenesis of UC is related to the dysfunction of immune system. B7-H5 is an important immune checkpoint molecule and is significant for the regulation of immune function. Ainis: To investigate the expression and clinical significance of B7-H5 in UC. Methods: A total of 65 UC tissue specimens were collected from Jan. 2010 to Dec. 2020 at the First Affiliated Hospital of Soochow University, and 5 healthy subjects were served as controls. Immunohistoehemistry and immunofluorescence were used to detect the expression of B7-H5, and its relationship with elinieopathologieal characteristics of UC patients was analyzed. Results: Expression of B7-H5 was significantly increased in UC patients than in controls (P 0. 05). Conclusions; The expression of B7-H5 in UC patients is significantly increased and is correlated with ESR and CRP, and can be used as a new marker for reflecting the severity of inflammation in UC patients.

Objective To explore the inhibitor of growth family member 4 (ING4) and hypoxia inducible factor-1α(HIF-1α)expression in colorectal cancer and the prognostic significance. Methods Immunohistochemistry was used to detect the ING4 and HIF-1α expression in 133 cases of colorectal cancer tissues and 76 cases of normal rectal tissues. Survival analysis was performed on the following data. Results ING4 in colorectal cancer tissues with positive rate (53.4%) was significantly lower than normal rectal tissue (85.5%) and the difference was statistically significant (P < 0.01); HIF-1α in colorectal cancer tissues with positive rate (69.9%) is higher than normal rectal tissue (42.1%), and the difference was statistically significant (P < 0.01); ING4 and HIF-1αexpression was related with tumor differentiation, Dukes stage and lymph node metastasis (P < 0.05); colorectal cancer tissues ING4 and HIF-1α expression was negatively correlated (r = -0.317, P < 0.001); By multivariate analysis, tumor differentiation, Dukes stage, lymph node metastasis, ING4 expression of HIF-1α expression has independent prognostic significance. Conclusion ING4 and HIF-1α may be involved in the occurrence and development of colorectal cancer , and combined detection could help determine the prognosis of colorectal cancer.

Objective To investigate the correlation between the expression of Ku70 and Ku80 in esophageal squamous cell carcinoma and chemotherapy sensitivity and prognosis.Methods The expression levels of Ku70 and Ku80 in esophageal squamous cell carcinoma were measured by immunohistochemistry.The relationship between the expression of Ku70 or Ku80 and clinical pathology parameters,recent curative effect of concurrent chemoradiation and prognosis of the patients were analyzed.Results The positive expression rate of Ku70 and Ku80 was 59.0% (69/117) and 50.4% (59/117) in esophageal squamous cell carcinoma.The expression of Ku70 and Ku80 was significandy related to recent curative effect of esophageal squamous cell carcinomas (P =0.017,0.012).The patients with complete response had lower positive expression rate of Ku70 and Ku80 [49.2%(32/65),40.0%(26/65)].But the expression had no relationship with the age,length of lesions in CT,lymph node metastasis of esophageal squamous cell carcinoma (P >0.05).There was no relationship between the expression of Ku70 and Ku80 neither (P =0.114).KaplanMeier analysis found Ku70 and Ku80 had worse prognosis in patients with positive expression (Ku70:P =0.004 ; Ku80:P =0.025).In Cox univariate analysis concluded the same result (Ku70:P =0.005,HR =1.971,95% CI:1.230-3.159; Ku80:P =0.028,HR =1.659,95% CI:1.057-2.605); but in Cox multivariate analysis,only Ku70 expression was the independent factor (P =0.033).Conclusions The upregulation of Ku70 and Ku80 is correlated to decreased concurrent chemoradiation sensitivity.In addition,the upregulation of Ku70 is correlated to worse prognosis.Ku70 and Ku80 can be a prediction index for predicting concurrent chemoradiation sensitivity in esophageal squamous cell carcinoma.

Objective To investigate the expression and location of Matrix metalloproteinase-9 (MMP-9) in human salivary gland tumors and the effect of MMP-9 in soft and neural invasion,and to find whether there will be a rational option to treat saliary gland tumors by inducing MMP-9,especially saliary malignant tumors.Methods A total of 140 cases of human salivary gland tumors,including 32 cases of plemorphic adenoma tissues,28 cases of basal cell,adenoma tissues,40 case of mucoepidermoid carcinoma tissues,16 case of carcinomatous metastasis,24 case of no metastasis,18 cases of neurotropism,22 of case non-neurotropism,adenoid cystic carcinoma tissues of 40 cases and normal human salivary tissues of 10 cases were detected the expression and location of MMP-9 by PV-9000 polymer detection system for immunohistoch emical staining mcthod.The relationship between the expression and clinical pathological behaviors was analyzed.Results The positive rate of MMP-9 expression in salivary gland malignant tumors,salivary gland benign tumors and normal salivary glands was 72.5%,30.0% and 10.0% ( x2 =32.12,P < 0.01 ).The positive rate of MMP-9 was 93.75% (15/16) in the mucoepidermoid carcinoma tissue,which was signicantly different from that of 66.67% (16/24) in the carcinomatous metastasis group ( x2=4.038,P < 0.05 ).The positive rate of MMP-9 in the normal human salivary tissues,adenoid cystic carcinoma tissues and mucoepidermoid carcinoma tissues was 10.0% (1/10),67.5% (27/40) and 77.5(31/40) (x2=16.263,P<0.005).Significant difference was found between groups of neurotropism and non-neurotropism on the positive rate of MMP-9 expression in adenoid cystic carcinoma(94.44% vs 63.63%,x2=5.389,P < 0.05 ).Conclusion MMP-9 is correlated with the invasion and metastasis of malignant salivary gland tumors,and can be used as an indicator of potential metastasis.The finding suggests that MMPs suppressor combined with antiangiogenic agents might be a new option to treat saliary gland tumors.

In order to investigate the role of the PTEN expression in carcinogenesis and develop-ment of endometrial carcinoma and clarify whether and how PTEN and PI3K/Akt pathway relate to endometrial carcinoma,the expression of PTEN and phospho-Akt was detected by semiquantitative reverse transcription-polymerase chain reaction (RT-PCR) methods and Western-blot from 24 cases of endomctrial carcinoma,10 cases of endometrial atypical hyperplasia,10 cases of endometrial hy-perplasia,and 10 cases of normal endometrium.SP immunohistochemical methods were used to measure levels of PTEN protein expression in following 5 study groups:31 cases of endometrium in proliferative phase,30 cases of endometrium in secretory phase,71 cases of endometrial hyperplasia,25 cases of atypical hyperplasia and 73 cases of endometrial carcinoma.Immunostaining score of PTEN was 3.39±0.15 in proliferative phase,1.90±0.21 in secretory phase,3.34~0.29 in endometrial hyperplasia,0.624±0.11 in atypical hyperplasia,and 0.74±0.19 in endometrial carcinoma,respectively.PTEN mRNA relative value in normal endometrium,endometrial hyperplasia,endometrial atypical hyperplasia,and endometrial carcinoma was 2.45±0.51,2.32±0.32,0.46±0.11,and 0.35±0.13 respec-tively.The expression levels of PTEN mRNA and protein in patients with endometrial carcinoma and atypical hyperplasia were significantly lower than in those of proliferative phase and with endo-metrial hyperplasia.The level of PTEN expression in patients with endometrial carcinoma was sig-nificantly related to tissue type (P<0.005),differentiation (P<0.05) and clinical stage (P<0.05),but not to depth of myometrium invasion (P>0.05).Western blot analysis revealed that Phospho-Akt level in PTEN negative cases was significantly higher,and there was a negative correlation between PTEN and phospho-Akt (r=- 0.8973,P<0.0001).It was suggested that loss of PTEN expression was an early event in endometrial tumorigenesis.The phosphorylation of Akt induced by the loss of PTEN took part in the tumorigenesis and development of endometrial carcinoma.

RESUMEN

Objective To investigate the clinicopathological features of glomus tumor and its differential diagnosis. Methods Clinical, morphologie and immunohistoehemical representations were described in 11 cases of glomus tumor. Results The tumors were located at the hyponychial in 7 cases, forearm subcutaneous in 2 cases, knee subcutaneous in 1 case, gastric antrum forewall in 1 cases. Localized pain was the primary symptom. 9 giomus tumors were histopathologically classified as solid type, and 2 glomangiomatous type. The prominent morphologic features of glomus tumor were as follows: the tumor cells were small, round, uniform, the tumor cells arranged around capillary vessel or expansion of the small vein showing nest pattern,some tumor cells in fasciation pattern; focally myxoid change in the stroma were seen. Immunohistochemistry: 11 cases were positive for SMA and Vimentin; while other markers including CK, CD117, CD34, CgA, Syn, Desmin, S-100 and HMB45, were all negative. All 11 cases were without any evidence of recurrence or metastasis in followed-up for 12-30 months. Conclusion Glomus tumor is extremely rare. The diagnosis depends on the histology and immunohistochemistry. Glomus tumor is rare in visceral sites. It may be misdiagnosed and needs to he differentiated from other tumors.

Objective To study the expression of NM-23 ,PCNA,p53 and S-100 in colorectal cancer tissues and their relationship.Methods Three hundred and seventy eases of elderly eolorectal cancer were labelled with NM-23,PCNA,p53 and S-100 and staged by pTNM classification.Results The positive rates of NM-23,PCNA,p53 and S-100 were 74.9% ,60.3% ,58.1% ,68.1% respectively.In elderly group,the expression of NM-23 had a neg-ative correlation with that of PCNA (r = -0.101 ,P = 0.026).However the expression of NM-23 had a positive corre-lation with that of p53 (r =0.111 ,P =0.016 ),the expression of S-100 had a positive correlation with that of p53 (r=0.112,P = 0.015 ),the expression of S-100 had a positive correlation with that of PCNA (r =-0.229,P =0.000).Conclusions In the elderly patients,NM-23,p53 and S-100 were significantly correlated with the inva-sion,metastasis and prognosis of eoloreetal cancer.PCNA may be a stimulative factor in the invasion and metastasis of colorectal cancer.NM-23 ,PCNA,p53 and S-100 can serve as effective markers in reflecting the invasion,metas-tasis and prognosis of colorectal carcinoma in the elderly.

Objective:To observe clinical and pathological features of pancreatic solid-pseudopapillary tumor ( SPPT) , and to find some useful immunohistochemical methods for its differential diagnosis.Methods: The clinical features of 14 SPPT patients were obtained. Each case underwent microscopic observation and immunohistochemical staining. The primary antibodies were CgA, Syn, E-cadherin, β-catenin and Cyclin Dl. These results were compared with 5 pancreatic well-differentiated tumors and well-differentiated carcinomas ( WET/WEC). Results: SPPT mainly involved young women, and the head of pancreas was the commonest location. Tumors were always in solid and cystic gross appearance.Although the tumor' s borderlines seemed clear, focal infiltrations could often be identified. The histological features of SPPT were similar in some aspects to those of WET/WEC, especially the solid pattern of WET/WEC. Both of them could express CgA and Syn. But all SPPTs lost E-cadherin membranous signals, and even had some nuclear signals (5/14) , while all WET/WECs remained the same staining pattern with normal pancreas cells, β-catenin positive signals in SPPTs were located both in nuclei and plas mas. WET/WECs' positive signals were all in membranes and plasmas, but negative ones in nuclei. Perinuclear dot-like signals could also be seen in the majority cells, which were similar to normal islet cells' staining pattern. SPPTs' nuclear positive rates of Cyclin Dl were usually more than 70% (12/14). WET/WECs' rates were all lower than 30%. Conclusion: Comprehensive analysis of patients'clinical, pathological features and immunohistoehemistry results, including E-cadherin, β-catenin and Cyclin Dl, was helpful to the diagnosis of SPPT and its differential diagnosis of WET/WEC.

RESUMEN

Objective To investigate the association between the expression of RASSF1A and Survivin proteins and clinicopathological characteristics in non-small cell lung cancer (NSCLC) and their clinical significance. Methods Expression of RASSF1A and Survivin proteins in the NSCLC tissue microarrays was detected by S-P Immunohistochemistry. Results The positive expression of RASSF1A in NSCLC (46.8 %) was significantly lower than that of in the normal lung tissues (92.9 %) (P < 0.001), but the positive expression of Survivin in NSCLC (78.5 %) was significantly higher than that of in the normal lung tissues (0%) (P<0.001). The percentage of RASSFI A protein expression in the stage Ⅰ and stage Ⅱ of NSCLC was evidently higher than that of in the stage Ⅲ (P<0.001, respectively), however, the percentage of Survivin protein expression in the stage Ⅰ and stage Ⅱ of NSCLC was significantly lower than that of in the stage Ⅲ (P=0.003, P=0.001). The percentage of RASSF1A in NSCLC with lymph node metastasis was observably lower than that of in cases without lymph node metastasis (P<0.05). Furthermore, there was observably negative correlation between expression of RASSF1A protein and that of Survivin protein in NSCLC (r=-0.780, P<0.001). Conclusion The loss expression of RASSF1A protein, over expression of Survivin protein and loss balance of expression of both RASSF1A and Survivin proteins in NSCLC might play important roles in the development and progression of NSCLC; RASSF1A and Survivin proteins might be acted as one helpful molecular marker to predict the lymph node metastasis and the prognosis of NSCLC.

Objective To investigate the expression and clinical significance of Matriptase and HAI-1 in bladder transitional cell carcinoma( BTCC), and to explore their relationship with the clinicopathologieal pa-rameters of BTCC. Methods Expressions of Matriptase and Hal-1 protein in specimens of BTCC and normal bladder tissues were detected by SP method of immunohistochemistry. The results were analyzed in relation to the clinicopathological data. Results The expression of Matriptase and HA1-1 were observed in both normal and tumor tissues of bladder;The protein levels of Matriptase and HAI-1 were significantly higher in normal bladder tissues as compared with BTCC tissues ( P < 0. 05 ) ; a progressive decrease in the protein levels of Hal-1 was observed with increasing of tumor grades (r = -0. 634,P < 0. 001) and clinical stages (r=-0. 521,P < 0. 001 ) ; the protein level of Matriptase was decreased with tumor grade increasing, and was significantly higher in high-grade tissues as compared with moderate and low-grade tissues( P < 0. 05 ) ; Matriptase protein levels showed no significantly difference between different tumor clinical stages ( P > O. 05 ). The correlation co- efficient of Matriptase and HAI-1 in normal tissues was 0. 772(P <0. 001 )and 0. 546 in tumor tissues of blad- der( P < 0. 001 ). Conclusion The abnormal and decreased expressions of Matriptase and HAI- 1 may be im- portant events during the genesis and progression of BTCC in humans; Hal-1 may be used as a new parameterfor assessing the malignancy and prognosis of BTCC; Matriptase protein in BTCC is related with differentiationpromotion, and may be an indicator of differentiation. The correlation between Matriptase and Hal- 1 decreasesin BTCC. The imbalance between HAI-1 and Matriptase may be contributed to the progression of BTCC.

RESUMEN

Objective To study the expression of Livin and its relalionship with expression of p53 in small cell hmg cancer(SCLC).Methods Immunohistochemical S-P method was used to detect the ex-pression of Livin and p53 protein in 30 SCLC tissues and 16 para-cancerous lung tissues.Results Livin protein was expressed in 17 of 30 SCLC tissues(56.67%),but Livin protein showed low levels in para-can-cerous lung tissues(12.50%),P<0.01.There was no significant correlalion between positive Livin protein expression and age,sex,TNM staging,lymph node metastasis and lumor diameter(P>0.05).p53 protein was expressed in 14 of 30 SCLC tissues(46.67%),but p53 protein was no expressed in para-caneerous lung tissues,P<0.01.The expression of Livin protein was positively related to the expression of p53 protein(P<0.01).Conclusions The aberrant expression of Livin may be a new target for diagnosis and gene treatment of SCLC.The aberrant expression of Livin and p53 may play synergetic role in process of carcinogenesis of SCLC.

RESUMEN

The depositions of immunoglobulins on the retina of diabetic rat were studied with immunohistochemistry,immunofluorecence and computer analysing system.In comparison with control group, depositions of IgG,IgA and IgM on the retina of diabetic rat induced by streptozotocin were significantly increased (all P<0.05 ).So the immunoglobulin depositions may contribute to the pathogenesis of diabetic retinopathy.
