RESUMEN

OBJECTIVE:To study the mechanism of improvement effects of Fupi rougan granule (FRG)on hepatic fibrosis model rats. METHODS :The rats were randomly divided into blank group ,model group ,Colchicine tablet group (chemical positive control ,0.2 mg/kg),Fuzheng huayu capsule group (TCM positive control ,0.415 g/kg),FRG low-dose ,medium-dose and high-dose groups (20,40,80 g/kg),with 10 rats in each group ,except for 11 rats in blank group and model group (one rat was used to judge whether the modeling was successful ). Except for blank group ,other groups were given intraperitoneal injection of 50% CCl4 olive oil solution and intragastric administration of 30% ethanol to induce hepatic fibrosis model. After modeling , administration groups were given relevant medicine intragastrically ;blank group and model group were given constant volume of normal saline intragastrically ,once a day ,for consecutive 4 weeks. After last administration ,morphology changes of liver tissue in rats were observed. The serum levels of HA ,LN,PCⅢ and Col Ⅳ in rats were detected ,and protein expression of Beclin- 1 and LC3-Ⅱin liver tissue were also determined. mRNA and protein expression of Akt ,AMPK,mTOR,p70S6K were detected in liver tissues of rats. RESULTS :Compared with blank group ,the structure of hepatic lobules in the model group was disordered ,the proliferation of fibrous tissue was obvious ,and some pseudolobules were formed ;the serum levels of HA ,LN,PCⅢ and Col Ⅳ, the protein expression of Beclin- 1 and LC 3-Ⅱ in liver tissue as well as mRNA and protein expression of Akt ,AMPK,mTOR and p70S6K were increased significantly (P<0.01). Compared with model group ,the liver injury of rats in FRG groups was significantly relieved ,and the levels of the above indexes in serum and liver tissue (except for LN and PC Ⅲ in FRG low-dose group) were significantly reduced (P<0.05 or P<0.01). CONCLUSIONS :FRG can improve hepatic fibrosis in rats ,the mechanism of which may be associated with down-regulating the expression of autophagy associated protein and Akt/AMPK/mTOR/ p70S6K signaling pathway related protein.

RESUMEN

Objective To study the effects of Huazhi Rougan granule in treatment of nonalcoholic fatty liver disease and its effect on level of adiponectin, interleukin-6 and tumor necrosis factor-α.Methods 88 patients with nonalcoholic fatty liver disease from October 2014 to September 2015 in our hospital were selected,and divided into observation group and control group according to the order of admission.The control group were taken silibinin meglumine tablets for treatment on the basis of aerobic exercise , dietary guidance.The observation group on the basis of aerobic exercise , dietary guidance were given Huazhi Rougan granule for treatment.The observation group were taken aerobic exercise and metformin for treatment.Two groups of patients with clinical efficacy were evaluated, blood parameters, adiponectin, interleukin-6 and tumor necrosis factor-αwere measured before and 30 days after treatment between the two groups of patients.Results After treatment, the total clinical efficacy of the observation group was significantly higher than the control group ( P<0.05 ) .The levels of adiponectin in the observation group were significantly higher than those in the control group (P<0.05).The level of IL-6, TNF-αin the observation group were significantly lower than those in the control group (P<0.05).Low density lipoprotein cholesterol and triglyceride in the observation group were significantly lower than those in the control group (P<0.05).Conclusion Huazhi Rougan granule can effectively reduce the levels of TNF-αand IL-6 and increase the level of adiponectin in nonalcoholic fatty liver patients, and the clinical curative effect is good.

RESUMEN

Objective To observe the curative effect of Rougan Granules on rats hepatocirrhosis induced by carbon tetra- chloride(CCl_4)with 10 % alcohol and high fat feed.Method The rat model was established by gastric garage of CCh (CCl_4:salad oil=1:1)1 mL/kg twice a week for two weeks,and with 10 % alcohol as drinking water,high fat feed as food.After administration of CCl_4,the rats were divided into 5 groups:the model group,high-,medium-and low-dosage of Rougan Granules groups,the eolchicine group.And normal control group was also set up.After adminis- tration for 6 weeks,hyaluronic acid,typeⅣcollagen and typeⅢprecollagen in serum were measured and liver histopathol- ogy examination was done.Result The contents of hyaluronic acid,typeⅣcollagen and typeⅢprecollagen in model rats were increased significantly,while Rougan Granules could decrease those increase significantly.Liver histopathology exami- nation showed medium or high fatty degeneration of hepatic cell,obvious hyperplasia of fibrous tissue with fibrous distance broaden and pseudolobuli formed in model group,but in Rougan Granules group the general pathological changes of liver were less.Conclusion Rougan Granules have an effect of experimental anti-hepatocirrhosis in rats.
