RESUMEN

Massoia (Cryptocarya massoia) is an Indonesian endemic plant containing essential oils with massoia lactone as adominant compound component. This study aims to clarify the inhalation effect of massoia lactone on body weight andlipid profile on Sprague Dawley rats. Chemical components of two fractionated portions among four fractions frommassoia oil were identified to be composed of 97% and 1.4% massoia lactone, thus named as massoia lactone rich andpoor fractions, respectively. The two fractions and massoia oil were further used for in vivo tests on four treatmentgroups (one group was negative control). The results showed that the experimental animals that inhaled massoialactone rich fraction had the lowest body weight analysis of variance (ANOVA p < 0.05), while the animals that inhaledmassoia oil and poor massoia lactone fraction had the lowest average cholesterol and triglyceride concentrations(ANOVA p < 0.05). These data indicate that the inhalation of fractionated massoia oil has the potential to decreasebody weight, restrain inclining of triglycerides and cholesterol concentration in the blood. Also, the number of livercells with a larger size than normal liver cells in rats with massoia lactone rich and poor fraction treatment was lessthan the negative control and treated with massoia oil.

Objective To calibrate contact parameters of liver tissue discrete element model. Methods Based on MATLAB image processing technology, the accumulation angle of liver tissues was measured. The ‘Hertz-Mindlin with JKR’ contact model was used to simulate the accumulation angle of liver tissues. The orthogonal experiment was designed with the coefficient of rolling friction and the energy of JKR surface as factors. The parameters of the contact model were calibrated by batch processing, and the optimal parameter combination was verified by secondary simulation calibration. Results The accumulation angle obtained by the physical test was 11.2°±0.86°. In the orthogonal experiment, the accumulation angle of the 6th set of parameter combinations was 11.8°, and the relative error was 5.1%. The simulation test and the physical test had a high similarity in accumulation angle and shape. The sequence of factors affecting the accumulation angle was the JKR surface energy between the tissue particles and the stainless steel plate > the rolling friction coefficient between the tissue particles and the stainless steel plate > the JKR surface energy between the tissue particles and the tissue particles=the rolling friction coefficient between the tissue particles and the tissue particles. Conclusions The optimization parameter could be used to further conduct the discrete element simulation between the tissue particles and the pulverizer, so as to reveal the accumulation and flow state of the tissue particles under the action of myoma pulverizer.

Objective To provide clues for the study on the mechanism of autoimmune liver disease (AILD) by exploring the existence of specific bacteria in liver tissues of AILD patients.Methods From August 2017 to August 2018,at Department of Gastroenterology arnd Hepatology,Tianjin Medical University General Hospital,a total of 12 patients diagnosed as AILD (four autoimmune hepatitis (AIH),four primary biliary cirrhosis (PBC) and four PBC-AIH overlap syndrome (OS)) and four patients with hepatic cyst (control group) were enrolled and all the patients underwent liver biopsy.16S rRNA gene sequencing was carried out in the obtained aseptic liver tissues.Linear discriminant analysis effect size was used to find out the specific bacteria.Spearman correlation analysis was performed to analyze the correlation between the liver microbiota and the disease.The metabolic function of the 16S rRNA gene sequences was also predicted.Results Bacteria were detected in the liver tissues of all the 16 patients.At the species level,the abundance of Planococcus rifietoensis of AIH group was 0.100%,which was higher than those of other three groups (0),and the difference was statistically significant (linear discriminant analysis (LDA) =3.31,P =0.034).The abundance of Anoxybacillus flavithermus of PBC group was 0.200%,which was higher than those of other three groups (0.100%),and the difference was statistically significant (LDA =3.34,P =0.014).The abundance of Pseudomonas aeruginosa PAO1,Bacillus firmus,Brevibacillus agri,Acinetobacter baumannii,Sphingomonas zeae and Salmonella enterica were significantly negatively correlated with serum level of γ-glutamyl transferase (r=-0.68,-0.68,-0.67,-0.68,-0.68 and-0.66,all P <0.01).Compared with that of the hepatic cyst group,the lipid metabolism of AILD patients decreased.The levels of serum low density lipoprotein and total cholesterol were significantly negatively correlated with the biosynthesis of unsaturated fatty acids (r =-0.55 and-0.65,both P < 0.05).Conclusions There exist specific bacteria in the liver tissues of AIH and PBC groups.The liver microbiota which is closely related with the pathogenesis of AILD might be a potential therapeutic target and diagnostic biomarker.

RESUMEN

PURPOSE: Many researchers have tried to develop animal models that mimic the human immune system, e.g. a humanized mouse model, to improve the engraftment of hematopoietic stem cells and develop human immune cells in an animal model. This study evaluated the feasibility of the cultured human umbilical cord blood (hUCB)-derived CD34(+) cells for cell expansion, in Rag2(-/-)gamma(c)(-/-) mice, and establish co-transplantation with human fetal thymus/liver tissue (Thy/Liv) under the kidney capsule. METHODS: Co-transplantation of hUCB-derived CD34(+) cells with Thy/Liv was performed. The hUCB-derived CD34(+) cells were prepared by freshly thawing (G1) and culturing for 7 days with two types of cytokine combinations (G2, G3). The CD45(+) cell populations were measured at 6, 8, 10 and 16 weeks in the peripheral blood. The splenocytes were cultured with mitogenic stimuli (PHA -L or IL-2) at 20 weeks post- transplantation, and the proliferation of human immune cells was evaluated. RESULTS: There were no significant differences in the human CD45(+) cell populations at 6, 8, 10 and 16 weeks post-transplantation between the groups. In the cultured splenocytes at 20 weeks post-transplant with PHA-L or IL-2, there was remarkable expansion of CD3(+) cells in the three groups. Although no CD19(+) cells were detected in the spleen, human Ig G was detected in the sera of these mice. CONCLUSION: The cultured and expanded hUCB-derived cells with cytokine combinations might be a feasible cell source in humanized mouse modeling. In addition, human immune cells can be reconstituted from the co-transplantation of Thy/Liv and cultured hUCB-derived CD34(+) cells.

Objective:To investigate the relationship between serum HBV DNA levels,aminotransferase and grading and staging of liver tissues in patients with chronic hepatitis B.Methods:Routine biochemical liver function tests and serum HBV DNA level was assayed by amplisensor quantitative polymerase chain reaction in 285 patients;and the liver biopsy was performed and the grading and staging of the liver were routinely obtained.Results:There was not obvious correlation between serum HBV DNA level and the grading and staging.A significant inverse relationship between the HBV DNA and hepatitis activity was demonstrated in 230 patients with positive HBeAg.The serum ALT level was consistent with hepatitis activity.Conclusion:There is no significant correlation between serum HBV DNA and grading and staging.In HBeAg-positive patients,serum HBV DNA levels showed a significant negative correlation with hepatic activity index.The higher ALT in serum shows the more severe hepatitis activity,and ALT could be helpful for assessing the hepatitis activity.
