RESUMEN

Objective To observe the effects of pioglitazone on secretion of E2 and expressions of P450 aromatase (P450arom), insulin-receptor substrate-1 (IRS-1), and insulin-receptor substrate-2 (IRS-2) mRNA in polycystic ovary syndrome (PCOS) ovarian granulosa cells.Methods In this study, granulosa cells that were fertilization-embryo transferred from 27 PCOS patients were primary cultured in vitro with different concentrations of pioglitazone (0, 10, 102, 103 and 104nmol/L) (Group A), different concentrations of pioglitazone+FSH (50ng/L, Group B) and different concentrations of pioglitazone+insulin-like growth factor I (IGF-I, 50ng/L, Group C).Estradiol concentrations in the culture supernatant were detected by radioimmunoassay;P450arom, IRS-1 and IRS-2 mRNA expressions on granulosa cells were detected by Real-time PCR.Results The levels of E2 secreted by granulosa cells and the expression of P450arom mRNA on granulosa cells of PCOS for 48 hours were different among Groups A, B and C (P<0.05).With increase in pioglitazone concentration, the level of E2 and the expression of P450arom mRNA declined, some of which correlated negatively with the concentration of pioglitazone.Among these groups, the level of E2 secretion and the expression of P450arom mRNA were higher in Group C than in Group B (P<0.01) and Group A (P<0.01) at the same concentration of pioglitazone.The level of E2 secretion and the expression of P450arom mRNA were higher in Group B than in Group A (P<0.05) at the same concentration of pioglitazone.The expressions of IRS-1 and IRS-2 mRNA on granulosa cells of PCOS under pioglitazone stimulation for 48 hour were different among the groups of different pioglitazone concentrations (P<0.05).With increase inpioglitazone concentration, the expression of IRS-1 mRNA on granulosa cells of PCOS was decreased, but the expression of IRS-2 mRNA on granulosa cells of PCOS was increased.Conclusion Pioglitazone may decrease estrogen production by inhibiting p450 aromatase and adjusting the expressions of IRS-1 and IRS-2 on granulosa cells of PCOS to play a role in ovulation induction and ameliorate insulin resistance in ovary of PCOS.Pioglitazone can inhibit IGF-I and FSH in inducing E2 secretion by ovarian granulosa cells.

RESUMEN

Objective To observe the therapeutic effect of Bushen huoxue huatan recipe (BHHR) on androgen-induced sterilized rats (ASR) and the expression of androgen synthase and metabolic enzymes in the ovary of ASR before and after treatment with BHHR, and to discuss the possible mechanism. Methods Female SD rats of 9-day old were injected subcutaneously with testosterone propionate (1. 25 mg) to create model. The model ratswere randomly divided into 3 groups: model group (treated with distilled water by gastrogavage, 10 mL/kg)' metformin therapy group (gastrogavage, 0. 1 g/kg) and BHHR therapy group (gastrogavage, 10 mL/kg)' with 13 animals in each group. Ten rats with normal estrous cycle served as normal controls. The body mass' sexual cycle recovery and ovary mass/body mass ratio were observed after 28-day treatment. Serum testosterone level was measured by radioimmunoassay; the expressions of 3(-hydroxylsteroid dehyrogenase (3β-HSD) cytochrome P450 17α-crhydroxylase/17, 20-lyase (CYP17) and P450 aromatase (P450arom) in ovary were detected by quantitative immunohistochemistry method. Results (1) Significantly more rats in the metformin and BHHR groups had sexual cycle recovery compared with that in themodtl group (P 0. 05). Conclusion BHHR can reduce serum testosterone levels in ASR rat, which might be through up-regulating the expression of the androgen metabolism enzyme P450arom.

0.05).Immunohistochemical staining for intratumoral aromatase in endometrial cancer was positive,21(42 %)in stromal cells.The expression of P450 aromatase in stromal cells correlated positively with advanced surgical stage and histologic grade,indepedent on muscular invasion and lymph node metastasis.Conclusion Aromatase P450 play an important function in establishment and devel- opment of the endometrioid endometrial cancer.The expression of P450 aromatase in intratumoral stromal cells correlated positively with the development and prognosis of endometrioid endometrial cancer.

RESUMEN

Objective To study the effects of dibutyl phthalate(DBP) on estradiol biosynthesis and the related mechanism in puberty female rats.Methods Thirty-two female SD rats aged 21 days were randomly divided into the control group(corn oil) and low-dose(250 mg/kg),moderate-dose(500 mg/kg),and high-dose(1 000 mg/kg) DBP exposed groups,the rats were treated with DBP through gavage with a dose of 5 ml/kg,once a day for 8 consecutive weeks.After 8 weeks of exposure,the animals were sacrificed at proestrus.Radioimmunoassay was used to detect estradiol in the serum,the expression of aromatase mRNA levels was detected by real-time fluorescent quantitative RT-PCR.Results Compared with the control group,the body weight of rats in the moderate-dose group was significantly higher(P
