RESUMEN

Objective To determine potential gene-gene interactions of TNF-? and VDR loci with outcomes of hepatitis B virus(HBV) infection.Methods A total of 391 chronic hepatitis B(HB) patients as a case group and 212 HBV self-limited infected subjects as a control group were recruited to conduct a case-control study.TNF-?-238G/A,-857C/T,-863C/A,VDR-TaqⅠT/C and FokⅠC/T gene polymorphisms were examined by polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP).The interactions between TNF-? and VDR genes were analyzed by multiple model.Results There were positive gene-gene interactions between TNF-?-238 GA and FokⅠ CT/CC(ORint=4.04),between-863 CC and-857 CC(ORint=1.26) and between-857 CC and FokⅠ CT/CC(ORint=1.37),respectively,which increase the risk of chronic hepatitis B.There were negative gene-gene interactions between TNF-?-238 GA and-857 CC(ORint=0.92)and between FokⅠ CT/CC and TNF-?-863 CC(ORint=0.95),which decrease the risk of chronic hepatitis B.Conclusion Interaction between TNF-? loci and VDR loci potentially increase the risk of chronic hepatitis B after HBV infection.

RESUMEN

Objective:To investigate the possibility of producing vaccine with higher effect of anti-hepatoma by means of freezing the tumour cells which have been modified by TNF-? genes before. Methods: H22 cells which had been modified by TNF-? genes were treated by means of freezing and 60Co radiation or of 60Co radiation, respectively. Then, they were put to use to produce two kinds of vaccines. Effects of these two kinds of vaccines on anti- plant-hepatoma in mice were compared. Results: Effects of increasing the inhibiting rates, the levels of lectin-induced IL-2 and TNF from splenocytes, and the splentic NK activity, were all obviously seen stronger in vaccine made up of cells treated by the method combined with freezing, than that in vaccine made up of cells by no freezing. Conclusion: Method of freezing, after procedure of TNF-a gene modifying, could raise up the immunogenicity of the tumour cells, thereby enhance the anti-plant-hepatoma effect in mice induced by the vaccine.

RESUMEN

To determine whether uremia per se can activate peripheral blood mononuclear cells (PBMCs). Methods Maintained hemodialysis patients with cuprophane, non-dialysis uremic patients and healthy volunteers were selected to investigate IL-1?、 TNF-? and 11,6 mRNA in PBMCs by RT-PCR and cells in situ hybridization. Results IL-1?,TNF-? and IL-6 mRNA in PBMCs were undetected in healthy volunteers, but these cytokine gene expression were detected in hemodialysis patients and non-dialysis uremic patients. IL-1?、TNF-? and IL-6 mRNA level was lower in non-dialysis uremic patients than that in hemodialysis patients. Conclusion Uremia per se or its related factors may activated PBMCs and the preactivation of PBMCs might be associated with the body resistance against infection in uremic patients.

RESUMEN

The tumor necrosis factor(TNF) secreted by gene modified tumor cells can lead to very effective tumor rejection. This effect of TNF on tumor growth is mediated mainly by the induction of an antitumor immune response. It requires a local and continuous presence of TNF at the tumor site. Tumor suppression induced by TNF is close dependent and a complete tumor eradication must not be accompanied by systemic toxic side effects. A complex pattern of tumor infiltrating cells has been observed in TNF producing tumors, consisting of macrophages, CD4+ and CD8~(+)T cells. For efficient tumor inhibition maccophages and CD8~(+)T cells are needed,whereas CD4~(+)T cells seem to be innocent bystander cells. TNF is also effective in T eell deficient mice, but in most cases for complete tumor elimination T ceils have to be present. Depending on the cell lines used or the levels of TNF secreted by transduced tumor cells, systemic toxicity of TNF has also been observed including cachexia or wasting of the experimental animals. In some cases, TNF gene transfected tumors did not show growth inhibition in vivo, but rather, their metastases were enhanced. Using TNF producing tumor cells as vaccine, no systemic protective immunity against a parental tumor cell challenge has been observed.

RESUMEN

Objective:To study single nucleotide polymorphisms in the promoter region of TNF-? gene in Chinese Han,Miao and Buyi population in Guizhou province.Methods:The five polymorphism sites at-1031,-863,-857,-308,-238 in TNF-? gene promoter region were detected using PCR-RFLP and SSP-PCR.Results:The allele frequencies of TNF-? gene in promoter region in these three main ethons of population were observed.In Chinese Han population,the-238 G was 96% and-238 A was 4%.Inaddition,-308 G was 92% and-308 A was 8%,-857 C was 65% and-857 T was 35%,-863 C was 80% and-863 A was 20%,-1031 T was 48% and-1031 C was 52%;In Miao people,the frequency at the loci above were 15.0%,8.0%,26.0%,4.0% and 50.0%;And the frequency of these five loci above were 14.0%,6.0%,11.0%,16.0% and 50.0% in Buyi population.Conclusion:Single nucleotide polymorphisms of TNF-? gene exhibits some specific characters in Chinese Han and Miao and Buyi population in Guizhou province.
