RESUMEN

Twenty-five years have passed since the initial observation of endemic zoonotic sporotrichosis in Rio de Janeiro, Brazil. Since then, this disease has spread throughout South America. Accompanying the emergence of this mycosis, some progress has been made, including the expansion of a research network in this field and higher visibility of sporotrichosis within government authorities and funding agencies. However, there are still some challenges to curbing the expansion of this disease in the coming years. These include the development of rapid and accurate diagnostic tests, new antifungal drugs, particularly for the treatment of extracutaneous manifestations of sporotrichosis, and more comprehensive care for cats with sporotrichosis. Including these actions in the sporotrichosis research agenda is required so as to change the development of this disease in the years to come.

A esporotricose é uma zoonose micótica emergente e subcutânea, que afeta a pele, o sistema linfático e outros órgãos de humanos e animais. Assim como outras doenças infecciosas fúngicas, se torna ainda mais grave quando acomete pacientes imunossuprimidos. Essa infecção possui distribuição global e é endêmica em algumas regiões do Brasil e de outros países tropicais e subtropicais, sendo um problema de saúde pública importante em nosso país. A doença é causada por um complexo de pelo menos quatro espécies patogênicas, incluindo o Sporothrix brasiliensis (S. brasiliensis). A resposta imunológica contra estas espécies ainda não é completamente elucidada, mas estruturas como as vesículas extracelulares (VEs) poderiam transportar componentes importantes que podem contribuir na modulação e no controle desta importante infecção. Assim, o objetivo deste trabalho, é analisar a participação das VEs de células dendríticas (DCs) naive e VEs de DCs previamente primadas com leveduras de S. brasiliensis e primadas com VEs do fungo, na resposta imune contra a esporotricose experimental em modelos murinos. Para isso, as DCs obtidas da medula óssea de camundongos, foram cultivadas com leveduras de S. brasiliensis ou com VEs do fungo e posteriormente, VEs totais das DCs foram purificadas a partir de ultracentrifugação e analisadas quanto a sua participação na modulação da resposta imunológica. Essas VEs foram utilizadas em protocolo profilático em modelos murinos, previamente a infecção subcutânea experimental. Foi observado o diâmetro médio das lesões no decorrer de 35 dias de infecção e a carga fúngica da lesão na pele. Os resultados obtidos mostram que as VEs de DCs naive, e VEs de DCs previamente cultivadas com leveduras do fungo ou com VEs fúngicas, são capazes de modular a carga fúngica. Os grupos que receberam VEs de DCs de forma profilática, de modo geral apresentaram diminuição significativa da carga fúngica em relação ao grupo controle. Na análise comparativa apenas dos grupos que receberam a profilaxia, observa-se que o uso de VEs de DCs naive, resultam em uma carga fúngica maior que o uso de VEs de DCs previamente ativadas, e quando as DCs são ativadas com levedura, essa carga fúngica é a menor. Quando analisamos o perfil de citocinas na pele de camundongos tratados com as VEs previamente a infecção, observamos aumento de IFN-γ, TNF-α, IL-17 e IL-10 principalmente nos animais previamente tratados com VEs de DCs que foram ativadas com leveduras. Em relação às citocinas produzidas, podemos sugerir até o momento, uma resposta imunológica mista, mas que de alguma maneira, ainda não esclarecida, devem contribuir para melhor controle do processo infeccioso in vivo. Em relação a linfoproliferação, observa-se principalmente um aumento de linfócitos T CD4+ quando acrescentamos VEs de DCs que não foram previamente ativadas, mostrando uma ação de uma resposta mais inespecífica. Vale ressaltar que todos os protocolos profiláticos foram capazes de modular e minimizar o crescimento fúngico, quando comparados ao controle, ou seja, as VEs contribuíram com o controle da infecção e agiram a favor do hospedeiro, demonstrando um caráter protetivo

This infection has a global distribution and is endemic in some regions of Brazil and other tropicals and subtropicals countries, being an important public health problem in our country. The disease is caused by a complex of at least four pathogenic species, including Sporothrix brasiliensis (S. brasiliensis). The immunological response against these species has not yet been completely elucidated, but structures such as extracellular vesicles (EVs) could carry important components that can contribute to the modulation and control of this important infection. Thus, the objective of this work is to analyze the participation of EVs from naïve dendritic cells (DCs) and EVs from DCs previously primed with S. brasiliensis yeast and primed with EVs from the fungus, in the immune response against experimental sporotrichosis in murine models. For this, DCs obtained from the bone marrow of mice were cultivated with S. brasiliensis yeast or EVs from the fungus, and subsequently, total EVs from the DCs were purified through ultracentrifugation and analyzed for their participation in modulating the immune response. These EVs were used in a prophylactic protocol in murine models, before experimental subcutaneous infection. The average diameter of the lesions over 35 days of infection and the fungal load of the lesion on the skin were observed. The results obtained show that EVs from naïve DCs, and EVs from DCs previously cultured with yeast or fungal EVs, are capable of modulating the fungal load. The groups that received EVs from DCs prophylactically generally showed a significant decrease in fungal load compared to the control group. In the comparative analysis of only the groups that received prophylaxis, it was observed that the use of EVs from naïve DCs results in a higher fungal load than the use of EVs from previously activated DCs, and when the DCs are activated with yeast, this load fungal is smaller. When we analyzed the cytokine profile in the skin of mice treated with EVs before infection, we observed an increase in IFN-γ, TNF-α, IL-17, and IL-10, mainly in animals previously treated with EVs from DCs that were activated with yeast. About the cytokines produced, we can so far suggest a mixed immunological response, but in some way, not yet clear, they should contribute to better control of the infectious process in vivo. About lymphoproliferation, an increase in CD4+ T lymphocytes is mainly observed when we add EVs from DCs that were not previously activated, showing a more nonspecific response. It is worth highlighting that all prophylactic protocols were able to modulate and minimize fungal growth, when compared to the control, that is, EVs contributed to the control of the infection and acted in favor of the host, demonstrating a protective character

Se describe el aislamiento de Sporothrix brasiliensis desde una biopsia de piel de un caso humano de esporotricosis linfocutánea, en la región de Valparaíso, Chile. Esta especie es la más virulenta del género y es de transmisión zoonótica, desde los gatos a los humanos. Hasta ahora, solo se había publicado un brote por esta especie en gatos domésticos y asilvestrados en el extremo sur de Chile, por lo que este aislamiento, en una mujer residente de un sector densamente poblado de la Región de Valparaíso, constituye una preocupación por su eventual diseminación hacia otros gatos y la población general.


The isolation of Sporothrix brasiliensis from a skin biopsy of a human case of lymphocutaneous sporotrichosis in the region of Valparaíso, Chile is described. This species is the most virulent of the genus and is zoonotic in transmission from cats to humans. Until now, only one outbreak of this species has been published in domestic and feral cats in the extreme south of Chile, so this isolation in a woman residing in a densely populated sector of the fifth region is a concern for its eventual spread to other cats and the general population.

ABSTRACT Mycetoma is a neglected tropical disease (NTD) declared by the World Health Organization (WHO) in 2016. It is characterized by the progressive growth of nodules and granulomatous lesions on the legs, arms, and trunk. It is potentially disfiguring and causes disability or amputations in working-age people from marginalized areas. The causative agents can be fungi (eumycetoma) or actinobacteria (actinomycetoma), the latter being the most common in America and Asia. Nocardia brasiliensis is the most important causal agent of actinomycetoma in the Americas. Taxonomic problems have been reported when identifying this species, so this study aimed to detect the 16S rRNA gene variations in N. brasiliensis strains using an in silico enzymatic restriction technique. The study included strains from clinical cases of actinomycetoma in Mexico, isolated from humans and previously identified as N. brasiliensis by traditional methods. The strains were characterized microscopically and macroscopically, then subjected to DNA extraction and amplification of the 16S rRNA gene by PCR. The amplification products were sequenced, and consensus sequences were constructed and used for genetic identification and in silico restriction enzyme analysis with the New England BioLabs® NEBcutter program. All study strains were molecularly identified as N. brasiliensis; however, in silico restriction analysis detected a diversity in the restriction patterns that were finally grouped and subclassified into 7 ribotypes. This finding confirms the existence of subgroups within N. brasiliensis. The results support the need to consider N. brasiliensis as a complex species.

ABSTRACT Acid-fast bacteria can be implicated in skin and soft tissue infections. Diagnostic identification can be challenging or not feasible by routine laboratory techniques, especially if there is no access to the Matrix Assisted Laser Desorption Ionization Time of Flight Mass Spectrometry (MALDI-TOF MS) technology. Here, we present two cases of skin and soft tissue infections caused by two different acid-fast bacteria, Nocardia brasiliensis and Mycobacterium marinum. They both grew on Löwenstein-Jensen medium, Sabouraud agar medium and blood agar medium. Both bacteria appeared acid-fast by Ziehl-Neelsen stain and Gram-positive by Gram stain. The identification was performed by MALDI-TOF MS and gene analysis. N. brasiliensis and nontuberculous mycobacterium M. marinum represent rare pathogens that cause severe skin and soft tissue infections. Failure to identify the causative agent and subsequent inappropriate or inadequate treatment may lead to severe complications or even disseminated disease, especially in immunocompromised individuals.

Sporothrix brasiliensisis one of themost virulent zoonosis which affects animals and humans. This fungus is responsible for subcutaneous infection and its contamination is possible through trauma to the skin. Sporotrichosis is highly prevalent in feline. And Rio de Janeiro appears to have the highest occurrence of cases. Objectives: This study aims to evaluate the in vitroefficacy of Sporothrix brasiliensisbiotherapic, with and without an association to allopathic medicine commonly used in the treatment.Methodology: Conidiumcells of Sporothrix brasiliensiswill be cultured in Potato dextrose agar (PDA) for 5 to 7 days and yeast cells in Brain heart infusion (BHI) for 3 to 5 days. After incubation, the cells will be scraped with a drigalski handle and filtered using cells strainer into a tube and centrifuge for 5 minutes at 3000 RPM. The cells will be resuspended with Phosphate buffered saline (PBS), centrifuge again,and finally resuspended in PBS. After preparing the inocule, the microplates will be prepared. There will be 5groups in vitro. The first one will be the control group, only fungi. The second will be the treatment of fungi with homeopathic medicine (Sporothrix brasiliensis30DH). The third group will be the homeopathic medicine in association with itraconazole. The fourth will be the treatment with itraconazole only. And the last group will be the fungi with dynamized distilled water 30DH. Sporothrix brasiliensis30DHwill be prepared according to Brazilian Homeopathic Pharmacopeia. Results and discussions: The experiments are still in progress and the results will be analyzed through Analysis of variance (ANOVA) to determine statistically significant differences. Previous articles based on biotherapic treatments demonstrated successful results, so our research group is conducting these experiments to evaluate the effect in this model. Conclusion: Experiments will be made to verify the efficacy of biotherapic on sporotrichosis treatments.

The productive potential of the rubber tree (Hevea brasiliensis) is dependent on its genetic composition, in addition to edaphoclimatic factors and management practices. However, as soil properties are not homogenous, knowing the spatial variability of soil attributes would be important to increase productivity and reduce production costs. In this context, the objective of this study was to determine the spatial variability of chemical attributes of the soil and its influence on the nutritional status and growth of rubber tree clones. Clones FX 3864, FDR 5788, CDC 312, and RRIM 600 were planted at Jaturnaíba Farm, in the municipality of Silva Jardim, Rio de Janeiro State, Brazil. The sampling sites were distributed at a spacing of 20 × 20 m on the northern and southern sides of the relief. The chemical attributes of the soil (pH, Ca2+, Mg2+, K+, P, Al3+, H+Al, sum of bases, cation exchange capacity, and base saturation) were evaluated at a depth of 0­20 cm in the different clone plantations. Additionally, the N, P, K, Ca, and Mg content as well as trunk circumference and total plant height, were also evaluated. Geostatistics was used to determine the spatial variability of the soil and clone attributes, while Ordinary Kriging was used to draw variability maps of the variables. A difference in the distribution of the variables, which was dependent on the slope of the relief, was detected through the maps. The southern side presented better conditions as some degradation was observed on the northern side. Certain soil characteristics influenced the distribution of the attributes of the planted clones; for example, the low concentration of Ca2+ in the soil caused Ca deficiency in the FX clone on the southern slope, indicating that liming did not supply enough nutrients for this clone. Our results showed that the variability in soil attributes influenced the nutritional status and growth of the rubber tree clones, indicating that variability maps can guide the planting and management of the rubber tree, providing more efficient management.

Abstract Sporothrix spp. are the major dimorphic fungus associated with a type of subcutaneous mycosis, sporotrichosis. The limitation of antifungal availability and the past reports of in vitro resistance of Sporothrix spp. clinical isolates makes it important to search for new compounds with antifungal activities. In this study, we therefore evaluate the in vitro activities of complexes coordinated with Co(II) and cobalt chloride hexahydrate against clinical isolates of Sporothrix spp. Broth microdilution test was performed as per M38-A2 from CLSI (2008) in duplicate for 31 clinical isolates of Sporothrix spp. (27 S. brasiliensis e 04 S. schenckii stricto sensu). The antifungal activities of the complexes coordinated with Co(II) and cobalt chloride hexahydrate were detected at a concentration range of 32-128 µg/mL for all isolates. None of the compounds demonstrated any cytotoxicity (to macrophage cells) at the concentration of 200 µg/mL. The activity against Sporothrix spp. recorded in this study instigate the continuity of experimental studies with Co(II) to search for the mechanisms of antifungal action as well as to evaluate its interaction with the commercial antifungal drugs.

BACKGROUND Sporothrix brasiliensis is the causative agent of zoonotic cases of sporotrichosis in Brazil and is associated with atypical and severe presentations in cats, dogs, and humans. Sporotrichosis treatment is usually time- and cost-consuming, sometimes with poor response and host toxicity. Schinus terebinthifolius has proven efficacy against bacteria and fungi of clinical interest. OBJECTIVE To determine the in vitro activity of S. terebinthifolius against S. brasiliensis. METHODS Five S. brasiliensis isolates and three reference strains were subjected to a hydroethanol extract derived from the leaves of S. terebinthifolius and its fractions. The minimal inhibitory concentration (MIC) was determined using the broth microdilution method according to the M38-A2 CLSI guidelines. Also, the fungicidal/fungistatic activity of the extract and fractions was studied. FINDINGS The crude extract of S. terebinthifolius inhibited the growth of S. brasiliensis (MIC: 0.5-1.0 µg/mL), while the partitioned extracts dichloromethane, ethyl acetate, and butanol demonstrated growth inhibition at 8 µg/mL due to a fungistatic activity. MAIN CONCLUSIONS Due to its in vitro efficacy against S. brasiliensis and its known pharmacological safety, S. terebinthifolius is a candidate to be tested using in vivo models of sporotrichosis.

Abstract This study evaluated the bone repair in surgical defects of rats treated with hyaluronic acid (HA) associated or not with Hevea brasiliensis fraction protein (F-1). Bone defect were created in 15 albino Wistar rats divided into 3 groups (n=5): Control group (1) - blood clot; HA group (2) - 0.5% hyaluronic acid; HAF1 group (3) - 0.1% F-1 protein fraction dissolved in 0.5% hyaluronic acid. After 4 weeks, the animals were euthanized and the bone repair was evaluated through histomorphometric analysis, zymography and immunohistochemistry. The neoformed bone area did not show a significant difference (p = 0.757), but there was a tendency for bone trabeculation to increase in the groups HA and HAF1. For immunohistochemically analysis, there was a difference in vascular endothelial growth factor (VEGF) labeling (p = 0.023), being higher in the groups HA and HAF1 than the control group. No significant difference in bone sialoprotein (BSP) (p = 0.681), osteocalcin (p = 0.954), however, significant difference in platelet endothelial cell adhesion molecule-1 (CD-31) (p = 0.040), with HAF1 group being significantly lower than the control. For zymographic analysis, there was no significant difference for metalloproteinase-2 (MMP-2) (p = 0.068), but there was a tendency to increase MMP-2 in the HA group. Resumo Este estudo avaliou o reparo ósseo em defeitos cirúrgicos de ratos tratados com ácido hialurônico (AH) associado ou não à fração proteica de Hevea brasiliensis (F-1). Foram criados defeitos ósseos em 15 ratos albinos Wistar divididos em 3 grupos (n = 5): Grupo controle (1) - coágulo sanguíneo; Grupo HA (2) - ácido hialurônico 0,5%; Grupo HAF1 (3) - fração proteica F-1 0,1% dissolvida em ácido hialurônico a 0,5%. Após 4 semanas, os animais foram submetidos à eutanásia e o reparo ósseo avaliado por meio de análise histomorfométrica, zimografia e imunohistoquímica. A área óssea neoformada não apresentou diferença significativa (p = 0,757), mas houve tendência de aumento da trabeculação óssea nos grupos HA e HAF1. Para a análise imunoistoquímica, houve diferença na marcação do fator de crescimento endotelial vascular (VEGF) (p = 0,023), sendo maior nos grupos HA e HAF1 do que no grupo controle. Nenhuma diferença significativa na sialoproteína óssea (BSP) (p = 0,681), osteocalcina (p = 0,954), no entanto, diferenças significativas foram encontradas para a molécula de adesão de células endoteliais plaquetárias-1 (CD-31) (p = 0,040), com o grupo HAF1 sendo significativamente inferior ao controle. Para a análise zimográfica, não houve diferença significativa para metaloproteinase-2 (MMP-2) (p = 0,068), mas houve tendência de aumento da MMP-2 no grupo HA. Apesar da influência sobre os fatores angiogênicos e da aparente tendência de maior trabeculação nos grupos HA e HAF1, não houve diferença significativa na área de tecido ósseo neoformado no período analisado.

Neuroparacoccidiodimycosis (NPDM) is an uncommon granulomatous disease, which more frequently affects immunocompromised male patients over 30 years of age in the course of chronic lung disease. Paracoccidioides brasiliensis (PB) is an endemic fungus in Brazil, and grows as thick-walled yeast (with round to oval bodies) measuring 10 µm to 60 µm in diameter. Neuroparacoccidiodimycosi may develop many years after transmission and/or primary lung involvement. The authors describe a case of NPDM affecting a male patient, 52 years of age, farmer, heavy smoker, with clinical complaint of headache, asthenia, seizures, and prostration in the previous nine months. Upon physical examination, the patient presented regular general condition, without other relevant physical alterations. Computed tomography (CT) showed multiple bilateral pulmonary nodules associated to enlargement of the mediastinal lymph node. Magnetic resonance imaging (MRI) and CTscans of the central nervous system showed six heterogeneous nodular lesions compromising the frontal and parietal lobes, the largest one measuring 3.8 3.2 3.2 cm. The hypothesis of a neoplastic process compromising the lung and brain was considered. A biopsy of the mediastinal lymph node showed epithelioid granulomas, which exhibited round, thin-walled fungal structures in Grocott silver stain. The stereotactic biopsy of the frontal lesion was constituted by necrotic tissue admixed with some round to oval, thin-walled fungi measuring 10 µm to 60 µm, compatible with PB (identified on Grocott silver stain/confirmed in culture). The diagnosis of NPDM was then established. The employed therapeutic regimen was intravenous amphotericin B, itraconazole, and sulfamethoxazole-trimetropin. After ninety days of clinical follow-up, no episodes of seizures/neurological deficits were identified, and a marked decrease in the number and size of the lung and brain lesions were found.

Anesthesia reduces the handling process duration and prevent fish injuries. The anesthetic effect and ideal concentrations of eugenol and tricaine methanesulfonate (MS-222) were tested for pearl cichlid (Geophagus brasiliensis) juveniles with an average weight of 4.4 g in water at 24ºC. The criterion for determining the optimal dose considered an induction time of one minute. Experiment 1 tested the concentrations of 25, 75, 150 and 300 mg L-1 of eugenol. The best results were obtained at doses of 150 and 300 mg L-1. Experiment 2 aimed to establish a more accurate result by testing the concentrations of 180, 210, 240 and 270 mg L-1, and led to an estimation of 217 mg L-1 of eugenol to induce anesthesia in one minute. Experiment 3 evaluated 200, 300, 400, 500 and 600 mg L-1 of tricaine, of which the concentration of 294 mg L-1 was estimated to induce anesthesia in one minute. No significant differences were observed for recovery times when using either of the anesthetics. No mortality was observed within 24 hours after the experiments for any concentration of the anesthetics. The present study recommends 217 mg L-1 of eugenol or 394 mg L-1 of tricaine for anesthesia of the pearl cichlid.

Triatoma brasiliensis is the main vector of the parasite in Brazil, and dogs are an important reservoir of the parasite. The aim of this study was to evaluate the insecticidal efect of furalaner (Bravecto®) on T. brasiliensis after a blood meal in treated dogs. Methods: Healthy mongrel dogs (n=8) were recruited from the Zoonoses Control Center (ZCC) in the city of Natal, Rio Grande do Norte, Brazil, and randomized into two groups, a furalaner (Bravecto®)-treated group (n=4) and a control group (n=4). Colony-reared third-, fourth- and ffth-instar nymphs of T. brasiliensis nymphs (n=10) were allowed to feed on dogs from both groups for 30­40 min, once monthly, for up to 12 months. Bug mortality was observed up to 5 days after each blood meal. Results: Mortality in triatomines which had a blood meal on furalaner (Bravecto®)-treated dogs was 100% for up to 7 months after treatment, with mortality decreasing to 66.4% after 8 months, 57% after 9 months, 35% after 10 months, 10% after 11 months and 0% after 12 months. The mortality of triatomines that fed on non-treated control dogs was always ≤ 2.5%. Conclusions: Our results suggest that furalaner (Bravecto®) treatment of dogs induces long-term mortality of T. brasiliensis after the blood meal. This is a potential approach to be used to control vector transmission of T. cruzi, the etiological agent of Chagas disease, especially in endemic areas.

The aim of this study was to analyze the effect of essential oils on the control of soft rot of kale. Clove essential oil at 0.25%, lemongrass and palmarosa essential oils at 0.5%, melaleuca and orange essential oils at 0.75%, bergamot, rosemary, sage and ginger essential oils at 1% were evaluated for the in vitro inhibition of Pectobacterium carotovorum subsp. brasiliensis (Pcb) and control of soft rot of kale, sprayed 72 hours before or seven hours after inoculation. Clove, citronella, bergamot, rosemary, palmarosa, sage, melaleuca, and lemongrass oils completely inhibited the growth of Pcb. Lemongrass oil (0.5%) caused 0% of disease incidence (INC), providing 100% of disease control in both periods of inoculation. Clove oil (0.25%) showed a lower INC (25%) when applied after inoculation, providing a control percentage of 71.42%. The lemongrass and clove essential oils were analyzed by GC/FID (Gas Chromatography ­ Flame Ionization Detector) and by GC/MS (Gas Chromatography /Mass Spectrometer). The major components were eugenol (91,9%) for clove oil and citral, isometric mixture of neral (34,1%) and geranial (42,9%) for lemongrass oil. The Minimum inhibitory concentration (MIC) of lemongrass, clove oils and their major components (citral and eugenol, respectively) was determined by using a broth macrodilution technique, as well as they were evaluated at different concentrations on the control of soft rot of kale, sprayed according descriptions above. The MIC was 0.03125% for citral, and 0.0625 and 0.125% for lemongrass and clove oils, respectively. Eugenol didn't show MIC. Lemongrass oil at 0.125% (post-inoculation) and citral at 0.125% (pre and post-inoculation) provided the highest percentages of disease control (33.33, 50, and 100%, respectively). Clove oil at 0.125% (post-inoculation) showed better effectiveness than eugenol (0.25%), providing a percentage of disease control of 16.67%. Lemongrass and clove essential oils were the most effective in control of soft rot of kale, suggesting that these oils have a potential to be used as antibacterial agents.


O objetivo do estudo foi avaliar o efeito de óleos essenciais no controle da podridão mole em couve. Os óleos essenciais de cravo a 0,25%, capim-limão e palmarosa a 0,5%, citronela, melaleuca e laranja a 0,75%, bergamota, alecrim, sálvia e gengibre a 1% foram avaliados na inibição in vitro de Pectobacterium carotovorum subsp. brasiliensis (Pcb) e controle da podridão mole em couve, pulverizados 72 horas antes ou sete horas após a inoculação. Os óleos essenciais de cravo, citronela, bergamota, alecrim, palmarosa, sálvia, melaleuca e capim-limão inibiram completamente o crescimento de Pcb. O óleo de capimlimão (0,5%) promoveu 0% de incidência (INC) da doença (percentual de controle de 100%), em ambos os períodos de inoculação. O óleo de cravo (0,25%) proporcionou menor INC (25%) quando aplicado após inoculação (percentual de controle de 71,42%). Os óleos essenciais de capim-limão e cravo foram analisados por GC/FID (cromatografia gasosa/detector por ionização de chama) e por GC/MS (cromatografia gasosa/ espectometria de massas). Os componentes majoritários foram eugenol (91,9%) no óleo de cravo e citral (neral34,1% e geranial- 42,9%) no óleo de capim-limão. A concentração inibitória mínima (CIM) dos óleos essenciais de capim-limão e cravo e de seus componentes majoritários (citral e eugenol, respectivamente) foi determinada por meio da técnica de macrodiluição em caldo, bem como foram avaliados, em diferentes concentrações, no controle da podridão mole em couve, pulverizados conforme descrito acima. A concentração inibitória mínima (CIM) foi de 0,03125% para o citral, e de 0,0625 e 0,125% para os óleos de capim-limão e cravo, respectivamente. O eugenol não apresentou CIM. O óleo de capim-limão a 0,125% (pós-inoculação) e o citral (0,125%), em ambos os períodos de inoculação, proporcionaram os maiores percentuais de controle (33,33; 50 e 100%, respectivamente). O óleo de cravo a 0,125% (pós-inoculação) mostrou maior eficiência que o eugenol (0,25%), promovendo um percentual de controle de 16,67%. Os óleos essenciais de capim-limão e cravo destacaram-se na eficiência de controle da podridão mole em couve, sugerindo que esses óleos têm potencial para serem utilizados como agentes antibacterianos.

Background and Objectives: Paracoccidioidomycosis (PCM) is a systemic disease caused by the dimorphic fungus Paracoccidioides brasiliensis found in the tropical and subtropical regions of Latin America. This study aimed to perform a retrospective analysis of PCM cases from the northern region of Rio Grande do Sul, Brazil. Methods: A total of 200 records of PCM cases diagnosed at the local reference pathology laboratory from 1995 to 2015 were analyzed Results: Of the patients, 185 were male and 15 female. Patients ranged in age from 31 to 80 years, the largest pro portion ( being aged between 51 and 60 years and living or working in the countryside. Clinical samples were mostly obtained from the oral cavity, followed by the oropharynx, lungs, brain, skin, and prostate. Conclusion: PCM is endemic in the south o f Brazil, as the local economy is largely based on agricultural activities, favoring the contact of the population with P. brasiliensis . Due to the great similarity between PCM symptoms and other respiratory diseases, the differential diagnosis is essentia l for the correct treatment of the disease and to avoid its progression.(AU)

La Esporotricosis es una micosis profunda causada por el hongo dimorfo Sporothrix schenckii, El advenimiento de técnicas moleculares como la reacción en cadena de la polimerasa ha hecho posible identificar varias especies dentro del complejo Sporothrix spp como S. brasiliensis, S. schenckii sensu stricto, S. globosa, S mexicana y S. albicans. Las diferencias son moleculares, pero también geográficas, macroscópicas, en asimilación de azúcares y crecimiento de colonias a 37º; determinando formas clínicas, evolución y respuestas terapéuticas diferentes. Comunicamos 2 casos de Esporotricosis diagnosticados en el Hospital Nacional de Paraguay, cuyos estudios de PCR del gen de la calmodulina hechos en el extranjero, determinaron ser producidos por S. brasiliensis y S. globosa respectivamente. El objetivo de la comunicación es resaltar la importancia de las técnicas moleculares para el diagnóstico preciso de la especie de Sporothrix spp, considerando los factores de riesgo asociados a la caracterización epidemiológica y a las diferencias clínico-evolutivas de los casos de esporotricosis.


Sporotrichosis is a deep mycosis caused by the dimorphic fungus Sporothrix schenckii. The advent of molecular techniques as the polymerase chain reaction has made it possible to identify several species within the Sporothrix spp complex such as S. brasiliensis, S. schenckii sensu stricto, S. globosa, S mexicana and S. albicans. The differences are molecular but also geographic, macroscopic, in sugar assimilation and colony growth at 37º, determining different clinical forms, evolution and therapeutic responses. We report 2 cases of sporotrichosis diagnosed in the National Hospital of Paraguay, whose studies of PCR of the calmodulin gene carried out abroad, determined to be produced by S. brasiliensis and S. globosa respectively. The objective of the communication is to highlight the importance of molecular techniques for the precise diagnosis of the species of Sporothrix spp, considering the risk factors associated with the epidemiological characterization and the clinical-evolutionary differences of the cases of sporotrichosis.

The rubber tree (Hevea brasiliensis Müell. Arg.) is a species of significant economic interest in the natural rubber industry in Brazil and the world. This species presents recalcitrance to rooting, and its cuttings are difficult to propagate. This study aimed to evaluate the effect of the pre-conditioning of rubber tree mini-cuttings with zinc on the improvement of the adventitious rooting of rootstocks. Mini-cuttings were standardized with 45 mm length and submitted to preconditioning by immersion of the mini-cutting base in solutions containing 0.00; 0.04; 0.08; 0.16; 0.32 and 0.64 mg L-1 of Zn, for 24 hours. The experimental design was randomized blocks with six treatments and four replicates of 10 mini-cuttings. The rubber tree mini-cuttings were placed in a fitotron-type growth chamber, at 25 °C, with 12-hour photoperiod, 5,000 K intensity, and 95% of relative air humidity, for 60 days. The survival rate, number of buds, percentage of mini-cuttings that had leaf abscission, the percentage of mini-cuttings with callogenesis in the root meristem, the percentage of rooted mini-cuttings, the number of primary roots and root length were evaluated. The highest values of survival rate, the number of buds, the number of primary roots, the percentage of mini-cuttings with callogenesis in the root meristem, the percentage of rooted mini-cuttings and root length were observed with 0.16 to 0.26 Mg L-1 of Zn. The use of zinc in the mini-cuttings of rubber tree reduces linearly the percentage of mini-cuttings that had leaf abscission and the formation of callogenesis in the root meristem.


A seringueira (Hevea brasiliensis Müell. Arg.) é uma espécie de importância econômica para a indústria da borracha natural do Brasil e do mundo. Esta espécie apresenta recalcitrância ao enraizamento e suas estacas são difíceis de se propagar. Este trabalho objetivou avaliar o efeito do pré-condicionamento de miniestacas de seringueira com zinco na melhoria do enraizamento adventício de porta-enxertos. As miniestacas foram padronizadas com 45 mm de comprimento e submetidas ao pré-condicionamento por imersão da miniestaca em soluções contendo 0.00; 0,04; 0,08; 0,16; 0,32 e 0,64 mg L-1 de Zn, por 24 horas. O delineamento experimental foi em blocos ao acaso, com seis tratamentos e quatro repetições de 10 miniestacas. As miniestacas de seringueira foram colocadas em câmara de crescimento tipo fitotron, a 25 °C, com fotoperíodo de 12 horas, intensidade de 5.000 K e umidade relativa do ar de 95% por 60 dias. Foram avaliadas a taxa de sobrevivência, o número de gemas, a porcentagem de miniestacas que apresentaram abscisão foliar, o percentual de miniestacas com calogênese no meristema radicular, o percentual de miniestacas enraizadas, o número de raízes primárias e o comprimento das raízes. Os maiores valores de taxa de sobrevivência, o número de gemas, o número de raízes primárias, a porcentagem de miniestacas com calogênese no meristema radicular, o percentual de miniestacas enraizadas e o comprimento radicular foram verificados com 0,16 a 0,26 Mg L-1 de Zn. O uso de zinco nas miniestacas de seringueira reduz linearmente a porcentagem de miniestacas que tiveram abscisão foliar e a formação de calogênese no meristema radicular

Objetivo: Devido à atual resistência bacteriana aos antimicrobianos e à necessidade de buscar novas substâncias com essa atividade, principalmente em produtos naturais, esse trabalho objetivou avaliar e comparar o potencial antibacteriano de extratos de folhas da planta Eugenia brasiliensis, ainda pouco estudada, obtidas por coletas sazonais. Métodos: As coletas foram realizadas trimestralmente no Jardim Botânico do RJ (JBRJ) e os extratos etanólicos preparados com o pó das folhas secas. Foram utilizadas cepas de referência ATCC (American Type Culture Collection) Gram positivas de Staphylococcus aureus: 25.923 (Beta Lactamase -) e 29.213 (Beta Lactamase +), e duas Gram negativas, Escherichia coli (28.922) e Pseudomonas aeruginosa (27.853). A técnica de escolha foi a microdiluição em placa de 96 poços, pois permite avaliar a concentração inibitória mínima (CIM) e posteriormente a concentração bactericida mínima (CBM). Resultados: A melhor atividade antibacteriana (1 mg/mL) foi identificada nos extratos obtidos nos meses mais quentes (menores CIM para bactérias Gram positivas). A CBM foi compatível com a CIM na maioria das estações/cepas bacterianas, indicando possível atividade bactericida dos extratos. Conclusão: A pesquisa demonstrou uma atividade antibacteriana promissora para Gram positivos e existência de variação sazonal, sugerindo um bom potencial para o uso de E. brasiliensis como antimicrobiano. Os resultados também indicam que, no futuro, outros estudos devem ser realizados, como a prospecção química destes extratos.


Objective: Due to the current bacterial resistance to antimicrobials and the need to search for new substances with this activity, mainly in natural products, this work aimed to evaluate and compare the antibacterial potential of Eugenia brasiliensis leaf extracts, still little studied, obtained by seasonal collections. Methods: Leaf collections were made quarterly at the Jardim Botânico do RJ (JBRJ) and ethanol extracts prepared with dry leaf powder. Reference strains ATCC (American Type Culture Collection) Gram positive from Staphylococcus aureus were used: 25923 (Beta Lactamase -) and 29213 (Beta Lactamase +), and two Gram negative strains, Escherichia coli (28.922) and Pseudomonas aeruginosa (27.853). The technique of choice was microdilution in a 96-well plate, as it allows the assessment of the minimum inhibitory concentration (MIC) and subsequently the minimum bactericidal concentration (CBM). Results: The best antibacterial activity (1 mg/mL) was identified in the extracts obtained in the warmer months (lower MICs for Gram positive bacteria). CBM was compatible with MIC in most seasons/ bacterial strains, indicating possible bactericidal activity of the extracts. Conclusion: The research demonstrated a promising antibacterial activity for Gram positive and seasonal variation, suggesting a good potential for the use of E.brasiliensis as an antimicrobial. The results also indicate that in the future, other studies should be carried out, such as the chemical prospecting of these extracts.
