RESUMEN

Lomefloxacin is a flouroquinolone antibiotic that is quite efficacious against many gram negative and gram positive pathogens. Lomefloxacin evince antibacterial effects by modifying DNA gyrase in gram negative pathogens and topoisomerase IV in gram positive pathogens. This study is designed to assess the immunomodulatory effects of lomefloxacin in male albino mice. Three doses of lomefloxacin 12.5 mg/kg, 25 mg/kg and 50 mg/kg were used and delayed type hypersensitivity assay, cyclophosphamide induced neutropenic assay, carbon clearance assay, heamagglutination assay and mice lethality test were performed to evaluate the effects of lomefloxacin on immune system of mice. DTH assay has depicted the significant immunosuppressant potential of lomefloxacin at 25 mg/kg and 50 mg/kg dose. Total leukocyte count have exhibited highly significant reduction (P<0.001) in leukocyte count after cyclophosphamide administration. Differential leukocyte count has shown significant (P<0.01) reduction in lymphocyte count, whereas, highly significant (P<0.001) increase in monocyte count and significant (P<0.05) increase neutrophil count has been observed. In carbon clearance assay, highly significant (P<0.01) increase in phagocytic index has been noted at 12.5 mg/kg and 25 mg/kg doses. Humoral immune system responses are suppressed in dose dependent manner by both heamagglutination assay (P<0.001) and mice lethality assay (P<0.001). Results clearly depict that lomefloxacin possess quite significant immunomodulatory potential

RESUMEN

AIM:To study the clinical effect of silica gel drainage tube combined with lomefloxacin hydrochloride eye ophthalmic gel in the treatment of lacrimal duct obstruction.METHODS:Totally 86 cases (138 eyes) of lacrimal duct embolism treated in our hospital from February to December 2015 were divided into 43 cases (68 eyes) as control group and 43 cases (70 eyes) of the observation group according to whether they were treated with lomefloxacin hydrochloride eye ophthalmic gel.Patients in the control group were treated with silica gel drainage, while the observation group was treated with lomefloxacin hydrochloride eye ophthalmic gel on the basis of the control group.The total effective rate, serum hypersensitivity C-reactive protein (h-CRP) level, complications and recurrence rate were observed and compared between the two groups.RESULTS:The total effective rate in the patients with obstructive nasolacrimal duct, common lacrimal duct obstruction, and lacrimal duct obstruction in observation group were 95.2%, 100.0%, 96.7%, higher than those in control group (P0.05).The serum levels of h-CRP in patients in the observation group at 3d and 7d after operation respectively were 2.40±0.84g/mL, 1.94±0.84g/mL, lower than those of control group at the same time (P<0.05).The complication rate of the observation group was 1.4%, which was lower than that of the control group (P<0.05).Follow up for 6-18mo, the recurrence rate was 11.4% in the observation group, which was significantly lower than the control group (P<0.05).CONCLUSION:The combination of silica gel drainage tube and lomefloxacin hydrochloride eye ophthalmic gel in the treatment of obstruction of lacrimal duct has good clinical efficacy, low inflammatory reaction, low complication rate and low recurrence rate.

RESUMEN

Objective:To evaluate the antimicrobial effectiveness of lomefloxacin hydrochloride eye drops from 13 domestic manu-facturers. Methods:The antimicrobial effectiveness of lomefloxacin hydrochloride eye drops was tested based on the antimicrobial ef-fectiveness testing in Chinese Pharmacopoeia(2015 edition,volume 4). Results:The samples from 5 manufacturers could reach level A,those from 3 manufacturers were at level B, and those from 5 manufacturers did not comply with the specification. Conclusion:Some problems exist in the usage of antimicrobial agents in lomefloxacin hydrochloride eye drops from some Chinese manufacturers,and these manufacturers should optimize the type and concentration of antimicrobial agents according to the requirements of Chinese Phar-macopoeia combined with the corresponding prescriptions under the principle of minimum concentration while best effect.

RESUMEN

Screening aptamers using nano-materials ( such as graphene oxide, gold nanoparticle, carbon nano-tube, etc. ) that can quench fluorescence and absorb single stranded DNA using hydrogen bond, π-πbond, charge transfer, and other non-covalent ways to combine with ssDNA, but without other conformational DNA, can excellently separate specific aptamers from non-specific ones. In this case, we can shorten the cycle numbers, enhance the success rate, and reduce the labour intensity of systematic evolution of ligands by exponential enrichment ( SELEX) . Especially for small molecular target, due to its difficulty in immobilization and small size, it is difficult to use traditional methods such as SPR-SELEX or affinity-SELEX for screening. In this experiment, polydopamine nanospheres ( MNPs@PDAs) were used to screen the Lomefloxacin. Also, we used magnetic separation technique to screen small molecular target rapidly. The interaction between aptamer candidates and the target could be monitored by recovery ratio of ssDNA and the whole MNPs@PDAs-SELEX process was performed through seven-round selection. As a result, we successfully obtained the aptamer named AF-3 which could recognize the lomefloxacin with high affinity (KD=(17. 57±0. 5) nmol/L). This screening method based on MNPs@PDAs makes it a promising reagent in the efficient aptamers selection of other targets.

RESUMEN

The present work describes a novel, accurate, sensitive and economic safe spectrophotometric method was developed by application of hydrotropy, using 8 M Urea solution as hydrotropic solubilizing agent, for the quantitative determination of poorly watersoluble lomefloxacin HCl in tablet dosage form. There were more than 43 times enhancements in the solubility of lomefloxacin HCl increases in hydrotropic solution as compared to solubilities in distilled water. Lomefloxacin HCl shows maximum absorbance at 281 nm. Urea and other tablets excipents did not show any absorbance above 230 nm, and thus no interference in the estimation was seen. Lomefloxacin HCl was obeyed Beer,s law in the concentration range of 5 to 25μg/ml (r2= 0.9998) in hydrotropic solvent with mean recovery ranging from 98.03±0.65 to 98.59±0.32%. Proposed method is new, simple, economic, safe, rapid, accurate and reproducible. The developed methods were validated according to ICH guidelines and values of accuracy, precision and other statistical analysis were found to be in good accordance with the prescribed values. The method can be used for routine analysis in both research laboratories, and pharmaceutical and chemical industries to analyze the drugs without the use of organic solvents thus make the environment eco-friendly.

OBJECTIVE:To establish the method for determining the content of Lomefloxacin in hollow type suppository of Lomefloxacin hydrochloride.METHODS:RP-HPLC was used with Eclipse XDB-C 18 column(4.6mm?250mm)and the mobile phase consisting of citric acid(0.05mol/L)-acetonitrile(82∶18)the pH of which was adjusted to4.0with triethy?lamine.The flow rate was1.0ml/min and the detection wavelength was287nm.RESULTS:The average recovery of Lome?floxacin was99.8%and RSD was0.6%.CONCLUSION:The method is rapid and accurate and can be used for determination of the content of Lomefloxacin in hollow type suppository.

OBJECTIVE:To prepare lomefloxacin hydrochloride and diethylstilbestrol oil and to establish the method for its quality control.METHODS:The content of lomefloxacin was determined by UV-spectrophotometry at the wavelength of283nm.RESULTS:The detected concentration of lomefloxacin hydrochloride was in linear relation to absorptivity in the range of2.5~16.0?g/ml(r=0.9995,n=6).The average recovery was99.8%(RSD=0.74%).CONCLUSION:The preparation technique is simple and the method for quality control is feasible.

RESUMEN

OBJECTIVE:To study the bacterial endotoxin test for lomefloxacin hydrochloride and glucose injection.METHODS:According to the assigned method and guiding principle for bacterial endotoxin test in CHINESE PHARMACOPOEIA 2000,the interference test was performed.RESULTS:Lomefloxacin and glucose injection could interfere with limulus test,however,the interference could be eliminated after adjusting the pH of injection to 6.8~7.0.The proper concentration of limulus test agent of bacterial endotoxin test was 0.25EU/ml.CONCLUSION:Bacterial endotoxin test can be used for quality control of lomefloxacin and glucose injection.

RESUMEN

OBJECTIVE:To investigate the permeability of lomefloxacin through blood-pancreatic barrier in rats.METHO-DS:Lomefloxacin(20mg/kg body weight) was injected through caudal vein.At the given time points,the samples were collected.The concentrations of lomefloxacin in the serum,pancreatic tissue and liver tissue were measured by HPLC.RESULTS:The concentration-time profiles of lomefloxacin could be described as two-compartment model in rats.The peak concentrations in serum,pancreatic tissue and liver tissue were 65.550?g/ml,48.801?g/g and 84.121?g/g at 5 min post-injection respectively.Then the concentrations decreased quickly in all of them.Concentrations in pancreatic tissue were higher than those in serum at 10 min and even at 480 min post-injection.The permeation ratio (PR) through blood-pancreatic barrier was 0.744 at 5 min and rose to 3.817 at 480min.CONCLUSION:After intravenous injection,lomefloxacin can permeate blood-pancreatic barrier satisfactory,therefore it is worthy of being recommended for prevention and treatment of pancreatic infections.

RESUMEN

OBJECTIVE:A fluorospectrophotometric method was established for the determination of lomefoxacin hydrochloride in injection and capsule.METHODS:Samples were diluted with distilled water and detected by fluorospectrophotometry at excitation wavelength of 285nm and emission wavelength of 445nm.RESULTS:Linear relationship was found in the lomefloxacin hydrochloride concentrations ranging from 0.5?g/ml to 2.5?g/ml.The correlation coefficient was 0.9 998.The recoveries of lomefloxacin in injection and capsule were 99.7%(RSD=0.6%,n=5) and 99.6%(RSD=0.8%,n=5),respectively.CONCLUSION:The method is simple,accurate and sensitive and can be used to determine lomefloxacin hydrochloride in injection and capsule.
