RESUMEN

Objective:To observe the effects of dentin proseeded with BMSCs or PDLCs incorporated with PDLCs sheets and calci-nated ceramic bovine bone(CBB)in the reconstruction of periodontal tissue.Methods:Root dentin slices were prepared and pre-seeded with BMSCs or PDLCs from beagle dog.After cultivated in osteoinduction medium(OST groups)orα-modified eagle's medi-um(MEM groups)for 3 weeks,the dentin slices observed by scanning electron microscope (SEM).Then the slices were wrapped successively with PDLCs sheets and CBB as new constructs(BMSCs or PDLCs/dentin/PDLCs sheets/CBB).The new constructs were transplanted into nude mice subcutaneously,8 week after transplantation,samples were havested and examined by HE staining.Re-sults:Enough cells and extracellular matrices were detected on the dentin slices by SEM in vitro.A little new immature cementum-like tissue without periodontal-like tissue on the surface of the new construcs was observed in OST groups.By contrast,in MEM groups,periodontal-like without immature cementum-like tissue formation was observed.Conclusion:Dentin proseeded with BMSCs or PDLCs incorporated with PDLCs sheets and CBB cultured by MEM can promote periodontal-like tissue regeneration.Cultured with osteooinduction medium can promote cementum-like tissue regeneration.

RESUMEN

The overall goal of our research projects is to develop effective immunotherapeutic regimens, particularly combining vaccine and gene therapy/ cell therapy strategies. For the development of clinically effective immunotherapy for brain cancers, the following issues are considered to be particularly important: 1) Induction of effective immune responses against tumors (afferent arm of the immune response), 2) Delivery of immune effector cells to the target tumor sites and maintaining the activity of the effector cells (efferent arm), 3) For specific and safe immunotherapy, specific brain tumor rejection antigens have to be identified, 4) Feasibility, safety and efficacy need to be tested in a series of clinical trials. The following presentation summarizes my research projects and demonstrates how each plan will fit in the whole schema of designing successful immunotherapeutic strategies for brain cancers. In this presentation, I would like to focus on our clinical and basic studies related to the vaccine strategies for patients with glioma, and modulation of tumor-microenvironment using bone-marrow derived stroma cells as vehicles for cytokine- gene delivery.

RESUMEN

Objective To observe the treatment effect of transplanted autogenous bone marrow stroma cells (BMS-C) absorbed in the type-Ⅰ collagen matrix in bone repair. Methods Thirty-two adult white New Zealand rabbits were randomly divided into Group Ⅰ and Group Ⅱ. The bone marrow was extracted from tubercle and greater trochanter of femur for culture. Then, the cells were amplified, implanted into type-Ⅰ collagen matrix and further cultured for two weeks. Two weeks later, the cells were implanted into the 1.5 cm radius defect. In the Group Ⅰ, management A (BMS-C+type-Ⅰ collagen) was given to the defect of one radius and management B (control) to the defect of the contralateral radius. In the Group Ⅱ, the defect of one radius was given management A and the defect of the contralateral radius management C (type-Ⅰ collagen).The rabbits were killed 8 and 12 weeks after implantation, the effect of three managements was compared. Results Twelve weeks after implantation, in the management A, all specimens had complete bony union; and in the management B, the specimens were filled with fibrous tissues; in the management C, a little scar formed. Conclusions BMS-C absorbed in the type-Ⅰ collagen can effectively repair bone defect.

RESUMEN

To observe the growth, expansion and differentiation of the cultured bone marrow stroma cell (BMSC) from Macaca Lrus, BMSC isolated from adult Macaca Lrus were cultured with the culture medium confected by ourselves and were induced with some cytokines such as LIF and bFGF. The results showed that the BMSC could proliferate and generate Nestin positive clones when they were cultured in vitro. After subculture, these cells could grow rapidly and differentiated into neuron like cells and astrocyte like cells further, which expressed GFAP or NSE antigen respectively. Therefore, these BMSC possess renewal and differentiation abilities. On the other hand, the culture method we used in this experiment is suitable for culture of BMSC in vitro. The BMSC might be used as the seed cells of the neural stem cells.
