RESUMEN

Objective To study the expression of cyclooxygenase2 in hepatic inflammatory reaction of rats with sepsis and its effect on metabolism.Methods Fifty-four Wistar rats were randomly divided into 3 groups:Sham(A)group,sepsis model(B)group,and NS398 intervention(C)group.The rats were subjected to cecal ligation and puncture(CLP)for sepsis model or sham operation.RT-PCR was used to determine COX-2 mRNA expression,serum prealbumin(PA)and transferrin(TF)by radioimmunity.At the same time,ALT,AST,albumin determination and liver pathological changes were determined.Results(1)The expression of COX-2 mRNA was low in A group,higher in B and C groups,and higher in B than C group at each time period(P

RESUMEN

ObjectiveTo explore the development and mechanism of hepatic cell apoptosis, and the effect of anti-apoptosis apoptosis of Se in the liver in obstructive jaundice rats. Methods(GPx) activity and (ROS) level were measured by biochemical methods,terminal-deoxynucleotidyl medicated nick end labeling (TUNEL)in different groups at different time points, and analysed by multivariate regression. Results Apoptosis cells appeared on day 3 after common bile duct(CBD) ligation ,significantly increased on day 7,reached the peak level on day 11, and began to decrease on day 14. The longer the time of obstructive jaundice in rats, the higher the level of ROS and apotosis index(AI), while the level of GPx decreased progressively. AI reached the peak 11-day after CBD ligation. RESUMEN

Objective To investigate the relationship between the intercellular adhesion molecule-1(ICAM-1) and lymphocyte function associated with antigen-1( LFA-1) and acute rejection of liver transplantation in rats. Methods SD rats and Wistar rats were randomly divided into normal control group, isograft group, and allograft group. Liver biopsies from the recipients were obtained on the day 2,4, and 7 postoperatively to examine the expression level of ICAM-1 and LFA-1 in liver grafts. Samples were firstly stained with specific monoclonal antibodies using immunohistochemistry stain and then applied to image semiquantitive analysis. Results The expression levels of ICAM-1 and LFA-1 in allograft group were obviously higher than those in both isograft group and normal group. Moreover, the expression levels of ICAM-1 and LFA-1 were correlated positively with the degree of acute liver rejection. Conclusions Detecting the expressions of ICAM-1 and LFA-1 in liver graft tissue may be helpful to the diagnosis of acute rejection after liver transplantation.

RESUMEN

Objective To explore the therapeutic effects and mechanisms of somatostatin on liver injury in acute hemorrhagic necrotizing pancreatitis(AHNP) in rats.Methods Murine model of AHNP was established by retrograde injection of artificial bile into the biliopancreatic duct.The animals were divided into sham operation group(S group), AHNP saline injection group(AHNP group), and AHNP octreotide treated group. RESUMEN

Objective To explore the regulating mechanism of inducible nitric oxide synthase(iNOS) in hepatic injury of obstructive jaundice (OJ) in vivo and in vitro experiments. Methods (1) Rat hepatocytes were isolated by in situ collagenase perfusion and primary culture. Hepatocytes were pretreated with various concentrations of iNOS inhibitor SMT for 20 min. After pretreatment, 50?M GCDC was added for an additional 24hr. Cells were next detected by FCM and TUNEL.(2) Experimental obstructive jaundice (BDL) was induced by double ligation of the bile duct in rats. After BDL for 3d、7d、14d、and 21d, the apoptotic status in liver of all rats were determined with TUNEL, and iNOS protein in liver of OJ was ditermined with immunohistochemistry method. Results (1) SMT decreased GCDC-induced apoptosis in a concentration-dependent manner. (2) The apoptotic rate of liver was related to length of time of OJ. Apoptosis index (AI) was highest from rats with 14d bile duct ligation. The stronger the iNOS expression, the higher was the number of apoptotic cells that was found in OJ. Conclusions iNOS is involved in the regulation and the occurrence and progression of hepatic injury of obstructive jaundice.
