RESUMEN

Epigenetic pathways play a critical role in the initiation, progression, and metastasis of cancer. Over the past few decades, significant progress has been made in the development of targeted epigenetic modulators (e.g., inhibitors). However, epigenetic inhibitors have faced multiple challenges, including limited clinical efficacy, toxicities, lack of subtype selectivity, and drug resistance. As a result, the design of new epigenetic modulators (e.g., degraders) such as PROTACs, molecular glue, and hydrophobic tagging (HyT) degraders has garnered significant attention from both academia and pharmaceutical industry, and numerous epigenetic degraders have been discovered in the past decade. In this review, we aim to provide an in-depth illustration of new degrading strategies (2017-2023) targeting epigenetic proteins for cancer therapy, focusing on the rational design, pharmacodynamics, pharmacokinetics, clinical status, and crystal structure information of these degraders. Importantly, we also provide deep insights into the potential challenges and corresponding remedies of this approach to drug design and development. Overall, we hope this review will offer a better mechanistic understanding and serve as a useful guide for the development of emerging epigenetic-targeting degraders.

Abstract: Background: Nutritional labeling enables consumers to make better food choices, helping to fight chronic non-communicable diseases. Objective: To determine the influence of the interpretation of the nutritional labeling, present in industrialized foods in Colombia, on the purchase decision of undergraduate students in the health area of the CES University. Material and Methods: A descriptive observational study was carried out in a population of 76 fifth-semester students enrolled in the undergraduate programs of the health area. Results: The frequency of nutrition labeling use was associated with sex (p=0.036), the type of undergraduate program (p<0.001), and knowledge of the function of the nutrition labeling (p=0.000). A good interpretation of the nutrition labeling does not condition the frequency of its use (p=0,095). Additionally, the level of interpretation of the nutritional labeling was not associated with sex, nor the type of undergraduate program. Finally, it was found that the purchase decision was influenced by custom (p=0.018) and advertising (p=0,008), and not by the interpretation of the nutrition labeling. Conclusions: Although the use of nutrition labeling is more frequent among women, among students of Nutrition and Dietetics, and among those who are aware of the role of nutrition labeling, the purchase decision is not influenced by its interpretation.

RESUMEN

Introduction: The bull shark, Carcharhinus leucas, is particularly vulnerable to anthropogenic actions because of its permanence in coastal ecosystems; populations depletion is registered in different places around the world. Aggregations of bull sharks have been reported in the North Pacific of Costa Rica, at Islas Murciélago, within the Guanacaste Conservation Area. Objective: To study the residency of bull sharks at San Pedrillo islet, Islas Murciélago. Methods: During the study period (June 2013 to February 2015) we used passive telemetry to tag 10 bull sharks. Results: All the sharks tagged were females, they were detected on 59 798 occasions by the acoustic receiver deployed in San Pedrillo. Acoustic signals from tagged sharks were received for a total period of 1 to 229 days (mean = 73.9 ± 71.3 days), with the last detections occurring on 9 January 2015. The Residency Index for each tagged shark across the entire monitoring period ranged from 0.41 to 1.00. The bull shark activity showed a significant pattern throughout the day at the receiver that specifically corresponded with the daily light cycle. Conclusions: This study concludes that San Pedrillo is an aggregation site (cleaning station) for bull sharks (C. leucas), possibly related to reproduction and not feeding behaviors.

Background@#Lower Gastrointestinal bleeding (LGIB) is a serious and urgent condition which can be assessed using several different modalities. Tc-99m tagged RBC scintigraphy has been established as a diagnostic tool in Nuclear Medicine but several other modalities, including CT-based imaging (i.e. angiography) currently exist. @*Objective@#The objective of this study is to compare Tc-99m tagged RBC scintigraphy with CT-based imaging studies in terms of clinical utility and diagnostic outcomes. @*Methods@#A systematic review of available literature was done, with the goal of creating a meta-analysis focusing on the reported diagnostic outcomes - mainly sensitivity and specificity on the presence of a LGIB. Aside from this, a systematic review of the clinical utility and the differences of each test were discussed, including non-quantifiable advantages. The literature search was conducted following the guidelines of PRISMA, with searches from PubMed, Medline, and other pertinent databases. Quality assurance was done using the QUADAS tool. Statistical analyses of sensitivity, specificity, and a summary receiver operating characteristics plot were computed for the meta-analysis.@*Results@#Pooled sensitivity and specificity for RBC scintigraphy were 0.886 and 0.119, respectively. Pooled sensitivity and specificity for CT-based imaging were 0.729 and 0.660, respectively. CT based imaging also showed higher localization and faster completion times. RBC scintigraphy had a longer acquisition window.@*Conclusion@#Both Tc99m-tagged RBC scintigraphy and CT-based imaging have important clinical utility, with each modality having different advantages that the other test cannot provide.

RESUMEN

@#In tbhe assessment of prosthetic joint pains, differentiating between aseptic loosening and infection can be challenging due to their similarities in clinical presentation and histopathology. Combined radiolabeled leukocyte and bone marrow scintigraphy and SPECT/CT is considered the most suitable nuclear imaging modality for this purpose. However, this is infrequently performed in our local setting. We present two cases where these studies were appropriately performed with different scan outcomes. The first case involved a 67-year-old male with painful right knee prosthesis and an alleged history of recurrent periprosthetic joint infection (PJI). The leukocyte tagging scan and marrow scan images were found to be congruent, which was more compatible with aseptic loosening. No emergent surgical intervention was done in our hospital, and the patient returned to this home territory for further management. The second case was a 72-year-old male with progressive pain in his left prosthetic hiip which began after surgery a year prior. Scan images were incongruent, suggestive of PJI; this was confired on joint aspiration and eventual surgical revision. Despite this imaging study's high sensitvity and high specificity, it has not been well-received in management algorihms of PJI. Collaboration between clinicans and nuclear imaging specilists is key in increasing the general use of these procedures.

RESUMEN

The protection of the habitats used by juvenile sharks is a management strategy that has recently caught the attention of fishery biologists. In the present study, we evaluated the population of the lemon shark (Negaprion brevirostris) from Los Roques Archipelago in order to identify the nursery area, describe the size composition, and examine the variation in nocturnal activity of the juvenile individuals. The data analysed came from three different sources: commercial shark fishery, tag-recapture sampling, and visual records. A total of 375 lemon sharks with total lengths between 55 and 281 cm were recorded during the study period. Overall data showed that the area occupied by juvenile lemon sharks was clearly partitioned into primary and secondary nurseries. Additionally, nighttime activity seemed to change according to the size of sharks in the primary nursery, suggesting a reduction of time activity overlapping among juveniles of distinct size/age. Results suggest that the strategy of utilization of the primary nurseries by the lemon shark may lead to important ecological benefits by reducing the competition, predation and natural mortality.

Photo-identification allows individual recognition of animal species based on natural marks, being an alternative to other more stressful artificial tagging/marking techniques. An increasing number of studies with different animal groups has shown that photo-identification can successfully be used in several situations, but its feasibility to study freshwater fishes is yet to be explored. We demonstrate the potential use of photo-identification for intraspecific recognition of individuals in the stream-dwelling loricariid Rineloricaria aequalicuspis . We tested photo-identification in laboratory and field conditions based on the interindividual variability in abdominal bony plates. Our test yielded high correct matches in both laboratory (100%) and field conditions (> 97%), comparable to other reliable techniques and to studies that successfully used photo-identification in other animals. In field conditions, the number of correct matches did not differ statistically between computer-assisted and naked-eye identification. However, the average time expended to conclude computer-assisted photo evaluations was about half of the time expended to conclude naked-eye evaluations. This result may be exacerbated when using database with large number of images. Our results indicate that photo-identification can be a feasible alternative technique to study freshwater fish species, allowing for a wider use of mark-recapture in ecological and behavioral studies.

In recent decades, Brazilian hydroelectric reservoirs have been stocked with hundreds of millions of juvenile migratory fish, but almost no research on stocking has been conducted. In order to study stocking it is essential to tag the fish to be stocked. In this investigation we determined handling-induced mortality, tag retention rate, and absolute growth rate of hatchery-reared juvenile curimbatá (Prochilodus lineatus) tagged with coded wire tags (CWT), a tag widely used on stocked fish. We injected CWT into the dorsal musculature just below the first dorsal fin ray. Despite the need for frequent tag injector maintenance, this tagging location proved to be suitable for tagging juvenile curimbatá. We found no short-term mortality of juvenile curimbatá handled for CWT tagging, high retention rate for CWT (≥ 98.2%) even after 174 d of tagging, and no influence on absolute growth rate by CWT. We conclude that CWT is effective at tagging juvenile curimbatá, and we expect that it will also be effective for many other Brazilian fish species used for stocking.

Abstract Olive Ridley sea turtle (Lepidochelys olivacea) have been amply studied on Costa Rican beaches that experience mass nestings, "arribadas", both in their nesting behavior and other aspects of their biology; however, very little published information exists about beaches where the Olive Ridley is nesting in solitary form, especially along the Southern Pacific Coast. For this reason, the objective of this study is to describe relevant nesting aspects, such as number of nest per season and nesting sites of L. olivacea on the Tortuga Beach in the South Pacific of Costa Rica, during three nesting seasons (2010, 2011, 2012). The beach was divided into 14 sectors of 100m each. Beach patrols were conducted every night from July to December (2010) and July to January (2011, 2012), with the purpose of observing and recording nesting behavior in the turtles as well as to protect their nests. The females observed were identified by means of metal tags applied to their rear flippers. Other data collected included: number of eggs deposited, nest location, hour, biometric data (width and length taken along the curve of the carapace), and the total of nests laid (found with or without the turtle present). After three seasons, 100 females of Olive Ridley Sea Turtle were successfully marked, and a total of 233 nests were counted. The frequency of re-nesting of an individual during a season was of every 14 to 20 days and four individuals re immigrated to nest again over the period of one year. The total population of nesting females during these three years was estimated at 117. The peak nesting happens in September, more sea turtles visited the beach during this period in all the seasons reported. With respect to biometry, the average value of the LCC was 69.91±1.05cm and the average value of the WCC was 70.476+/-1.767cm. A total of 18 711 eggs were collected and 9 858 were successfully hatched. The Tortuga Beach was proven to be a nesting beach of the species L. olivacea; the population of nesting females on the beach can be considered significantly reduced in comparison with other nesting beaches of the same species along the Southern Pacific Coast of Costa Rica. Furthermore, marking the turtles with metal tags has proven a useful tool towards the characterization of the nesting aspects of the sea turtles, such as the frequency and interval of re-nesting, and other variables such as the re immigration which requires a period of time greater than the time period of the study to date. RESUMEN

Objective To study the automatic word segmentation scheme suitable for traditional Chinese medical record literature. Methods Hierarchical Hidden Markov Model was used as segmentation model. Totally 300 ancient medical record literature and 300 modern medical record literature were set as experimental subjects to establish the dictionary of traditional Chinese medicine and the test corpus, with a purpose to segment the words and evaluate of the results. Results Without using dictionary of traditional Chinese medicine, the word segmentation accuracy of two kinds of medical record literature was about 75%;the part-of-speech tagging accuracy of ancient medical literature was 56.74%, the modern medical literature accuracy was 64.81%. By using dictionary of tradition Chinese medicine, the word segmentation accuracy of ancient medical record literature was 90.73%, the modern medical record literature accuracy was 95.66%;the part-of-speech tagging accuracy of ancient medical record literature was 78.47%, the modern medical record literature accuracy was 91.45%, which was obviously higher than that of ancient medical record literature. Conclusion The current word segmentation scheme has initially solved the problem of word segmentation of traditional Chinese medical record literature and part-of-speech tagging of modern medical record literature. Part of speech tagging is basically correct, but part-of-speech tagging of ancient medical record literature tagging needs further study for many influencing factors.

RESUMEN

Tag, similar to a keyword and belonging to a non-controlled term, is a marking language, and can be freely marked by its users, thus playing a role in organization of information resources.Social tagging, a tag created by network users and added to the resources they concerned, has become one of the important methods for the network information resources organizing system to organize and mange resources.Suggestions were thus put forward in this paper for improving the information retrieval function of future social tagging system by comparing its 3 functions and assessing its efficiency.

RESUMEN

We review recent work on three major lines of memory research: a) the possible role of the protein kinase M-zeta (PKMzeta) in memory persistence; b) the processes of “synaptic tagging and capture” in memory formation; c) the modulation of extinction learning, widely used in the psychotherapy of fear memories under the name of “exposure therapy”. PKMzeta is a form of protein kinase C (PKC) that apparently remains stimulated for months after the consolidation of a given memory. Synaptic tagging is a mechanism whereby the weak activation of one synapse can tag it with a protein so other synapses in the same cell can reactivate it by producing other proteins that bind to the tag. Extinction, once mistakenly labeled as a form of forgetting, is by itself a form of learning; through it animals can learn to inhibit a response. We now know it can be modulated by neurotransmitters or by synaptic tagging, which should enable better control of its clinical use.

Approximately 12 species of shark inhabit at Coco Island National Park (PNIC); some are migratory, such as the hammerhead shark (Sphyrna lewini) and whale shark (Rincodon typus), and others are resident, like the whitetip shark (Triaenodon obesus). The whitetip shark is a species related to coral reefs, it inhabits coastal environments and is nocturnal and, it is very common in tropical waters, but there are few studies worldwide. In fact, only short researches have been done about their behavior at PNIC. Therefore, this study aimed to give data about the abundance, size and population structure of the whitetip shark at Chatham Bay in Coco Island National Park, providing baseline information for monitoring its population which is also very vulnerable to climate change. In order to accomplish this aim, a mark-recapture method was used, as well as underwater visual counts to determine their relative abundance and visual recaptures at Chatham Bay. The most effective hours for tagging were between 18:00 and 22:00 (sunset), when the whitetip sharks were more active probably related to foraging behavior. In addition, the depth was a relevant factor for tagging, sharks were caught in shallow water (10.9±1.9m). This species prefers to feed in shallow water and this behavior is probably related to the distribution and occurrence of rock and coral reefs in Chatham Bay. Based on the results, whitetip shark do not have a defined spatial ontogenic segregation, since in the same sites both juveniles and adults were found. The study confirmed that whitetip shark use specific residence sites: during the tagging trips, different individuals were recaptured; and during the underwater counts, at least 32.76% of tagged sharks were observed. The average length of the tagged sharks was 101.8±12.1cm (130.0cm maximum and 71.0cm minimum). The relative abundance of the whitetip shark in Chatham Bay observed was estimated on 49.5±10.4 sharks/hour. Furthermore, using the recapture data obtained during the night tagging expeditions, a population of 408 (IC = 181-1050) sharks was estimated.

In the present study, a novel plant transformation system for Doritaenopsis and Phalaenopsis has been developed. The pollen-mediated activation tagging system was established by artificial pollination. The pollens, co-cultured with Agrobacterium tumefaciens strain EHA105 harbouring an activation tagging vector (pTAG-8), were used for pollination. In order to optimize the transformation efficiency, several factors (concentration of A. tumefaciens, concentration of acetosyringone during co-cultivation and the duration of co-cultivation) known to influence Agrobacterium-mediated DNA transfer were examined. A concentration of 0.5-1 x 10(8) CFU/ml for A. tumefaciens, 0.1 mM acetosyringone, and 6 hrs of co-culture period were found to be the optimal condition for high transformation efficiency. Integration of T-DNA into the genome of putative transgenic plants was confirmed by PCR and DNA blot analyses. Single copy of the transgene was observed in all transgenic plants analyzed. Most of the transgenic plants had a morphologically normal phenotype and the overall capsule formation efficiency was similar to control plant. Our results showed a new approach of genetic transformation in orchids and this method can be employed for genetic improvement of the orchids.
