RESUMEN

Objective To explore the expression of matrix metalloproteinase-9 (MMP-9) in breast carcinoma and the relationship between the expression and clinical pathological characterization.Methods Specimens taken from 50 fresh resected breast carcinoma tissue and 16 normal breast tissue were selected.Tissue sections were stained by EnVisionTM imnunohistochemistry with MMP-9 as monoclonal antibody.The expression of MMP-9 in breast carcinoma and the relationship between the expression and clinical pathological characterization were studied.Results The positive rate of MMP-9 breast cancer carcinoma was 59.32%,which was significantly higher than that of normal breast tissue 9.62%,with statistical significance (P < 0.01).The average rank and score of MMP-9 in the metastatic tissue of breast carcinoma were higher than that of no metastatic breast cancer tissue (P < 0.01).Conclusions MMP-9 may be related to the destruction of extracellular matrix and basement membrane near the surface of breast cancer cells,thereby promoting the invasion and metastasis of breast cancer.

Objective To explore the expression and correlation of Fascin-1 and epidermal growth factor receptor (EGFR) in hormone receptor-positive breast cancer.Methods The immunohistochemical technique,EnVision method,was used to evaluate the expression of Fascin-1 and EGFR in 294 cases of hormone receptor-positive breast cancer,which contains 290 cases of estrogen receptor (ER) positive and 244 cases of progestrone receptor (PR) positive.According to ER,PR,Epidermal growth factor receptor 2 (HER2),and Ki-67 status,all cases of hormone receptor-positive breast cancer were categorized into 2 subtypes:160 cases of luminal A and 134 cases of luminal B.Results Fascin-1 and EGFR protein positive rates in hormone receptor-positive breast cancer was 13.9% (41/294) and 30.6% (90/294),respectively.Fascin-1 positive rate was significantly higher in EGFR positive cases (30.0%,27/90) than in EGFR negative cases (6.9%,14/204) (x2 =27.857,P =0.000).In the ER positive and PR positive cases,Fascin-1 positive rates were both significantly higher in EGFR positive cases than in EGFR negative cases (x2 =29.23,P =0.000;x2 =27.596,P =0.000,respectively).In the Luminal A and Luminal B subtype,Fascin-1 positive rates were also both significantly higher in EGFR positive cases than in EGFR negative cases (x2 =23.247,P=0.000;x2 =5.325,P=0.021,respectively).Conclusions EGFR signal pathway may positive regulate Fascin-1 expression in hormone receptor-positive breast cancer.

RESUMEN

The breast cancer is the most common malignant tumor of women, and its treatments were widely explored.A lot of studies have confirmed that the abnormal activation of WNT/ beta-catenin signaling pathway and the abnormal expression of miRNA are closely related to the occurrence and development of breast cancer.miRNA could target the regulation of WNT/beta-catenin signaling pathway and affect the occurrence and development of many human tumors as an endogenous non-coding small molecule RNA.Therefore, the mechanism of WNT/beta-catenin signaling pathway in breast cancer and its interaction with miRNA may provide new ideas for the treatment of breast cancer.The article reviewed the composition of the WNT/beta-catenin signaling pathway and the role of this signaling pathway and its interaction with miRNA in the pathogenesis and progression of breast cancer.

Objective To investigate the expression and clinical significance of serum neutrophil gelatinase-associated lipocalin (NGAL), kidney injury molecule-1 (KIM-1) and serum cystain C (CysC) in patients with renal tumor. Methods A total of 120 patients with renal tumor diagnosed in International Hospital of Zhejiang University from January 2016 and March 2016 was selected as the research object, and 60 patient medical volunteers were selected as control. The levels of NGAL and KIM-1 were determined by enzyme-linked immunosorbent assay ( ELISA) method, and the levels of serum CysC were determined by electrochemical luminescence method. The levels of NGAL, KIM-1, and CysC were compared, and their relevance was analyzed. Results The levels of NGAL, KIM-1, and CysC in research object were signifi-cantly higher than the control group; the levels of NGAL, KIM-1, and CysC in malignant tumor patients were significantly higher than the benign tumor;the levels of NGAL, KIM-1, and CysC in one level of ser-um tumor were lower than the two levels, three levels, and four levels of tumor patients. With the more Furhman grading, the levels of NGAL, KIM-1, and CysC were higher, the differences were statistically sig-nificant (P<0. 05). The levels of serum NGAL, KIM-1, and CysC were significantly related to renal dam-age degree. The levels of serum NGAL were significantly positively related to KIM-1 and CysC ( r= 0. 812 and 0. 765 ) . The levels of serum KIM-1 expression were significantly positively related to CysC ( r =0. 832) (P<0. 05). Multiple regression analysis showed that the levels of serum NGAL, KIM-1, and CysC were significantly related to patients with renal tumor Furhman classification ( P<0. 05 ) . Conclusions The levels of NGAL, KIM-1 and CysC were significantly increased in patients with renal tumor renal tumor, the higher the clinical stage, the higher expressive level, and were significantly related to renal damage de-gree. NGAL, KIM-1, and CysC can be used as the diagnostic markers of renal tumor.

Objective To investigate the expression of forkhead box C2 (FOXC2),Vimentin and E-cadherin protein in colorectal adenocarcinoma and the relationship of FOXC2,Vimentin and E-cadherin with the clinicopathological parameters,and the role of epithelial-mesenchymal transition (EMT) in colorec tal adenocarcinoma and the correlation between FOXC2 and EMT.Methods The expressions of FOXC2,Vimentin and E-cadherin in 102 cases of colorectal adenocarcinoma tissues and 30 cases of adjacent noncancerous tissues were detected by immunohistochemical method,and the correlation between FOXC2 and EMT and the clinicopathological parameters of colorectal adenocarcinoma were analyzed.Results The positive rates of FOXC2,Vimentin and E-cadherin in colorectal adenocarcinoma tissues were 53.9%,40.2% and 26.5%,respectively.The positive rates of FOXC2,Vimentin and E-cadherin in adjacent noncancerous tissues were 6.7%,0 and 63.3%.Compared to paracancerous tissue,the expression of FOXC2 and Vimentin in colorectal adenocarcinoma was significantly increased,while the expression of E-cadherin was significantly decreased.The positive rates of FOXC2 and Vimentin in lymph node metastasis group were 79.1% and 67.4%,respectively,which were significantly higher than those without lymph node metastasis (35.6%,P <0.01;20.3%,P <0.01).The positive rate of E-cadherin in lymph node metastasis group was 16.3%,which was significantly lower than that without lymph node metastasis (33.9%,P < 0.01).The positive rates of FOXC2 and Vimentin in T1 +T2 phase were 41.7% and 25.0%,respectively,which were significantly lower than those in T3 + T4 stage (83.3%,P <0.01;76.7%,P <0.01).The positive rate of Ecadherin in T1 + T2 stage group was 33.3%,which was significantly higher than that in T3 + T4 stage (10.0%,P < 0.01).There was a positive correlation between FOXC2 and Vimentin in colorectal adenocarcinoma (P <0.01),and negatively correlated with E-cadherin expression (P < 0.01).Conclusions EMT may promote the development and metastasis of colorectal adenocarcinoma,FOXC2 may be involved in colorectal cancer EMT and through the EMT to promote the malignant process of colorectal adenocarcinoma.

Objective To explore the expressions of glutathione reductase in gastric cancer,to investigate the relationship between glutathione reductase (GSR) and clinical pathological characteristics of gastric cancers,to identify the role of GSR in evaluation of the prognosis of gastric cancer patients,and to investigate the role of GSR in the development of gastric cancer.Methods The gastric cancer datasets were searched and downloaded from The Cancer Genome Atlas (TCGA),and chip data were analyzed with clinical information.Gene set enrichment analysis (GSEA) was conducted to explore the gene sets enriched in samples with high GSR expression.Results The expression of GSR was down-regulated in high grade tumors (P < 0.01).No significant difference was found between different age,Shortest tumor diameter,American Joint Committee on Cancer (AJCC) M stage,Barrett's esophagus,family history of gastric cancer,and Helicobacter pylori (H.pylori) infection.Higher expression of GSR indicated poor prognosis in gastric cancer.GSEA indicated that GSR regulates gene sets associated with oxidative phosphorylation,metabolism of nucleotides,mitochondrial protein import,and mitotic G1 S phases.Conclusions GSR can be used as an indicator to predict the prognosis of gastric cancer patients and a target for the treatment of gastric cancer.

Objective To explore the changes of serum interleukin-6 (IL-6),and interleukin-8 (IL-8) in patients with gastric cancers and the clinical relationships with Helicobacter pylori (Hp) infection.Methods Totally 62 cases of patients who were diagnosed gastric cancer in our hospital from January 2013 to September 2015 were selected as the study group,who did not accept anti-tumor therapy.The patients were divided into the Hp positive group (with 53 cases) and Hp negative group (with 9 cases) according to whether the merger of Hp infection.Another 60 normal healthy volunteers of the same age in our hospital for health examination during the same period were selected as the control group.The serum IL-6 and IL-8 levels were detected,and the changes of the factors in the study group and control group were compared and analyzed,as well as the U values of Hp infection.The clinical relationships between the expression levels of the two in patients with gastric cancer and the U values of Hp infection were confirmed.Results The serum IL-6 and IL-8 levels and the U values of Hp infection of the study group were significantly higher than those of the control group (P < 0.05),which in the Hp positive group were significantly higher than those in the Hp negative group (P < 0.05).The levels of serum IL-6 and IL-8 in patients with gastric cancer showed positive correlations with the U value of Hp infection (r =0.457,0.531,P < 0.05).Conclusions The serum IL-6 and IL-8 levels in patients with gastric cancer are much higher than those in normal healthy people,and there are significant positive relationship with Hp infection.It can improve the curative effect of gastric cancer patients by improving the eradication rate of Hp.

Objective To explore the difference of the expression level of FK506 Binding Protein 51 (FKBP51) in colorectal adenocarcinoma and normal colorectal tissues,and the correlation between FKBP51 expression level and clinicopathological characteristics,and to clarify whether FKBP51 is involved in the occurrence and development of colorectal cancer.Methods By immunohistochemical staining [streptavidin-peroxidase (SP) method] and Western blotting methods tested 31 cases of colorectal cancer tumor tissues and normal colorectal tissues far from tumor 5 cm,and explored the expression level difference of FKBP51.Combined with clinical data of patients,results were analyzed by statistical method x2 test of four case table data.Results The high expression rate of FKBP51 in tumor tissues was 74.19% (23/31 cases),while the high expression rate of FKBP51 in normal tissue was 9.68% (3/31 cases).The difference was significant.The expression level of FKBP51 in patients with colorectal carcinoma had no obvious correlation with gender (P =0.771),age (P =0.474),tumor location (P =0.213),degree of differentiation (P =0.318),lymph node metastasis (P =0.124),distant metastasis (P =0.318) and clinical stage (P =0.171);and the tumor size (P =0.049),depth of invasion related (P =0.031),the difference was statistically significant.Conclusions The expression of FKBP51 in colorectal cancer was strong,while weak expression in normal colorectal tissues.With the increase of tumor infiltration and deepening,the expression of FKBP51 became stronger,which indicated that FKBP51 participated in the genesis and development of colorectal cancer,and it might become a new target for individual therapy of colorectal cancer.

Objective To explore the incidence of brain metastases from esophageal cancer,survival-related factors after brain metastases,and the result of different treatment methods.Methods From January 2000 to October 2016 treated 11 338 cases of esophageal cancer patients,84 cases of brain metastases,were collected with complete clinical data.Results Eighty four cases of patients with brain metastases included esophageal squamous carcinoma in 74 cases,adenocarcinoma in 6 cases,and small cell undifferentiated carcinoma 4 cases;single brain metastases in 19 cases,65 cases of multiple brain metastases included 59 cases with brain metastasis symptoms,58 with extracranial metastases,primary tumors and brain metastases interval time less than 6 months in 31 cases,24 cases of 6-12,16 cases for 12-24 months.Prognostic logrank univariate analysis of survival rate found 24-36 months survival for 13 patients.All patients were followed for a median of 19.31 months after brain metastases,the median survival time was 5.43 months.Conclusions About 0.7% of brain metastases from esophageal cancer occurs.Esophageal carcinoma metastatic to the brain:There was no extracranial metastasis,recursive partitioning analysis classification (RPA),Karnofsky (KPS) score,brain metastases different treatment methods is a poor prognostic factor for esophageal cancer brain metastasis.

Objective To analyze the changes of serum fatty acid synthase (FAS),carcinoembryonic antigen (CEA) and carbohydrate antigen 72-4 (CA72-4) in patients with gastric cancer and its clinical diagnostic value.Methods Forty five patients with gastric cancer and 45 patients with benign gastric cancer treated in our hospital from January 2016 to December 2016 were enrolled in this study.Forty five healthy subjects were enrolled in this study.The levels of FAS,CEA,and CA72-4 in three groups were analyzed.Results The levels of FAS,CEA,and CA72-4 in patients with gasttric cancer [(12.73 ± 5.48) mg/L,(31.36 ± 14.51) ng/ml,and (39.32 ± 18.76) U/ml] were significantly higher than those in benign gastric cancer group [(2.26 ± 1.15) ng/L,(3.24 ± 1.47) ng/ml,and (3.75 ± 1.69) U/ml],and normal control group [(1.83 ± 0.92) mg/L,(2.71 ± 1.54) ng/ml,and (3.13 ± 1.82) U/ml] (P < 0.05).FAS,CEA,and CA72-4 levels in patients with lymph node metastasis of gastric cancer [(13.58 ± 6.09) mg/L,(6.25 ± 11.54) ng/ml,and (41.31 ± 13.67) U/ml] were significantly higher than those without lymph node metastasis [(9.21 ± 5.42) mg/L,(28.38 ± 9.72) ng/ml,and (26.75 ± 11.86) U/ml] (P < 0.05).The sensitivity of FAS,CEA,and CA72-4 in patients with gastric cancer was significantly lower than that in combined detection,the specificity of FAS,CEA,and CA72-4 in patients with gastric cancer was significantly higher than that in combination test (P < 0.05).The sensitivity of FAS,CEA,and CA72-4 in patients with lymph node metastasis was significantly lower than that in combination test,and the specificity of FAS,CEA and CA72-4 was significantly higher than that of combined detection (P < 0.05).Conclusions FAS,CEA,and CA72-4 can be used as indicators of gastric cancer and diagnosis of lymph node metastasis.Combined detection of three indexes can improve the diagnostic sensitivity and have good clinical significance.

Objective To investigate the relationship between miR-411 and breast cancer,and the expression of miR-411 and its effects and mechanism research on proliferation and metastasis in breast cancer.Methods Real-time polymerase chain reaction (RT-PCR) was used to detect the expression of miR411 in breast cancer cells and tissues.Methyl thiazolyl tetrazolium (MTT),clone formation assay and Transwell assay were used to detect the effect of miR-411 expression on the proliferation,migration and invasion in breast cancer cells.The effect of miR-411 on growth factor receptor-bound protein 2 (GRB2) expression in breast cancer cells was detected by RT-PCR and Western blot.The direct effect of miR-411 target on GRB2 was detected by dual luciferase reporter assay.MTT,clone formation assay and Transwell assay detect the effect of GRB2 expression on the proliferation and invasion in breast cancer cells.Detection the effect high expression of miR-411 on GRB2 downstream signaling pathway related molecules expression in breast cancer cell with Western blot.Results The expression of miR-411 in breast cancer tissues was significantly lower than that in adjacent non-cancerous tissues (P < 0.05).The expression of miR-411 in breast cancer cells SK-BR-3,BT-549 and MDA-MB-231 was significantly lower (P < 0.05).Compared to the negative control group,the transfection of miR-411 mimic inhibited the proliferation,migration and invasion of MDA-MB-231 breast cancer cells (P < 0.05).Targetscan showed that miR-411 could bind to GRB2 3'UTR at position 741-747.Compared with the negative control group,GRB2 3'UTR wild-type plasmid and miR-411 co-transfection reduced the fluorescence activity (P < 0.05).Transfection of siGRB2 significantly reduced the expression of GRB2 protein in MDA-MB-231 breast cancer cells (P < 0.05).Compared to the negative control group,the inhibition of GRB2 expression reduced the proliferation and the number of colony formation of MDA-MB-231 breast cancer cells (P < 0.05).Transwell assay showed that transfection of siGRB2 significantly reduced the number of invasive cells in MDA-MB-231 breast cancer cells (P < 0.05).Conclusions miR-411 is related closely to the occurrence and development of breast cancer,miR-411-GRB2-Ras axis is expected to become a new target for biological treatment of breast cancer.

Objective To investigate the expression and clinical significance of lactate dehydrogenase A (LDHA) in breast cancer.Methods Tissue samples of 76 breast cancers and corresponding paired adjacent normal tissues were collected and made into tissue micrcarrays (TMAs).Immunohistochemistry (IHC) analysis was performed to detect the expression of LDHA and further analyzed the correlation of LDHA expression and clinicopathological variables and prognosis of breast cancers.Results LDHA was frequently upregulated in breast cancer tissues compared to the normal breast tissues (P < 0.05).High LDHA expression was associated with distant metastasis (P < 0.05) and worse patient prognosis (P < 0.05).Conclusions LDHA is closely related to the occurrence and clinical progress of breast cancers.LDHA might be a potential novel molecular marker for diagnosis,prognosis and therapy in breast cancers.

Objective To investigate the expression of paired box 2 (Pax2) and E-cadherin in human renal cell carcinoma (RCC) and their correlation with clinicopathologic parameters.Methods The RCC tissue microarrays containing 85 renal cell carcinoma specimens and 35 normal kidney tissue specimens were used to detect Pax2 and E-cadherin expressions by immunohistochemistry.Results The positive expression rate of Pax2 was 77.6% (66/85) in RCC specimens,which was significantly higher than that in 35 normal kidney tissue specimens (P < 0.01).The positive expression rate of E-cadherin was 30.6% (26/85) in RCC specimens,which was significantly lower than that in 35 normal kidney tissue specimens (P < 0.01).The expression of Pax2 in RCC was significantly related to histological classification and pathological grade (P < 0.05),while it was not related to the size of tumor and clinical stage (P > 0.05).The expression of E-cadherin in RCC was significantly related to the size of tumor,histological classification,pathological grade and clinical stage (P < 0.05).The positive expression rate of E-cadherin in survival group was significantly higher than that in death group (P < 0.05).Conclusions The abnormal expressions of Pax2 and E-cadherin may play a crucial stage in development of human RCC.Detection of the expressions of Pax2 and E-cadherin can be used in the evaluation of malignant degree and prognosis of RCC.

Objective To investigate the expression of microRNA-451 (miR-451) in female invasive ductal carcinoma (IDC) and its roles in tumor cell invasion.Methods Forty five IDC tissues and matched tumor adjacent tissues were collected between January and December in 2014.The expressions of miR-451 in tissues were detected by quantitative real-time polymerase chain reaction (qRT-PCR).The relationship between long noncoding RNA taurine regulated genes 1 (lncRNA-TUG1) and clinical features was analyzed by student-t test.miR-451 mimics was transfected into MDA-MB-231 cells.Transwell assay was used to measure cell invasion ability.The expression of c-myc,a potential target of miR-451,and its downstream genes,matrix metalloproteinase (MMP)-2 and MMP-9 were detected by qRT-PCR and Western-blot.Resuts The expression of miR-451 was significantly lower in IDC tissues than in matched tumor adjacent tissues (P < 0.05).Low expression of miR-451 was positively associated with lymphatic metastasis (P < 0.05) and advanced tumor node metastasis (TNM) stage (P < 0.05).Up-regulation of miR-451 in MDA-MB-231 cells could significantly suppress cell invasion ability (P <0.05).c-myc expression was down-regulatedwhen miR-451 was transfected (P <0.05).As downstream genes of c-myc,the expressions of MMP-2 and MMP-9 were suppressed.Conclusions miR-451 is down-regulated in IDC tissues and associated with cell invasion.miR-451 might inhibit IDC progression by inhibiting c-myc/MMP axis.

Objective To investigate the chemokine 12 (CXCL12) and chemokine receptor 4 (CXCR4) expressions in hypopharyngeal carcinoma and its place in the disease development,invasion and metastasis of significance.Methods Immunohistochemistry was used to detect the expressions of CXCL12 and CXCR4 in 35 cases of hypopharyngeal cancer tissues and in 28 cases of tumor-adjacent non-tumor tissues.Results The expressions of CXCL12 and CXCR4 in the hypopharynx carcinomas were significantly higher (P < 0.05).Both expressed in hypopharyngeal carcinomas was significantly positively correlated (P < 0.01).Both hypopharynx cancer in lymph node metastasis group were significantly higher than the expression of cervical lymph node metastasis group,the difference was significant (P < 0.05).Conclusions CXCL12 and CXCR4 are involved in hypopharynx cancer development,invasion and metastasis,and there is a positive feedback regulation mechanism between two factors.Moreover,CXCL12 and CXCR4 have synergistic effect in development,invasion and metastasis of hypopharynx cancers.

RESUMEN

Objective To explore the role of human papillomavirus (HPV) in squamous cell carcinoma of the larynx.Methods A cohort of 152 laryngeal squamous cell carcinoma (LSCC) patients,40 premalignant lesions of the larynx,and 40 vocal fold polyp patients was evaluated for the expression of HPV16 E7 protein by immunohistochemistry (IHC).E7 protein expression was correlated with clinicopathological features and outcomes.Results The expression of E7 protein in vocal fold polyp was negative,and it's positive expression in precancerous lesions and laryngeaI carcinomas was 2.5%,and 32.9%,respectively.There were no other significant clinical or demographic differences between pl6-positive and-negative cases.There was no difference in overall survival (OS) between E7-positive and-negative patients with 5-year survival of 79% and 66% for p16 positive and negative patients,respectively (P =0.581).There was no statistically significant difference in failure-free survival (FFS) with 5-year FFS of 57.8% and 42.3% for p16 positive and negative patients,respectively (P =0.579).Conclusions E7 expression is infrequent in laryngeal carcinomas.There are no statistically significant correlations between E7 IHC status and OS or FFS.

RESUMEN

Objective To investigate the relationship between p62 expression,and occurrence and metastasis of nasopharyngeal carcinomas.Methods Immunohistochemical method was used to analyze p62 expression in 123 nasopharyngeal carcinoma (NPC) cases and 30 chronic nasopharyngitis cases.The clinical pathological characteristics were analyzed.Results The positive rate of p62 protein in chronic nasopharyngitis nasopharyngeal epithelium,non-metastatic NPC tissue,and metastatic NPC tissues was 13.3%,66.67%,and 84.72%,respectively,and the difference was statistically significant.The expression of p62 protein in nasopharyngeal carcinoma patients with lymph node metastasis or distant metastasis was significantly higher than non-metastatic NPC patients (P < 0.01 or P < 0.05).However,the expression of p62 was not related to age,gender,tumor size,and TNM stage (P > 0.05).Conclusions High p62 protein expression in nasopharyngeal carcinoma tissue is closely related to the occurrence and metastasis of nasopharyngeal carcinomas.It provides good reference value to predict NPC malignancy and metastases.

Pituitary adenoma is a serious disease that affects human health through interfering hypothalamus-pituitary-target organ axis systems.Proteomics is an effective approach to elucidate molecular mechanisms of a pituitary adenoma and discover effective biomarkers for a pituitary adenoma.A great progress has been made in the field of pituitary adenoma proteomics in the past ten years:(1) the use of laser capture microdissection,(2) functional pituitary adenoma proteomics (such as prolactinoma),(3) proteomics analysis of invasive characteristics of nonfunctional pituitary adenoma,(4) protein post-translational modifications including phosphorylation and tyrosine nitration,(5) the use of protein antibody array,(6) proteomics analysis of hormone isoforms,(7) serum proteomics and peptidomics,(8) integration of proteomics and other omics data,and (9) proposal of multi-parameter systematic strategy for a pituitary adenoma.
