RESUMEN

Aims: This study is aimed at determining the concentration of two widely used BFRs; Decabromodiphenyl Ether (BDE-209) and Tetrabromo Bisphenol-A (TBBPA) in sediment and leachate samples. Place and Duration of Study: Field sampling were carried out from five major dumpsites around Warri Municipality, Delta State, Nigeria. Analyte extraction was done in 2017 at the Science laboratory, Federal University of Petroleum Resources, Effurun Delta State, Nigeria and quantification done in Switzerland by Bachema Analytical Laboratories in 2017. Methods: Three soil samples were collected from each site 15cm from the soil surface. Also, three leachate samples from three different trial pits done for each site. Collected soil samples were stored in glass bottles and labelled. While the leachate samples are stored using glass containers and labelled. The BFRs were extracted using Aceton and cyclohexane for each soil matrix and cyclohexane for the leachate samples, then the extract was analysed using GC coupled with an ECD supplied by Thermo Trace GC Ultra, Italy. Results: The results showed the average concentration for TBBPA in the sediments was 0.0234 g/kg and that of the BDE-209 was recorded as 0.1828 g/kg. Results from the leachate sample were below the detectable range of the analytical equipment, TBBPA (0.02 g/kg) and BDE (0.1 g/kg). There is no statistical difference between the mean concentration of TBBPA for the sediment in each of the locations (P>.05) and no difference (P>.05) for BDE-209 for the sediment in each of the locations (P>.05). Conclusion: Findings from this study holds that the concentration of TBBPA and BDE-209 in sediment is higher when compared with concentrations presented in other literatures studied in this report and this calls for immediate action due to the health risk associated with exposure in these municipalities.

BACKGROUND: Tetrabromobisphenol A (TBBPA), one of the most widely used brominated flame-retardants, is a representative persistent organic pollutants group. Studies on TBBPA toxicity have been conducted using various target cells; however, few studies have investigated TBBPA toxicity in bone cells. Therefore, this study investigated the in vitro effects of TBBPA on osteoclasts, a cell type involved in bone metabolism. METHODS: RAW264.7 cells were cultured in medium containing 50 ng/mL receptor activator of nuclear factor kappa B ligand (RANKL) and varying concentrations of TBBPA. To evaluate the effects of TBBPA on the differentiation and function of osteoclasts, osteoclast-specific gene expression, tartrate-resistant acid phosphatase (TRAP) activity, bone resorbing activity, mitochondrial membrane potential (MMP) and mitochondrial superoxide were measured. RESULTS: The presence of 20 μM TBBPA significantly increased TRAP activity in RANKL-stimulated RAW264.7 cells, the bone resorbing activity of osteoclasts, and the gene expression of Akt2, nuclear factor of activated T-cells cytoplasmic 1, and chloride channel voltage-sensitive 7. However, TBBPA treatment caused no change in the expression of carbonic anhydrase II, cathepsin K, osteopetrosis-associated transmembrane protein 1, Src, extracellular signal-related kinase, GAB2, c-Fos, or matrix metalloproteinase 9. Furthermore, 20 μM TBBPA caused a significant decrease in MMP and a significant increase in mitochondrial superoxide production. CONCLUSION: This study suggests that TBBPA promotes osteoclast differentiation and activity. The mechanism of TBBPA-stimulated osteoclastogenesis might include increased expression of several genes involved in osteoclast differentiation and reactive oxygen species production.

RESUMEN

OBJECTIVE: To develop a method for simultaneous detection of 4 kinds of bisphenol A analogs in human urine by high performance liquid chromatography and tandem quadrupole mass-spectrometry. METHODS: The urine samples were extracted by solid phase extraction( Bond Elut Plexa) after hydrolyzed using β-glucuronidase and sulfatase. The gradient elution was carried out with acetonitrile-water. The bisphenol A-d16 and tetrabromobisphenol A-d10 were used as internal standard substances to conduct the quantitative analysis using the combined mode of liquid chromatography-mass spectrometry and mass spectrometry with the negative electrospray ionization,and the multiple reaction monitoring scan mode. RESULTS: The linear range of all the 4 kinds of bisphenol A analogs( Bisphenol F, Bishpenol A,Tetrachlorobisphenol A and Tetrabromobisphenol A) in urine was found to be 0. 50-100. 00 μg / L, and the linear correlation coefficients were 0. 998 6,0. 999 1,0. 999 6 and 0. 999 7,respectively. The limits of detection were 0. 21,0. 25,0. 01 and 0. 14 μg / L. The recovery rate was 69. 2%-99. 9%. The relative standard deviations of within-run and between-run precisions were 5. 5%-22. 8% and 2. 4%-14. 4%,respectively. The samples could be kept for at least 7 days at room temperature. The method was applied to detect urine samples of 38 children and 20 medical workers and the results were statistically analyzed. The detection rates of bisphenol A, bisphenol F, tetrachlorobisphenol A and tetrabromobisphenol A for children group were 94. 7%,94. 7%,0. 0% and 34. 2%,respectively,and those of the medical workers group were 100. 0%,100. 0%,5. 0% and 75. 0%,respectively. CONCLUSION: The method is effective for the detection of 4 kinds of bisphenol A analogs in human urine with simple operation,high sensitivity and good selectivity.

RESUMEN

A reliable UPLC-MS method was developed for the simultaneous determination of 4 chemicals (Sudan Ⅰ, tetrabromobisphenol A, tris ( 1, 3-dichloroisopropyl ) phosphate and tris ( chlorisopropyl ) Phosphate) in children products. The samples were ultrasonic extracted with acetonitrile, and then the four chemicals were separated on a C18 column in 3 min. Results showed that the limit of quantification of the method was between 5 and 500μg/kg. The calibration curves were linear within 2-3 orders of magnitude with typical correlation coefficient above 0 . 9995 . The recoveries ranged from 83 . 7% to 97 . 8% with three addition levels. The sensitivity, recovery and selectivity of the method could fully meet the requirements of practical work.

A stimulate method for determination of polybrominated diphenyl ethers ( PBDEs) and derivatives (OH-PBDEs and MeO-PBDEs), tetrabromobisphenol A (TBBPA), hexabromocyclododecane (HBCD) in egg samples was developed by gel permeation chromatography ( GPC) and dispersive solid phase extraction ( DSPE) combined with liquid chromatography tandem mass spectrometric ( HPLC-MS/MS) and gas chroma-tography-negative chemical ionization mass spectrometry ( GC-NCI/MS ) . The analytes were extracted with mixture of hexane and dichloromethane (1∶1, V/V) by accelerated solvent extraction (ASE), and purified by 100 mg C18 dispersive solid phase extraction ( SPE) sorbents followed with gel permeation chromatography (GPC) , and then analyzed by liquid chromatography tandem mass spectrometric (HPLC-MS/MS) and gas chromatography-negative chemical ionization mass spectrometry (GC-NCI/MS), respectively. The quantita-tion was carried out external standard method. The recoveries of objects were 64. 5%-97. 2% and 65. 6%-109 . 2% ( except BDE85 was 54 . 8%, OH-BDE-137 was 47 . 4%) spiked at 1 . 0 μg/kg or 5 . 0 μg/kg in egg white and egg yolk, respectively. The relative standard deviations (RSDs) were less than 20. 2%. The limits of quantitation (LOQ) for the object were 0. 01-0. 2 μg/kg.

RESUMEN

An analysis method was developed for the determination of free and bound fractions of polybromi-nated ethers (PBDEs) and tetrabromobisphenol A (TBBPA) in sediment. The free PBDE and TBBPA were extracted with acetone/hexane ( 1:1) mixed solvent,and the bound fraction was released from the sediment by saponification reaction at 80℃. PBDEs and TBBPA were separated via adjusting pH; PBDEs were determined by GC-negative chemical ionization (NCl)-MS after cleaned up by multilayer silica column,and TBBPA was derivative before pre-separated by acid silica column and finally determined by GC-EI-MS. All of the target compounds were quantitative by internal standard method. The limits of detection of eight low bromina-ted congeners (BDE28,-47,-66,-100,-99,-154,-153 and -183),deca-BDE ( BDE209),and TBBPA were 0.6 - 12.5 pg/g,172 pg/g and 4. 2 pg/g,respectively;This method is reliable and stable. The recoveries of all compounds were ranged from 74% to 106% and the relative standard deviations were below 10%. This method is suitable for the measurement of different forms of PBDEs and TBBPA in sediment under reliable quality assurance and quality control.

Tetrabromobisphenol A(TBBPA)is supposed to have potential thyroid disrupting activities due to its similar structure to thyroid hormones.TBBPA has been proved to show thyroid disrupting effects in vivo and in vitro studies.TBBPA might disrupt thyroid hormone system through transthyretin- and thyroid receptor-mediated pathways.The ability of TBBPA inducing the production of reactive oxygen species might be the extension of its thyroid disrupting activities.The thyroid disrupting effects of TBBPA might be closely related to its oxidative stress,reproduction toxicity and neurotoxicity.More experiments are required for the effects of TBBPA on the aquatic and amphibian animals.

RESUMEN

Tetrabromobisphenol-A (TBBP-A) is the most widely used brominated flame retardant in the world. It is a kind of potential environmental endocrine disruptor, similar to the persistent organic pollutants. It can accumulate in the environment and biota and it has adverse effects on the environment and the living creature, such as disrupting the action of hormones related to the skeleton and brain development. Research progresses on TBBP-A, including toxicology, analysis methods, were reviewed in this paper.
