RESUMEN

Myrcia hatschbachii D. Legrand (Myrtaceae) is a native and endemic species from Brazil. This study investigated the essential oil composition, phytotoxic potential, in vitro toxicity, antioxidant properties and antibacterial activity of species. Chromatographic analysis of essential oil identified trans-calamanene, (E)-caryophyllene and spathulenol as major components. Antioxidant capacity was determined by the DPPH• scavenging method and phosphomolybdenum complex formation assay. Antibacterial activity was evaluated using the Minimal Inhibitory Concentration test, demonstrating that the essential oil was active toward Enterococcus faecalis and Staphylococcus aureus. Phytotoxic activity of essential oil was analyzed by testing interference on germination and growth of Lactuca sativa, demonstrating significant inhibition of the hypocotyls and radicles of seeds. Preliminary toxicity studies were determined using Artemia salina, resulting in an LC50 of 409.92 µg/mL, and through hemolytic activity. The results of the phytotoxic activity point to a possible application for Myrcia hatschbachii in the development of natural herbicides and the in vitro toxicity assays suggests the performance of antitumoral activity tests, having in mind the prospection of antineoplastic drugs.

RESUMEN

The essential oil from the fresh leaves of Kyllinga pumila (Michx) was obtained by hydrodistillation and characterized by gas chromatography-mass spectrometry (GC-MS). Twenty-eight volatile compounds were identified, major constituents of the oil were Methyl E,E-10,11-epoxyfarnesoate (43.8%), ß-elemene (12.5%), Z-caryophyllene (11.3%), germacrene D (7.1%) and E-caryophyllene (5.6%). Repellent and fumigant activities of the oil against Tribolium castaneum Herbst (Coleoptera: Tenebrionidae), were done using the area preference method. Additionally, we studied their antioxidant and phytotoxic effects. Essential oils exhibited a dose- dependent repellent activity, with values 90% at the applied concentration (0.01%), for both two and four hour's exposure. Essential oil from K. pumila showed 92% mortality at 500 µL L-1 air against T. castaneum on 24 hours of exposure. The value LC50 was 153.4 µL L-1. With moderate selective phytotoxic effects on L. perenne root growth (±70% inhibition). Kyllinga pumila shows high antioxidant potential (91.5%), an effect that is comparable with ascorbic acid (92.9%) used as a standard. The results indicated that K. pumila essential oil could be a promising alternative to new natural antioxidants, repellents, and biocides.

The effects of thymol and carvacrol and the essential oil of Lippia gracilis on caulinary shoots of heliconia were evaluated. After disinfection, the shoots were inoculated into MS medium and subjected to the treatments with 420 µL L-1 of essential oil (EO) of L. gracilis; 420 µL L-1 of thymol; 420 µL L-1 of carvacrol; 210 µL L-1 of thymol and 210 µL L-1 of carvacrol. The control treatment consisted of the MS medium without any phytoregulators. The main components of EO from L. gracilis are carvacrol, ρ-cimene, and thymol. Seven days after the initiation of the experiments, 36.3% of the control treatment shoots were necrotized, but 90% of the caulinary shoots exposed to EO, thymol, or carvacrol appeared necrotized. Transmission electron microscopy of the shoots revealed that the treatment with EO, thymol, or carvacrol caused the destruction of the plasma cell membranes, and the cell organelles and the nucleus were hardly evident. The EO and its main constituent were toxic to caulinary shoots of heliconia.

The effect of different concentrations of nickel nitrate (0.25, 0.50, 1.00 and 2.00 mM) uptake by the roots, on root growth of onion (Allium cepa) and the transport of Ni2+, Fe2+, Mn2+, Zn2+, K+, Na+ and H+ ions were investigated spectrophotometrically. The uptake of Ni2+, Fe2+, Mn2+ and Zn2+ was monitored by flame atomic absorption spectrometry with a 24-h period for 7 days and the amounts of K+ and Na+ were determined in solutions by flame photometer. The mineral content of the solution, instead of the root material was measured. Ni2+ ions showed inhibitory effect on the root growth at all concentrations during the entire treatment. The EC50 (effective concentration that reduced root growth by 50%) was found at 0.25 mM Ni2+. No significant change in inhibitory effect was observed after at 0.50 mM Ni2+ concentrations. A large amount of Ni2+ was translocated into the roots. The kinetics of metal ion transport followed a pseudo-first order reaction in all metal ion concentrations. Ni2+, Zn2+ Fe2+ Mn2+ and H+ ions transferred together into plant, but Na+ and K+ ions transferred to the solution from the plant. The amount of H+ in the solution decreased at all Ni2+ concentrations.
