RESUMEN

The present study aimed to investigate the effect of Xianglian Pills(XLP) on lipid metabolism in obese mice and explore the underlying mechanism based on network pharmacology and intestinal flora. Firstly, network pharmacology was used to predict the possible effect of XLP on obesity. Secondly, an obese mouse model induced by a high-fat diet was established to observe changes in mouse body weight, adiposity index, liver and adipose tissue pathology. Lipid profiles, liver and kidney function markers, insulin content, and the expression of recombinant uncoupling protein 1(UCP-1) and PR structural domain protein 16(PRDM16) were measured. The 16S rRNA gene sequencing technology was used to analyze the changes in the intestinal flora. Kyoto Encyclopedia of Genes and Genomes(KEGG) pathway enrichment analysis showed that XLP mainly played a role in improving obesity by regulating lipolysis, type 2 diabetes mellitus, and insulin resistance. The results of animal experiments showed that XLP significantly reduced body weight, adiposity, blood lipid levels, and serum insulin levels in obese mice, while enhancing the expression of UCP-1 and PRDM16 in adipose tissue without causing damage to the liver or kidneys. The 16S rRNA gene sequencing results showed that XLP decreased the Firmicutes/Bacteroidetes(F/B) ratio at the phylum level, increased the relative abundance of Akkermansia and Bacteroides at the family and genus levels, and reduced the abundance of Allobaculum. Therefore, XLP can effectively improve lipid metabolism disorders in high-fat diet-induced obese mice, and the mechanism is related to the improvement of brown adipose function, the browning of white fat, the accelerated lipid metabolism, and the improvement of intestinal flora. However, its effect on promoting the conversion of white adipose to brown adipose still needs to be further studied.

Se describe el caso clínico de una paciente de 32 años de edad, con antecedentes de esquizofrenia paranoide, atendida en la consulta de Ortopedia y Traumatología del Hospital General Docente Dr. Juan Bruno Zayas Alfonso de Santiago de Cuba, por presentar aumento de volumen en la cara interna del muslo izquierdo que le dificultaba la marcha. Al examen físico se observó un tumor de aproximadamente 20 cm, no doloroso a la palpación, así como blando y pastoso, con límites imprecisos y adherido a planos profundos. Según los estudios imagenológicos efectuados se trataba de tejido graso, por lo que no existía correlación con los elementos aportados al examen físico. Teniendo en cuenta lo anterior se discutió el caso con los imagenólogos y se decidió realizar la exéresis del tumor. Se tomó muestra para biopsia cuyo resultado informó la presencia de un hibernoma. La fémina evolucionó satisfactoriamente y se reincorporó a sus actividades habituales a los 2 meses.


The case report of a 32 years patient with history of paranoid schizophrenia is described. She was assisted in the Orthopedics and Traumatology Service of Dr. Juan Bruno Zayas Alfonso Teaching General Hospital in Santiago de Cuba, due to an increase of volume in the internal face of the left thigh that difficulted her from walking. A tumor of approximately 20 cm was observed with the physical exam, that was not painful to the palpation, but soft and doughy, with imprecise limits and adhered to deep planes. According to the imaging studies it was an adipose tissue, reason why there was no correlation with the elements obtained with the physical exam. Taking into account the above-mentioned the case was discussed with the imaging specialists and the decision was removing the tumor. A sample for biopsy was taken whose result informed the presence of a lipoma. The woman had a favorable clinical course and she returned to her usual activities 2 months later.

RESUMEN

Hibernoma is a rare, benign soft tissue tumor described in <200 case reports/case series. It is slow-growing, painless, and commonly mistaken for lipoma or liposarcoma. Histopathological diagnosis is must for confirmation. Total excision is the treatment of choice. We present a rare case of hibernoma of thigh, which was initially misdiagnosed as atypical lipomatous tumor/well-differentiated liposarcoma on imaging, which turned out to be a hibernoma on histopathological examination.

Objective To assess the feasibility of quantitative measurement of subcutaneous and supraclavicular adipose tissue with iterative decomposition of water and fat with echo asymmetry and least square estimation-iron quantification sequence (IDEAL-IQ).Methods Totally 87 normal young female volunteers (20-35 years old) were recruited and divided into body mass index (BMI)<24 kg/m2 group (n=72) and BMI≥24 kg/m2 group (n=15).Fat fraction (FF) and T2* relaxation rate (R2*) of supraclavicular adipose tissue,chest wall adipose tissue,abdominal wall adipose tissue and liver were measured,respectively.The differences of FF and R2* value of chest wall,subcutaneous and supraclavicular adipose tissue were compared between two groups,and the correlation between FF,R2* value of supraclavicular adipose tissue and BMI was respectively analyzed.Results FF of supraclavicular adipose tissue ([80.99 ± 7.73]%) was lower than that of chest wall subcutaneous adipose tissue ([93.04 ± 1.55] %,P<0.001).R2* of supraclavicular adipose tissue ([65.52±23.59]Hz) was higher than that of chest wall subcutaneous adipose tissue ([38.82±7.11]Hz,P<0.001).The differences of FF and R2* values of supraclavicular adipose tissue,chest wall,abdominal wall subcutaneous adipose tissue and liver were significant between BMI<24 kg/m2 group and BMI≥24 kg/m2 group (all P<0.05).There was positive correlation (r=0.601,P<0.001) between FF of supraclavicular adipose tissue and BMI and negative correlation (r=-0.409,P =0.001) between R2* of supraclavicular adipose tissue and BMI.Conclusion IDEAL-IQ technique can quantitatively assess the difference between subcutaneous and supraclavicular adipose tissue in healthy young women.FF and R2 * value has significant difference between the above mentioned positions.

RESUMEN

Hibernomas are uncommon benign tumours that arise from the remnants of fetal brown adipose tissue. They are usually asymptomatic and have a slow growth pattern. Intrathoracic and pleural locations are exceptional for localization of hibernoma. A review of the English language medical literature revealed more than 110 cases, 20 of which were intrathoracic. In the article below, we discuss a 40-year old male patient who had pleural involvement and was treated by surgical resection. Following resection, the patient has remained problem-free for nine years.


Los hibernomas son tumores benignos poco frecuentes que surgen de restos del tejido adiposo marrón fetal. Son generalmente asintomáticos y tienen un patrón de crecimiento lento. Las localizaciones intratorácicas y pleurales son excepcionales para la localización del hibernoma. Una revisión de la literatura médica en lengua inglesa reveló más de 110 casos, 20 de los cuales se trataban de hibernomas intratorácicos. En el siguiente artículo, discutimos el caso de un paciente de 40 años de edad, que tenía una afección pleural y fue tratado con una resección quirúrgica. Después de la resección, el paciente ha permanecido sin problemas durante nueve años.

RESUMEN

A 35 year old woman presented with 18 months history of a painless, soft, well-circumscribed, vulvar subcutaneous lump which measured approximately 7 cm. The lesion was completely excised and microscopically was consistent with a hibernoma. According to the available literature in English, this is the first reported case of hibernoma of the vulva.

RESUMEN

Of the two variants of adipose tissue, white fat is traditionally known as a lipid rich tissue which undergoes pathological expansion in obese conditions. To counter the excess accumulation of white fat in states of energy imbalance, the second and unique type of brown fat plays a key role by burning extra energy into heat through a special metabolic pathway. In addition brown fat also plays a vital role in thermoregulation in animals and newborn humans and infants. Recent progress in research areas of these two types of fat tissue has provided compelling evidence to show that they secrete a large number of chemicals that play an important role in body weight control that involves several mechanisms. Brown fat was considered absent in the adult humans until recently. But new techniques have provided ample support for its active existence. Based on the very recent data it has been suggested that brown fat can be a target organ in the treatment of obesity which can lead to exciting and informative outcomes in the future.

RESUMEN

PURPOSE: Hibernoma is a very rare benign soft tissue tumor of the hypervascularized brown fat tissue with no malignant potential. MATERIALS AND METHODS: However, it is difficult to differentiate a hibernoma from other malignant tumors, such as liposarcoma using computed tomography and magnetic resonance imaging, and a surgical resection with histological confirmation is the treatment of choice. RESULTS: Histopathologically, hibernoma is composed of brown adipose cells that are polygonal and multivacuolated with a centrally located nucleus and granular cytoplasm, unlike white adipose cells. CONCLUSION: This article describes a patient with a histologically-confirmed hibernoma of the right shoulder.

RESUMEN

AIM: To evaluate the effect of cold acclimation on activity and concentration of UCP in mitochondria inner membrane of rat brown fat. METHODS: Using -labeled GTP binded UCP in mitochondria, radioactivity was measured by liquid aglimmer instrument, the dissociation constand(Kd) and maximal binding(Bmax) were determined by Scatchard plot. RESULTS: In cold acclimation pretreated rats, brown fat weight increased, Bmax increased 1.9-fold,Kd decreased 1.4-fold compared with control. CONCLUSION: Cold acclimation not only increase concentration of UCP in mitochondria inner membrane of rat brown fat,but also increase its activity.

RESUMEN

Using submitochondrial particles, from rat brown adipose tissue mitochondria treated by Lubrol and 3 mol/L urea, as a model, the relationship between different pH and pKd of binding of purine nucleotides to uncoupling protein was estimated. When it was shown as plots of pKd against pH, it was found that the pKd peak of binding of GDP or ADP to uncoupling protein was about pH 5.5. The greater pH deviated, the less pKd became. The highest pKd of binding of GTP or ATP to uncoupling protein was also near pH 5.5, below which pKd remained constant, but above which pKd dropped rapidly. At a certain pH, pKd of binding of GDP or GTP to uncoupling protein was higher than that of binding of ADP or ATP, respectively.

RESUMEN

Using brown adipose tissue submitochondrial particles from different kinds of animals, binding of uncoupling protein to different purine nucleotides was estimated. The results indicate that the dissociate constants (Kds) of uncoupling protein from hamsters to purine nucleotides are more than the Kds of uncoupling protein from guinea pigs, which are more than the Kds of that from rats, presumably because of different amino acid residue sequences of uncoupling protein polypeptide from different kinds of animals.
