RESUMEN

Objective To prepare a novel extravascular human epithelial growth factor receptor 2 (HER2)-targeted polylactic-co-glycolic acid (PLGA)-COOH ultrasound contrast agent with liquid perfluorocarbons(PFH-PLGA-tra),and to observe it's general properties and it's effects on ultrasound imaging in vitro. Methods Polymeric nanocapsules of liquid perfluorocarbons were prepared by the single emulsion technique and conjugated with trastuzumab monoclonal antibody by EDC/NHS.The general characteristics were observed. The conjugation was demonstrated by immunofluorescence and the targeting performance of the agent was checked in human MCF-7 cells line in vitro. The encapsulation of liquid fluorocarbons was detected by heating in vitro. The effects on ultrasound imaging were observed in vitro. Results The mean diameter of agents was (261 ± 28)nm.The targeted nanocapsules were positive in immunofluorescence. In the targeted study,it was shown that a number of targeted nanocapsules conjugated with MCF-7 cells in vitro.Droplet-to-bubble transition experiment in vitro showed that there were no nano/microbubbles formed by heating in control groups at 80℃,while there were a lot of nano/microbubbles appeared in nanocapsules formulations at 80 ℃.The agents presented a good ultrasound imaging in vitro. Conclusions The HER2-targeted polymeric nanocapsules of liquid perfluorocarbons are successfully prepared, which may become a novel extravascular targeted ultrasound contrast agents.

RESUMEN

Objective To investigate the clinical effect by early mechanical ventilation and perfluorocarbons gasification inhala-tion on patients with severe acute pancreatitis (SAP) complicated with acute respiratory distress syndrome (ARDS) .Methods 34 cases of SAP and ARDS patients accepted early mechanical ventilation and vaporization of perfluorocarbon inhalation therapy (ob-servation group) ,and 34 patients received mechanical ventilation for early treatment alone (control group) ,The efficacy ,oxygenation index(PaO2/FiO2 ) and inflammatory cytokines IL-1 ,6 ,8 ,and TNF-a level of two groups were compared .Results The total effi-ciency of the observation group was 94 .12% ,which was significantly higher than the control group that was 76 .47% (P<0 .05) . After treatment ,the arterial oxygen saturation (SaO2 ) ,arterial oxygen pressure (PaO2 ) ,PaO2/FiO2 of the observation group in-creased significantly ,and higher than the control group(P<0 .05) .After treatment ,IL-1 ,6 ,8 ,TNF-αof the observation group were significantly lower than the control group(P<0 .05) .Conclusion The treatment of early mechanical ventilation with perfluorocar-bon inhalation gasification could control the inflammation in patients with SAP complicated with AROS ,which could improve the cure rate .

RESUMEN

AIM:To investigate the effects of perfluorocarbon and ligustrazine on lung injury during liver transplantation in pigs with hepatopulmonary syndrome. METHODS:A hepatopulmonary syndrome (HPS) model of pig was established by chronic bile duct ligation. The animals were assigned randomly to 2 groups:(1) Perfluorocarbon in combination with ligustrazine treatment groups (PFCL group):the pigs were treated with intratracheal instillation of perfluorocarbon and ligustrazine; (2) The conventional mechanical ventilation group (MV group):all animals were subjected to mechanical ventilation and orthotopic liver transplantation. After 5 h the lungs were harvested for further analysis. RESULTS:The lung wet to dry weight radio, pulmonary permeation index and leukocyte count in bronchoalveolar lavage fluids (BALF) in PFCL group significantly decreased compared to MV group (P<0.05). Contents of tumor necrosis factor-α (TNF-α) and interferon-γ (IFN-γ) in the lung tissue, plasma and BALF of pigs in PFCL group were significantly lower than those in MV group (P<0.05). Moreover, the activation of NF-κB was inhibited markedly by PFCL. CONCLUSION:Perfluorocarbon in combination with ligustrazine effectively reduces the PMN accumulation in the lungs, inhibits TNF-α and IFN-γ production and protects against lung injury during liver transplantation in pigs with hepatopulmonary syndrome.
