RESUMEN

Objective To investigate the application of MRI targeting contrast agent ( P-Gd-Probe) for P selectin in evaluating the severity grades of acute pancreatitis (AP).Methods L-arginine solution (100mg/100kg) were intraperitoneally injected for 3 times once an hour to establish AP model in SD rats. Control group was injected with normal saline in a equal volume .All rats were divided into acute edematous pancreatitis (AEP) group (6 h and 12 h after model establishment)and acute necrotizing pancreatitis (ANP) group (24 h and 48 h after model establishment ) based on the pathology .The rats were examined by T 1 WI plain and enhanced scanning at 6 h, 12 h, 24 h and 48 h after injection , and P-Gd-probe was as the enhanced contrast agent. The signal noise ratio ( SNR) of the pancreas were measured before and after enhancement .Then the pancreas tissues were harvested for pathological examination and P selectin expression in pancreatic tissue was detected using immunohistochemical analysis .Results Inflammation was observed in pancreatic tissue at 6h after establishment , and became more serious as the modeling time extended .P selectin expression was increased as pancreatitis inflammation became more serious .The SNR of control group was stable before and after enhancement. The SNR before and after enhancement was 17.22 ±1.35 and 37.38 ±1.66 in AEP group, and 16.29 ±1.39 and 58.18 ±1.03 in ANP group. The SNR after enhancement was higher than that before enhancement, and the differences were significantly different ( t=-49.59 and -86.09, P<0.001 ). Conclusions Monoclonal antibody MRI contrast agent targeting P selectin is helpful in evaluating the severity of AP.

RESUMEN

Objective To determine the changes of patients with severe pulmonary contusion and the relationship among the serum level of alpha-granular membrane protein (CD62p), the severity of injury and prognosis. Methods The serum level of CD62p, was measured in 45 patients with severe pulmonary contusion on hour 2, hour 12,day 1,3,7 after injured. Forty-five patients were divided into two groups: the survive group and the death group. The levels of CD62p between two groups were compared. Results The serum levels of CD62p in patients with severe pulmonary contusion on 12 hours, 1,3 days after injury were higher than those on 2 hours (P<0.05). The serum levels of CD62p in the survive group and death group on 2 h, 12 hours after injury were higher than those in the control group(P<0.05), and the serum levels of CD62p in the death group were higher than those in the survive group. Conclusion It was helpful in judging the severity of head in jury and prognosis to determine the serum levels of CD62p in patients with severe pulmonary contusion.

RESUMEN

Objective To explore the relationship between P-selection,nitric oxide(NO),high-sensitive-C-reactive protein(hs-CRP)and type 2 diabetes mellitus(T2DM)with macroangiopathy. Methods In sernm of 125 T2DM subjects(60 subjects with macroangiopathy and 65 subjects without macroangiopathy)and 55 normal controls, the activity of P-selection,NO and hs-CRP was detected by ELISA,Nitrate Reductase Assay and scattering immunoturbidimetric method, respectivley. Results The activity of serum P-selection and hs-CRP was significantly higher in diabetic macroangiopathie group than in non macroangiopathic group and control normal.Serum NO was significantly lower in diabetic rnacroanglopathic group than in non macroangiopathic group and control normal.Serum P-selection was positively correlated with hs-CRP-systolic blood pressure(SBP),LDL-C,and was negatively correla- ted with NO:P-selection,hs-CRP,LDL-C and SBP were independently risk factors for T2DM with macroangiopathy. Conclusion Determination of P-selection and NO is very important for prevention and diagnosis of diabetic mac- roangiopathy in early stage.

RESUMEN

P-selection is an adhesive molecule in vascular endothelicyte and platelet, exerting key function on immuno-recognition,inflammation reaction and thrombus formation.Measuring the expression of P-selection is of great meaning to explore mechanism, observe disease progress, guide clinical treatment and prospect prognosis of kidney disease.

RESUMEN

Objective To study the clinical significance of P - selection expression in children with viral encephalitis and the correlation between this expression and the cerebral infarction with critical viral encephalitis. Methods Flow cytometric was employed to detect the expression of P- selection on the surface of platelet membrane in 44 children with viral encephalitis(20 light patients and 24 critical patients) and 20 healthy control children. The area of the cerebral infarction was determined by computed tomographic scan in 20 patients with critical viral encephalitis. The correlation between the two variables was analyzed. Results The expressions of P - selection on the surface of platelet membrane on less than 5 days and on 2 weeks after the onset of viral encephalitis were significantly higher in critical patients than those in normal control children and light patients( P

RESUMEN

ObjectiveTo investigate the correlations between Herpes simplex virus (HSV) infection and acute myocardial infarction(AMI).MethodsSpecific antibodies and DNA to HSV-1 were measured in 51 patients with AMI(AMI group),42 patients with old myocardial infarction (OMI group) and 31 normal controls (NC group) who had no angiographic evidence of coronary artery disease. Moreover, the changes of and relationships between fibrinogen(Fg),6-Keto-prostaglandin-F 1? (6-Keto-PGF 1? ),Thromboxane B 2 (TXB 2 ),P-selection(GMP 140 ) and HSV-1 infection were determined in the three groups.ResultsThe prevalence of HSV-1IgG antibody was associated with AMI(OR4.266).Adjustment for the major cardiovascular risk factors didn't attenuate this correlation(OR3.321).Those who were HSV-1 positive in the AMI group had significantly higher levels of Fg,TXB 2 and GMP 140 and a significantly lower level of 6-Keto-PGF 1? than those who were HSV-1 negative in the same group.Correlation analysis revealed that the average concentration of IgG antibody to HSV-1 was positively associated with the levels of Fg,TXB 2 and GMP 140 but negatively associated with the level of 6-Keto-PGF 1? in the HSV-1 positive patients of the AMI group.ConclusionThere is a significant correlation between HSV-1 infection and AMI.HSV-1 infection is associated with Fg,TXB 2 ,6-Keto-PGF 1? and GMP 140 .

RESUMEN

AIM To study the action of danshengsuan (Dan) on expression of CD11b, P selectin, ICAM 1, VCAM 1 and E slectin in vitro. METHODS Adheshion molecule expression on the cell surface was measured by flow cytometry. RESULTS Treatment of neutrophils with N fomyl met leu phe(fMLP), resulted in a marked increase of CD11b expression. Dan inhibited the effects of fMLP in a concentration dependent manner. Dan did not inhibit the increase of P selectin expression of thrombin actived human blood platelets. Treatment of cultured human umbilical vein endothelial cells (HUVEC) with TNF ? resulted in an increase of ICAM 1, VCAM 1 and E slectin. Dan inhibited the increase of VCAM 1 and E slectin expression, not ICAM 1. CONCLUSION Dan inhibited expression of adhesion molecules CD11b, ICAM 1, VCAM 1 in human neutrophils and HUVEC.
