Objective To explore the expression level and clinical significance of serum interleukin-2 (IL-2),IL-4,IL-6,IL-10,interferon-γ (IFN-γ) and tumor necrosis factor-α (TNF-α) in patients with chronic periodontitis and chronic periodontitis complicated with chronic obstructive pulmonary disease (COPD).Methods From August 2015-August 2016,31 COPD patients combined with chronic periodontitis were set as group A,31 patients with chronic periodontitis were set as group B,and another 31 healthy subjects were selected as the control group in Chaoyang Hospital.Each group extracted fasting venous blood 4 ml,serum inflammatory factors levels (IL-2,IL-4,IL-6,IL-10,IFN-γ and TNF-α) were measured by cytometric bead array (CBA),periodontal condition [plaque index (PLI),clinical attachment loss (CAL),probing depth (PD),bleeding index (BI)] and lung function index [first 1 s forced expiratory volume% of predicted value (FEV1%),first 1 s forced expiratory volume occupies the percentage of vital capacity (FEV1/FVC)] were compared.Results The PLI,CAL,PD and BI levels in group B were higher than those in control group,indicators of group A were higher than group B,the difference was statistically significant (P < 0.05).There was no significant difference in FEV1 % and FEV1/FVC between group B and control group,indicators of group A were less than group B and control group,the difference was statistically significant (P < 0.05).The serum levels of IL-4,IL-6,IFN-γ and TNF-α in group B were higher than those in control group,indicators of group A were significantly higher than group B,the levels of IL-2 and IL-10 in group B were significantly lower than those in control group,indicators of group A were significantly less than group B (P < 0.05).Conclusions Periodontal status and lung function of chronic periodontitis and chronic periodontitis with COPD patients is not good,the serum levels of IL-4,IL-6,IFN-γ,and TNF-α were increased,and the serum levels of IL-2 and IL-10 were decreased,which might be important risk factors for chronic periodontitis and COPD happen and progress.Therefore,treatment regimen can be adjusted by detecting the levels of above indicators.

Objective To investigate the changes of T cells and dendritic cells (DCs) and their related factors in children with immune thrombocytopenia (ITP) before and after therapy,and to analyze their clinical significance.Methods T-cells and DCs subsets were determined by flow cytometry both in 64 children with ITP (ITP group) before and after therapy and the control group.The serum levels of interleukin (IL)-4,interferon-γ (IFN-γ),transforming growth factor beta 1 (TGF-β1),and IL-27 were detected by enzyme linked immunosorbent assay (ELISA).Results Treatments of glucocorticoid or IVIg were effective in 41 cases of 64 ITP children.Compared to the control group,helper T cells (Th),Th/suppressor T cells (Ts),T regulatory cells (Treg),plasmacytoid DC (pDC),pDC/myeloid DC (mDC),and TGF-β1 in ITP patient group before treatment were significantly lower,while IFN-γ and IFN-γ/IL-4 were significantly higher (P <0.05).In ITP group,Th,Th/Ts,Treg,pDC,pDC/mDC,TGF-β1,and IL-27 were significantly increased,while IFN-γ and IFN-γ/IL-4 were decreased in children with ITP after therapy and achieved response (P < 0.05).However,there was no significant difference between before and after therapy in ITP children without treatment response (P > 0.05).Conclusions T cells and DCs subsets disorder and abnormal cytokine levels are observed in children with ITP,which can be corrected by immunosuppressive therapy,indicating that Th1 overactivity and the decrease of Treg and pDC both in quantity and function may be related to the pathogenesis of ITP in children.

Objective To explore the correlation between the serum levels of interferon (IFN)-γ,interleukin (IL)-18,IL-10 and recurrent spontaneous abortion (RSA).Methods Enzyme-linked immunosorbent assay was used to detect the serum levels of IFN-γ,IL-18 and IL-10.There were 40 patients with RSA (RSA group) and 40 normal early pregnant women (control group) enrolled in this study.After 2 months,those serum cytokines in two groups of non-pregnant state were detected again.Results Compared to control group,the serum levels of IFN-γ and IL-18 of RSA group were significantly higher (P <0.01),the serum level of IL-10 was significantly lower (P < 0.01).Two months after termination of pregnancy,there was no significant difference in the serum levels of IFN-γ and IL-10 between the two groups.The serum level of IL-18 maintained a high level in the RSA group after termination of pregnancy.Conclusions In patients with RSA,the serum levels of IFN-γ and IL-18 were elevated,the serum level of IL-10 was decreased.The serum levels of IFN-γ,IL-18 and IL-10 were closely related with RSA.

ObjectiveTo explore the changes of the frequency and cytokine production of CD19 + CD5 + B cells in Endometriosis before and after treatment.MethodsFlow cytometry and quantitative PCR were used to assess the frequency,cytokine expression of CD19 + CD5 + B cells in the peripheral blood,peritoneal fluids and endometrium biopsies of 35 patients with Endometriosis and 29 patients with myoma of uterus (control subjects).Changes in the CD19+ CD5+ B cell compartment in the peripheral blood were compared before and after treatment.ResultsAll 35 endometriosis patients had higher frequencies(all P <0.01 ) of CD19+ CD5 + B cells in the peripheral blood,peritoneal fluids and endometrium biopsies [ (13.1 ± 1.9)%,(12.1 ±2.0)%,(11.7 ± 1.7)%] than 29 control subjects[ (2.9 ±0.8)%,(2.6 ±0.9)%,(2.8 ±1.1 )% ].CD19 + CD5+ B cells from endometriosis patients expressed higher IFN-γ[ (7.2 ± 1.0) × 103,(7.9±1.3) ×103,(7.4±1.1) ×103 copies] than control subjects [ (1.9±0.7)×103,(2.2±0.8)×103,(2.0 ±0.5) × 103 copies).The endometriosis patients also had higher IL-10 production of CD19 + CD5 + B cells [ (6.4 ±0.9) × 103,(6.8 ± 1.2) × 103,(6.1 ±0.8) × 103 copies] than control subjects [ ( 1.7 ±0.5) × 103,(2.1 ±0.9) × 103,( 1.6 ±0.4) × 103 copies].Compared with control subjects,the endometriosis patients had a clinical response to treatment demomtrated a significant decrease in CD19 + CD5 + B cells in the peripheral blood[ ( 13.1 ± 1.9)% vs (7.3 ± 1.2)% ],expressed lower IFN-γ [ (7.2 ± 1.0)×103 copies vs (3.3 ±0.6) × 103 copies],produced less IL-10 [(6.4 ±0.9) × 103 copies vs (3.2 ±0.7) × 103 copies].ConclusionsCD19+ CD5 + B cells might play an important role in the pathogenesis of endometriosis through higher IFN-γand IL-10 expression.
