RESUMEN

Porcine proliferative enteropathy (PPE) is one of the most common enteric diseases in growing and finishing pigs. PPE is characterized by reduced growth performance, accompanied or not by diarrhea. PPE is highly prevalent in several countries of the Americas, Europe and Asia, causing high economic losses in swine herds. The most common form of PPE control in pigs is antibiotic therapy. The objective of this study was to evaluate a new product based on tylosin injectable (Eurofarma Laboratórios S.A.) to control PPE in experimentally inoculated animals. Sixty 5-week-old pigs with mean weight of 9.5kg were divided into two experimental groups of 30 animals: medication and control. All pigs were challenged with Lawsonia intracellularis, the etiologic agent of PPE, on day zero. Fecal score, body condition score, and behavior were daily evaluated. Pigs were weighted on days -2, 13 and 21 of the experiment. Pigs in the Medication Group received tylosin injectable 13 days after inoculation, in three doses with a 12-hour interval between them. Pigs in the Control Group received injectable saline solution following the same protocol. In the Control Group, 23pigs presented with diarrhea before day 13. After day 13, the number of diarrheic animals in this group was reduced to 17. In the Medication Group, 26 pigs presented with diarrhea in the initial period, and in the period after medication, only 11 animals had diarrhea. The score of gross intestinal PPE lesions in the Medication Group was lower than that in the Control Group (p=0.031). The Medication Group also showed lower score for Lawsonia intracellularis antigen-labeling by immunohistochemistry compared with that of the Control Group (p=0.032), showing lower level of infection. These results demonstrate that tylosin injectable (Eurofarma Laboratórios S.A.), administrated in three doses (1mL/20kg) every 12 hours, was effective for the control of PPE in experimentally inoculated pigs.(AU)


Enteropatia proliferativa suína (EPS), causada pela bactéria Lawsonia intracellularis, é uma das doenças entéricas mais comuns em suínos de recria e terminação. A EPS caracteriza-se por redução no desempenho dos animais, acompanhada ou não por diarreia. É uma doença altamente prevalente em diversos países da América, Europa e Ásia, provocando elevados prejuízos econômicos nos rebanhos suínos. A forma de controle da EPS mais adotada em rebanhos suínos é a antibioticoterapia. O objetivo deste estudo foi avaliar um novo produto à base de tilosina (Eurofarma Laboratórios S.A.) na forma injetável para controlar a EPS em animais experimentalmente inoculados. Foram utilizados 60 leitões, de cinco semanas de idade, com peso médio de 9,5kg, divididos em dois grupos experimentais (n=30), medicados e não medicados. Todos os leitões foram desafiados com Lawsonia intracellularis no dia zero. Avaliações clínicas de escore fecal, escore corporal e comportamento foram realizadas diariamente além da pesagem individual dos animais realizada nos dias -2, 13 e 21 do experimento. Os leitões do grupo medicado receberam tilosina injetável 13 dias após a inoculação em três doses com intervalo de 12 horas cada. Já os leitões do grupo não medicado receberam solução salina injetável com o mesmo protocolo. O grupo não medicado apresentou 23 animais com diarreia antes do dia 13 e 17 após este período. No grupo medicado, 26 animais apresentaram diarreia previamente à medicação e apenas 11 após a medicação a partir do dia 13. Os leitões medicados apresentaram extensão de lesão macroscópica, caracterizada por espessamento de mucosa intestinal, menor em comparação com o grupo não medicado (p=0,031). A imunomarcação para Lawsonia intracellularis foi menor no grupo medicado (p<0,032), mostrando redução no grau de infecção por L. intracellularis nos animais medicados. Estes resultados demonstram que a tilosina injetável (Eurofarma Laboratórios S.A.) (1mL/20kg) em três doses, a cada 12 horas, foi eficaz no tratamento da enteropatia proliferativa suína em animais experimentalmente inoculados.(AU)

Objective. To compare four different dry cow treatments (DCT) and establish their effectiveness in reducing intramammary infections (IIM). Materials and methods. DCTs included systemic tylosin (12g) alone or accompanied by cefapirine intramammary infusions and or an internal teat sealant. A total number of 278 cows at the end of lactation period were randomly assigned to one of 4 dry cow treatment groups: CESE Group (n=89), intramammary cephapirin and teat sealant. TYCESE Group (n=84), intramammary cephapirin, tylosin 12 g intramuscular and teat sealant. TYSE Group (n=86), 12 g intramuscular tylosin and teat sealant; TY Group (n=76) 12 g intramuscular tylosin only. Milk samples for culture were collected at dry-off and 1 and 2 weeks after calving. Somatic cell counts (SCC) were taken from Dairy Herd Improvement Association (DHI) tests at dry-off, and the first two test days after calving. Results. Bacteria cure rate for Gram-positive intramammary infections (IMI) for TYCESE group was 93.6%, CESE group 78.9%, TYSE group 88.2%, and TY group 78.1%. All four groups showed a decrease in the SCC upon the first and second test after calving. Conclusions. The use of systemic tylosin in combination with intramammary antibiotics increased DCT effectiveness improving the Gram-positive cure rate IMI. Furthermore, systemic tylosin alone plus teat sealant is as effective as cephapirin plus teat sealant when used as DCT.


Objetivo. Comparar 4 tratamientos para el periodo seco (TPS) y establecer su efectividad en reducir infecciones intramamarias (IIM). Materiales y métodos. Los TPS incluían tilosina sistémica (12 g) solamente o acompañada con infusiones intramamarias de cefapirina y/o un sellante interno de pezones. Un total de 278 vacas al final de la lactancia fueron asignadas al azar a 1 de 4 grupos de tratamiento. Grupo CESE (n=89), cefapirina y sellante interno de pezones. Gupo TYCESE (n=84), cephapirina intramamaria, tilosina intramuscular y sellador interno de pezones. Grupo TYSE (n=86), tilosina intramuscular y sellante interno de pezones. Grupo TY (n=76) tilosina intramuscular solamente. Muestras de leche de cada cuarto productivo fueron tomadas al momento del secado, y 1 y 2 semanas después del parto. Los recuentos individuales de células somáticas (RCS) fueron tomados del control lechero (DHI) en sus pruebas de antes del momento de secado, y las primeras dos pruebas después del parto. Resultados. Las tasa de cura bacteriológica para IIM causadas por Gram positivos en TYCESE fue de 93.6%, para CESE 78.9%, TYSE 88.2% y TY 78.1%. Todos los grupos de tratamiento mostraron una disminución en RCS a la primera y segunda prueba despues del parto en comparación con la de secado. Conclusiones. El uso de tilosina en combinación con cefapirina incremento la efectividad del TPS, mejorando las tasas de cura de IIM por Gram positivos. Además TYSE fue tan efectivo como CESE en la reducción de IIM.

RESUMEN

Se validó la técnica para la cuantificación de tilosina en polvo, para lo cual previamente se estandarizó la técnica recomendada por la United States Pharmacopeia versión 30 del 2007, con el fin de ajustarse y dar cumplimiento a la normativa internacional. La muestra se compuso del 10 por ciento de tilosina fosfato y 90 por ciento de Carrier 40-60. El microorganismo sometido a prueba fue Micrococcus luteus ATCC 9341 y se empleó el método de difusión en agar. Los resultados de las pruebas se sometieron al programa de validación microbiológica "Validation Manager". El método de difusión en agar cumplió con los valores esperados, y así quedó validada la técnica.


The quantification technique of powdered tylosin was validated. To this end, the technique recommended by the United States Pharmacopeia, 30th version, 2007 was standardized to adapt it to and to comply with the international regulations. The sample was composed of 10 percent tylosin phosphate and 90 percent Carrier 40-60. The agar diffusion was used to test the micro-organism Micrococcus luteus ATCC 9341 and all results were microbiologically validated by the program "Validation Manager". The agar diffusion method met the prospective values, thus validating the technique.

RESUMEN

Las espiroquetas intestinales del género Brachyspira ocasionan enfermedades importantes en porcinos y aves. Se ha evidenciado un problema de incremento en la presentación de cepas resistentes a los antimicrobianos utilizados normalmente para tratar las espiroquetosis intestinales en porcinos, y esto podría ser aplicable a los aislamientos de aves. Hay muypocos reportes de sensibilidad antimicrobiana in vitro de cepas de Brachyspira spp. aisladas en aves. En este estudio se evaluó la sensibilidad de doce aislamientos de Brachyspira pilosicoli obtenidos de granjas de ponedoras comerciales a los agentes antimicrobianos tiamulina, tilosina y lincomicina, y se estableció la concentración mínima inhibitoria (CMI)mediante la técnica de dilución en agar. Todas las bacterias analizadas fueron sensibles a tiamulina (CMI≤0,1 μg/ml) y lincomicina (CMI 1 μg/ml) y resistentes a tilosina (CMI 5 μg/ml).


Intestinal Spirochaetes of the genus Brachyspira cause important diseases in swine and poultry. An increasing problem in the presentation of resistant strains to the antimicrobial drugs usually used to treat the intestinal spirochaetosis in swine has been evidenced and this could be applicable to the isolations from poultry. There are very few reports of in vitro antimicrobialsusceptibility of Brachyspira spp. isolated from birds. In this study the antimicrobial susceptibility of twelve Brachyspira pilosicoli isolates obtain from commercial layers was evaluated against tiamulin, tylosin and lincomycin establishing the Minimum Inhibitory Concentration (MIC) by agar dilution technique. All bacteria analyzed were sensitive to tiamulin (MIC ≤0,1μg/ml), and lincomycin (MIC 1μg/ml) and resistant to tylosin (MIC 5μg/ml).

Con la finalidad de determinar la eficacia y eficiencia de la antibioterapia sistémica en la terapéutica de las lesiones podales agudas y sub-agudas, se desarrollo una investigación durante los meses de julio y agosto del año 2000, en dos explotaciones lecheras del estado Zulia, Venezuela, en la cual se evaluaron 29 vacas cojas determinando el grado de claudicación presente y clasificando las diferentes lesiones encontradas en los dígitos. Los animales fueron asignados en tres grupos: dos experimentales (sistémico y local) y uno control, tratando de mantener el mismo reto en cuanto a claudicaciones y lesiones en cada uno, los datos clínicos fueron registrados en fichas individuales. La metodología establecida permitió el chequeo clínico a las 0; 48 y 120 horas con la finalidad de determinar la eficacia comparativa de cada uno de los tratamientos, basado en dos variables respuesta: Grado de claudicación (GC) y Características clínicas de la Lesión (CL). Los resultados mostraron que ambos tratamientos fueron capaces de lograr una reducción en el grado de claudicación (GC) y en las características clínicas de la lesión (CL) de los animales, al compararlos con el grupo control (P < 0,05). Sin embargo, en relación a la eficiencia (costos del tratamiento) el uso del tratamiento local resultó superior al tratamiento sistémico, recomendándose su difusión y aplicación para la terapéutica de las lesiones podales.


With the goal to determine the efficacy and efficiency of the systemic antibiotherapy on the acute and sub acute hoof lesions a research was developed in two milking farms at Zulia state, Venezuela. In a period within July and August 2000, 29 cows were evaluated to determine the degree of lameness and to classify the different lesions founds at the digits. The experimental animals were group in three lots: two experimental groups (treated either intramuscular or local) and one control group. As far as it was possible the three groups were maintained with the same challenge in relation of lameness and lesions in each one. The clinical data were recorded individually. The established method allowed to monitor the animals at 0; 48: 120 h with the goal to determine the efficacy and efficiency of each treatment supported by two variable responds: grade of lameness (GC) and the clinical characteristics of lesions (CL). The results showed that both treatments were able to diminish the degree of lameness (GC) and over the clinical characteristics of lesions (CL) of experimental animals when it was compared with the control group (P < 0.05). However in relation to efficiency (treatment cost), the use of local treatment was better than the systemic treatment. Therefore its promotion and application is recommended for the treatment of hoof lesions.
