RESUMEN

Objective To study the effect of exogenous nitric oxide donor sodium nitroprusside(SNP)on antioxidant en?zymes activities and lipid peroxidation of mice infected with Trichinella spiralis. Methods BALB/c mice were infected with T. spiralis separated by the digestion method. Forty?two days post?infection,the peripheral blood and hepatic tissue from the in?fected or normal mice were collected. Then 4 groups were set:liver homogenate from infected mice+SNP(Group A),liver ho?mogenate from normal mice+SNP(Group B),peripheral blood from infected mice+SNP(Group C),and peripheral blood from normal mice+SNP(Group D). The final concentrations of SNP in each group were set as 0(blank control),2,5,10μmol/L and 30μmol/L,respectively. After reacting with SNP at 37℃for 30 min,the superoxide dismutase(SOD),glutathi?one peroxidase(GSH?Px),catalase(CAT)activities,and malondialdehyde(MDA)concentration were examined and com?pared. Results The levels of SOD,CAT,GSH?Px and MDA concentration in the liver and the blood from the mice infected with T. spiralis were significantly higher than those of the normal ones(all P < 0.05). When reacted with 10 μmol/L and 30 μmol/L SNP,the SOD,GSH?Px,and CAT activities in Group A and B decreased significantly(all P<0.05),while the liver MDA concentration reacted with 2-30μmol/L SNP increased obviously(all P<0.05). As reacted with 30μmol/L SNP,the ac?tivities of blood SOD,GSH?Px,and CAT in Group C and D decreased,while the MDA concentration in blood still increased (all P<0.01). When the SNP concentration was in the range of 2-30μmol/L,there were a negative correlation between the SNP concentrations and SOD,GSH?Px,and CAT activities,as well as a positive correlation with the MDA concentration in the liver and blood from the mice infected with T. spiralis(all P<0.05). Conclusions T. spiralis infection could cause oxidative damage to mice,and increase SOD,GSH?Px,and CAT activities. Nitric oxide released from SNP can decrease antioxidase activ?ities,and inhibit the antioxidant capacity of mice infected with T. spiralis.

RESUMEN

OBJECTIVE:To investigate the preventive effect of Gynostemma pentaphyllum water extract on model rats with stress-induced gastric ulcer. METHODS:60 rats were randomly divided into normal group,model group,positive group (Sanjiu weitai granules,3.69 g/kg)and G. pentaphyllum water extract high-dose,medium-dose,low-dose groups(4.0,2.0,1.0 g/kg,cal-culated by crude drug),10 in each group. All rats were intragastrically administrated,once a day,for one week. After 2 h of ad-ministration,except for normal group,rats were induced to stress gastric ulcer model by water immersion method in other groups. After 3 h of modeling,ulcer index (UI),ulcer inhibition rate were detected;superoxide dismutase (SOD) and malondialdehyde (MDA)in serum,tumor necrosis factor-α(TNF-α),prostaglandin E2(PGE2),nitric oxide(NO)and endothelin-1(ET-1)levels in gastric mucosal tissue were determined. RESULTS:Compared with normal group,SOD level in model group was decreased, while MDA level was increased in serum(P<0.05);PGE2,NO levels were decreased,TNF-α,ET-1 levels were increased in gas-tric mucosal tissue (P<0.05). Compared with model group,UI in administration groups were obviously decreased,above-men-tioned indexes were obviously improved,which showed certain dose-dependence. And the levels of PEG2,ET-1 of gastric mucosal tissue in G. pentaphyllum water extract high-dose group improved more obviously than positive group,with statistical significances (P<0.05). CONCLUSIONS:G. pentaphyllum water extract plays a role in preventing gastric ulcer by regulating serum SOD-MDA balance,gastric mucosal tissue NO-ET-1 balance and reduce the degree of gastric mucosa injury,etc.

AIM:To observe the effect of simvastatin on myocardial tissue after renal ischemia-reperfusion in-jury and its mechanism .METHODS:A rat model of renal ischemia-reperfusion injury was prepared by clamping the bilat-eral renal arteries for 45 min.The rats (n=36) were randomly divided into sham operation group , renal ischemia-reperfu-sion (I/R) group and simvastatin group with 12 rats in each group.The content of serum creatinine (SCr), blood urea ni-trogen ( BUN) and myocardial tissue malondialdehyde ( MDA) , the myocardial activity of lactate dehydrogenase ( LDH) , creatine kinase (CK) and superoxide dismutase (SOD), and the myocardial protein expression of Bcl-2 and Bax were de-tected.RESULTS:Compared with sham operation group , the content of SCr , BUN and myocardial MDA , and the myo-cardial activity of LDH and CK in I/R group were significantly increased (P<0.05), and the activity of SOD was signifi-cantly decreased (P<0.05).Compared with I/R group, the content of SCr, BUN and myocardial MDA, and the myocar-dial activity of LDH and CK in simvastatin group were significantly decreased ( P<0.05 ) , while SOD activity was en-hanced (P<0.05).The protein expression of Bcl-2 and Bax in sham operation group was less than that in I/R group (P<0.05), and the protein level of Bax in simvastatin group was significantly lower than that in I /R group (P<0.05), while the protein level of Bcl-2 was increased (P<0.05).CONCLUSION:Simvastatin has a protective effect on the my-ocardium of the rats with renal ischemia-reperfusion injury , and the protective mechanism may be related to the elimination of free radicals by simvastatin , increase in the protein expression of Bcl-2 and decrease in the protein expression of Bax .

Aim To investigate the effects and possible mechanisms of kaemperol in the rats with chronic cere-bral ischemia.Methods Chronic cerebral hypoperfu-sion model was produced by permanent occlusion of bi-lateral common carotid arteries (2VO)in rats.After KAE treatment,the rats underwent Morris water maze and prehensile traction test.Neuronal morphology was observed using Nissl and HE staining.The activity of SOD and the content of MDA in brain tissue were de-termined.The DJ-1 protein expression was assayed by Western blot.Results Compared with 2VO model group,KAE significantly improved learning and memo-ry and the grasping ability.In addition,KAE signifi-cantly reduced brain tissue pathological injury induced by 2VO. Furthermore, KAE significantly increased SOD activity and enhanced antioxidant protein DJ-1 ex-pression in brain tissue.Conclusions KAE could sig-nificantly attenuate the cognitive impairment,limb bal-ance dysfunction and pathological injury in rats with chronic cerebral ischemia.The mechanism may be re-lated to improving the antioxidant system in vivo.

RESUMEN

OBJECTIVE: To assess maternal circulating levels of lipid peroxides, superoxide dismutase and total antioxidants of women with pregnancy-induced hypertension. METHODS: Cross-sectional study consisting of 18 preeclamptic, 21 uncomplicated pregnant and 22 healthy non-pregnant women. Fasting venous blood samples were collected during the 3rd trimester of antepartum period and maternal circulating levels of malondialdehyde as a lipid peroxidation product, superoxide dismutase activity and total antioxidants were measured. RESULTS: In the preeclamptic group, lipid peroxides were significantly increased, otherwise the activity of superoxide dismutase in the erythrocytes was significantly decreased compared to normal pregnant women. The value of serum total antioxidants was similar in both groups. Strong correlation was detected between malondialdehyde and blood pressure in the pregnant women. CONCLUSION: Preeclampsia is associated with decrease of antioxidant enzyme activity while lipid peroxidation was increased during the 3rd trimester of pregnancy. Therefore, a significant elevated lipid peroxidation and reduced superoxide dismutase activity may contribute to pathophysiology and pathogenesis of preeclampsia via vascular endothelial cell damage.
