RESUMEN

Purpose To explore the expression of bone morphogenetic protein-4 (BMP-4) in prostate cancer tissues and to analyze the effects of its down-regulation on proliferation and invasion of prostate cancer cells. Methods The expression of BMP-4 protein in prostate tissues of 58 patients with prostate cancer and 40 patients with benign prostatic hyperplasia (BPH) was detected by immunohistochemical En Vision method. The BMP-4 protein expression of different prostate cancer cells (LNcap, PC-3, Du145) and BPH was analyzed by Western blot.BMP-4 siRNA and control siRNA were transfected into LNcap respectively and then divided into three groups: untreated group, control siRNA group and BMP-4 siRNA group. Western blot was used to detect the expression levels of BMP-4 protein, MMP-2and MMP-9 in LNcap cells among the three groups. The proliferation of three groups was detected by CCK 8. The invasion ability of three groups was detected by Transwell chamber. Results The positive proportion of BMP-4 protein in prostate cancer tissues was higher than that in benign prostatic hyperplasia (P < 0.05) , and the expression level of BMP-4 protein in three kinds of prostate cancer cells was higher than that in BPH cell. The expression of BMP-4 protein in BMP-4 siRNA group was lower than that in untreated group and control siRNA group.The cell proliferation ability and cell invasion ability in BMP-4siRNA group were lower than those in untreated group and control siRNA group. The expression levels of MMP-2 and MMP-9proteins in BMP-4 siRNA group were lower than those in untreated group and control siRNA group. Conclusion BMP-4 is highly expressed in prostate cancer tissues and is associated with proliferation and invasion of prostate cancer cells, and it may become a potential therapeutic target for prostate cancer.

Cognitive impairment in diabetes ( CID) is one of the complications of diabetes .The features of mild or moderate cognitive disorder and the decread abilities in memory and studying are the main symptoms of CID .But it′s mechanism is still un-known .NF-κB is the original signal activator , it can activate other signal pathway of cell dysfunction .With the influence of hyperglyce-mia and oxidative stress, the expression of NF-κB is enhanced, that leads to the changes of the expression of NOS , MMP-9, TNF-α, then, these changes lead to the apoptosis of hippocampus neuron cell .This review focuses on NF-κB in order to provide evidences in studying CID by reviewing the relationship between NF-κB and it′s relative factors of NOS, MMP-9, TNF-αand CID.

Objective To explore the differences of serum expression of cystatin C,macrophage migration inhibitory factor (MIF)and matrix metalloproteinase-1 (MMP-1)in elderly patients with coronary artery disease.Methods 120 cases of senile cor-onary heart disease patients in the department of cardiovascular medicine from July 2010 to August 2012 were selected and divided into the control group (n=30),stable angina pectoris group(SAP group,n= 34),unstable angina group (UA group,n=28)and a-cute myocardial infarction group (AMI group,n=28)according to the results of coronary arteriography combined with the electro-cardiogram and myocardial enzyme spectrum test.The serum levels of cystatin C,MIF,MMP-1 and C reactive protein (hs-CRP) were compared among the four groups,and correlations of the four indicators were analysed as well.Results There were significant differences in the serum levels of cystatin C,MIF,MMP-1 and hs-CRP among the four groups(P 0.05).The significant positive correlations were observed among cystatin C,MIF and MMP-1.Conclusion The serum levels of cystatin C,MIF and MMP-1 have a gradually increasing trend with disease progression in pa-tients with coronary heart disease,which could be important indicators of disease evaluation.

RESUMEN

Objective: To study the influence and curative effect of carvedilol on serum matrix metalloproteinase -2, 9 and 10 (MMP-2, 9 and 10) in the patients with chronic congestive heart failure (CHF). Methods:Totally 76 patients with CHF were select-ed and randomly divided into the observation group and the control group. All the patients were given routine medical treatment, inclu-ding cardiotonic treatment ( digoxin) , diuresis ( thiadiazide) , vascular dilation ( nitrate ester) and so on. The patients in the observa-tion group were additionally given carvedilol for 6 months. The changes of serum MMP-2, 9 and 10 levels in the patients before the treatment and 6 months after the medical treatment were observed, and the curative effect and safety were studied as well. Results:Af-ter the 6-month medical treatment, the serum MMP-2, 9 and 10 levels in all the patients were obviously declined(P<0. 05), and the declining rates in the observation group were much higher than those in the control group(P<0. 05). Meanwhile, the total clinical effi-ciency of the observation group was 94. 74%, which was higher than that of the control group (78. 95%) (P<0. 05). Conclusion:Carvedilol has favorable curative effect and safety in the treatment of CIF, and the underlying mechanism may be relative to the de-crease of serum MMP-2, 9 and 10 levels.

Background Previous studies showed that after herpes simplex virus-1 (HSV-1) infection of the cornea,matrix metallo proteinase-2 (MMP-2) (produced by corneal cells and corneal epithelial cells) plays an important role in the development of HSK.Focal adhesion kinase (FAK)plays an important role on the expression,release and activation of MMPs.This study explored the expressions of MMP-2 and FAK,which induced by HSV-1 infected human corneal epithelial cells.Objective To investigate MMP-2 and FAK expression in HSV-1 infected human corneal epithelial cells.Methods Human corneal epithelial cells were infected with HSV-1 in vitro to establish cell model of viral infection.The expression of MMP-2 and FAK were detected by reverse transcription PCR (RT-PCR),Western blot,immunohistochemical method and immunofluorescence method at 2 hours,20 hours and 40 hours after infection.Results At 2 hours,20 hours and 40 hours after infection,the expressionis of MMP-2 mRNA and FAK mRNA were significantly increased in comparison with uninfected cells (MMP-2 mRNA:Ftime =0.968,P=0.436 ; Fgroup =47.649,P =0.000 ; Fi ion =0.757,P =0.536.FAK mRNA:Ftime =0.658,P =0.631 ; Fgroup =35.182,P=0.000;Finteraction =1.386,P=0.137).Western blot assays showed that there were no significant differences in p-FAK,FAK or MMP-2 expressions between infected cells and control cells after 2 hours (P>0.05),but the expression levels of infected cells were significantly increased at 20 hours and 40 hours (both at P < 0.05).Immunohistochemistry results showed that longer infection time was associated with an increased number of cells staining for MMP-2,FAK and p-FAK.Conclusions At the initial stage of HSV-1 infected,p-FAK plays an important role in the process of virus invading and MMP-2 activation.

About 2/3 patients with chest tumor requires radiation therapy .Normal lung tissues inevitably receive a certain amount of radiation during the treatment ,which results in the most frequent complication in tho-racic tumor radiotherapy:radiation-induced lung injury(radiation-induced lung injury,RILI).Usually,radio-logical changes are more common than clinical manifestations .RILI has become the bottleneck in radiotherapy which restricts the improvement of efficacy .At present ,there is no accurate and reliable method to predict the oc-currence of RILI .Once RILI occurs ,the condition is irreversible and treatment in recent decades has been stag-nant.In the present review ,we summarized the RILI predictors and RILI treatment methods .

Objective To approach on the expression pattern of MMP-2 and E-Cadherin in the effects of gastrin on gastric canc-er cells ,so as to investigate their role in the metastasis and invasion of gastric cancer .Methods Genomic DNA modified by sodium bisulfite was used as a template .RQ-MSP detect the methylation status of the gene promoter .The relative amount of Methylation status was performed with comparative threshold cycle (2- △ t ) method .Results MMP-2 and E-Cadherin genes in gastric cancer cells were at hemimethylated status in gastrin before and after the intervention .Gastrin can reduce the methylation levels of MMP-2 gene(P0 .05) .Gastrin can pro-mote the methylation levels of MMP-2 gene ,but proglumide decrease that effect (P<0 .05) .Conclusion Gastrin plays a certain role in the biological behavior of gastric cancer impact by changeing on the expression pattern of MMP-2 and E-Cadherin in the effects of gastrin on gastric cancer cells .

Objective:To observe the effects of fuyuan capsule on change of morphology of synovium,expression of IL-1?、MMP-13 and iNOS in synovium in experimental osteoarthritis of rabbits,and investigate the mechanism about the reparation and treatment of fuyuan capsule.Methods:Sixty New Zealand rabbits were divided into five groups randomly:normol group,model contrast group,low dose group, middle dose group and high dose group of fuyuan capsule.OA model was induced in rabbits by improved Hulth'S method.Three Chinese herb groups were treated with fuyuan capsule(0.727,1.453,4.795 g(/kg?d)),continuously for 4,8 weeks and 12 weeks,model contrast group was treated with 0.9%NS,ig,at the same time.Synovium sections were analyzed by immunohistochemistry for the IL-1?,MMP-13 and iNOS,observation and measurement of the change of morphology of synovium by light microscopic and transmission electron microscopic.After the experiment,the differences of these indexes between Three Chinese herb groups and model contrast group were compared.Results:There are found significant differences about measurement of morphology of synovium between Three Chinese herb groups and model contrast group after the experiment(P

RESUMEN

Objective:The aim of the current study was to investigate the expression of nuclear factor-?B(NF-?B) and metalloproteinase-9(MMP-9) in the small intestine and to explore the potential role of NF-?B and MMP-9 in the damage of gut mucosal barrier after traumatic brain injury.Methods:The trauma was produced by a free-falling weight on the exposed dura of right parietal lobe.The rats were randomly divided into control group and traumatic brain injury groups at hours 3,12,24 and 72,and on day 7.NF-?B binding activity in the small intestine was studied by electrophoretic mobility shift assay(EMSA),and the expression of MMP-9 was studied by immunohistochemistry.Results:The results showed that NF-?B binding activity and MMP-9 expression in the small intestine was progressively increased,reached the maximum at 72 h and kept at high level up to 7 d after TBI.Concomitant upregulation of NF-?B~()and MMP-9 was observed.MMP-9 positively immunostained cells were mainly located at villous interstitium,lamina propria,crypt and submucosal layer,including endothelial cells,lymphocytes and neutrophils.Conclusion:It was concluded that cortical contusion trauma could induce a concomitant and persistent upregulation of NF-?B binding activity and MMP-9 expression in the small intestine which might play a central role in the~()damage of gut mucosal barrier.

AIM: To investigate the role of PPAR ? or ? ligands in regulating the expression of extracellular matrix metalloproteinase inducer(EMMPRIN).METHODS: THP-1 monocytes were induced into macrophages and foam cells in vitro then interfered with clofibrate and pioglitazone.The cells and supernatant were collected after 24 h,respectively.EMMPRIN gene and its protein were assessed by real-time PCR and Western blotting in different interferences.The concentration of matrix metalloproteinases(MMP-9) was measured with ELISA method and the activity of MMP-9 was detected with gelatin zymography.RESULTS: Two known PPAR ? or ? ligands,colfibrate and pioglitzaone,were found,both of which inhibited EMMPRIN expression in macrophages and foam cells.The inhibition was correspondent to the secretion and activity of MMP-9 simultaneously.CONCLUSION: Both PPAR ? and ? ligands inhibit EMMPRIN expression,which may account for their effect on inhibition of MMPs.

Objective To study the role of TNF-?,MMP-9 and TIMP-1 in the lesions of psoriasis vulgaris.Methods SABC immunohistochemical technique was used to examine the expression of TNF-?,MMP-9 and TIMP-1 in lesional skin of 40 cases and uninvolved skin of 20 cases of psoriasis and in skin of 20 cases normal controls.Results The expression of TNF-? and MMP-9 was significantly higher in the lesional skin than that in nonlesional skin and normal controls(P0.05).There was a positive correlation between the expression of TNF-? and MMP-9(r=0.471,P
